Antenatal Glucocorticoid Exposure Results in Sex-Specific and Transgenerational Changes in Prefrontal Cortex Gene Transcription that Relate to Behavioural Outcomes.

Synthetic glucocorticoids (sGC) are administered to women at risk for pre-term delivery to reduce respiratory distress syndrome in the newborn. The prefrontal cortex (PFC) is important in regulating stress responses and related behaviours and expresses high levels of glucocorticoid receptors (GR). Further, antenatal exposure to sGC results in a hyperactive phenotype in first generation (F1) juvenile male and female offspring, as well as F2 and F3 juvenile females from the paternal lineage. We hypothesized that multiple courses of antenatal sGC modify gene expression in the PFC, that these effe

Transcriptomic effects of paternal cocaine-seeking on the reward circuitry of male offspring.

It has been previously established that paternal development of a strong incentive motivation for cocaine can predispose offspring to develop high cocaine-seeking behavior, as opposed to sole exposure to the drug that results in drug resistance in offspring. However, the adaptive changes of the reward circuitry have not been fully elucidated. To infer the key nuclei and possible hub genes that determine susceptibility to addiction in offspring, rats were randomly assigned to three groups, cocaine self-administration (CSA), yoked administration (Yoke), and saline self-administration (SSA), and

Overview of the Genetic Basis and Epigenetic Mechanisms that Contribute to FASD Pathobiology.

Prenatal alcohol (ethanol) exposure (PAE) is the underlying cause for a variety of birth defects and neurodevelopmental deficits referred to as "Fetal Alcohol Spectrum Disorders (FASD)". The more visible phenotypes caused by PAE include growth retardation, and characteristic craniofacial abnormalities associated with functional and structural damage to the central nervous system. Ethanol is a teratogenic agent itself; but it can also alter gene expression. These changes may contribute to the spectrum of effects and different phenotypes that are dependent on alcohol metabolism, as well as the t

Parental occupational exposure to low-frequency magnetic fields and risk of leukaemia in the offspring: findings from the Childhood Leukaemia International Consortium (CLIC).

OBJECTIVES: Previously published studies on parental occupational exposure to extremely low-frequency magnetic fields (ELF-MF) and risk of acute lymphoblastic leukaemia (ALL) and acute myeloid leukaemia (AML) in their offspring were inconsistent. We therefore evaluated this question within the Childhood Leukemia International Consortium. METHODS: We pooled 11 case-control studies including 9723 childhood leukaemia cases and 17 099 controls. Parental occupational ELF-MF exposure was estimated by linking jobs to an ELF-MF job-exposure matrix (JEM). Logistic regression models were used to estima

Paternal Preconception Hepatitis B Virus Infection and Risk of Congenital Heart Disease in Offspring.

IMPORTANCE: Previous evidence suggests that maternal hepatitis B virus (HBV) infection during prepregnancy or pregnancy is associated with congenital heart diseases (CHDs) in offspring. However, the association of paternal HBV infection with CHDs is not well examined. OBJECTIVE: To explore the association of paternal preconception HBV infection with CHDs in offspring. DESIGN, SETTING, AND PARTICIPANTS: This retrospective cohort study used propensity score matching of data from the Chinese National Free Preconception Checkup Project (NFPCP) from January 1, 2010, to December 31, 2018. Male par

Paternal scrotal heat stress epigenetically impairs spatial memory in F1 male offspring in mice.

Increasing evidence indicates that the maternal preconception environment and lifestyle affect offspring health; however, the effects of paternal preconception environment and lifestyle have rarely been examined. Therefore, we aimed to examine whether paternal infertility-related factors affect offspring produced via assisted reproductive technology in mice. Scrotal heat impairment, which can result from factors such as varicoceles, sauna use, physical inactivity, or occupations such as welding, is a common risk factor associated with reduced male fertility. To model these effects, we exposed

Paternal morphine alters offspring circulating beta-endorphin and corticosterone responses to oxycodone and cocaine.

BACKGROUND: The opioid epidemic is leading to increased opioid use in adolescent populations. A growing body of evidence suggests that taking opioids during adolescence can disrupt normal development and impact future offspring. This study investigates the impact of paternal morphine exposure during adolescence on the hypothalamic-pituitary-adrenal (HPA) axis and release of endorphins in the offspring. METHODS: Male rats were administered morphine once a day from postnatal day (PND)30-39 using an increasing dosing regimen (5-25 mg/kg/day increasing every other day). They were mated during adu

A dangerous boomerang: Injunctive norms, hostile sexist attitudes, and male-to-female sexual aggression.

This study examined the interactive effects of injunctive norm exposure and hostile and benevolent sexist attitudes on men's sexually aggressive responses during a behavioral analogue paradigm in which they interacted online with a bogus female partner. Heterosexual adult men (n = 201), recruited from an online sample, read fictional information regarding other men's approval of misogynistic, paternalistic, or egalitarian treatment of women, or non-gender-relevant control information. Through a media preference survey, men then learned that their female partner disliked sexual content in films

Paternal preconception modifiable risk factors for adverse pregnancy and offspring outcomes: a review of contemporary evidence from observational studies.

BACKGROUND: The preconception period represents transgenerational opportunities to optimize modifiable risk factors associated with both short and long-term adverse health outcomes for women, men, and children. As such, preconception care is recommended to couples during this time to enable them to optimise their health in preparation for pregnancy. Historically, preconception research predominately focuses on maternal modifiable risks and health behaviours associated with pregnancy and offspring outcomes; limited attention has been given to inform paternal preconception health risks and outco

Environmental Impact on Male (In)Fertility via Epigenetic Route.

In the last 40 years, male reproductive health-which is very sensitive to both environmental exposure and metabolic status-has deteriorated and the poor sperm quality observed has been suggested to affect offspring development and its health in adult life. In this scenario, evidence now suggests that epigenetics shapes endocrine functions, linking genetics and environment. During fertilization, spermatozoa share with the oocyte their epigenome, along with their haploid genome, in order to orchestrate embryo development. The epigenetic signature of spermatozoa is the result of a dynamic modulat

Paternal nicotine taking elicits heritable sex-specific phenotypes that are mediated by hippocampal Satb2.

Nicotine intake, whether through tobacco smoking or e-cigarettes, remains a global health concern. An emerging preclinical literature indicates that parental nicotine exposure produces behavioral, physiological, and molecular changes in subsequent generations. However, the heritable effects of voluntary parental nicotine taking are unknown. Here, we show increased acquisition of nicotine taking in male and female offspring of sires that self-administered nicotine. In contrast, self-administration of sucrose and cocaine were unaltered in male and female offspring suggesting that the intergenera

Focus on and venting of emotion mediate the pathway from maternal child abuse to adulthood depression severity.

BACKGROUND: Exposure to parent-perpetrated child abuse is a risk factor for adulthood major depressive disorder (MDD). Coping strategies may be a plausible mechanism underlying this relationship. The current study examined whether positive reinterpretation (PR), focus on and venting of emotion (FVE), and behavioral disengagement (BD) mediated the relationship between maternal or paternal abuse and adulthood MDD symptoms. METHOD: Data from 3294 community-dwelling adults were collected across three waves, with each interval nine years apart. Structural equation mediation modeling examined wheth

Influence of increased paternal BMI on pregnancy and child health outcomes independent of maternal effects: A systematic review and meta-analysis.

OBJECTIVES: The influence of maternal body mass index (BMI) on pregnancy and child health outcomes is well characterised, however less is known about paternal BMI. This systematic review investigated the independent effects of increased paternal BMI on conception and pregnancy as well as neonatal and childhood outcomes. METHODS: Our systematic search (Sept. 2018) of PubMed, Embase, Cinahl, Web of Science, ProQuest, and OpenThesis resulted in 11,045 hits from which 17 studies met the inclusion criteria (Participants: men Exposure: BMI or waist circumferences Outcomes: associations with time to

Literature on the aetiology of hypospadias in the last 5 years: molecular mechanism and environmental factors.

Year-by-year, there has been an increasing number of literature on hypospadias, and most of them are mainly focused on two aspects, namely surgical techniques and aetiology, including the molecular mechanism and associated environmental factors. Surgical techniques and nursing levels are being continuously improved. However, in stark contrast, the study of aetiology still lags behind. Up to now, there is still no consensus on the aetiology of hypospadias, including the molecular mechanism and associated environmental factors. To obtain an overall and latest result on the aetiology, we reviewed

Microsatellite mutations in the offspring of irradiated parents 19 years after the Cesium-137 accident.

In September of 1987, a radiotherapy unit containing 50.9 TBq of Cs(137)Cl was removed from an abandoned radiotherapy clinic. This unit was subsequently disassembled leading to the most serious radiological accident yet to occur in the Western hemisphere. This event provides an opportunity to assess the genetic effects of ionizing radiation. We surveyed genetic variation of 12 microsatellite loci in 10 families of exposed individuals and their offspring and also in non-exposed families from the same area of Goias state. We found an increase in the number of new alleles in the offspring of the

PALS (pregnancy and lifestyle study): association between occupational and environmental exposure to chemicals and reproductive outcome.

A prospective study examined the reproductive outcome, live birth, miscarriage or 9 months infertility, in 585 participating couples. Examination of the data relating to environmental and occupational exposure to chemicals and radiation revealed the following associations. Infertility was significantly associated with male factors of age, occupational exposure to dusts and occupation of labourer in men aged 35 or older. Female factors associated with infertility were age and home renovating if aged 35 or older. First trimester spontaneous miscarriage was associated with male factors of age, X-
