Stem cell regulatory gene expression in human adult dental pulp and periodontal ligament cells undergoing odontogenic/osteogenic differentiation.

INTRODUCTION: During development and regeneration, odontogenesis and osteogenesis are initiated by a cascade of signals driven by several master regulatory genes. METHODS: In this study, we investigated the differential expression of 84 stem cell-related genes in dental pulp cells (DPCs) and periodontal ligament cells (PDLCs) undergoing odontogenic/osteogenic differentiation. RESULTS: Our results showed that, although there was considerable overlap, certain genes had more differential expression in PDLCs than in DPCs. CCND2, DLL1, and MME were the major upregulated genes in both PDLCs and DP…

Enamel matrix proteins; old molecules for new applications.

Emdogain (enamel matrix derivative, EMD) is well recognized in periodontology, where it is used as a local adjunct to periodontal surgery to stimulate regeneration of periodontal tissues lost to periodontal disease. The biological effect of EMD is through stimulation of local growth factor secretion and cytokine expression in the treated tissues, inducing a regenerative process that mimics odontogenesis. The major (>95%) component of EMD is Amelogenins (Amel). No other active components have so far been isolated from EMD, and several studies have shown that purified amelogenins can induce the …

Functional analysis of a mutation in PAX9 associated with familial tooth agenesis in humans.

Pax9 is a paired domain-containing transcription factor that plays an essential role in the patterning of murine dentition. In humans, mutat … t 219InsGPAX9 likely alter the selective activation and/or repression of PAX9 effector genes during odontogenesis. This loss-of-function of PAX9 most likely results in its haploinsufficiency during the patterning of dentition and the subsequent loss of posterior teeth.

Molecular signaling and pulpal nerve development.

The purpose of this review is to discuss molecular factors influencing nerve growth to teeth. The establishment of a sensory pulpal innervat … form terminals near odontogenic sites. In some species, local axons have an initiating function in odontogenesis, but it is not known if this is also the case with mammals. In diphyodont mammals, the primary dentition is replaced by a permanent dentition, which involves a profound remodeling of terminal pulpal axons. The molecular signals underlying this remod …

Glomerular epithelial protein-1 promotes reparative dentin formation in vivo.

Glomerular epithelial protein-1 (Glepp1/PTPRO), a member of the R3 subtype family of receptor-type protein tyrosine phosphatases that, regulates cellular processes, like proliferation, differentiation, and apoptosis, involved in odontogenesis, with its overexpression causing thicker dentin. However, its in vivo impact during reparative dentin formation remains unexplored. This study evaluated an alternative therapeutic approach of enhancing natural dentin repair by overexpressing Glepp1 in an animal model. A pulp access cavity was prepared on the upper right first molar of eight-week-old male …

The role and molecular mechanism of Shikonin in promoting odontogenic differentiation of human dental pulp stem cells.

BACKGROUND: In tissue engineering applications, human dental pulp stem cells (hDPSCs) are an optimal biological resource due to their ability to regenerate the pulp-dentin complex through odontogenic differentiation. Shikonin (SHI), a red-purple naphthoquinone compound extracted from the root of Lithospermum erythrorhizon, has shown promise in this context. However, comprehensive studies on SHI's capacity to promote odontogenic differentiation in hDPSCs are limited. This study aims to investigate the effects of SHI on the odontogenic differentiation of hDPSCs and explore the underlying mechani

Mechanical control in dental and jaw morphogenesis and remodeling.

Tooth and maxillomandibular bone development is a synergistic process precisely and coordinately regulated by genetic programs, molecular signaling networks, and external environmental factors. Recent rapid progress in in situ biomechanical measurement techniques, cellular mechanical imaging, and molecular tracing technologies has greatly promoted research in developmental biomechanics. Accumulating evidence indicates that mechanical stimuli can be converted into biochemical signals via cellular mechanotransduction pathways, thereby modulating key cellular processes, including proliferation, d

Origins of Alterations to Rankl Null Mutant Mouse Dental Root Development.

The purpose of the present study was to assess the early stages of development of mouse first molar roots in the osteopetrotic context of RANKL invalidation in order to demonstrate that the radicular phenotype observed resulted not only from defective osteoclasts, but also from loss of cell-to-cell communication among dental, periodontium and alveolar bone cells involving RANKL signaling. Two experimental models were used in this study: Rankl mutants with permanent RANKL invalidation, and C57BL/6J mice injected during the first postnatal week with a RANKL neutralizing antibody corresponding to

[The pattern and regulatory mechanism of mammalian diphyodont tooth replacement].

Although the dental lamina of permanent teeth in human being has been developed as early as the embryo stage, the replacement of the deciduous teeth by permanent teeth does not take place untill the age of 6 to 12 years old. The molecular mechanism of the initiation of permanent teeth is still unclear. The rodent species are usually used for the tooth development research in the past. However, this animal model is not suitable for the tooth replacement study because of the absence of tooth replacement in rodents. After 10 years of efforts, our team has established the animal model of miniature

Treatment options: apexogenesis and apexification.

This article will describe requirements for case selection and review the procedures for apexogenesis and apexification in immature permanent teeth. Nonclinical and clinical data will be presented to support the recommendations, and outcomes will be presented from clinical studies. The dental pulp is an ectomesenchymally derived connective tissue with certain unique properties such as being encased in hard tissues, which limits its collateral circulation. The pulp provides a matrix for binding of its cells and provides support allowing communication between the cells. In addition to immune cel

Prevention of third molar tooth development in neonate rat with a long pulse diode laser.

BACKGROUND AND OBJECTIVES: Third molars (wisdom teeth) in humans have limited functional value and cause a high frequency of morbidity. A window of opportunity exists to stop their development. The purpose of this study is to evaluate whether a long pulse diode laser can selectively prevent third molar development in a rat animal model. STUDY DESIGN/MATERIALS AND METHODS: The study calculated theoretical diode laser parameters and irradiated 2- to 4-day-old rat pups in the posterior aspects of the oral cavity near the time of tooth bud initiation. Rats were sacrificed at 35-50 days and examin

Enamel mineralization and the role of ameloblasts in calcium transport.

Amelogenesis is a dynamic and unique process of cell-matrix interactions in that matrix synthesis, degradation and resorption all proceed simultaneously, coupled with mineral depositions in a compartment between ameloblasts and dentin or dental papilla. Accumulation of data suggest the role of ameloblasts in tooth morphogenesis and matrix formation, but no fully acceptable explanation has been given concerning the role of ameloblasts in calcium transport. In this article, old and new points of issue raised regarding the role of ameloblasts in calcium acquisition are reviewed and possible mecha

Experimental odontomas in osteopetrotic op/op rats.

Osteopetrosis is an autosomal recessive disease in several mammalian species. Osteopetrotic op/op rats suffer from complete failure of tooth eruption related to reduced bone resorption. In our earlier studies, op/op rats grafted with bone marrow cells 3 days after birth were cured of the disease and their molar eruption was restored. However, the incisors failed to erupt and their proliferating ends were distorted, forming odontomas. The purpose of the present investigation was to study the odontogenic tissues in the odontomas, using the correlated techniques of radiography and microradiograph

The development and fate of the dental lamina of the mandibular first molar tooth in the rat.

The lamina of the first mandibular molar teeth of rats, age range 13 d intrauterine (i.u.) to 16 d postnatal (p.n.), was examined by light and transmission electron microscopy to establish histological baselines of its development and fate. All material was obtained from animals anaesthetised with ether, killed by cervical dislocation and prepared by routine methods for both types of examination. Contrary to earlier reports that the lamina remains intact throughout development, mesenchymal elements disrupt the lamina. These were seen first at 19 d i.u., as collagen-filled bays in the basal epi
