Sulfinamide Formation from the Reaction of Bacillithiol and Nitroxyl.

Bacillithiol (BSH) replaces glutathione (GSH) as the most prominent low-molecular-weight thiol in many low G + C gram-positive bacteria. BSH plays roles in metal binding, protein/enzyme regulation, detoxification, redox buffering, and bacterial virulence. Given the small amounts of BSH isolated from natural sources and relatively lengthy chemical syntheses, the reactions of BSH with pertinent reactive oxygen, nitrogen, and sulfur species remain largely unexplored. We prepared BSH and exposed it to nitroxyl (HNO), a reactive nitrogen species that influences bacterial sulfur metabolism. The prof

Directed evolution of Oenococcus oeni strains for more efficient malolactic fermentation in a multi-stressor wine environment.

High concentrations of ethanol, low pH, the presence of sulfur dioxide and some polyphenols have been reported to inhibit Oenococcus oeni growth, thereby negatively affecting malolactic fermentation (MLF) of wine. In order to generate superior O. oeni strains that can conduct more efficient MLF, despite these multiple stressors, a continuous culture approach was designed to directly evolve an existing ethanol tolerant O. oeni strain, A90. The strain was grown for ∼350 generations in a red wine-like environment with increasing levels of stressors. Three strains were selected from screening expe

Metabolism of SO₂ binding compounds by Oenococcus oeni during and after malolactic fermentation in white wine.

Sulfur dioxide SO₂ is the key additive for the preservation of wines. Carbonyl and keto compounds in wine can bind to SO₂ and decrease its efficacy, resulting in higher total SO₂ requirements. Increased consumer demand for low sulfite and organic wines pose production challenges if SO₂ binders have not been properly managed during vinification. Malolactic fermentation (MLF) has been known to reduce bound SO₂ levels but detailed time course studies are not available. In this work, the kinetics of major SO₂ binding compounds and malic acid were followed during MLF in wine with 12 commercially av

Sorption of copper and zinc to the plasma membrane of wheat root.

Sorption of Cu(2+) and Zn(2+) to the plasma membrane (PM) of wheat root (Triticum aestivum L cv. Scout 66) vesicles was measured at different pH values and in the presence of organic acids and other metals. The results were analyzed using a Gouy-Chapman-Stem model for competitive sorption (binding and electrostatic attraction) to a negative binding site. The binding constants for the two investigated cations as evaluated from the sorption experiments were 5 M(-1) for Zn(2+) and 400 M(-1) for Cu(2+). Thus, the sorption affinity of Cu(2+) to the PM is considerably larger than that of Ca(2+), Mg(

Relative Bioavailability Assessment of Solid Forms by An Artificial Stomach and Duodenum Apparatus.

In this work, the ability of the artificial stomach and duodenum (ASD) model to predict bioavailability in rats was investigated using a poorly soluble model compound, BI-639667. A solution and four suspensions of different solid forms of BI-639667 were tested both in an ASD and rats. Rank order of the bioavailability estimated from an ASD apparatus is consistent with that of in vivo result in rats, i.e., solution > salicylic acid cocrystal > malate salt > maleate salt > monohydrate, which correlates with the ability of the different solid forms to maintain supersaturation with respect to the

Using our understanding of interactions between helminth metabolism and host immunity to target worm survival.

Helminths can adapt to environmental conditions in the host, utilising anaerobic processes like fermentation and malate dismutation to produce energy from carbohydrate. Although targeting carbohydrate metabolism is an established therapeutic strategy to combat helminth infection, questions remain over the metabolic pathways they employ as adults to survive and evade host immunity. Helminths also use amino acid, polyunsaturated fatty acid (PUFA), and cholesterol metabolism, a possible strategy favouring the production of immunomodulatory compounds that may influence survival in the host. Here,

Two-dimensional electrophoresis and characterization of antigens from Paracoccidioides brasiliensis.

Paracoccidioides brasiliensis is a fungal pathogen of humans. To identify antigens from P. brasiliensis we fractionated a crude preparation of proteins from the fungus and detected the IgG reactive proteins by immunoblot assays of yeast cellular extracts with sera of patients with paracoccidioidomycosis (PCM). We identified and characterized six new antigens by amino acid sequencing and homology search analyses with other proteins deposited in a database. The newly characterized antigens were highly homologous to catalase, fructose-1,6-biphosphate aldolase (aldolase), glyceraldehyde-3-phosphat

Brugia malayi: status of host during different stages of infection.

The status of glycogen, protein, lipid components, lipid peroxides and a few enzymes of energy metabolism was studied in liver of Mastomys natalensis during the development of Brugia malayi infection. Glycogen and lipid contents were decreased during the patent phase of infection while total protein was slightly altered in latent animals. Phospholipid and triglyceride contents declined at prepatent and patent phase of infection. The levels of lactate and malate dehydrogenases, as well as of adenosine triphosphatases (Na+K+, Mg2+, Ca2+), were significantly elevated and monoamine oxidase activit

Differentiation of hydatid cyst from cysticercus cyst by proton MR spectroscopy.

The metabolite patterns obtained by ex vivo proton MR spectroscopy of fluid from different locations of hydatid cysts of sheep and humans (n = 16) and cysticercus cysts of swine and humans (n = 25) were compared with an objective of differentiating the two parasites on the basis of their metabolite pattern. The spectra from hydatid fluid differed from cysticercus cyst by the absence of creatine in the former. When the hydatid cyst was fertile, malate and/or fumarate was also observed, which was absent in cysticercus cyst. The most likely explanation for the presence of creatine only in the cys

Experimental host-induced selection in Schistosoma mansoni strains from Guadeloupe and comparison with natural observations.

Allelic frequency variation at the malate dehydrogenase (E.C.1.1.1.37) polymorphic locus (Mdh-1) was analysed during several successive generations in four strains of Schistosoma mansoni from Guadeloupe, maintained experimentally on mice. A rapid evolution of the frequency of the Mdh-1a allele is interpreted as being the result of an interaction between experimental drift and selection induced by the murine laboratory host. These experimental results are compared to the genetic structures observed among the corresponding natural populations of S. mansoni in Guadeloupe (West Indies). They stren

Ammonium inhibition of fatty acid oxidation in rat liver mitochondria. A possible cause of fatty liver in Reye's syndrome and urea cycle defects.

NH4C1 inhibited oxygen consumption (State 3, ADP induced) by rat liver mitochondria respiring on palmitoyl-L-carnitine or octanoic acid but not on succinate or malate + glutamate. The inhibition became apparent at 0.02 mM reaching a plateau (40%) at 2 mM NH4C1. Similar inhibition was observed with uncoupled (in the presence of 2, 4-dinitrophenol) mitochondria. The inhibition of uncoupled mitochondria was reversible as the rate of respiration with palmitoyl-L-carnitine was further increased by succinate and the total rate was unaffected by NH4C1. Therefore, NH+4 inhibition of mitochondrial resp

Setaria cervi: enzymes in microfilariae and in vitro action of antifilarials.

Microfilariae of bovine filarial parasite Setaria cervi are equipped with the enzymes of glycolysis, pentose phosphate and PEP-succinate pathways and thus resemble the adult form in its metabolic pattern. Malate dehydrogenase was the most active enzyme in microfilariae followed by lactic dehydrogenase and fumarase, while phosphoglucoisomerase, PEP-carboxykinase and FDP-aldolase were comparatively less active. The very low ratio of PK/PEPCK in S. cervi microfilariae indicates active fixation of CO2 into PEP to produce oxalacetate. Centperazine and diethylcarbamazine significantly inhibited PEP-

An anoxygenic photosynthesis-inspired dual biohybrid for light-enhanced chemical production.

Integrating light-harvesting materials with non-photosynthetic microbes offers a promising route to convert sunlight into chemicals. However, many existing biohybrid systems lack a clear architectural basis linking external light capture to intracellular redox processes. Inspired by the modular architecture of anoxygenic photosynthesis, we report a dual biohybrid system for light-enhanced chemical production in Escherichia coli. The system combines extracellular conjugated polymer nanoparticles as light-harvesting antenna with intracellularly biomineralized CdS nanoparticles as reaction-center

Methanotroph Methylotuvimicrobium alcaliphilum 20Z-3E as a fumarate producer: transcriptomic analysis and the role of malic enzyme.

The halotolerant obligate methanotroph Methylotuvimicrobium alcaliphilum 20Z is a promising biotechnological strain that has been repeatedly tested as a producer of high-added-value polycarbon compounds. The mutant M. alcaliphilum 20Z-3E lacking two fumarases and a malic enzyme is a potential fumarate producer. The analysis of strand-specific 3'-end sequencing of mRNA did not reveal any effects of the mutations on the central metabolism of the methanotroph; however, it showed a dramatic change in the expression of putative iron transport genes, as well as some genes associated with stress resp

Effect of dipeptides on the growth of Oenococcus oeni in synthetic medium deprived of amino acids.

Oenococcus oeni has numerous amino acid requirements for growth and dipeptides could be important for its nutrition. In this paper the individual or combined effect of dipeptides on growth of O. oeni X2L in synthetic media deficient in one or more amino acids with L-malic acid was investigated. Utilization of dipeptides, glucose, and L-malic acid was also analyzed. Dipeptides were constituted by at least one essential amino acid for growth. Dipeptides containing two essential amino acids, except leucine, had a more favorable effect than free amino acids on the growth rate. Gly-Gly was consumed

[Metabolic state of the other eye in post-traumatic uveitis].

Biochemical analysis of the paired eye lacrimal fluid lipid peroxides (diene conjugates, Schiff's bases, TBA-active products), antioxidant enzymes (catalase, superoxide dismutase), dehydrogenases (lactate and malate dehydrogenases), and lactic and pyruvic acids was carried out during the early and remote posttraumatic period in 468 patients after perforating injuries to the eye. Signs of activation of lipid peroxidation, antioxidants, dehydrogenases, and of glycolysis intensification were detected. These changes corresponded to the results of analysis of lacrimal fluid of the injured eye and d

Enzyme polymorphism in Schistosoma mattheei from cattle in the Eastern Transvaal Lowveld.

Enzyme electrophoresis was conducted on 10 Schistosoma mattheei adult worm samples, comprising 270 individuals, collected from cattle in the Eastern Transvaal Lowveld. Glucose-6-phosphate dehydrogenase (G6PDH) was studied in all the samples and phosphoglucomutase (PGM) and malate dehydrogenase (MDH) in five populations each. Only one population was polymorphic for G6PDH. In this population, in addition to the allele found in all the other samples, a second allele occurred with a similar Rf value to S. haematobium. The two alleles were in Hardy-Weinberg equilibrium. MDH-1 exhibited two alleles.
