Various technical procedure were applied to the purification of T and B lymphocytes subpopulation in sarcoidosis. Patients were selected according to the presence of a characterized deficit in cell-mediated immune functions. These purified lymphocytes populations were further analysed for their ability to release lymphokines in the presence of Con-A-sepharose or specific antigens. Among these factors, MIF, LIF and mitogenic factor (MF) were looked for. For the determination of MIF and LIF, a newly elaborated technique was deviced which allows a semi-quantitative approach of the lymphokine prod

PURPOSE OF REVIEW: Macrophages differentiate from peripheral-blood monocytes. Both monocytes and synovial macrophages are key players in rheumatoid arthritis. These cells are involved in the initiation and perpetuation of inflammation, leukocyte adhesion and migration, matrix degradation and angiogenesis. Macrophages express adhesion molecules, chemokine receptors and other surface antigens. They also secrete a number of chemokines, cytokines, growth factors, proteases and other mediators.
RECENT FINDINGS: Macrophage migration-inhibitory factor has drawn significant attention recently. This c

Phenotyping cells by flow cytometry is a powerful way to identify cell type and any morphological changes during cell culture. The staining procedure used in this chapter enables the characterization of mouse macrophages by a flow cytometry antibody panel which can be used for both bone marrow-derived macrophages (BMM) and macrophages derived from other tissues, such as the mouse spleen or peritoneal cavity. The surface and intracellular staining methods are versatile and can be applied to flow cytometry staining of several different cell types by changing the surface markers used with knowled

Many children with chronic cicatricial stenosis of the larynx and trachea showed a high activity of the factor stimulating macrophage migration in vitro (MSF). The high MSF level is typical of patients with large scars in tissues rich in cellular elements. The MSF predominance was to a certain extent correlated with abnormal responses to surgical intervention. This was indicated by poor results of surgical treatment if the MSF was low before and immediately after it. With normal MSF the treatment was effective in at least 50% of cases. The therapeutic effectiveness was correlated with MSF vari

Hapten-specific sensitivity of guinea-pigs sensitized to dinitrophenyl (DNP) contactants and to DNP-protein conjugates was investigated by skin test and by antigen-induced elaboration of migration inhibition (MIF) and macrophage agglutination factors (MAF) From lymph node cells. The delayed contact reaction was highly specific for low doses of contactant and markedly less so for conjugates; lymph node cells elaborted both lymphokines in response to brief exposures to dinitrofluorobenzene (DNFB) or prolonged exposures to DNP conjugates. Elicitation of MAF by DNFB or DNP conjugate was inhibited

Well defined cell-mediated immune responses were detectable following experimental immunization of hamsters with Entamoeba histolytica antigen, using glucan as an adjuvant. Peritoneal cells from amoeba antigen-glucan sensitized animals, upon incubation with specific antigen in vitro, were found to release into the supernatant a macrophage migration inhibition factor (MIF). Such supernatant fluids inhibited the migration of macrophages from non-sensitized hamsters. The production of MIF was found to be greatly increased if glucan is added to amoeba antigen when sensitizing animals. The optimal

Using a standard heterologous assay in which lymphokines alter guinea pig peritoneal macrophage migration, it was found that regional lymph node cells (RLNCs) from patients with primary breast cancer elaborate soluble factors which variably affect such migration. Variation of migration resulted when soluble factors employed were obtained from different nodes in the same patient as well as from nodes from different patients. Some nodes from a patient elaborated migration inhibition factors (MIF) and other migration enhancing factors (MEF). The findings are in keeping with others obtained by us

Solid phase peptide synthesis is a method used to generate peptides through the sequential addition of amino acids attached to an immobilized resin. Due to the procedure followed, excess reagents and byproducts are removed while synthesis is ongoing, resulting in a final product that is reproducibly obtained at high purity. Herein, we describe the synthetic procedure of peptides derived from the pleotropic protein macrophage migration inhibitory factor (MIF). Notably, this protocol is valid for the generation of peptides regardless of their sequence and length. Whereas MIF has diverse activiti

Liver injury is a common complication during infection of Toxoplasma gondii. However, the Toxoplasma effector proteins involved remain unknown. Herein, we identified that T. gondii macrophage migration inhibitory factor (TgMIF) is a critical pathogenic factor of liver injury in acute toxoplasmosis mouse model induced by a less virulent strain, which is widely prevalent in humans. We show that TgMIF is a novel activator of nucleotide-binding oligomerization domain-like receptor pyrin domain containing 3 (NLRP3) inflammasome in hepatocytes, resulting in subsequent pyroptosis. Furthermore, T. gon

Immunological reactivity in alcoholic hepatitis has bben attributed to alcoholic hyalin, the histological hallmark of this disease. A purified isolate of alcoholic hyalin with electron microscopic, biochemical, and serological characteristics documented previously was added to lymphocytes from healthy subjects and patients with alcoholic hepatitis or other hepatic disorders. Production of migration inhibition factor (MIF) in response to this material was used as an index to lymphocyte reactivity. MIF was significantly increased in lymphocytes obtained from patients with alcoholic hepatis, as c

Heart failure (HF) is a complex clinical syndrome resulting from various cardiac diseases and a significant medical issue worldwide. Although the role of inflammation in HF pathogenesis is well-known, the specific cell types and regulatory molecules involved remain poorly understood. Here, we identified key cell types and novel biomarkers via an analysis of single-cell and bulk RNA sequencing data obtained from patients with two major HF types of ischemic cardiomyopathy and dilated cardiomyopathy. Myeloid cells were identified as the primary cell population involved in HF through cellular frac

The macrophage migration inhibition factor assay was used as a specific measure of cellular hypersensitivity to peripheral nervous system antigen in a large group of Guillain-Barre patients and control subjects. Lymphocytes from 34 patients with Guillain-Barre syndrome, 33 with other peripheral nervous system disease, and 33 normal controls were assayed for production of macrophage migration inhibition factor. A mean of 101 +/- 7.2 was obtained in the control group, 70 +/- 16.3 in the Guillain-Barre syndrome group, and 96 +/- 11.3 in those with other peripheral nervous system disease. Twenty-s

PHA-induced mononuclear production of lymphokines altering leukocyte migration was studied in patients with psoriasis, mycosis, and scabies. The findings evidence that T-cells synthesize leukocyte migration inhibition factor (LMIF) in mycosis and scabies, whereas in psoriasis they produce both LMIF and leukocyte migration stimulation factor (LMSF). Blood leukocytes of mycosis and scabies patients were more sensitive to LMIF than the cells of psoriasis patients. The 'skin fenestra' cells (granulocytes) of these patients were functionally active only in respect of LMIF, even when mononuclears sy

To determine the role of endogenous migration-inhibitory factor (MIF) in the development of protective immunity against cutaneous leishmaniasis, we analyzed the course of cutaneous Leishmania major infection in MIF gene-deficient mice (MIF(-/-)) and wild-type (MIF(+/+)) mice. Following cutaneous L. major infection, MIF(-/-) mice were susceptible to disease and developed significantly larger lesions and greater parasite burdens than MIF(+/+) mice. Interestingly, antigen-stimulated lymph node cells from MIF(-/-) mice produced more interleukin-4 (IL-4) and gamma interferon (IFN-gamma) than those

Seventy-nine cytokines, chemokines, and growth factors were measured by protein array analysis in the cerebrospinal fluid of patients with meningitis and controls. Several factors were found to be regulated, which have not been studied in the CNS before, e.g., macrophage inflammatory protein-1delta (CCL15) and neutrophil-activating peptide-2 (CXCL7). In pneumococcal meningitis, other new observations were an increase of macrophage migration inhibitory factor, monocyte chemoattractant protein-2 (CCL8), pulmonary and activation-regulated chemokine (CCL18), and macrophage inflammatory protein-3al

Liver damage has been induced in animal experiments using carbon tetrachloride (CCl4), a potent hepatotoxin. CCl4 is activated by cytochrome P450 2E1, which results in the formation of various metabolites including phosgene. Although D-dopachrome tautomerase (DDT) is abundant in the liver, its role currently remains unclear. The biological activity of DDT, for which the N-terminal proline is a key site, has been detected in various tissues. We herein incidentally detected a 333 Da modification to the N-terminal proline of DDT in rat livers damaged by CCl4. We identified that this modification

The clinical spectrum of cutaneous leishmaniasis (CL), an intracellular parasitic pathogen, ranges from a single sore healing to chronic crusty lesions with a manifestation of treatment resistance. The complicated interaction between Leishmania bodies and the early immune response, including innate and adaptive mechanisms, determines the evolution of nodules. This study examined the levels of the chemoattractant interleukin 8 (IL-8), pro-inflammatory nitric oxide (NO), and immunoregulatory macrophage inhibitory factor (MIF) in the serum of subjects recently diagnosed with cutaneous leishmanias
