In vitro characterization of calcium phosphate biomaterial loaded with linezolid for osseous bone defect implantation.

Osteomyelitis is a severe bone infection frequently caused by Staphylococcus aureus, which shows significant resistance to methicillin. One therapeutic treatment would be to insert a bone substitute loaded to an antibiotic, which would enable the bone to be filled while the illness is being treated. Linezolid is an oxazolidinone antibiotic with a large spectrum of action. It is effective against most Gram-positive bacteria and displays a specific mode of action. The aim of this work was to study the association of linezolid with a calcium phosphate-deficient apatite matrix. Granules containing…

Enhanced activity of rifalazil in combination with levofloxacin, linezolid, or mupirocin against Staphylococcus aureus in vitro.

Rifalazil is a potent second-generation ansamycin that kills bacterial cells by inhibiting the beta subunit of RNA polymerase. Rifalazil has several improved properties compared with rifampicin, but retains rifampicin's propensity to develop resistant mutants at high frequency. To explore strategies to overcome resistance development, we studied the effects of rifalazil in combination with several different antibiotics in an in vitro time-kill model, against both log phase and stationary phase Staphylococcus aureus cells. Experiments were carried out at high initial cell density so that the fr

The safety and efficacy of linezolid in orthopaedic practice for the treatment of infection due to antibiotic-resistant organisms.

Linezolid is the first of a new class of antibacterial agents, the oxazolidinones. It is particularly effective against Gram-positive infections and little resistance has been reported, even amongst methicillin- and vancomycin-resistant bacteria. The compound's excellent oral bioavailability and reasonable safety profile, along with the increasing incidence of resistant infections, means that linezolid offers a valuable alternative to more traditional therapies such as vancomycin. Although no large randomised trials have been carried out in patients with orthopaedic infections such as osteomye…

Linezolid versus vancomycin in the treatment of known or suspected resistant gram-positive infections in neonates.

BACKGROUND: Gram-positive infections caused by susceptible and resistant strains of Staphylococcus aureus, coagulase-negative staphylococci and enterococci are increasing problems in neonates. Linezolid, a new oxazolidinone, is active against these pathogens and has recently been approved by the Food and Drug Administration for treating Gram-positive infections in pediatric patients. OBJECTIVE: To compare the clinical efficacy and safety of intravenous and oral linezolid with vancomycin (10 to 15 mg/kg every 6 to 24 h) in neonates (age 0 to 90 days). METHODS: Hospitalized infants with known …

Efficacy of linezolid plus rifampin in an experimental model of methicillin-susceptible Staphylococcus aureus endocarditis.

The efficacy of linezolid, alone or in combination with rifampin, against methicillin-susceptible Staphylococcus aureus in rabbits with experimental endocarditis was investigated. Linezolid (50 or 75 mg/kg of body weight), rifampin, and linezolid (25, 50, or 75 mg/kg) plus rifampin produced statistically significant reductions in bacterial counts compared with those in untreated controls. Plasma or valvular vegetation levels of linezolid in the groups treated with the linezolid-rifampin combination were similar to those in the respective linezolid-only treatment groups. At therapeutic levels o…

A validated chiral HPLC method for the enantiomeric separation of Linezolid on amylose based stationary phase.

Two chiral HPLC methods namely method A and method B were developed for the separation of enantiomers of Linezolid. The mobile phases containing hexane, 2-propanol and trifluoro acetic acid (TFA) in the ratio (80:20:0.1, v/v/ v); hexane, ethanol and TFA in the ratio (65:35:0.1, v/v/v) were used in method A and method B, respectively. The assay results of the two methods were checked in terms of F-test variance ratio and found to be less than the table value, confirming their good precision. The enantiomeric separation of Linezolid on different chiral stationary phases was investigated. The two…

The use of linezolid in the treatment of vancomycin-resistant enterococcal septicaemia in two patients with burn injuries.

Vancomycin-resistant enterococci (VRE) are multi-resistant micro-organisms that have emerged as important nosocomial pathogens during the la … icaemia during their in-patient stay. Both patients were successfully treated using the antibiotic, linezolid. Linezolid is the first in a new class of antibiotics known as the oxazolidinones whose mode of action inhibits early bacterial protein synthesis. Linezolid: pharmacokinetic and pharmacodynamic evaluation of coadministration with pseudoephedrine HCl, phenylpropanolamine HCl, and dextromethorpan HBr.

Linezolid is a novel oxazolidinone antibiotic with mild reversible monoamine oxidase inhibitor (MAOI) activity. The potential for interaction with over-the-counter (OTC) medications requires quantification. The authors present data evaluating the pharmacokinetic and pharmacodynamic responses to coadministration of oral linezolid with sympathomimetics (pseudoephedrine and phenylpropanolamine) and a serotonin reuptake inhibitor (dextromethorphan). Following coadministration with linezolid, minimal but statistically significant increases were observed in pseudoephedrine and phenylpropanolamine pl…

Comparative outcomes of daptomycin and linezolid in vancomycin-resistant enterococcal intra-abdominal infections with bacteraemia.

OBJECTIVE: We set out to see if daptomycin or linezolid (8-12 mg/kg) is effective for vancomycin-resistant enterococci (VRE) intra-abdominal infections (IAIs) because of its efficacy for VRE bloodstream infections (BSIs). METHODS: We conducted a multicentre observational study. Between April 2010 and October 2024, we screened all patients with VRE-BSIs and included those with a documented VRE-IAI treated with daptomycin or linezolid. Limiting enrolment to patients with concurrent bloodstream infections reduced misclassification bias. The primary outcome was 28‑day in‑hospital mortality. RESU…

Serotonin syndrome associated with concomitant tramadol and linezolid therapy: a case report and literature review.

Serotonin syndrome (SS) is a potentially life-threatening adverse drug reaction resulting from excessive serotonergic activity in the central and peripheral nervous systems. Tramadol produces analgesia through μ-opioid receptor agonism while also inhibiting the reuptake of serotonin and norepinephrine. Linezolid an oxazolidinone antibacterial agent, possesses reversible, non-selective monoamine oxidase inhibitory activity. Through distinct pharmacological mechanisms, both agents enhance serotonergic neurotransmission and may theoretically increase the risk of SS when used concomitantly. We rep…

Clinical Impact of Linezolid Therapeutic Drug Monitoring on the Tolerability of Prolonged Courses in the Outpatient Setting.

BACKGROUND: Linezolid is a gram-positive oral antibiotic whose duration-dependent risk of thrombocytopenia has historically constrained durations of therapy to <3 weeks. This study aimed to evaluate whether therapeutic drug monitoring (TDM)-based management of linezolid can support prolonging the course of therapy for complex infections. METHODS: This single-center, retrospective cohort study included patients ≥18 years old discharged on linezolid for a planned duration of ≥3 weeks between January 1, 2023, and April 1, 2025. The primary outcome was early discontinuation of linezolid due to th…

Black hairy tongue: an early adverse effect of linezolid in treatment of multidrug resistant tuberculosis.

A man in his 20s with microbiologically confirmed multidrug-resistant pulmonary tuberculosis was initiated on a regimen comprising bedaquiline, pretomanid, linezolid and moxifloxacin. Within 2-3 weeks, he developed altered taste sensation followed by black discolouration of the tongue. In accordance with clinical examination and temporal association linezolid was considered the likely causative agent and was discontinued, while the remaining regimen was continued. The patient was advised on oral hygiene measures with which complete resolution occurred within 8-10 days, with no recurrence on fo…

The combinations of cefazolin with linezolid and cefazolin with clindamycin are indifferent against methicillin-susceptible Staphylococcus aureus.

Antimicrobial combinations resulting in synergy against Staphylococcus aureus are desirable for improving clinical efficacy in the treatment of severe infections caused by this pathogen. Despite an existing biological rationale for the combination of cefazolin and linezolid for methicillin-susceptible S. aureus (MSSA), this combination has yet to be evaluated for in vitro synergy. We employed the checkerboard and E-test methods of synergy assessment to evaluate cefazolin in combination with linezolid against 19 clinical MSSA isolates from two centers and compared the findings to cefazolin in c…

Genomic and clinical characterization of linezolid resistance in Enterococcus species from cancer patients in China.

BACKGROUND: Immunocompromised cancer patients represent a susceptible population for enterococcal infections. While linezolid remains the primary therapeutic option for vancomycin-resistant Enterococcus (VRE) infections, the emergence of linezolid-resistant Enterococcus (LRE) worldwide has not only exacerbated public health risks but also posed significant challenges to clinical treatment. This study aimed to analyze the prevalence of LRE isolates among cancer patients in China and to investigate the mechanisms of linezolid resistance. METHODS: A total of 656 non-repetitive Enterococcus isola…

Penetration of linezolid and tedizolid in cerebrospinal fluid of mouse and impact of blood-brain barrier disruption.

Penetration of antimicrobial treatments into the cerebrospinal fluid is essential to successfully treat infections of the central nervous system. This penetration is hindered by different barriers, including the blood-brain barrier, which is the most impermeable. However, inflammation may lead to structural alterations of these barriers, modifying their permeability. The impact of blood-brain barrier disruption on linezolid and tedizolid (antibiotics that may be alternatives to treat nosocomial meningitis) penetration in cerebrospinal fluid (CSF) remains unknown. The aim of this study is to ev…
