An electropharmacological analyses of the effects of some drugs on neuromuscular transmission in the vas deferens of the guinea-pig.

1 The effects of several drugs upon the excitatory junction potential (e.j.p.) in the guinea-pig vas deferens have been investigated. 2 Amiodarone, a noradrenergic neurone blocker which also blocks both alpha and beta-adrenoreceptors, did not reduce the e.j.p. 3 The alpha-adrenoreceptor antagonists, azapetine, piperoxan and prazosin, only enhanced the e.j.p. irrespective of their relative potencies at alpha 1 and alpha 2-adrenoreceptors. 4 Sotalol, a beta-adrenoreceptor antagonist, was without effect upon the e.j.p. 5 Clonidine and lysergic acid diethylamide, alpha-adrenoreceptor agonists, pro…

The initial phase of the dextran-induced anaphylactoid reaction in the rat: a comparison of inhibitors of the blood pressure fall.

The histamine H2-receptor antagonist cimetidine (25-100 mg/kg) caused a partial inhibition of the pronounced blood pressure fall induced by dextran (Macrodex, 40-100 mg/kg) in the rat. The inhibition by cimetidine could not be distinguished from the inhibition achieved by the serotonin D-receptor antagonist bromolysergic acid diethylamide (BOL, 1-4 mg/kg). Doses of cimetidine and BOL that gave submaximum inhibitory effects separately, showed approximately additive effects when combined. The combined effect of these drugs never exceeded the maximum effects of the drugs separately, whereas injec…

Psychological Therapy Quantity and Depressive Symptom Reduction in Psychedelic-Assisted Therapy: A Systematic Review and Meta-Analysis.

IMPORTANCE: Psychedelic-assisted therapy (PAT) is a novel intervention for depressive symptoms, typically delivered with additional psycholo … ith depressive symptoms who received PAT using classic serotonergic psychedelics (eg, psilocybin or lysergic acid diethylamide). Within these trials, psychedelic dosing sessions were embedded in therapeutic sessions before (preparation) and after (integration). Studies were excluded if they used microdosing as the primary intervention, involved naturalistic or …

Classic psychedelics and personality: An updated systematic review.

BACKGROUND: The impact of classic psychedelics (5-HT2A receptor agonists) on personality traits has been studied for decades, and it has bee … abel, and observational studies. Interventions included the consumption of any classic psychedelic (lysergic acid diethylamide, psilocybin, ayahuasca/dimethyltryptamine, mescaline), and the studies were required to use some instrument for personality assessment. We followed the Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines to con …

Existing evidence for the use of psychedelics in patients with cancer and other serious illness: A narrative review.

OBJECTIVES: Mood disorders and existential distress impact those with cancer or a serious illness at higher rates than the general population. There have been limited pharmacological advances in recent years, and available psychological interventions vary in degree of impact and durability as a treatment modality in this population. A recent renaissance in psychedelic research has suggested that this class of medications might offer an alternative treatment model for anxiety, depression, and existential and psychological distress that often accompanies the diagnosis of a serious illness. Modulation of long-term potentiation following microdoses of LSD captured by thalamo-cortical modelling in a randomised, controlled trial.

BACKGROUND: Microdosing psychedelics is a phenomenon with claimed cognitive benefits that are relatively untested clinically. Pre-clinically, psychedelics have demonstrated enhancing effects on neuroplasticity, which cannot be measured directly in humans, but may be indexed by non-invasive electroencephalography (EEG) paradigms. This study used a visual long-term potentiation (LTP) EEG paradigm to test the effects of microdosed lysergic acid diethylamide (LSD) on neural plasticity, both acutely while on the drug and cumulatively after microdosing every third day for six weeks. Healthy adult ma…

Psychedelic therapy in depression and substance use disorders.

Psychoactive substances obtained from botanicals have been applied for a wide variety of purposes in the rituals of different cultures for thousands of years. Classical psychedelics from N,N'-dimethyltryptamine, psilocybin, mescaline and various lysergamides cause specific alterations in perception, emotion and cognition by acting through serotonin 5-HT2A receptor activation. Lysergic acid diethylamide, the first famous breakthrough in the field, was discovered by chance by Albert Hoffman in the Zurich Sandoz laboratory in 1943, and studies on its psychoactive effects began to take place in th…

The promises and perils of psychedelic pharmacology for psychiatry.

Psychedelic drugs including psilocybin, N,N'-dimethyltryptamine (DMT) and lysergic acid diethylamide (LSD) are undergoing a renaissance as potentially useful drugs for various neuropsychiatric diseases, with a rapid onset of therapeutic activity. Notably, phase II trials have shown that psilocybin can produce statistically significant clinical effects following one or two administrations in depression and anxiety. These findings have inspired a 'gold rush' of commercial interest, with nearly 60 companies already formed to explore opportunities for psychedelics in treating diverse diseases. Add…

Psychedelic drug abuse potential assessment research for new drug applications and Controlled Substances Act scheduling.

New medicines containing classic hallucinogenic and entactogenic psychedelic substance are under development for various psychiatric and neurological disorders. Many of these, including psilocybin, lysergic acid diethylamide (LSD), and 3,4-methylenedioxymethamphetamine (MDMA) are Schedule I controlled substances of the United States Controlled Substances Act (US CSA), and similarly controlled globally. The implications of the CSA for research and medicines development, the path to approval of medicines, and their subsequent removal from Schedule I in the US are discussed. This entire process o…

Flashback phenomena after administration of LSD and psilocybin in controlled studies with healthy participants.

BACKGROUND: LSD and psilocybin are increasingly used in phase I trials and evaluated as therapeutic agents for mental disorders. The phenomenon of reoccurring drug-like experiences after the acute substance effects have worn off was described for both substances and especially attributed to LSD. According to the DSM-V, the persisting and distressing manifestation of these experiences is called hallucinogen-persisting perception disorder (HPPD). Data on both conditions is very limited. OBJECTIVE: This study aims to provide descriptive data on reoccurring drug-like experiences after the adminis

Animal Behavior in Psychedelic Research.

Psychedelic-assisted psychotherapy holds great promise in the treatment of mental health disorders. Research into 5-hydroxytryptamine 2A receptor (5-HT2AR) agonist psychedelic compounds has increased dramatically over the past two decades. In humans, these compounds produce drastic effects on consciousness, and their therapeutic potential relates to changes in the processing of emotional, social, and self-referential information. The use of animal behavior to study psychedelics is under debate, and this review provides a critical perspective on the translational value of animal behavior studie

Psychedelics: Where we are now, why we got here, what we must do.

The purpose of this commentary is to provide an introduction to this special issue of Neuropharmacology with a historical perspective of psychedelic drug research, their use in psychiatric disorders, research-restricting regulatory controls, and their recent emergence as potential breakthrough therapies for several brain-related disorders. It begins with the discovery of lysergic acid diethylamide (LSD) and its promising development as a treatment for several types of mental illnesses during the 1940s. This was followed by its abuse and stigmatization in the 1960s that ultimately led to the pl…

Modern Clinical Research on LSD.

All modern clinical studies using the classic hallucinogen lysergic acid diethylamide (LSD) in healthy subjects or patients in the last 25 years are reviewed herein. There were five recent studies in healthy participants and one in patients. In a controlled setting, LSD acutely induced bliss, audiovisual synesthesia, altered meaning of perceptions, derealization, depersonalization, and mystical experiences. These subjective effects of LSD were mediated by the 5-HT2A receptor. LSD increased feelings of closeness to others, openness, trust, and suggestibility. LSD impaired the recognition of sad…

Confirmation of LSD intoxication by analysis of serum and urine.

Serum and urine specimens of 31 patients with suspected lysergic acid diethylamide (LSD) intoxication were analyzed for LSD by both radioimmunoassay (RIA) and high-pressure liquid chromatography (HPLC). The RIA assay, using 0.1 ng/mL as the limit of detection instead of the manufacturer's recommendation of 0.5 ng/mL, was positive for LSD in 13 blood and urine specimens from 14 patients. Results were compared to HPLC analysis using methysergide instead of lysergol as the internal standard and a limit of detection of 0.5 ng/mL. HPLC detected LSD in 9 of 13 serum specimens and 11 of 13 urine spec…

Bromolysergide and methysergide protection against ECS-induced retrograde amnesia.

Bromolysergide (BOL 148) and methysergide (UML 491), 2 mg/kg intraperitoneally, and saline were administered to rats 45 min before one-trial passive-avoidance conditioning followed by electroconvulsive shock (ECS) or sham-ECS (ECS). On test session (24 h later), the groups treated with both BOL 148 and UML 491 exhibited a clear-cut retention in comparison to saline-ECS rats. On the other hand, all drugged groups, regardless of their submission to ECS, showed a little less pronounced consolidation than saline-ECS rats. The antiamnestic effect brought about by the two drugs was discussed in term
