Exogenous Strigolactone (GR24) Positively Regulates Growth, Photosynthesis, and Improves Glandular Trichome Attributes for Enhanced Artemisinin Production in Artemisia annua.

Artemisia annua is a medicinal plant particularly known for the production of a sesquiterpene lactone artemisinin; a specialty metabolite kn … rate. Moreover, the activity of various enzymes related to photosynthesis like carbonic anhydrase, nitrate reductase, and RuBisCO was escalated. The GR24 also improved certain attributes related to glandular trichomes, with a significant enhancement in content and yield of artemisinin as compared to untreated plants.

NO and N2 O transformations of diverse fungi in hypoxia: evidence for anaerobic respiration only in Fusarium strains.

Fungal denitrification is claimed to produce non-negligible amounts of N2 O in soils, but few tested species have shown significant activity. We hypothesized that denitrifying fungi would be found among those with assimilatory nitrate reductase, and tested 20 such batch cultures for their respiratory metabolism, including two positive controls, Fusarium oxysporum and Fusarium lichenicola, throughout the transition from oxic to anoxic conditions in media supplemented with NO 2 - . Enzymatic reduction of NO 2 - (NIR) and NO (NOR) was assessed by correcting measured NO- and N2 O-kinetics for ab…

Modulation of nitrosative stress by S-nitrosoglutathione reductase is critical for thermotolerance and plant growth in Arabidopsis.

Nitric oxide (NO) is a key signaling molecule in plants. This analysis of Arabidopsis thaliana HOT5 (sensitive to hot temperatures), which is required for thermotolerance, uncovers a role of NO in thermotolerance and plant development. HOT5 encodes S-nitrosoglutathione reductase (GSNOR), which metabolizes the NO adduct S-nitrosoglutathione. Two hot5 missense alleles and two T-DNA insertion, protein null alleles were characterized. The missense alleles cannot acclimate to heat as dark-grown seedlings but grow normally and can heat-acclimate in the light. The null alleles cannot heat-acclimate a

These ions specifically inhibited molybdenum cofactor-dependent nitrate reductase activity in Enterobacteriaceae metabolism, thereby creating a favorable ecological niche for probiotics. Consequently, the retained BS promoted beneficial microbes and metabolites, reversing colitis-associated metabolic disorders and synergistically …

Sodium Hydrosulfide (NaHS) Triggers Jasmonate and Reactive Oxygen Species to Boost Rice (Oryza sativa L.) Growth, Flowering, and Grain Yield.

Hydrogen sulfide (H2S) functions as a pivotal gaseous signaling molecule in plants, yet its role in promoting crop yield remains elusive. Here, we demonstrate that sodium hydrosulfide (NaHS) application, a donor of hydrogen sulfide (H2S), significantly accelerates growth, promotes flowering, and enhances grain yield in rice (Oryza sativa L.). Optimal NaHS treatment increased plant height, root length, and biomass accumulation, concomitant with elevated sucrose, starch, chlorophyll contents, and nitrate reductase activity. Integrated transcriptomic and proteomic analyses revealed that NaHS repr…

Glucose-6-Phosphate Dehydrogenase Modulates Shiraia Hypocrellin A Biosynthesis Through ROS/NO Signaling in Response to Bamboo Polysaccharide Elicitation.

Hypocrellin A (HA), a photodynamic perylenequinone pigment from Shiraia fruiting bodies, functions as an efficient photosensitizer for clini … y NADPH oxidase induction, while NO generation was attributed to elevated nitric oxide synthase and nitrate reductase activities. Critically, the G6PDH inhibitor glucosamine (1.0 mM) suppressed both H2O2 and NO production. These ROS/NO signals upregulated key HA biosynthetic (PKS, Omef) and transport (MFS) genes. Our findings establish G6PDH as a central regulator …

NO-dependent H2O2 signaling regulates melatonin-induced resistance to Fusarium oxysporum f. sp. niveum race 1 in watermelon.

Fusarium wilt, caused by Fusarium oxysporum f. sp. niveum (FON), is a widespread and destructive fungal disease in watermelon. While melatonin helps plants defend against various pathogens, its role and mechanism in combating FON are not well understood. Here, we demonstrated that overexpressing the melatonin biosynthesis gene ClCOMT1 enhances watermelon (Citrullus lanatus) resistance to FON race 1 (FON1), accompanied by increased production of nitric oxide (NO) mediated by nitrate reductase 1 (ClNR1) and H2O2 mediated by respiratory burst oxidase homologue D (ClRBOHD). Conversely, knockout of…

CRISPRi screen identifies FprB as a synergistic target for gallium therapy in Pseudomonas aeruginosa.

With the rise of antibiotic-resistant bacteria, non-antibiotic therapies like gallium gain increasing attention. Intravenous gallium nitrate is under Phase II clinical trials to treat chronic Pseudomonas aeruginosa infections in cystic fibrosis patients. However, its clinical efficacy is constrained by the achievable peak concentration in human tissue. To address this limitation, we apply a genome-wide CRISPR interference approach (CRISPRi-seq) to identify potential synergistic targets with gallium. We classify the essential genes by response time and growth reduction, pinpointing the most vul

TiO2 nanomaterial promotes plant growth and disease resistance.

TiO2 nanomaterials can promote plant growth and enhance disease resistance. However, the underlying mechanism remains unclear. This study applied TiO2 to promote the growth of wheat, soybean, tobacco, cucumber, and corn. Genetic analysis using macro-element transporter rice mutants in rice revealed that growth promotion induced by TiO2 was dependent on potassium transporter (AKT1), nitrate transporter 1.1B (NRT1.1B), ammonium transporter 1 (AMT1), and phosphate transporter 8 (PT8). TiO2 also enhanced chlorophyll accumulation, and growth promotion was inhibited in the chlorophyll biosynthesis r

A self-amplifying NO-H2S loop mediates melatonin-induced CBF-responsive pathway and cold tolerance in watermelon.

Melatonin is a pivotal bioactive molecule that enhances plant cold stress tolerance, but the precise mechanisms remain enigmatic. Here, we have discovered that overexpressing melatonin biosynthetic gene ClCOMT1 or applying exogenous melatonin activates the C-repeat binding factor (CBF)-responsive pathway and enhances watermelon cold tolerance. This enhancement is accompanied by elevated levels of nitric oxide (NO) and hydrogen sulfide (H2S), along with upregulation of nitrate reductase 1 (ClNR1) and L-cysteine desulfhydrase (ClLCD) genes involved in NO and H2S generation respectively. Converse…

The compound risk of decabromodiphenyl ethane and cadmium to soil nutrient cycling in agroecosystems: Linking the enzyme, microbial community, gene networks to plant stress.

Soil serves as the habitat for numerous organisms and is increasingly threatened from co-pollution of novel brominated flame retardant (NBFRs) and heavy metals (HMs). Focusing on Deca-bromodiphenyl ethane (DBDPE) and cadmium (Cd) as the targets, we constructed a soil-lettuce-earthworm microcosm to explore co-pollution effects in rhizosphere soils. Results showed that DBDPE increased bioavailable Cd fraction to amplified its ecological risks. Co-pollution significantly inhibited the expression of soil functional genes, especially C/N cycling genes, including pgu and abfA in C-cycling and narG,
