Paternal uniparental disomy of chromosome 6 and transient neonatal diabetes mellitus.

Transient neonatal diabetes mellitus occurs in growth-retarded infants, has an incidence of 1 in 400000 live births and has been associated with both paternal uniparental disomy of chromosome 6 and paternal duplications of 6q. We analysed samples from our cohort of patients with transient neonatal diabetes mellitus for uniparental disomy of chromosome 6 using polymorphic microsatellite repeat analysis. We report here the fifth case of paternal uniparental disomy of chromosome 6 associated with classic transient neonatal diabetes mellitus and estimate that uniparental disomy of chromosome 6 acc

Paternal transmission of X-linked placental dysplasia in mouse interspecific hybrids.

It has previously been shown that abnormal placental development, i.e., hyper- and hypoplasia, occurs in crosses and backcrosses between different mouse (Mus) species. These defects are caused mainly by abnormal growth of the spongiotrophoblast. The precise genetic basis for these placental malformations has not been determined. However, a locus that contributes to the abnormal development (Ihpd: interspecific hybrid placental dysplasia) has been mapped to the X chromosome. The X-chromosomal location of Ihpd and its site of action, that is the spongiotrophoblast, mean that normally only the ma

Autism or atypical autism in maternally but not paternally derived proximal 15q duplication.

Duplications of proximal 15q have been found in individuals with autistic disorder (AD) and varying degrees of mental retardation. Often these abnormalities take the form of a supernumerary inverted duplicated chromosome 15, more properly described as an isodicentric chromosome 15, or idic(15). However, intrachromosomal duplications also have been reported. In a few cases, unaffected mothers, as well as their affected children, carry the same duplications. During the course of the genotyping of trios of affected probands with AD and their parents, at the positional candidate locus D15S122, an

Genetic counselling and prenatal diagnosis of osteogenesis imperfecta caused by paternal mosaicism.

In a family with recurrent osteogenesis imperfecta (OI) caused by paternal mosaicism, prenatal diagnosis was made using restriction enzyme analysis for a mutation in COL1A2. Parental mosaicism is important to consider in genetic counselling for OI. Prenatal diagnosis of OI is available currently by means of collagen or gene analyses in the first trimester or by ultrasonography in the second trimester.

Mechanisms and targets involved in maternal and paternal age effects on numerical aneuploidy.

Trisomy in the human appears to be predominantly associated with maternal age. The maternal-age effect, however, shows considerable variability across affected chromosomes. Chromosome-specific variation has been reported in the shapes of the maternal-age-effect curves, including very small effects for the large chromosomes (groups A and B), linear increases (chromosome 16), and exponential increases (chromosome 21). There is also variation among chromosomes in whether the segregation errors occur predominantly at maternal meiosis I, meiosis II, and/or postfertilization mitotic divisions. There

Paternal exposures and cardiovascular malformations. The Baltimore-Washington Infant Study Group.

Possible associations between paternal exposures and cardiovascular malformations were evaluated in the Baltimore-Washington Infant Study, a population based case-control investigation of congenital heart disease and environmental factors. Home interviews of case and control parents elicited information on parental home and occupational exposures. Analysis focused on twelve cardiac diagnostic groups and paternal exposures incurred during the six months preceding the pregnancy. Associations were identified between jewelry making and atrial septal defect (Odds ratio: 12.6; 95% confidence interva

Paternal exposure to radiation and offspring cancer in mice: reanalysis and new evidence.

Parental exposure to radiation could induce various kinds of tumors in the next generation. In ICR mice, a large and significant increase of adult types tumor was observed in the F1 offspring after X-ray exposure at spermatozoa and spermatid stages, and less clear increase was observed after spermatogonial exposure. Mature oocytes were resistant upto 1 Gy, but very sensitive to tumor induction at higher doses. While there was no difference in the tumor incidence between acute and fractionated (0.36 Gy at 2 hr intervals) irradiation at post-gonial stages, a large reduction of tumor incidence wa

Familial occurrence of Duchenne dystrophy through paternal lines in four families.

In a survey of 454 families with patients affected with Duchenne muscular dystrophy (DMD) we have found 4 genealogies with 2 or more affected patients who were related through paternal lines. In 1 of these families, 2 affected cousins showed different DNA deletions suggesting 2 independent mutations; in the other 2, in which only the propositus could be studied DNA deletions were also detected in the dystrophin gene. In the last one, with 3 affected patients, no DNA deletions were detected but immunohistochemical study of muscle biopsies showed a negative dystrophin pattern typical of DMD. Alt

Fetus as an allograft: noncytotoxic maternal antibodies to HLA-linked paternal antigens.

A cellular enzyme-linked immunospecific assay (CELISA) was used to monitor maternal humoral responses in human pregnancy. Non-cytotoxic IgG antibodies to paternal lymphocytes were detected in sera from 6 of 20 normal first trimester primigravidae and 6 of 13 multiparae. No antibody activity against lymphocytes from their partners was detected in sera from any of the 15 nulliparous women. The differences in antibody response between primigravidae and nulliparae (P = 0.024) and between multiparae and nulliparae (P = 0.005) were statistically significant. Lymphocytotoic antibodies to T- and B-lym

Complete hydatidiform moles combine maternal mitochondria with a paternal nuclear genome.

The parental origin of mitochondria in hydatidiform moles has been investigated by analysis of genetic variants of mtDNA restriction enzyme patterns. In six complete moles the mtDNA was found to be maternal in origin, with no contribution from the sperm mitochondria, while the nuclear genome was shown to be exclusively paternal in five cases. The occurrence of mtDNA variation in the healthy population was investigated using white blood cells and placentae, and the most common variation occurred at the Ava II restriction sites. The variants exhibited by molar mtDNA were the same as those found

Paternal age and Down's syndrome: data from prenatal diagnoses (DFG).

From prenatal diagnosis data obtained on mothers aged 35 years and above in the Federal Republic of Germany (DFG data), older fathers are demonstrated to have an increased risk of having trisomy 21 offspring. For paternal ages of 41 years upward, the age effect is quite strong. The risk for a fetus to have any de novo chromosomal aberration increases more with advancing paternal age for older mothers than for younger ones. Thus the ages of both parents have to be taken into account as an indication for prenatal diagnosis. Risk figures for trisomy 21 and for any de novo chromosomal aberration a

[Depressive reactions in children following the loss of a paternal figure, by death or divorce].

A review of different viewpoints on the theme "Death and the child" is made. When the child loses a loved object, he has to accept it in the outside world and manage the changes in his inner world. If the lost object is very needed, his ability to comprehend death is obstructed. When and how it is possible to help the child to understand death, is also examined. Depending of the age, development stage, previous relationships with the dead person, each child reacts differently. Effects of divorce on children are studied. A method that provides better avenues for ellaboration to children is pres

Seizure disorders in offspring of parents with a history of seizures - a maternal-paternal difference?

If the patterns of seizure disorders in parents and offspring could be assessed accurately, some insight might be gained into the relative role of genetic and environmental factors in the development of convulsive disorders. In this study 908 children born in a Rochester, Minnesota hospital from 1922 through 1972 whose mother or father had a verified and classified diagnosis of seizure have been followed from birth for evidence of any convulsive episode or seizure disorder. The observed numbers of various types of convulsions in the offspring are compared to the expected the number based on lo

Older paternal age and fresh gene mutation: data on additional disorders.

Older paternal age has previously been documented as a factor in sporadic fresh mutational cases of several autosomal dominant disorders. In this collaborative study, an older mean paternal age has been documented in sporadic cases of at least five additional dominantly inheritable disorders; the basal cell nevus syndrome, the Waardenburg syndrome, the Crouzon syndrome, the oculo-dental-digital sysdrome, and the Treacher-Collins syndrome. It was also found to be a factor in acrodysostosis and progeria, suggesting a fresh mutant gene etiology for these two conditions in which virtually all case

Dynamic interplay of maternal and paternal contributions to offspring phenotype in Eurasian perch.

BACKGROUND: Parental contributions to offspring phenotype extend beyond genetic inheritance, encompassing non-genetic factors that influence early development. However, the interplay between maternal and paternal effects remains poorly understood. This study investigates these contributions in Eurasian perch (Perca fluviatilis) - a valuable model to study parental effect in finfishes - by analyzing early life traits and transcriptomic profiles of larvae resulting from crosses between wild and domesticated parents. RESULTS: Maternal effects dominated key developmental traits, including hatchin

Risk of neurodevelopmental disorders associated with paternal use of valproate during spermatogenesis.

BACKGROUND: Regulatory authorities have raised concerns about potential teratogenic and neurodevelopmental disorders (NDDs) risks associated with paternal valproate use during spermatogenesis, leading to restrictions. We aimed to investigate the association between paternal valproate exposure during spermatogenesis and the risk of NDDs in offspring in Sweden and Norway. METHODS: We conducted a population-based cohort study of two nationwide cohorts from Sweden and Norway, including all singleton live births (≥22 gestational weeks) from 2007 to 2020. Paternal monotherapy exposure to valproate,

Health consequences of paternal incarceration using a future-treated control group.

BACKGROUND: Paternal incarceration is a now a common experience in the life course for young people in the United States that likely shapes health. This study examines the effect of paternal incarceration on adolescents’ health care access/utilization and health behaviors during adolescence. This study employs an innovative approach, which we refer to as the future treated control method, to adjust for unobserved heterogeneity by exploiting plausibly exogenous variation in the timing of paternal incarceration to develop a future treated control group. In this approach youth who have experience

Paternal exposure to hydrocarbon solvents in the workplace and cancer risk in children and adolescents.

BACKGROUND: According to the International Agency for Research on Cancer, certain hydrocarbons are known or suspected carcinogens. OBJECTIVE: Our study aimed to estimate cancer risks in children and adolescents associated with paternal exposure to groups of aliphatic/alicyclic, aromatic, and chlorinated hydrocarbons, and the individual chemicals, toluene, methylene chloride, trichloroethylene, and 1,1,1-trichloroethane, from 3 months preconception to birth. METHODS: For this population-based case-control study of Danish children and adolescents (born 1968-2013) < 20 years of age when diagnos
