Exploring gadolinium deposition in maternal and offspring mice: Impacts of gestational and lactational exposure.

To investigate the gadolinium deposition induced by repeated administration of gadolinium-based contrast agents (GBCAs) in multi-organ/tissue of mother and pup mice during pregnancy and lactation, two hundred and seventy ICR mice were divided into three groups (non-pregnant, pregnant, and lactating; n = 90/group) and received gadodiamide, gadoterate meglumine, or saline intravenously (2.5 mmol Gd/kg once every two days for a total of 10 doses) throughout the entire gestation or lactation period. Gadolinium concentration detection, histological analyses, and transmission electron microscopy wer

Gene-by-Environment Interactions Involving Maternal Exposures with Orofacial Cleft Risk in Filipinos.

BACKGROUND/OBJECTIVES: Maternal exposures are known to influence the risk of isolated cleft lip with or without cleft palate (CL/P)-a common and highly heritable birth defect with a multifactorial etiology. METHODS: To identify new risk loci, we conducted a genome-wide gene-environment interaction (GEI) analysis of CL/P with maternal smoking and vitamin use in Filipinos (Ncases = 540, Ncontrols = 260). Since GEI analyses are typically low in power and the results can be difficult to interpret, we applied multiple testing frameworks to evaluate potential GEI effects: a one degree-of-freedom (1

Perinatal psychiatry: balancing maternal-fetal exposures to mental disorders and drug treatments.

INTRODUCTION: The potential effects of psychotropic medications during pregnancy must be balanced against those of untreated maternal mental illness, which has been under-appreciated as an exposure that impacts development. METHODS: This is an expert, non-systematic literature review of the effect of maternal stress and psychotropic medication on fetal development. RESULTS: Studies on the outcomes of pregnancy and child development following exposures to stress, mental disorders and psychotropic medications during pregnancy are reviewed. Alterations in pharmacokinetics due to the physiology

To identify new CL/P risk loci, we conducted a genome-wide gene-environment interaction (GEI) analysis of CL/P on a sample of 540 cases and 260 controls recruited from the Philippines, incorporating the interaction effects of genetic variants with maternal smoking and vitamin use. As GEI analyses are typically low in power and the results can be difficult to interpret, we used multiple testing frameworks to eva

Exposure to Dengue Virus During Pregnancy: Incidence and Impact on Maternal and Child Outcomes.

Dengue virus (DENV) is the most common arbovirus globally, with its incidence growing dramatically in recent decades. Although the effects of DENV infection during pregnancy are unclear, reported associations with adverse health outcomes include miscarriage, prematurity, and low birth weight. In this study, we used an IgG ELISA to identify mothers exposed to DENV during pregnancy by testing samples obtained from a previous study that followed a cohort of pregnant women in Kenya to investigate parasitic infections during pregnancy. We compared adverse pregnancy and infant health outcomes betwee

Maternal pesticides exposure in pregnancy and the risk of wheezing in infancy: A prospective cohort study.

INTRODUCTION: Pesticide exposure in pregnancy may have health effects in the offspring. We studied whether maternal pesticides exposure during pregnancy is associated with infant wheezing. METHODS: The study involved 5997 children from the Italian NINFEA birth cohort, whose mothers were recruited during pregnancy between 2005 and 2016. We used questionnaires completed during pregnancy and 6 months after delivery to derive the following indirect measures of exposure: i) Self-reported pesticide use during the first and the third trimester of pregnancy; (ii) Agricultural activities during the sa

Pregnancy outcomes following maternal or paternal exposure to teriflunomide in the Danish MS population.

Background Teriflunomide (TFL) is an oral drug used for treatment of multiple sclerosis (MS). Due to possible teratogenicity it is contraindicated in women of childbearing potential. Additionally, a low risk of male-mediated embryo-fetal toxicity has been described as the drug can be transmitted via semen. This study investigated the association of adverse perinatal outcomes in newborns with either maternal or paternal exposure to TFL. Methods The Danish Multiple Sclerosis Registry was used to identify all patients treated with teriflunomide from 1st of September 2013 to 31st of December 2018.

Associations between maternal and environmental exposures on atopic disease in the offspring of mothers with asthma.

BACKGROUND AND OBJECTIVE: Available data on the impact of perinatal and environmental factors on atopic diseases in the offspring are sparse and conflicting. We, therefore, investigated the impact of these factors on the risk of atopic diseases in the offspring of women with asthma. METHODS: Pregnant women referred to give birth at Copenhagen University Hospital-Hvidovre, Denmark, have since 2007 been invited to participate in the Management of Asthma during Pregnancy program. Women with diagnosed asthma, currently prescribed asthma medication, first visit to the respiratory out-patient clini

Maternal exposure to sulfur dioxide before and after conception and the risk of hypospadias in offspring.

As far as we know, there have been no studies exploring the association between maternal sulfur dioxide (SO2 ) exposure and the risk of hypospadias in offspring. We aimed to evaluate this association during the 3 months before conception and the first trimester. A population-based case-control study was conducted in male infants, consisting of 348 cases of hypospadias and 4023 controls. Maternal exposure to SO2 , particulate matter with aerodynamic diameters ≤10 μm (PM10 ), and nitrogen dioxide (NO2 ) was assessed by averaging the concentrations recorded at all stations in the mother's city of

Neural processing of infant and adult face emotion and maternal exposure to childhood maltreatment.

Face processing in mothers is linked to mother-infant social communication, which is critical for parenting and in turn for child development. Neuroimaging studies of child maltreatment-exposed (CME) mothers are sparse compared to studies of mothers with postpartum depression, which have suggested blunted amygdala reactivity to infant stimuli. We expected to see a similar pattern in CME mothers. Based on broader studies in trauma-exposed populations, we anticipated increased amygdala reactivity to negative adult face stimuli in a comparison task in CME mothers given heightened evaluation of po

Incidence of Neonatal Neutropenia and Leukopenia After In Utero Exposure to Chemotherapy for Maternal Cancer.

OBJECTIVE: The main purpose of this article was to report the incidence of neonatal neutropenia or leukopenia after chemotherapy exposure during pregnancy according to the time elapsed between treatment during pregnancy and birth. BACKGROUND: A single study reports 33% of infants exposed to chemotherapy within the last month of pregnancy are born with neutropenia, which can place the newborn at risk for nosocomial infections. On the basis of this report, chemotherapy is typically stopped by 34 weeks of pregnancy to avoid maternal or neonatal myelosuppression at delivery. Such a pause in treat

Exposure to maternal odor enhances intake of a taste that mimicks the sensory attributes of ethanol.

Early exposure to ethanol increases subsequent acceptance of this drug. Little attention, however, has been devoted to the interaction of the taste of the drug with other, familiar or non-familiar, odors contingent with ethanol access, particularly early in ontogeny. This study assessed the influence of exposure to maternal odor on intake and grasp responses to an artificial nipple providing a solution (a sucrose-quinine mix) that emulates the taste of alcohol, in 4-day-old rat pups. The results showed that the mother's odor enhanced intake from and seeking responses to an artificial nipple th

Exposure to maternal obesity programs sex differences in pancreatic islets of the offspring in mice.

AIMS/HYPOTHESIS: Obesity during pregnancy increases offspring type 2 diabetes risk. Given that nearly half of women of child-bearing age in many populations are currently overweight/obese, it is key that we improve our understanding of the impact of the in utero/early life environment on offspring islet function. Whilst a number of experimental studies have examined the effect of maternal obesity on offspring islet architecture and/or function, it has not previously been delineated whether these changes are independent of other confounding risk factors such as obesity, postnatal high-fat-feedi

Exposure to early and persistent maternal depression impairs the neural basis of attachment in preadolescence.

Maternal depression increases child vulnerability to psychopathology, loneliness, and social maladjustment; yet, its long-term effects on the social brain are unknown. In this prospective longitudinal study we examined the impact of early and persistent maternal depression on the neural basis of attachment in preadolescence. A community cohort was followed in two groups; children exposed to maternal depression from birth to 6 years and healthy controls. At 9 months and 6 years, mother-child interactions were coded for maternal sensitivity and affect synchrony and salivary oxytocin levels were
