An Engineered Adeno-Associated Virus Capsid Mediates Efficient Transduction of Pericytes and Smooth Muscle Cells of the Brain Vasculature.

Neurodegeneration and cerebrovascular disease share an underlying microvascular dysfunction that may be remedied by selective transgene delivery. To date, limited options exist in which cellular components of the brain vasculature can be effectively targeted by viral vector therapeutics. In this study, we characterize the first engineered adeno-associated virus (AAV) capsid mediating high transduction of cerebral vascular pericytes and smooth muscle cells (SMCs). We performed two rounds of in vivo selection with an AAV capsid scaffold displaying a heptamer peptide library to isolate capsids th

Tissue plasminogen activator disrupts the blood-brain barrier through increasing the inflammatory response mediated by pericytes after cerebral ischemia.

Pericytes, important elements of the blood-brain barrier (BBB), play critical roles in maintaining BBB integrity and modulating hemostasis, angiogenesis, inflammation and phagocytic function. We investigated whether pericytes are involved in the recombinant tissue plasminogen activator (rt-PA)-induced inflammatory response, which disrupts the BBB, and investigated the potential mechanisms. Middle cerebral artery occlusion (MCAO) and oxygen-glucose deprivation (OGD) were employed to mimic hypoxic-ischemic conditions. Rt-PA was intravenously injected into mice 1 h after 1 h MCAO, and Rt-PA was a

The Angiopoietin-1 Variant COMP-Ang1 Enhances BMP2-Induced Bone Regeneration with Recruiting Pericytes in Critical Sized Calvarial Defects.

Craniofacial bone defects are observed in a variety of clinical situations, and their reconstructions require coordinated coupling between angiogenesis and osteogenesis. In this study, we explored the effects of cartilage oligomeric matrix protein-angiopoietin 1 (COMP-Ang1), a synthetic and soluble variant of angiopoietin 1, on bone morphogenetic protein 2 (BMP2)-induced cranial bone regeneration, and recruitment and osteogenic differentiation of perivascular pericytes. A critical-size calvarial defect was created in the C57BL/6 mouse and COMP-Ang1 and/or BMP2 proteins were delivered into the

Transcriptional analyses identify pericyte-centered signaling programs altered by sex and brain region in Alzheimer's Disease.

Pericytes are critical components of the neurovascular unit (NVU), regulating endothelial cell stability, blood-brain barrier (BBB) integrity, and neuroimmune signaling. Their role in Alzheimer's Disease (AD), particularly regarding sex differences and brain region specificity, remains poorly defined. Using single-nucleus RNA sequencing analysis and cell-cell communication tools (LIANA, Tensor-cell2cell), we characterize pericyte transcriptional and signaling changes across the middle temporal gyrus (MTG) and dorsolateral prefrontal cortex (DLPFC) of AD and non-AD donors stratified by sex. We

Spatial and multi-omic profiling reveals pericyte-derived CCL19 as a key prognostic factor in CNS lymphoma.

Biological mechanisms underlying clinical heterogeneity in central nervous system lymphoma (CNSL) are largely unknown. While previous studies suggest the chemokine CLL19 as a crucial factor for the formation of CNSL in murine models, its role in human disease remains elusive. Here, we performed in-depth genetic and transcriptomic profiling of 82 CNSL specimens and identified distinct genetic aberrations and tumor cell compositions in lymphomas with high CCL19 expression, both of which were associated with immunosuppressive and anti-apoptotic signatures. CCL19 levels varied widely across CNSL p

EphB4/EphrinB2 signaling regulates pericyte-like function of DPSCs in angiogenesis via transcriptional factor SRF.

BACKGROUND: During angiogenesis, pericytes (PCs) drive vascular stabilization and maturation, a process largely regulated by the transcription factor serum response factor (SRF). Dental pulp stem cells (DPSCs) possess the potential to acquire PC-like properties and regulate angiogenesis. While EphB4/EphrinB2 signaling is reported to be involved in DPSC-mediated angiogenesis, its role in modulating PC-like function remains unexplored. Therefore, this study aims to investigate how EphB4/EphrinB2 signaling regulates the PC-like properties of DPSCs. METHODS: DPSCs were transfected with lentivirus

FOXF2 regulates pericyte-endothelial signaling required for vascular homeostasis after neonatal hyperoxic lung injury.

Pulmonary vascular development is essential for alveolarization, and disruption of this process contributes to pathogenesis of bronchopulmonary dysplasia (BPD). Proper vascular development requires an orchestration of many cell types within the lung. However, the transcriptional mechanisms by which pericytes support the endothelium in the postnatal lung remain poorly understood. Herein, we identify FOXF2 as a critical transcription factor that governs pericyte maturation and function during postnatal lung development and regeneration. FOXF2 expression in pericytes increases postnatally and is

The microcirculation, the blood-brain barrier, and the neurovascular unit in health and Alzheimer disease: The aberrant pericyte is a central player.

High fidelity neuronal signaling is enabled by a stable local microenvironment. A high degree of homeostatic regulation of the brain microenvironment, and its separation from the variable and potentially neurotoxic contents of the blood, is brought about by the central nervous system barriers. Evidence from clinical and preclinical studies implicates brain microcirculation, cerebral hypoperfusion, blood-brain barrier dysfunction, and reduced amyloid clearance in Alzheimer pathophysiology. Studying this dysregulation is key to understanding Alzheimer disease (AD), identifying drug targets, deve

Pericyte loss: a key factor inducing brain Aβ40 accumulation and neuronal degeneration in cerebral amyloid angiopathy.

Cerebral amyloid angiopathy (CAA) is a cerebral small vessel disease common among the elderly. Pericyte loss is one of the earliest characteristics of CAA. Although pericyte loss correlates with neuronal loss, the molecular mechanisms by which pericyte loss contributes to neurodegeneration and CAA progression remain poorly understood. This study aimed to explore the role and the mechanism of pericyte loss in CAA using in vivo APP/PS1 mice models and an in vitro neurovascular unit (NVU) model. The findings showed that pericyte coverage and mRNA expression levels of pleiotrophin (PTN) were signi

Systemic nimodipine affects pericyte calcium signaling, resting hemodynamics and neurovascular coupling in healthy mouse brain.

Nimodipine is a L-type voltage gated calcium channel blocker commonly given to patients after a sub-arachnoid hemorrhage. It is known to dilate cerebral arteries and affect brain pericytes that express voltage gated calcium channels. Here, we systemically administered nimodipine (1 mg/kg; i.p.) and measured brain pericyte calcium transients and single-vessel hemodynamics in the brains of mice by two-photon microscopy. We find that nimodipine reduces calcium transients in all types of pericytes, from ensheathing to thin-strand cells, at different locations in the vascular network. This induces

SARS-CoV-2 Spike S1 Subunit Triggers Pericyte and Microvascular Dysfunction in Human Pancreatic Islets.

UNLABELLED: The COVID-19 pandemic has profoundly affected human health; however, the mechanisms underlying its impact on metabolic and vascular systems remain incompletely understood. Clinical evidence suggests that SARS-CoV-2 directly disrupts vascular homeostasis, with perfusion abnormalities observed in various tissues. The pancreatic islet, a key endocrine miniorgan reliant on its microvasculature for optimal function, may be particularly vulnerable. Studies have proposed a link between SARS-CoV-2 infection and islet dysfunction, but the mechanisms remain unclear. Here, we investigated how

An In Vitro Partial Lesion Model of Differentiated Human Mesencephalic Neurons: Effect of Pericyte Secretome on Phenotypic Markers.

Parkinson's disease (PD) is characterised by the progressive degeneration of dopaminergic (DA) neurons in the substantia nigra pars compacta. Post-mortem data suggests that the loss of DA markers may long precede the cell death, leaving a window to rescue the DA phenotype. Screening for potential neuroprotective or restorative therapies, however, requires that partial lesions of DA neurons can be modelled in vitro. In order to establish a partial lesion model of DA neurons in vitro, we evaluated the effects of different exposure times to 1-methyl-4-phenylpyridinium (MPP+) and 6-hydroxydopamine

Loss of capillary pericytes and the blood-brain barrier in white matter in poststroke and vascular dementias and Alzheimer's disease.

White matter (WM) disease is associated with disruption of the gliovascular unit, which involves breach of the blood-brain barrier (BBB). We quantified pericytes as components of the gliovascular unit and assessed their status in vascular and other common dementias. Immunohistochemical and immunofluorescent methods were developed to assess the distribution and quantification of pericytes connected to the frontal lobe WM capillaries. Pericytes with a nucleus were identified by collagen 4 (COL4) and platelet-derived growth factor receptor-β (PDGFR-β) antibodies with further verification using PD

Role of capillary pericytes and precapillary arterioles in the vascular mechanism of betahistine in a guinea pig inner ear model.

AIMS: Betahistine is a histamine analogue that is used for the treatment of Menière's disease. Animal studies showed that it increases local blood flow in the stria vascularis. In terms of its mode of action, recent studies have prompted discussion of whether betahistine actively affects cochlear microcirculation by dilations of pericytes or of precapillary arterioles or by mere downstream effects. Hence, we investigated the effects of betahistine on cochlear capillary pericytes and precapillary arterioles. MAIN METHODS: The stria vascularis was visualized in 12 guinea pigs by in vivo fluores

PanIN-associated pericyte, glial, and islet remodeling in mice revealed by 3D pancreatic duct lesion histology.

Pericytes and glial cells are accessory cells of neurovascular networks, which have been reported to participate in scar formation after tissue injury. However, it remains unclear whether similar reactive cellular responses occur in pancreatic intraepithelial neoplasia (PanIN). In this study we developed three-dimensional (3D) duct lesion histology to investigate PanIN and the associated pericyte, glial, and islet remodeling. Transparent mouse pancreata with a Kras(G12D) mutation were used to develop 3D duct lesion histology. Deep-tissue, tile-scanning microscopy was performed to generate pano

Involvement of matrix metalloproteinase-9 in amyloid-β 1-42-induced shedding of the pericyte proteoglycan NG2.

Deposition of amyloid-β (Aβ) 1-42, the major component of senile plaques characteristic of Alzheimer disease, affects brain microvascular integrity and causes blood-brain barrier dysfunction, increased angiogenesis, and pericyte degeneration. To understand the cellular events underlying Aβ1-42 effects on microvascular alterations, we investigated whether different aggregation forms of Aβ1-42 affect shedding of the pericyte proteoglycan NG2 and whether they affect proteolytic cleavage mediated by matrix metalloproteinase (MMP)-9. We found decreased levels of soluble NG2, total MMP-9, and MMP-9

CO2 inhalation enhances pericyte-mediated neurovascular protection in ischemic stroke and remains safe in hemorrhagic stroke.

Therapeutic options for acute ischemic stroke beyond reperfusion therapy are limited by narrow time windows and associated risks. Carbon dioxide (CO2), a potent and adjustable vasodilator, may preserve ischemic brain tissue by enhancing cerebral blood flow (CBF) and stabilizing neurovascular components. We investigated whether CO2 preconditioning prior to arterial occlusion confers neuroprotection in a transient middle cerebral artery occlusion (tMCAO) model and evaluated its safety in a collagenase-induced intracerebral hemorrhage (ICH) model. Adult male rats received 20% CO2 before tMCAO or

Impact of NA-1 on Pericyte-Driven Vasoconstriction and Its Role in No-Reflow During Cerebral Ischemia-Reperfusion.

BACKGROUND: The no-reflow phenomenon in the ischemic brain following arterial recanalization leads to poor prognosis. Previous studies suggest that the clinically proven NA-1 drug, with disruption of PSD95 in the neuronal terminals alongside the cerebral microvasculature, may inhibit pericytic contraction of microvessels by reducing endothelin-1 secretion. METHODS: A 1.5-h tMCAO model using Balb/c mice was employed. In vivo two-photon imaging and immunofluorescence staining were employed to assess the constriction effect of pericytes on capillaries. The impact of NR2B9c/NA-1 administration (i

Globally patterned locus coeruleus-norepinephrine neuron-pericyte coupling orchestrates brain-wide vascular dynamics.

The brain regulates its blood supply at both local and global scales to ensure proper neural functions. While local cerebral blood flow (CBF) is regulated by regional neural activities via neurovascular coupling, how CBF is globally coordinated remains largely unclear. Leveraging in vivo whole-brain monitoring of multiple neurovascular components in larval zebrafish, we demonstrate that the locus coeruleus-norepinephrine (LC-NE) system, a key arousal node in vertebrates, functions as a central hub in global neurovascular communication through driving capillary-to-arteriole patterned activities

Mutated LRRK2 induces a reactive phenotype and alters migration in human iPSC-derived pericyte-like cells.

BACKGROUND: Pericytes play a crucial role in controlling inflammation and vascular functions in the central nervous system, which are disrupted in Parkinson's disease (PD). Still, there is a lack of studies on the impact of pericytes on neurodegenerative diseases, and their involvement in the pathology of PD is unclear. Our objective was to investigate the molecular and functional differences between healthy pericytes and pericytes with the LRRK2 G2019S mutation, which is one of the most common mutations associated with PD. METHODS: Our study employed pericyte-like cells obtained from induced
