Role of herbal bioactives as a potential bioavailability enhancer for Active Pharmaceutical Ingredients.

The current review emphasizes on the herbal bioenhancers which themselves do not possess inherent pharmacological activity of their own but when co-administered with Active Pharmaceutical Ingredients (API), enhances their bioavailability and efficacy. Herbal bioenhancers play a crucial role in enhancing the bioavailability and bioefficacy of different classes of drugs, such as antihypertensives, anticancer, antiviral, antitubercular and antifungal drugs at low doses. This paper highlights various natural compounds that can be utilized as an efficient bioenhancer. Several herbal compounds inclu

Sinomenine, theophylline, cysteine, and levamisole: Comparisons of their kinetic effects on mineral formation induced by matrix vesicles.

The effects of sinomenine (SIN, an alkaloid extracted from the Chinese medicinal plant Sinomenium acutum used for centuries to treat rheumatic disease, including rheumatoid arthritis) on apatitic nucleation and matrix vesicle (MV)-induced mineral formation were compared with those of cysteine, levamisole, and theophylline. We found that SIN was not an inhibitor of tissue non-specific alkaline phosphatase (TNAP), a marker of biological mineralization, but confirmed that cysteine, levamisole, and theophylline were. Further, none of these four molecules directly affected the nucleation of hydroxy

Activation of mu-opioid receptors are required for the conditioned enhancement of NK cell activity.

The type of opioid receptors involved in the conditioned enhancement of natural killer (NK) cell activity is identified in the present study. In our previous observations, we have demonstrated that the conditioned enhancement of NK cell activity was dependent on beta-endorphin and methionine-enkephalin, but not dynorphin. Based on the interaction of opioids with their homologous receptors, we concluded that mu- and delta-opioid receptors might be involved. To further classify the type(s) of opioid receptors involved in eliciting the conditioned NK cell activity, three opioid receptor antagonis

Voluntary respiratory effects of butorphanol and fentanyl following barbiturate induction: a double-blind study.

Thirty-one healthy women undergoing outpatient gynecologic surgery were given either butorphanol or fentanyl following barbiturate induction, and various respiratory parameters were measured at intervals after administration of the study drugs. Both fentanyl and butorphanol caused comparable decrements in tidal volume and other respiratory parameters. However, seven of the 15 patients receiving fentanyl had significant respiratory depression that required assisted ventilation. One of the butorphanol patients experienced a 45-second self-limited respiratory depression. One of three butorphanol

In vivo opiate receptor binding of oripavines to mu, delta and kappa sites in rat brain as determined by an ex vivo labeling method.

The relative in vivo receptor affinities of three oripavine drugs given subcutaneously were determined at the mu, delta and kappa type of opiate binding sites in rat brain. The oripavines include the agonist etorphine, the antagonist diprenorphine and the mixed agonist-antagonist buprenorphine. With the use of mu, delta and kappa specific labeling conditions in brain homogenates immediately after sacrifice (ex vivo labeling), the method relies on the assay of those receptor sites that remain unbound in vivo. Because of the slow receptor binding kinetics of the oripavines, little or no dissocia

A double blind comparison of buprenorphine and morphine in conscious subjects following administration by the intramuscular route.

Buprenorphine was studied to determine the respiratory depressant effect in a double blind comparison with morphine. Buprenorphine produced a more prolonged displacement of the carbon dioxide response curve than morphine. Because of the marked difference in the time effect curves the relative potency was determined at the time of peak effect--3 hours post-administration for buprenorphine (0.15 and 0.3 mg) and 1 hour post administration for morphine (5 and 10 mg). The data from the subjects who received higher doses of buprenorphine (0.6 and 1.2 mg) showed that there was a linear relationship o

Sinomenine hydrochloride-loaded liposomes-exosomes membrane fusion biohybrid attenuating collagen-induced rheumatoid arthritis via NF-κB pathway.

Sinomenine hydrochloride (SH) exhibits anti-inflammatory potential for rheumatoid arthritis (RA) but is hindered by suboptimal bioavailability. The NF-κB pathway, a central mediator of RA inflammation and immune dysfunction, remains a critical therapeutic target. Here, we engineered Sinomenine Hydrochloride-Loaded Liposomes-Exosomes Membrane Fusion Biohybrid (SHLE) to enhance SH delivery and achieve targeted NF-κB pathway modulation. Sinomenine hydrochloride-loaded liposomes (SHL) were prepared via thin-film dispersion and fused with exosomes using ultrasonic technology. In a collagen-induced

Sinomenine restrains the proliferation and hyperactivation of B lymphocytes partly by inhibiting interferon regulatory factor 5.

ETHNOPHARMACOLOGICAL RELEVANCE: B lymphocytes are central drivers of pathogenic immune responses in autoimmune diseases and represent a promising target for therapeutic intervention. Sinomenine (SIN), a natural alkaloid derived from Sinomenium acutum, exhibits broad immunomodulatory properties and has long been clinically used for treating autoimmune diseases, however, its direct effects on B cells and underlying mechanisms are largely unexplored. AIM OF THE STUDY: To investigate the effects of SIN on the proliferation and hyperactivation of B lymphocytes both in vitro and in vivo, and elucid

A Novel Dual Therapy for Diabetic Ischemic Heart Injury: Sinomenine and Liraglutide Restore Mitochondrial Homeostasis and Suppress Pyroptosis.

Type 2 diabetes mellitus (T2DM) significantly amplifies the risk and severity of myocardial ischemia-reperfusion (IR) injury, contributing to the disproportionately high cardiovascular mortality observed in diabetic patients. This study investigated the combined cardioprotective effects of sinomenine and liraglutide in a rat model of T2DM with myocardial IR injury and elucidated the underlying molecular mechanisms. Male Sprague Dawley rats were induced to T2DM with a high-fat diet and low-dose streptozotocin. After the induction of diabetes, the rats underwent 30 min of occlusion of the left a

Comparison of effect and mechanism between nalfurafine hydrochloride and narrow-band ultraviolet B phototherapy in the treatment of pruritus in hemodialysis patients.

OBJECTIVES: Pruritus in hemodialysis patients (HDP) is one of the serious complications associated with the quality of life and psychiatric disorder of patients. The narrowband ultraviolet B phototherapy (NB-UVB) treatment showed a statistically significant reduction of itching when performed more than 4 times compared to conservative treatment for itching, and the difference was confirmed to increase with repetition. We aim to compare newly developed Nalfuranfine HCL, kappa-opioid receptor agonist, with NB-UVB in HDP. METHODS: Twenty HDP were enrolled from Wonju severance Christian hospital.

Nalfurafine promotes myelination in vitro and facilitates recovery from cuprizone + rapamycin-induced demyelination in mice.

The kappa opioid receptor has been identified as a promising therapeutic target for promoting remyelination. In the current study, we evaluated the ability of nalfurafine to promote oligodendrocyte progenitor cell (OPC) differentiation and myelination in vitro, and its efficacy in an extended, cuprizone-induced demyelination model. Primary mouse (C57BL/6J) OPC-containing cultures were treated with nalfurafine (0.6-200 nM), clemastine (0.01-100 μM), T3 (30 ng/mL), or vehicle for 5 days. Using immunocytochemistry and confocal microscopy, we found that nalfurafine treatment increased OPC differen

Milonine attenuates the lipopolysaccharide-induced acute lung injury in mice by modulating the Akt/NF-κB signaling pathways.

Acute lung injury is an inflammation that triggers acute respiratory distress syndrome with perialveolar neutrophil infiltration, alveolar-capillary barrier damage, and lung edema. Activation of the toll-like receptor 4 complex (TLR4/MD2) and its downstream signaling pathways are responsible for the cytokine storm and cause alveolar damage. Due to the complexity of this pulmonary inflammation, a defined pharmacotherapy has not been established. Thus, this study evaluated the anti-inflammatory potential of milonine, an alkaloid of Cissampelos sympodialis Eichl, in an experimental model of lung

A non-convulsant delta-opioid receptor agonist, KNT-127, reduces cortical spreading depression and nitroglycerin-induced allodynia.

OBJECTIVE: The aim of this study was to determine if the non-convulsant delta-opioid receptor (DOR) agonist, KNT-127, could inhibit migraine-associated endpoints. BACKGROUND: The DOR has been identified as a therapeutic target for migraine. However, the development of delta agonists is limited as some ligands have seizurogenic properties, which may be related to their ability to induce receptor internalization. While both pro- and non-convulsant delta agonists can reduce migraine-associated allodynia, only the proconvulsant agonist, SNC80, has been shown to decrease cortical spreading depress

Nalfurafine suppresses pruritogen- and touch-evoked scratching behavior in models of acute and chronic itch in mice.

The kappa-opioid agonist, nalfurafine, has been approved in Japan for treatment of itch in patients with chronic kidney disease. We presently investigated if systemic administration of nalfurafine inhibited ongoing or touch-evoked scratching behavior (alloknesis) following acute intradermal injection of histamine or the non-histaminergic itch mediator, chloroquine, in mice. We also investigated if nalfurafine suppressed spontaneous or touch-evoked scratching in an experimental model of chronic dry skin itch. Nalfurafine reduced scratching evoked by histamine and chloroquine. Following acute hi

Sinomenine potentiates degranulation of RBL-2H3 basophils via up-regulation of phospholipase A2 phosphorylation by Annexin A1 cleavage and ERK phosphorylation without influencing on calcium mobilization.

Sinomenine (SIN), an alkaloid derived from the Chinese medicinal plant Sinomenium acutum, is the major component of Zhengqing Fongtong Ning (ZQFTN), a pharmaceutical drug produced by Hunan Zhengqing Pharmaceutical Co. Ltd. in China for the treatment of rheumatoid arthritis and other autoimmune diseases. Some clinic reports indicate that ZQFTN may induce an anaphylactic reaction via potentiating the degranulation of immune cells. In the current study, we aimed to examine whether SIN is capable of inducing the degranulation of basophilic leukemia 2H3 (RBL-2H3) cells to elucidate how the anaphyla
