Dysregulation of ErbB4 Signaling Pathway in the Dorsal Hippocampus after Neonatal Hypoxia-Ischemia and Late Deficits in PV+ Interneurons, Synaptic Plasticity and Working Memory.

Neonatal hypoxic-ischemic (HI) injury leads to deficits in hippocampal parvalbumin (PV)+ interneurons (INs) and working memory. Therapeutic hypothermia (TH) does not prevent these deficits. ErbB4 supports maturation and maintenance of PV+ IN. Thus, we hypothesized that neonatal HI leads to persistent deficits in PV+ INs, working memory and synaptic plasticity associated with ErbB4 dysregulation despite TH. P10 HI-injured mice were randomized to normothermia (NT, 36 °C) or TH (31 °C) for 4 h and compared to sham. Hippocampi were studied for α-fodrin, glial fibrillary acidic protein (GFAP), and

Polarity and subfield specific effects of transcranial direct current stimulation on hippocampal plasticity.

An increasing number of studies using human subjects substantiate the use of transcranial direct current stimulation (tDCS) as a noninvasive approach to treat various neurological symptoms. tDCS has been tested in conditions from motor to cognition dysfunctions. Performance enhancement of healthy subjects using tDCS has also been explored. The underlying physiological mechanism for tDCS effects is hypothesized to be through changes in neuroplasticity and we have previously demonstrated that in vivo anodal tDCS can enhance neuroplasticity of hippocampal CA1 neurons. The purpose of this study wa

mGluR2/3 receptor agonist LY379268 alleviates PTSD-like fear memories by downregulating the CaMKII/CREB signaling pathway in the hippocampus and prefrontal cortex.

Post-traumatic stress disorder (PTSD) is a mental health condition marked by fear memories triggered by life-threatening traumatic events. The formation and consolidation of these fear memories are central to PTSD, making it important to understand the underlying neurobiological mechanisms that regulate memory plasticity. In this context, LY379268, a selective mGluR2/3 agonist, induces presynaptic long-term depression (LTD) and modulates excitatory transmission, potentially affecting memory consolidation. Propranolol, a β-adrenergic antagonist, similarly influences fear memory through synaptic

Direct assessment of presynaptic modulation of cortico-striatal glutamate release in a Huntington's disease mouse model.

Glutamate is the main excitatory neurotransmitter in the brain, and impairments in its signaling are associated with many neurological disorders, including Huntington's disease (HD). Previous studies in HD mouse models demonstrate altered glutamate receptor distribution and signaling at cortico-striatal synapses, and some studies suggest that glutamate release is altered; however, traditional methods to study synaptic glutamate release are indirect or have poor temporal resolution. Here we utilize iGluSnFR, a modified green fluorescent protein reporter for real-time imaging of glutamate transm

Amorphous Dielectric-Integrated Retina-Inspired HfS2 Devices with Enhanced Synaptic Plasticity for Neuromorphic Vision Assistance.

Realizing neuromorphic functionalities in field-effect transistors (FETs) requires precise modulation of multilevel conductance states with robust retention. Two-dimensional (2D) semiconductor FETs, however, often suffer from a limited synaptic weight dynamic range and short retention times because of inadequate charge dynamics in the channel. This results from the low-k dielectric nature of the channel, which provides limited and unstable carrier storage sites and insufficient capacitive coupling. This study expands the weight dynamic range by forming a weight-control dielectric through oxyge

Nanoscale synaptic remodeling at corticostriatal circuits predicts flexible action control.

Synaptic plasticity is a fundamental substrate of behavioral adaptation, yet the underlying molecular dynamics remain poorly defined. We tested the hypothesis that, within striatal circuits, flexibility relies on nanoscale remodeling of synaptic machinery coupling anterograde glutamatergic transmission to retrograde endocannabinoid signaling, a process disrupted in states of rigidity and aging. In the dorsolateral striatum, we found cell-type-specific facilitation of metabotropic glutamate receptor 5 (mGlu5)-dependent, endocannabinoid-mediated long-term depression at cortico-striatal synapses

Inactivation of adenosine A2A receptors reverses working memory deficits at early stages of Huntington's disease models.

Cognitive impairments in Huntington's disease (HD) are attributed to a dysfunction of the cortico-striatal pathway and significantly affect the quality of life of the patients, but this has not been a therapeutic focus in HD to date. We postulated that adenosine A(2A) receptors (A(2A)R), located at pre- and post-synaptic elements of the cortico-striatal pathways, modulate striatal neurotransmission and synaptic plasticity and cognitive behaviors. To critically evaluate the ability of A(2A)R inactivation to prevent cognitive deficits in early HD, we cross-bred A(2A)R knockout (KO) mice with two

Hippocampal Hap1 down-regulation impairs glucocorticoid receptor nuclear translocation and exacerbates Alzheimer's disease-related neuropathology in APP/PS1 mice.

Impaired nuclear translocation of glucocorticoid receptor (GR) has been implicated in hippocampal vulnerability in Alzheimer's disease (AD), yet the molecular basis of this defect remains poorly understood. This study identified Huntingtin-associated protein 1 (Hap1) as a critical regulator of GR nuclear translocation in the hippocampus. Specifically, Hap1 expression progressively declined in the hippocampus of APP/PS1 mice with advancing age and pathological burden. Hippocampal Hap1 knockdown induced pronounced cognitive deficits and synaptic deterioration, as indicated by reduced dendritic a

Inflammation associated with chronic heart failure leads to enhanced susceptibility to depression.

Epidemiological and clinicopathological studies indicate that there is a high risk for chronic heart failure (CHF) in patients suffering from neuropsychiatric disorders, such as depression. However, it is unclear whether CHF causes depression, and the underlying mechanisms of this association remain largely unknown. In this study, mice with myocardial infarction and CHF were used to investigate behavioral alterations as well as changes in the brain-heart axis. During the first 6 months, abnormalities in neuropsychiatric behaviors were detected in mice with CHF. Using the sucrose preference tes

Astrocyte signaling controls spike timing-dependent depression at neocortical synapses.

Endocannabinoid mediated spike timing-dependent depression (t-LTD) is crucially involved in the development of the sensory neocortex. t-LTD at excitatory synapses in the developing rat barrel cortex requires cannabinoid CB(1) receptor (CB(1)R) activation, as well as activation of NMDA receptors located on the presynaptic terminal, but the exact signaling cascade leading to t-LTD remains unclear. We found that astrocytes are critically involved in t-LTD. Astrocytes gradually increased their Ca(2+) signaling specifically during the induction of t-LTD in a CB(1)R-dependent manner. In this way, as

Long-Term Treadmill Exercise and Voluntary Running Pre-Training Attenuates Vascular Dementia-Related Pathology by Regulating Hippocampal Structural Synaptic Plasticity in a Rat Model.

BACKGROUND: Clinical and basic research suggests that exercise is a safe behavioral intervention and effective in improving cognition in vascular dementia (VD). However, despite global efforts, there is still no effective method to completely cure VD. This study aimed to investigate the effects of long-term exercise pretreatment on typical VD pathology in a rat model, and further compare the neuroprotective impacts of different exercise modalities on VD rats. METHODS: Forty-eight SD adult male rats (200-240 g) were randomly allocated to a control group (Con, n = 12), a vascular dementia group

Alleviation of N-Methyl-D-Aspartate Receptor-Dependent Long-Term Depression via Regulation of the Glycogen Synthase Kinase-3β Pathway in the Amygdala of a Valproic Acid-Induced Animal Model of Autism.

The amygdala plays crucial roles in socio-emotional behavior and cognition, both of which are abnormal in autism spectrum disorder (ASD). Valproic acid (VPA)-exposed rat offspring have demonstrated ASD phenotypes and amygdala excitatory/inhibitory imbalance. However, the role of glutamatergic synapses in this imbalance remains unclear. In this study, we used a VPA-induced ASD-like model to assess glutamatergic synapse-dependent long-term depression (LTD) and depotentiation (DPT) in the amygdala. We first confirmed that the VPA-exposed offspring exhibited sociability deficits, anxiety, depressi

Phrenic long-term depression evoked by intermittent hypercapnia is modulated by serotonergic and adrenergic receptors in raphe nuclei.

Intermittent hypercapnia evokes prolonged depression of phrenic nerve activity (phrenic long-term depression, pLTD). This study was undertaken to investigate the role of 5-HT and α2-adrenergic receptors in the initiation of pLTD. Adult male urethane-anesthetized, vagotomized, paralyzed, and mechanically ventilated Sprague-Dawley rats were exposed to a protocol of acute intermittent hypercapnia (AIHc; 5 episodes of 15% CO2 in air, each episode lasting 3 min). The experimental group received microinjection of the selective 5-HT1A receptor agonist 8-hydroxy-2-(dipropylamino)tetralin hydrobromide

Hydrogen Sulfide Prevents Synaptic Plasticity from VD-Induced Damage via Akt/GSK-3β Pathway and Notch Signaling Pathway in Rats.

Our previous study has demonstrated that hydrogen sulfide (H2S) attenuates neuronal injury induced by vascular dementia (VD) in rats, but the mechanism is still poorly understood. In this study, we aimed to investigate whether the neuroprotection of H2S was associated with synaptic plasticity and try to interpret the potential underlying mechanisms. Adult male Wistar rats were suffered the ligation of bilateral common carotid arteries. At 24 h after surgery, rats were administered intraperitoneally with sodium hydrosulfide (NaHS, 5.6 mg·kg(-1)·day(-1)), a H2S donor, for 3 weeks in the VD+NaHS

Neuronal Calcium Signaling and Cytoskeletal Dynamics in Neurodegeneration.

Neuronal function relies on the precise coordination between intracellular calcium (Ca2+) signaling and the cytoskeletal architecture that underpins synaptic transmission, plasticity, and structural stability. Disruption of this calcium-cytoskeleton interplay has been noted in numerous neurodegenerative diseases. We discuss how Ca2+-dependent cytoskeletal remodeling governs long-term potentiation and depression, dendritic spine morphology, and presynaptic function, highlighting the functions of end-binding proteins, STIM (Stromal Interaction Molecule)/Orai-mediated store-operated calcium entry

Postactivation depression changes after robotic-assisted gait training in hemiplegic stroke patients.

Postactivation depression is decreased in patients with spasticity and partially restored by physical exercise in spinal cord injured patients. Up until now, the possibility to modulate postactivation depression with motor training has never been explored in subjects with spasticity following brain lesions. Postactivation depression, assessed as frequency related depression of soleus H-reflex, was investigated before and after robotic-assisted gait training in a group of seven subjects with spastic hemiparesis following hemispheric stroke. Patients received three sessions per week of robotic-a

Chronic co-treatment with geniposide and shanzhiside methyl ester elicits antidepressant effects via PACAP signaling in stressed male mice.

Yueju, an herbal medicine, has demonstrated rapid antidepressant effects, primarily attributed to its iridoid constituents geniposide (GP) and shanzhiside methyl ester (SM). This study investigates the sustained antidepressant efficacy of chronic co-treatment with GP and SM (GS) and elucidates the underlying molecular mechanisms, with a focus on hippocampal neurogenesis and PACAP signaling. Using a 4-week chronic unpredictable stress (CUS) mouse model, the effects of chronic co-treatment with GS on depression-like behaviors were examined. Adult male C57/BL6J mice received either vehicle or GS

Inhibition of sodium-glucose cotransporter 2 prevents sorafenib-induced depression-like behaviors by preserving hippocampal-dependent long-term potentiation.

The sodium-glucose cotransporter-2 (SGLT-2) inhibitor empagliflozin has been reported to exert significant neuroprotective effects on neurobehavioral deficits; however, its effects on chemotherapy-induced neuroplastic changes have not yet been investigated. The present study investigates the behavioral changes and hippocampal cellular responses through which empagliflozin exerts antidepressant effects against chemotherapy-induced pathological alterations. We employed a mouse model of depression induced by oral administration of the chemotherapeutic agent sorafenib for 28 days. Sorafenib-induce

Prenatal Alcohol Exposure Inhibits Transient Expression of Autophagy and Synaptic Proteins in Developing Brain.

INTRODUCTION: Neuronal apoptosis and consequent inhibition of autophagy, with loss of synaptic connections are central events in the genesis of fetal alcohol spectrum disorders (FASD). However, studies of molecular mechanisms of autophagy in human fetal brain are limited. Recently, prenatal exposure to EtOH was associated with reduced miRNA-9 levels in fetal brain-derived exosomes (FB- Es) isolated from maternal plasma, which correlated with small eyes, an anatomical hallmark of fetal alcohol syndrome (FAS). Since miR-9 targets several genes that regulate synaptic plasticity, EtOH-induced inhi
