Excess molybdenum impairs growth, nitrogen metabolism and nutrient translocation of soybean in Bradyrhizobium symbiosis, with parallels to tungsten stress.

Molybdenum (Mo) serves an important biological role as part of cofactors of various enzymes in all domains of life. In plants, it is critical for nitrogen metabolism and integral to rhizobial enzymes such as nitrogenase, which is essential for plant-rhizobia symbiosis. However, like other transition metals, Mo can be toxic at excess concentrations. Its chemical analog tungsten (W) has no biological function in eukaryotes and is known to be toxic, primarily by inhibiting molybdoenzymes. Previously, we found that soybean (Glycine max) in symbiosis with N2-fixing rhizobia (Bradyrhizobium japonicu…

Molybdenum Disulfide Supported on Metal-Organic Frameworks as an Ultrasensitive Layer for the Electrochemical Detection of the Ovarian Cancer Biomarker CA125.

Metal-organic frameworks (MOFs) are composed of metal ions/clusters and organic ligands, showing accessible functional sites, ultra-high porosity, and large specific surface area. Tricopper benzene-1,3,5-tricarboxylate (CuBTC), as a three-dimensional MOF architecture with an open and robust micro-/nanoconfiguration, possesses excellent catalytic performance and superior electric conductivity as compared to bulk MOF. In this study, CuBTC was used as a substrate on which molybdenum disulfide (MoS2) was in situ constructed by a hydrothermal reaction to enhance the electron- and ion-transfer capab…

Effects of molybdenum deficiency and defects in molybdate transporter MOT1 on transcript accumulation and nitrogen/sulphur metabolism in Arabidopsis thaliana.

Molybdenum (Mo) is a micronutrient essential for plant growth, as several key enzymes of plant metabolic pathways contain Mo cofactor in their catalytic centres. Mo-containing oxidoreductases include nitrate reductase, sulphite oxidase, xanthine dehydrogenase, and aldehyde oxidase. These are involved in nitrate assimilation, sulphite detoxification, purine metabolism or the synthesis of abscisic acid, auxin and glucosinolates in plants. To understand the effects of Mo deficiency and a mutation in a molybdate transporter, MOT1, on nitrogen and sulphur metabolism in Arabidopsis thaliana, transcr…

Mild renal failure induced by subchronic exposure to molybdenum: urinary kallikrein excretion as a marker of distal tubular effect.

In this study we investigated the effect of two molybdenum (Mo) doses (40 and 80 mg/kg/d) on renal function. Neither dose of Mo was able to induce significant hypertension in treated animals. Subchronic exposure to high doses of Mo resulted in a delay in body weight gain associated with mild renal failure marked by a decrease in glomerular filtration. An increase in diuresis and urinary kallikrein excretion associated with unchanged glycosuria and proximal tubular enzymuria (alanine aminopeptidase and gamma-glutamyl transpeptidase) evoked a preferential mild effect at the distal tubule.

Absence of hepatic molybdenum cofactor: an inborn error of metabolism leading to a combined deficiency of sulphite oxidase and xanthine dehydrogenase.

Five patients with a combined deficiency of xanthine dehydrogenase, sulphite oxidase and, possibly, also of aldehyde oxidase are described. This remarkable coincidence of three inborn errors of metabolism in a single individual was demonstrated to result from a deficiency of the 'molybdenum cofactor', an essential constituent of all three enzymes. The main biochemical findings in these patients included: hypouricaemia, xanthinuria, an increased excretion of sulphite, thiosulphate and S-SUL-sulphocysteine and a decreased excretion of inorganic sulphate. Plasma molybdenum was normal. The ultimat…

Molybdenum and cadmium co-activation of TLR4/MyD88/NF-κB: the hidden driver of lymphocyte inflammation in sheep spleen.

Environmental accumulation of excessive molybdenum (Mo) and cadmium (Cd) exerts detrimental effects on organisms. The TLR4/MyD88/NF-κB signaling cascade is classified as a core pathway responsible for regulating inflammatory responses in organisms. To elucidate the mechanism by which combined Mo and Cd exposure induces sheep spleen injury and clarify how the TLR4/MyD88/NF-κB pathway modulates this pathological process, we constructed both in vivo and in vitro models. Mo and/or Cd exposure induces splenic histological changes and mitochondrial ultrastructural damage. Notably, mRNA transcript an…

Accumulation of molybdenum in major organs following repeated oral administration of bis-choline tetrathiomolybdate in the Sprague Dawley rat.

Molybdenum is an essential dietary trace element required for several critical enzyme systems. High intake is associated with toxicity in ruminants and animal studies. The proposed therapeutic use of molybdenum-based drugs poses a potential risk for accumulation through chronic administration of therapeutic doses of this element. The current experiment was designed to study the effect of daily dosing of a molybdenum compound, bis-choline tetrathiomolybdate (TTM), in Sprague Dawley rats using laser ablation inductively coupled plasma time-of-flight mass spectrometry (LA-ICP-ToF-MS) and two dosi…

The use of XAFS to determine the nature of interaction of iron and molybdenum metal salts within PS-b-P2VP micelles.

The poly(styrene)-block-poly(2-vinylpyridine) (PS-b-P2VP) micelle route is a well established method for the preparation of bimetallic nanoparticles used for the catalysis of carbon nanotubes and other applications like ultrahigh density storage devices, yet to date no information is available concerning the internal structure of the P2VP-metal salt complex. For the first time, XAFS measurements were performed on micelles loaded with either iron(III) chloride or molybdenum(V) chloride and a combination of both. Analysis of the data revealed that iron is tetrahedrally coordinated within the cor…

Molecular characterization of human xanthine oxidoreductase: the enzyme is grossly deficient in molybdenum and substantially deficient in iron-sulphur centres.

XOR (xanthine oxidoreductase) purified from human milk was shown to contain 0.04 atom of Mo and 0.09 molecule of molybdopterin/subunit. On the basis of UV/visible and CD spectra, the human enzyme was approx. 30% deficient in iron-sulphur centres. Mo(V) EPR showed the presence of a weak rapid signal corresponding to the enzyme of low xanthine oxidase activity and a slow signal indicating a significant content of desulpho-form. Resulphuration experiments, together with calculations based on enzymic activity and Mo content, led to an estimate of 50-60% desulpho-form. Fe/S EPR showed, in addition

Molybdenum and vanadium do not replace tungsten in the catalytically active forms of the three tungstoenzymes in the hyperthermophilic archaeon Pyrococcus furiosus.

Three different types of tungsten-containing enzyme have been previously purified from Pyrococcus furiosus (optimum growth temperature, 100 degrees C): aldehyde ferredoxin oxidoreductase (AOR), formaldehyde ferredoxin oxidoreductase (FOR), and glyceraldehyde-3-phosphate oxidoreductase (GAPOR). In this study, the organism was grown in media containing added molybdenum (but not tungsten or vanadium) or added vanadium (but not molybdenum or tungsten). In both cell types, there were no dramatic changes compared with cells grown with tungsten, in the specific activities of hydrogenase, ferredoxin:N…

Myco-mediated synthesis of molybdenum oxide nanoparticles using Fusarium sp. strain JK-2 and their anticancer and antibacterial applications.

Oral cancer is among the six most prevalent cancers globally, with conventional treatments such as chemotherapy and radiotherapy limited by systemic toxicity, drug resistance and poor selectivity. Although molybdenum trioxide nanoparticles (MoO3NPs) have shown promising biomedical applications, studies on myco-mediated MoO3NP synthesis and their specific relevance to oral cancer therapy remains limited, highlighting an important knowledge gap. In this study, we report the first myco-fabrication of Fu@MoO3NPs using an endophytic fungus, Fusarium sp. strain JK-2, isolated from Justicia tranqueba…
