Mercury bioaccumulation and biomagnification in mesopelagic biota.

This study investigated the distribution and biomagnification pathways of total mercury (THg) concentrations in three mesopelagic species in the Tasman Sea: two fish (Diaphus hudsoni, Metelectrona ventralis) and one predatory squid (Lycoteuthis lorigera). THg concentrations in muscle tissue ranged from 0.06 to 0.26 μg g-1 dry weight (DW) and were consistently higher in liver tissue (0.15-0.96 μg g-1 DW) across all species. Species-specific variations in THg bioaccumulation and the liver-to-muscle THg ratio suggested differences in mercury metabolism. Stable isotope ratios of carbon (δ13C) vari

NMR resonance assignments of an engineered neomycin-sensing riboswitch RNA bound to ribostamycin and tobramycin.

The neomycin-sensing riboswitch is an engineered riboswitch developed to regulate gene expression in vivo in the lower eukaryote Saccharomyces cerevisiae upon binding to neomycin B. With a size of only 27nt it is the smallest functional riboswitch element identified so far. It binds not only neomycin B but also related aminoglycosides of the 2'-deoxystreptamine class with high affinity. The regulatory activity, however, strongly depends on the identity of the aminoglycoside. As a prerequisite for the structure determination of riboswitch-ligand complexes we report here the (1)H, (15)N, (13)C a

Sediment organic matter properties facilitate understanding nitrogen transformation potentials in East African lakes.

East African lakes include the most productive and alkaline lake group in the world. Yet, they generally receive fewer nutrient inputs than the densely populated subtropical and temperate lakes in the northern hemisphere. In these lakes with insufficient supplies of inorganic nitrogen, the mineralization of benthic organic matter can play an important role in driving the nutrient cycle and nitrogen loss. Using a suite of stable 15N isotope dilution and tracer techniques, we examined five main processes of the sediment nitrogen cycle in 16 lakes and reservoirs of Tanzania and Kenya, East Africa

Impact of Glaciers on Trophic Dynamics and Polyunsaturated Fat Accumulation in Southern Greenland Fjord Ecosystems.

The primary production of fjords across the Arctic and Subarctic is undergoing significant transformations due to the climatically driven retreat of glaciers and ice sheets. However, the implications of these changes for upper trophic levels remain largely unknown. In this study, we employ both bulk and compound-specific stable isotope analyses to investigate how shifts at the base of fjord food webs impact the carbon and energy sources of consumers. Focusing on two rapidly changing fjords in Southern Greenland, we used the migratory Arctic char as an indicator species, sampling populations al

1H, 13C and 15N chemical shift assignments of Rubella virus macro domain in the free and in the ADPr bound state.

Prokaryotes, eukaryotes, and certain viruses with positive single-stranded RNA genomes are among the forms of life that have been found to possess macro domains (MDs). There are claims that viral MDs inhibit the immune response mediated by PARPs, such as PARP12 and PARP14, and are involved in the formation of the viral replication transcription complex (RTC). Rubella virus (RuV) is included in this group of viruses. Its MD acts as an "eraser" of the posttranslation modification (PTM) ADP-ribosylation by binding to and hydrolyzing ADP-ribose (ADPr) from ADP-ribosylated substrates including prot

Mandibular bone mass density in a medieval population and its relationship with stable isotopes δ13C and δ15N.

The aim of this study was to compare the level of bone mass in digital orthopantomograms in two populations (medieval and current) using two radiomorphometric indexes, and to correlate the mandibular bone mass value, in the medieval mandible population, with stable isotope data δ13C and δ15N. An observational, cross-sectional, and analytical study on mandibles from two diachronic groups, 15 mandibles from the medieval settlement of La Torrecilla (Granada, Spain) and 15 mandibles from current patients at the Faculty of Dentistry of the University of Granada (Spain), matched by age and sex was c

Effects of chemical preservation on bulk and amino acid isotope ratios of zooplankton, fish, and squid tissues.

RATIONALE: It is imperative to understand how chemical preservation alters tissue isotopic compositions before using historical samples in ecological studies. Specifically, although compound-specific isotope analysis of amino acids (CSIA-AA) is becoming a widely used tool, there is little information on how preservation techniques affect amino acid δ15 N values. METHODS: We evaluated the effects of chemical preservatives on bulk tissue δ13 C and δ15 N and amino acid δ15 N values, measured by gas chromatography/isotope ratio mass spectrometry (GC/IRMS), of (a) tuna (Thunnus albacares) and squi

A protocol for pressurized liquid extraction and processing methods to isolate modern and ancient bone cholesterol for compound-specific stable isotope analysis.

RATIONALE: Bone lipid compound-specific isotope analysis (CSIA) and bone collagen and apatite stable isotope ratio analysis are important sources of ecological and paleodietary information. Pressurized liquid extraction (PLE) is quicker and utilizes less solvent than traditional methods of lipid extraction such as soxhlet and ultrasonication. This study facilitates dietary analysis by optimizing and testing a standardized methodology for PLE of bone cholesterol. METHODS: Modern and archaeological bones were extracted by PLE using varied temperatures, solvent solutions, and sample weights. The

Assessment of trace elements, POPs, (210)Po and stable isotopes ((15)N and (13)C) in a rare filter-feeding shark: The megamouth.

With less than 60 records being reported worldwide, the megamouth (Megachasma pelagios) is today one of the least known shark species inhabiting our oceans. Therefore, information concerning the biology and ecology of this enigmatic organism is very scarce and limited to feeding behaviour and preferred habitat. The present work reports new data on the concentrations of trace elements, organic mercury, POPs and (210)Po in hepatic and muscular tissues of a specimen found stranded in the southeastern coast of Brazil. Additionally, we provide new evidence based on stable isotope analysis (δ(15)N a

Mercury bioaccumulation and trophic transfer in sympatric snapper species from the Gulf of Mexico.

Consumption of marine fish is a major route of toxic methyl mercury (MeHg) exposure to ocean apex predators and human populations. Here we explore the influence of trophic structure on total mercury (Hg) accumulation in red snapper (RS, Lutjanus campechanus) and gray snapper (GS, Lutjanus griseus) from the coastal Louisiana region of the Gulf of Mexico, west of the Mississippi River. The objectives of this investigation were to: (1) determine the effectiveness of the use of offshore recreational fishing charter boats and marinas as sources of fish samples and (2) compare species differences in

Deposition and cycling of sulfur controls mercury accumulation in Isle Royale fish.

Mercury contamination of fish is a global problem. Consumption of contaminated fish is the primary route of methylmercury exposure in humans and is detrimental to health. Newly mandated reductions in anthropogenic mercury emissions aim to reduce atmospheric mercury deposition and thus mercury concentrations in fish. However, factors other than mercury deposition are important for mercury bioaccumulation in fish. In the lakes of Isle Royale, U.S.A., reduced rates of sulfate deposition since the Clean Air Act of 1970 have caused mercury concentrations in fish to decline to levels that are safe f

Synthesis of Bis(1,2,3-Triazole) Functionalized Quinoline-2,4-Diones.

Derivatives of 3-(1H-1,2,3-triazol-1-yl)quinoline-2,4(1H,3H)-dione unsubstituted on quinolone nitrogen atom, which are available by the previously described four step synthesis starting from aniline, were exploited as intermediates in obtaining the title compounds. The procedure involves the introduction of propargyl group onto the quinolone nitrogen atom of mentioned intermediates by the reaction of them with propargyl bromide in N,N-dimethylformamide (DMF) in presence of a potassium carbonate and the subsequent formation of a second triazole ring by copper catalyzed cyclisation reaction with

Well supplied in life, set aside in death: A multi-isotope study of Justinian plague victims from Saint-Doulchard (France, 7th-8th centuries AD).

OBJECTIVES: Justinian plague and its subsequent outbreaks were major events influencing Early Medieval Europe. One of the affected communities was the population of Saint-Doulchard in France, where plague victim burials were concentrated in a cemetery enclosure ditch. This study aimed to obtain more information about their life-histories using the tools of isotope analysis. MATERIALS AND METHODS: Dietary analysis using carbon and nitrogen isotopes was conducted on 97 individuals buried at Le Pressoir in Saint-Doulchard, with 36 of those originating from the enclosure ditch. This sample set in…

Deciphering natural and anthropogenic nitrate and recharge sources in arid region groundwater.

Recently, the subsoils of ephemeral stream (arroyos) floodplains in the northern Chihuahuan Desert were discovered to contain large naturally occurring NO3- reservoirs (floodplain: ~38,000 kg NO3-N/ha; background: ~60 kg NO3-N/ha). These reservoirs may be mobilized through land use change or natural stream channel migration which makes differentiating between anthropogenic and natural groundwater NO3- sources challenging. In this study, the fate and sources of NO3- were investigated in an area with multiple NO3- sources such as accidental sewer line releases and sewage lagoons as well as natur

Coupling a Knudsen reactor with the short lived radioactive tracer (13)N for atmospheric chemistry studies.

A Knudsen cell flow reactor was coupled to an online gas phase source of the short-lived radioactive tracer (13)N to study the adsorption of nitrogen oxides on ice at temperatures relevant for the upper troposphere. This novel approach has several benefits over the conventional coupling of a Knudsen cell with a mass spectrometer. Experiments at lower partial pressures close to atmospheric conditions are possible. The uptake to the substrate is a direct observable of the experiment. Operation of the experiment in continuous or pulse mode allows to retrieve steady state uptake kinetics and more

M9 medium composition alters E. coli metabolism during recombinant expression.

M9 minimal media and its enhanced variants (M9 + and M9++) are widely used for recombinant protein expression in Escherichia coli, particularly for isotopic labeling required in structural biology techniques such as NMR spectroscopy. This study investigates how different compositions of M9-based media (M9, M9+, and M9++) influence bacterial growth, metabolic stress, and central carbon metabolism during recombinant expression of the protein. Using 1D ¹H NMR spectroscopy and multivariate statistical analysis, we observed distinct media-dependent metabolic shifts. Standard M9 exhibited limited ba

Nitrate assimilation, dissimilatory nitrate reduction to ammonium, and denitrification coexist in Pseudomonas putida Y-9 under aerobic conditions.

The specific nitrate reduction pathway in Pseudomonas putida Y-9 under aerobic conditions was studied. Strain Y-9 removed 82% of the nitrate accompanied by an accumulation of ammonium and a decrease of total nitrogen. Ammonium inhibited nitrate transformation (removal efficiency was 22.65%), illustrating that nitrate assimilation exists in strain Y-9. The detectable ammonium in the supernatant during the nitrate reduction process came from intracellular locations in strain Y-9. The nirBD that encodes nitrite reductase had an important role in strain growth and ammonium production. A 15N isotop

Triple oxygen isotope analysis of nitrate using the denitrifier method and thermal decomposition of N2O.

Interaction with ozone transfers its anomalous (non-mass-dependent) 17O enrichment to atmospheric nitrogen oxides and nitrate. The 17O anomaly (Delta17O) in nitrate can be used to identify atmospheric nitrate inputs into terrestrial and aquatic environments as well as to study the role of ozone in the atmosphere's reactive nitrogen cycle. We report here on an online method for analysis of the 17O anomaly, using a strain of denitrifiers to convert nitrate to N2O, which decomposes quantitatively to N2 and O2 in a gold furnace at 800 degrees C, followed by gas chromatographic separation and isoto

High-efficiency simultaneous ammonia and nitrate removal in iron-sulfur coupled system under carbon limitation: Multi-omics insights into metabolic regulation and microbial interactions.

Iron‑sulfur coupling is a promising process for efficient nitrate removal, but its potential for simultaneous ammonia removal remains poorly recognized. This study established a biofilter (ISBF) using sponge iron and elemental sulfur as mixed fillers. Over 228 days of operation, ISBF achieved excellent nitrate (98.1%) and ammonia (89.7%) removal within 2 h. In-situ batch tests and ¹⁵N isotope tracing revealed synergistic nitrogen removal via autotrophic denitrification (56.6%), Feammox (9.1%), and Anammox (30.9%). X-ray diffraction confirmed FeOOH formation within biofilm, providing highly ava
