Expression and localization of FGF-1 in the developing rat olfactory system.

Primary olfactory axons project from the nasal olfactory neuroepithelium to glomeruli in the olfactory bulb where they synapse with mitral cells, the second-order olfactory neurons. We have shown that the heparin-binding growth factor FGF-1 is expressed by olfactory nerve ensheathing cells which surround fascicles of primary olfactory axons en route to the olfactory bulb. These cells are believed to modulate olfactory axon growth between the olfactory neuroepithelium and the olfactory bulb. During late embryogenesis, FGF-1 expression is turned on in the mitral cells, and the FGF-1 peptide beco

Beat- and Side-family cell-surface molecules are expressed combinatorially in the partner neurons of the olfactory circuit in Drosophila.

Over the past decades, many molecular players have been uncovered to control distinct steps of olfactory circuit assembly in Drosophila. Among these, multi-member gene families encoding cell-surface proteins are of interest as they can act as neuron-specific recognition tags in combinations and contribute to circuit assembly in complex brains. Recently, a multi-protein interactome has been described between Beat and Side families of IgSF proteins. Here, we use newly generated gene trap transgenic driver lines to probe the spatial expression pattern of beat/side genes in olfactory receptor neur

Coding odor modality in piriform cortex efficiently with low-dimensional subspaces: a shared covariance decoding approach.

A fundamental question in neuroscience is how sensory signals are decoded from noisy cortical activity. We address this question in the olfactory system, decoding the route by which odorants arrive into the nasal cavity: through the nostrils (orthonasal), or through the back of the throat (retronasal). We recently showed with modeling and novel experiments on anesthetized rats that orthonasal versus retronasal modality information is encoded in the olfactory bulb (OB, a pre-cortical region). However, key questions remain: is modality information transmitted from OB to anterior piriform cortex

Terminal nerve in the mouse-eared bat (Myotis myotis): ontogenetic aspects.

As in other mammals, ontogenesis of the terminal nerve (TN) in the mouse-eared bat (Myotis myotis) starts shortly after the formation of the olfactory placode, a derivative of the ectoderm. During development of the olfactory pit, proliferating neuroblasts thicken the placodal epithelium and one cell population migrates toward the rostroventral tip of the telencephalon. Here they accumulate in a primordial terminal ganglion, which successively divides into smaller units. Initial fibers of the TN can be distinguished from olfactory fibers in the mid-embryonic period. The main TN fiber bundle (m

Olfactory responses in a gustatory organ of the malaria vector mosquito Anopheles gambiae.

The proboscis is an important head appendage in insects that has primarily been thought to process gustatory information during food intake. Indeed, in Drosophila and other insects in which they have been identified, most gustatory receptors are expressed in proboscis neurons. Our previous characterization of the expression of AgOR7, a highly conserved odorant receptor (OR) of the Afrotropical malaria vector mosquito Anopheles gambiae in the labellum at the tip of the proboscis was suggestive of a potential olfactory function in this mosquito appendage. To test this hypothesis, we used electro

Convergence of olfactory inputs from both antennae in the brain of the honeybee.

Electrical activities of the olfactory neurones in the brain of the honeybee were investigated. Odorous stimuli were given to each antenna separately or to both simultaneously. The inputs from the antennae affected both the impulse frequency and the latency of the olfactory interneurones in the protocerebrum. The predominant response was to the stimulation of the ipsilateral antenna. Input from the contralateral antenna produced mainly excitatory effects, although a few inputs gave inhibitory effects. No particular relationships between the loci of the units in the brain and the types of respo

Role and Mechanism of Olfactory Stem Cells in the Treatment of Olfactory Disorders.

Olfactory dysfunction is one of the most prevalent diseases in otorhinolaryngology, particularly since the coronavirus 2019 (COVID-19) pandemic, with a potential impact on daily life. Several etiological factors can contribute to olfactory dysfunction owing to the complexity and specificity of the olfactory transmission pathway. However, current treatments for olfactory dysfunction are limited and their efficacy is unsatisfactory. Olfactory stem cells are multifunctional stem cells in the olfactory mucosa that comprise both horizontal and global basal stem cells (HBCs and GBCs, respectively).

Sense of smell: recognition and transduction of olfactory signals.

The primary processes in odour perception, i.e. recognition and transduction of olfactory stimuli, are mediated by the chemosensory olfactory neurons. Interaction of odorous compounds with suitable receptor proteins in the membrane of a subset of cells elicits chemo-electrical transduction pathways, including second messenger cascades and ion channels, that modulate the excitability of the sensory neurons, i.e. converting the chemical stimulus into electrical impulses. The encoded information is conveyed via the axons onto distinct glomeruli in the olfactory bulb. Olfactory sensory cells expre

Up regulated hippocampal insulin pathway and oxidative stress are related to opposite changes in olfaction and episodic memory in a compensatory metabolic syndrome model in rats.

Insulin resistance, a major component of metabolic syndrome (MetS), is involved in phosphorylated tau and beta amyloid buildup, linking MetS to neurodegenerative processes, e.g., Alzheimer's disease. One of the initial signs of neurodegeneration, in diseases such as Parkinson's and Alzheimer's, is olfactory dysfunction. Accordingly, in previous reports we found that high carbohydrate diet based on 30% sucrose in drinking water, increased visceral fat and insulin resistance, leading to an augment of oxidative stress and reduced brain energy metabolism, upregulating amyloidogenic genes (APP and

Proteomic Characterization of the Olfactory Molecular Imbalance in Dementia with Lewy Bodies.

Olfactory dysfunction is one of the prodromal symptoms in dementia with Lewy bodies (DLB). However, the molecular pathogenesis associated with decreased smell function remains largely undeciphered. We generated quantitative proteome maps to detect molecular alterations in olfactory bulbs (OB) derived from DLB subjects compared to neurologically intact controls. A total of 3214 olfactory proteins were quantified, and 99 proteins showed significant alterations in DLB cases. Protein interaction networks disrupted in DLB indicated an imbalance in translation and the synaptic vesicle cycle. These a

Senescence-like cells recruit γδ T cells to drive prolonged hyposmia after SARS-CoV-2 infection in mice.

Persistent hyposmia is a hallmark of post COVID-19 conditions, yet the mechanisms sustaining olfactory dysfunction after viral clearance remain poorly understood. Here, using mouse models of SARS-CoV-2 infection, we show that virus-induced senescence-like changes in uninfected olfactory mucosal fibroblasts persist long after viral clearance and drive prolonged olfactory dysfunction. These senescence-like cells secrete SASP factors, including IFNγ, CXCL9, and CXCL11, thereby recruiting γδ T cells to the olfactory mucosa. The accumulated γδ T cells produce excessive IL-17A, which acts on IL-17 r

Effects of prenatal alcohol exposure on the olfactory system development.

Fetal Alcohol Spectrum Disorders (FASD), resulting from maternal alcohol consumption during pregnancy, are a prominent non-genetic cause of physical disabilities and brain damage in children. Alongside common symptoms like distinct facial features and neurocognitive deficits, sensory anomalies, including olfactory dysfunction, are frequently noted in FASD-afflicted children. However, the precise mechanisms underpinning the olfactory abnormalities induced by prenatal alcohol exposure (PAE) remain elusive. Utilizing rodents as a model organism with varying timing, duration, dosage, and administr

Structural Basis for Histaminergic Regulation of Neural Circuits in the Mouse Olfactory Bulb.

Odor information is modulated by centrifugal inputs from other brain regions to the olfactory bulb (OB). Neurons containing monoamines, such as serotonin, acetylcholine, and noradrenaline, are well known as centrifugal inputs; however, the role of histamine, which is also present in the OB, is not well understood. In this study, we examined the histaminergic neurons projecting from the hypothalamus to the OB. We used an antibody against histidine decarboxylase (HDC), a synthesizing enzyme of histamine, to identify histaminergic neurons and assess their localization within the OB and the ultras

Human male superiority in olfactory sensitivity to the sperm attractant odorant bourgeonal.

Recent studies have shown that sperm chemotaxis critically involves the human olfactory receptor OR1D2, which is activated by the aromatic aldehyde bourgeonal. Given that both natural and sexual selection may act upon the expression of receptors, we hypothesized that human males are more sensitive than human females for bourgeonal. Using a 3-alternative forced-choice test procedure, olfactory detection thresholds were determined for a total of 500 subjects, 250 males, and 250 females between 18 and 40 years of age. We found that male subjects detected bourgeonal at significantly lower concentr

Translating basic research into clinical practice or what else do we have to learn about olfactory ensheathing cells?

Olfactory ensheathing cells (OECs) are Schwann cell-like glial cells of the olfactory system that have been shown to promote axonal regeneration and remyelination in a variety of different lesion paradigms. It is still a matter of debate in how far OECs differ from Schwann cells regarding their regenerative potential and molecular setup. The fact that OECs have been already used for transplantation in humans may imply that the need of the hour is the fine-tuning of clinical application details rather than to cross the bridge between laboratory animal and man. Considering the therapeutic transp

Olfactory ensheathing cell tumor with neurofibroma-like features: a case report and review of the literature.

A 32-year-old man had seizure attack since April 2008 and radiographic examination revealed a heterogeneous enhancing mass at the left subfrontal region. He underwent craniotomy for tumor removal on October 1, 2008. The tumor, which was grayish white with glistening appearance and rubbery consistency, was traced to the proximal part of left olfactory tract. Histopathological examination revealed a hypocellular tumor with dense hyalinization in most areas. The tumor cells had ovoid to elongate and often comma-shaped nucleus. Myxoid change of the stroma was apparent in places. Most of the tumor

Retinoic acid enhances the rate of olfactory recovery after olfactory nerve transection.

In the olfactory system, retinoic acid (RA) plays an important role in development and may affect growth in the adult animal. To explore the potential effects of RA on recovery after injuries, adult mice were trained in a buried food paradigm and were given a single oral supplement of RA after olfactory nerve transection. Results demonstrate that RA accelerates the recovery of olfactory functions after injury.

[Teratogenic effect of ammonium metavanadate in Swiss albino mice].

Far as it is known to us, there is only one report in the medical literature dealing with the teratogenous effect of vanadium on neural structures (1): while trying to experimentally induce anencephaly in the offsprings of swiss-albino mice through the administration of ammonium metavanadate via drinking water, during different stages of their lives; a neural tube defect consisting of arrhinencephaly was obtained; various degrees of hypoplasia or aplasia of both the olfactory bulbs and tracts were evidenced, as well as congenital lesions of variable intensity in other rhinencephalic structures

Transient expression of somatostatin immunoreactivity in the olfactory-forebrain region in the chick embryo.

The tissue distribution of somatostatin (SST) immunoreactivity was studied in the nasal and forebrain region in the chick embryo. On embryonic day (ED) 3, SST-immunoreactive (ir) cells were first detected in the cells migrating from the olfactory placode. Then, at ED3.5, SST-ir cells and -ir fibers appeared in the olfactory epithelium and olfactory nerve bundles. At ED6-8, one component of the SST-ir fibers was found to separate from the olfactory nerve and it entered the parenchyma of the medial forebrain surface. These SST-ir fibers extended dorsocaudally toward the preseptal area. During th

Functional subdivisions of the olfactory system correlate with lectin-binding properties in Xenopus.

Soybean agglutinin (SBA) is known to selectively label a portion of neurons in amphibian and mammalian primary olfactory systems. Hitherto, no other distinctive features have been found to correlate with the two neuronal populations. Investigating SBA-HRP binding in olfactory mucosa and CNS of Xenopus, we noted that labelled and unlabelled structures can readily be assigned to different olfactory subsystems. The SBA negative one is utilized to detect air-borne odors, whereas major SBA-positive structures serve a role in the perception of water dissolved molecules. Some labelled fibers by-pass
