The brainstem connects the forebrain to the spinal cord. It consists of three layers, the ventral basis, the intermediate tegmentum, and the dorsal tectum. Caudo-rostrally, the brainstem is divided into the medulla oblongata or myelencephalon, the metencephalon, and the mesencephalon. The tegmentum is the most conserved part of the brainstem and contains the motor and sensory nuclei of cranial nerves (CN) III-XII. They are arranged in a mediolateral direction, and, unlike the spinal cord, their afferent and efferent fibers are not macroscopically separated. The tegmental reticular formation, t

The thalamus is a neural processor and integrator for the activities of the forebrain. Surprisingly, little is known about the roles of the "cerebellar" thalamus despite the anatomical observation that all the cortico-cerebello-cortical loops make relay in the main subnuclei of the thalamus. The thalamus displays a broad range of electrophysiological responses, such as neuronal spiking, bursting, or oscillatory rhythms, which contribute to precisely shape and to synchronize activities of cortical areas. We emphasize that the cerebellar thalamus deserves a renewal of interest to better understa

Primary dystonia has traditionally been viewed as a basal ganglia disorder, but recent studies suggest that the cerebellum plays a crucial role in the disease. Primary dystonia is associated with several genotypes. Among those, DYT1 and DYT6 are inherited in autosomal dominant fashion with reduced penetrance. Extensive structural and functional imaging studies have been performed on manifesting and non-manifesting carriers of these mutations. The results suggest that primary dystonia can be viewed as a neurodevelopmental circuit disorder, involving the cortico-striato-pallido-thalamo-cortical

Chorea is one of the major types of involuntary movement disorders originating from dysfunctional neuronal networks interconnecting the basal ganglia and frontal cortical motor areas. The syndrome is characterised by a continuous flow of random, brief, involuntary muscle contractions and can result from a wide variety of causes. Diagnostic work-up can be straightforward in patients with a positive family history of Huntington's disease or acute-onset hemichorea in patients with lacunar stroke, but it can be a challenging and complex task in rare autoimmune or genetic choreas. Principles of man

We used the juxtacellular recording and labeling technique of Pinault (1996) in the uvula/nodulus of the ketamine anesthetized rat in an attempt to link different patterns of spontaneous activity with different types of morphologically identified cerebellar cortical interneurons. Cells displaying a somewhat irregular, syncopated cadence of spontaneous activity averaging 4-10 Hz could, upon successful entrainment and visualization, be morphologically identified as Golgi cells. Spontaneously firing cells with a highly or fairly regular firing rate of 10-35 Hz turned out to be unipolar brush cell

A review was conducted of the current knowledge of fetal and postnatal development of autonomic bladder function in animals. Studies of fetal and neonatal bladder development have been done in many animal species. Development of normal bladder function requires coordination of a number of different systems and processes, and continues after birth during the early neonatal period. In many neonatal animals, micturition occurs only after stimulation of a perineal-to-bladder reflex triggered when the mother licks the perineal region, and bladder distension fails to stimulate micturition. Various possible pathophysiological mechanisms to explain the pain in cluster headache are reviewed. Several locations for the initiation of pain along the primary afferent pathways in cranial nerves are discussed, together with the local mechanisms that might be responsible. These include neurogenic inflammation of large cranial vessels, irritation of vascular pain fibres by compression of dilated vessels in bony canals, and irritative foci in or around cranial sensory ganglia. In particular, recent neuroanatomical and pharmacological findings on nociceptive innervation of the intracranial se

Autism spectrum disorders (ASD) are diagnosed on the basis of three behavioral features namely deficits in social communication, absence or delay in language, and stereotypy. The susceptibility genes to ASD remain largely unknown, but two major pathways are emerging. Mutations in TSC1/TSC2, NF1, or PTEN activate the mTOR/PI3K pathway and lead to syndromic ASD with tuberous sclerosis, neurofibromatosis, or macrocephaly. Mutations in NLGN3/4, SHANK3, or NRXN1 alter synaptic function and lead to mental retardation, typical autism, or Asperger syndrome. The mTOR/PI3K pathway is associated with abn

Understanding how neurons of the striatum are formed and integrate into complex synaptic circuits is essential to provide insight into striatal function in health and disease. In this review, we summarize our current understanding of the development of striatal neurons and associated circuits with a focus on their embryonic origin. Specifically, we address the role of distinct types of embryonic progenitors, found in the proliferative zones of the ganglionic eminences in the ventral telencephalon, in the generation of diverse striatal interneurons and projection neurons. Indeed, recent evidenc

Dyspnoea is the result of a complex interaction of physiological, psychosocial, social, and environmental factors. Although several sensory receptors located throughout the respiratory system are considered to be responsible for generation of dyspnoea, there is no afferent receptor solely responsible for the sensation of dyspnoea. Afferent information from the sensory receptors is processed at the cortex along with the respiratory motor command from the cortex and brainstem, and a mismatch between the motor command and the incoming afferent information may result in dyspnoea. Dyspnoea is not a

Many electrophysiological studies have been conducted on the 'limbic system', which is known to play a crucial role in mnemonic, emotional, and olfactory functions. Because of the difficulty in accessing such deep brain structures under in vivo conditions, in vitro brain slice preparations are often used. Recertly, an isolated whole brain preparation in which multi-synaptic circuits and the intracellular activity they generate are well preserved has also become available. In this paper, we described about the studies of the olfactory circuits by using this unique preparation. This experimental

The kidneys play a central role in cardiovascular homeostasis by ensuring a balance between the fluid taken in and that lost and excreted during everyday activities. This ensures stability of extracellular fluid volume and maintenance of normal levels of blood pressure. Renal fluid handling is controlled via neural and humoral influences, with the former determining a rapid dynamic response to changing intake of sodium whereas the latter cause a slower longer-term modulation of sodium and water handling. Activity in the renal sympathetic nerves arises from an integration of information from th

Two classes of GABAergic cell bodies have been described. They probably can be divided into GABAergic local interneurons and GABAergic projection neurons. GABAergic cell bodies receive few terminals which is in contrast to non-GABAergic somata, which receive many synaptic contacts. GABAergic dendrites that originate from GABAergic cell bodies, however, receive numerous terminals, both GABAergic and nonGABAergic. It can therefore be concluded that somatic inhibition is not present on GABAergic neurons, but does occur on nonGABAergic neurons. Furthermore, dendrites traverse large parts of the NO

Neurons in the visual cortex were first found to be exquisitely selective for particular properties of visual stimuli in anesthetized animals, including mice. Studies of alert mice in an apparatus that allowed them to stand or run revealed that locomotion causes a change in cortical state that dramatically increases the magnitude of responses in neurons of the visual cortex without altering selectivity, effectively changing the gain of sensory responses. Locomotion also dramatically enhances adult plasticity in the recovery from long-term visual deprivation. We have studied the elements and op

The anterior hypothalamic area (AHA) has been implicated in a diverse array of functions ranging from defensive behavior selection to autonomic control. However, new methods that allow systematic characterization of the anatomical organization, connectivity, activity patterns, and molecular diversity have revealed the AHA as a hub for the convergence of innate and learned threat responding. Threat information is relayed to the AHA by synaptic inputs and circulating factors to convey information relevant for the regulation of organ system physiology and behavior. This structure is a critical hu

Gilles de la Tourette syndrome (GTS) is increasingly conceptualized as a neurodevelopmental network disorder. In this chapter, we have synthesized two decades of neuroimaging findings spanning gray- and white-matter morphology, structural connectivity, and functional network dynamics in GTS across childhood and adulthood. Structural MRI studies using voxel-based morphometry (VBM) have reported heterogeneous volume differences-both decreases and increases-across cortico-striatal-thalamo-cortical regions, motivating the use of meta-analytic approaches to identify more consistent loci, including

Schizophrenia (SZ) negative symptoms constitute a significant clinical challenge in everyday clinical practice, as most of them are persistent and unresponsive to standard psychopharmacotherapy. Recently, the role of brain network functional connectivity disturbances has been emphasized as a potential underlying mechanism for their occurrence and severity. This review aims to synthesize selected findings on altered brain network functional connectivity patterns associated with negative symptoms of SZ. Several distinctive functional dysconnectivity patterns have been identified in relation to p

Alzheimer's disease (AD) is the primary cause of dementia, and as the population ages, the number of individuals affected by the disease continues to rise. However, effective treatments for AD are still lacking. In the initial stages of AD, many brain regions are susceptible to its characteristic pathological changes, such as the Edinger-Westphal nucleus (EW nucleus) located in the midbrain region, which functions primarily as the parasympathetic oculomotor nucleus regulating pupil constriction. In this review, we outline the characteristics of EW nucleus alterations in AD and their potential

The parabrachial nucleus (PBN) is responsible for integrating both internal and external sensory information and controlling/regulating a wide range of physiological processes, such as feeding, thermogenesis, nociceptive and pruritic sensations, and respiration. Recently, the PBN has been found to be involved in mediating wakefulness maintenance, sleep-wake transition, exogenous neuromodulation of awakening, and arousal-promoting process triggered by drastic changes in the internal environments, such as hypercapnia, hypoxia, and hypertension. Multiple neural pathways and subpopulations of neur…
