Studies coordinated by the World Health Organization were conducted in seven clinical centres on the chemotherapy of human echinococcosis with mebendazole, albendazole, and flubendazole. The first phase of these ended with the following conclusions.Treatment of 85 patients with mebendazole for cystic (Echinococcus granulosus) echinococcosis was successful in 8 patients and partially successful in 4 others. Flubendazole was effective in only one case of lung echinococcosis. Albendazole was successful in 5 of 30 patients treated and partially successful in 4 others. Further studies on new drugs …

Mebendazole was given to four patients with hepatic hydatid disease. In three patients hydatidosis had remained after surgery, and in the fourth it could not be treated surgically. Mebendazole was given orally in maximum doses of 400-600 mg three times a day during courses lasting 21 to 30 days. Ultrasonic echotomography showed a complete regression of the intrahepatic cysts after four to 13 months in all four cases. In three patients the course of treatment had to be repeated. Mebendazole also induced clinical improvement and a progressive lowering of the concentration of specific IgE of Echi…

Iron deficiency and helminth infections are two common conditions of children in developing countries. The consequences of helminth infection in young children are not well described, and the efficacy of low dose iron supplementation is not well documented in malaria-endemic settings. A 12-mo randomized, placebo controlled, double-blind trial of 10 mg daily iron and/or mebendazole (500 mg) every 3 mo was conducted in a community-based sample of 459 Zanzibari children age 6-71 mo with hemoglobin > 70 g/L at baseline. The trial was designed to examine treatment effects on growth, anemia and appe…

The continuing heavy toll of the COVID-19 pandemic necessitates development of therapeutic options. We adopted structure-based drug repurposing to screen FDA-approved drugs for inhibitory effects against main protease enzyme (Mpro) substrate-binding pocket of SARS-CoV-2 for non-covalent and covalent binding. Top candidates were screened against infectious SARS-CoV-2 in a cell-based viral replication assay. Promising candidates included atovaquone, mebendazole, ouabain, dronedarone, and entacapone, although atovaquone and mebendazole were the only two candidates with IC50s that fall within thei…

BACKGROUND: Mebendazole and albendazole are the drugs of choice for the treatment of alveolar echinococcosis. In this prospective study we present and evaluate the outcome of the long-term treatment with both drugs.
PATIENTS AND METHODS: Forty-four patients were treated with either mebendazole or albendazole and they were followed up for an average of 42 months. Success of treatment was defined as non-progression for more than 1 year.
RESULTS: The overall success-rate was approximately 80% (35/44). An initial regimen was recurrence-free in 64% of cases under mebendazole and in 73% of cases u…

BACKGROUND: Trichuriasis, caused by the nematodes Trichuris trichiura and Trichuris incognita, affects an estimated 267 million people worldwide, particularly children. Currently available treatments (albendazole and mebendazole) fail to achieve sufficient cure rates. Oxantel pamoate is a drug known for its trichuricidal activity. In this study, we aimed to assess whether a newly developed, child-friendly oxantel pamoate formulation is superior to mebendazole, one of WHO's recommended treatments for trichuriasis.
METHODS: We conducted a randomised, single-blind, phase 2 trial in a primary sch…

This randomized, double-blind, placebo-controlled study evaluated the efficacy and safety of a new chewable, rapidly-disintegrating mebendazole (MBZ) 500 mg tablet for Ascaris lumbricoides and Trichuris trichiura infection treatment. Pediatric patients (1-15 years; N = 295; from Ethiopia and Rwanda) excreting A. lumbricoides and/or T. trichiura eggs were enrolled. The study had a screening phase (3 days), a double-blind treatment phase (DBP, 19 days), and an open-label phase (OLP, 7 days). Patients received MBZ or placebo on day 1 of DBP and open-label MBZ on day 19 ± 2 after stool sample coll…

Outcomes for pediatric acute myeloid leukemia (AML) have improved significantly in recent years. However, relapsed and refractory disease remains a significant problem. The chemotherapy burden experienced by these patients makes the translational development of non-genotoxic experimental therapies attractive. We previously reported that the anti-helminth drug mebendazole induces degradation of the transcription factor MYB and has potent anti-AML activity. In the present study, we use CRISPR drop-out screening to identify genes encoding the proapoptotic regulators BAK and NOXA as hits conferrin…

There is no consensus on the benefits of treatment with any specific anthelminthic compound on muscle-stage trichinosis. A double-blind, placebo-controlled comparison was done of 3 antiparasitic drugs during an outbreak of trichinosis in Chiangrai Province, northern Thailand. Forty-six adults were randomized to receive 10 days of oral treatment with mebendazole (200 mg twice a day), thiabendazole (25 mg/kg twice a day), fluconazole (400 mg initially, then 200 mg daily), or placebo. All patients received treatment to eradicate adult intestinal worms. Trichinella spiralis infection was proved pa…

Attempts to reduce the standard treatment of intestinal capillariasis of 200 mg mebendazole twice daily for 20 days were not successful. Two of 13 patients given mebendazole at single daily dosages of 500 mg per day for 16 days relapsed 3 to 6 months after treatment. Attempts to reduce the treatment to 14 days were even less satisfactory in that 4 of 8 patients treated experienced relapses 2 to 9 months later. Since intestinal capillariasis can be fatal it is recommended to continue the use of the standard schedule of 400 mg mebendazole daily for 20 days for new cases and 30 days for relapsed …

A cross-sectional study was conducted from March to September, 2018 on the efficacies of albendazole and mebendazole against ancylostomiasis in school children of district Swat, Pakistan. Faecal samples were collected from primary school children and preserved in 10% formalin. The samples were then sent to the Laboratory of Parasitology, in the University of Malakand for microscopic analysis. On the basis of drug availability, the Ancylostoma dueodenale infected students were divided into two groups. Group A was treated with Albendazole 400-450mg while group B was orally treated with Mebendazo…

A case of severe Strongyloides stercoralis infection is described in a patient with renal failure due to bilateral cortical necrosis. Because thiabendazole, the drug of choice in strongyloidiasis, is primarily excreted by the kidneys, alternative therapy with a mebendazole was instituted. Although the patient eventually died as a result of the renal failure, successful therapy with mebendazole was well demonstrated.

A randomized controlled trial comparing the efficacy of pyrantel-oxantel (10 mg/kg) with mebendazole (500 mg) was performed on 1329 schoolchildren aged 6-9 years on Pemba Island in September-October 2000 to evaluate alternative single-dose drugs for regular treatment of intestinal nematode infections. Both mebendazole and pyrantel-oxantel were very effective in eliminating Ascaris lumbricoides infection, inducing cure rates of more than 96% and reducing the mean egg counts by more than 95%. Both drugs had a moderate efficacy against Trichuris trichiura infection, but pyrantel-oxantel had a hig…

CONTEXT: More than a quarter of the human population is likely infected with soil-transmitted helminths (Ascaris lumbricoides, hookworm, and Trichuris trichiura) in highly endemic areas. Preventive chemotherapy is the mainstay of control, but only 4 drugs are available: albendazole, mebendazole, levamisole, and pyrantel pamoate.
OBJECTIVE: To assess the efficacy of single-dose oral albendazole, mebendazole, levamisole, and pyrantel pamoate against A lumbricoides, hookworm, and T trichiura infections.
DATA SOURCES: A systematic search of PubMed, ISI Web of Science, ScienceDirect, the World He…

In an experimental patent infection of Ancylostoma ceylanicum in golden hamsters the efficacy of seven anthelmintics was tested; bephenium hydroxynaphthoate, tetramisole, tetrachlorethylene, phenylene 1,4-diisothiocyanate, thiabendazole, parbendazole and mebendazole were studied. Mebendazole was the most effective, with parbendazole, phenylene 1,4-diisothiocyanate, thiabendazole and bephenium hydroxynaphthoate also satisfactory. Tetrachlorethylene and tetramisole were the least effective in our screen.

This article discusses the anthelmintics now in common use and describes their mode of action and toxic side effects. The drugs reviewed include: pyrantel pamoate, mebendazole, piperazine, thiabendazole, niclosamide, praziquantel, niridazole, oxamniquine and diethylcarbamazine. Most of the common roundworm intestinal infections respond to treatment with pyrantel pamoate, which has the advantage of single-dose therapy. Trichuriasis requires therapy with mebendazole, while the filariases are treated with diethylcarbamazine. There is no specific therapy for trichinellosis, but its symptoms can be…

Childhood infections with some helminths have been reported to reduce the risk of acquiring some immune-mediated diseases like sensitization to allergens, asthma and inflammatory bowel disease. Denmark keeps a medical registry that collects data on disease prevalence and medication use throughout the country. Mebendazole frequently is used to treat pinworm (Enterobius vermicularis) infection. In this population-based analysis of a cohort of 924,749 children, prescriptions for Mebendazole were used as a surrogate marker for pinworm infection. Children who received this medication were compared …

Four cases of multivesicular hepatic hydatidosis are reported. Two of them were affected by exogenous growth. All patients had undergone a long-term treatment with mebendazole at the usual doses. We are contrasting our experience in the use of such drug and the previous reports by other authors. In our opinion, as mebendazole acts on the cyst in a double way (both inhibiting glucose uptake by the larva and modifying the structure of the cyst membranes) ir induces the cyst multivesiculation which is a sign of suffering or resistance of the parasite. Despite the fact that previous reports, and a…
