Axonal transport declines with age in two distinct phases separated by a period of relative stability.

Axonal transport is critical for supplying newly synthesized proteins, organelles, mRNAs, and other cargoes from neuronal cell bodies into axons. Its impairment in many neurodegenerative conditions appears likely to contribute to pathogenesis. Axonal transport also declines during normal aging, but little is known about the timing of these changes, or about the effect of aging on specific cargoes in individual axons. This is important for understanding mechanisms of age-related axon loss and age-related axonal disorders. Here we use fluorescence live imaging of peripheral nerve and central ner

Phase segregation of polymerizable lipids to construct filters for separating lipid-membrane-embedded species.

Supported lipid bilayer (SLB) platforms have been developed to transport and separate membrane-embedded species in the species' native bilayer environment. In this study, we used the phase segregation phenomenon of lipid mixtures containing a polymerizable diacetylene phospholipid, 1,2-bis(10,12-tricosadiynoyl)-sn-glycero-3-phosphocholine (DiynePC), and a nonpolymerizable phospholipid, 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC), to create filter barrier structures in SLBs. Upon exposing the phase segregated samples to UV light, the DiynePC-rich domains could become crosslinked and remain

[Chiral separation of glycerin monosulfides and bisulfides with a stationary phase of amylose-tris(3,5-dimethylphenylcarbamate)].

Amylose-tris(3,5-dimethylphenylcarbamate) (ADMPC) and aminopropylated silica gel(APS) were prepared after the reported methods. ADMPC was immobilized on APS from tetrahydrofuran solution with a coating amount of 15%. The chiral stationary phase was packed in a stainless-steel column(150 mm x 4.6 mm i.d.) by slurry method. The column was used for the enantioseparation of 6 glycerin monosulfides and 4 glycerin bisulfides. Mixtures of hexane and 2-propanol were used as mobile phases. The enantiomers of the monosulfides could be separated quite well, while those of the bisulfides could not at all.

Separation of toxic peptides (microcystins) in capillary electrophoresis, with the aid of organic mobile phase modifiers.

A capillary electrophoretic (CE) method incorporating sodium dodecyl sulphate (SDS)-organic modifier solvents in the CE buffer was developed for the detection of toxic cyclic heptapeptide toxins (microcystins) produced by blue-green algae (cyanobacteria). The applicability of these run buffers for the analysis of microcystins was evaluated and optimum conditions for separation were determined. The migration times, elution order and selectivity of the toxic peptides were influenced by modifying the composition of the electrophoretic buffer with organic solvents [0 to 20% (v/v)]. At maximum addi

Macromolecular association creates dispersity of components that determines liquid-liquid phase separation of oppositely charged polyions.

Developing insight into liquid-liquid phase separation in complex polyelectrolyte mixtures has acquired new urgency considering the role this process plays in the formation of biomolecular condensates, which are involved in diverse processes including, stress response, signal transduction, metabolism, gene regulation, and many human diseases. Condensates can be viewed as a biological membraneless structure situated, in scale and function, between macromolecules and cells. The physical properties of, and the physico-chemical environments within, condensates are very important determinants of th

Separation of phosphodiester impurities from fully phosphorothioate-modified oligonucleotides by ion-pair reversed-phase liquid chromatography.

Phosphorothioate (PS)-modified oligonucleotides are widely used in therapeutic applications; however, the separation of phosphodiester (PO)-substituted impurities remains a significant analytical challenge because these species differ only by the replacement of sulfur with oxygen within the phosphate backbone. Although ion-pair reversed-phase liquid chromatography (IP-RPLC) is widely employed for oligonucleotide analysis, the factors governing PS/PO selectivity have not been fully elucidated. In this study, we systematically investigated the effects of alkylamine structure, hexafluoroisopropan

Ionically Counter-Programmable Phase-Separation of Superstable Star-Biopolyamide Coacervates for Electrothermally-Responsive Pseudosolid Smart Windows.

Thermoresponsive polymers are highly anticipated for passive thermochromic smart windows, yet the instability and too-high mobility of their light-scattering states for separated phases remain significant hurdles. To address these challenges, we report a design concept for pyrrolidone-based star-polyamides synthesized from itaconic acid and cadaverine, whose architecture provides exceptional sensitivity to external stimuli. In a stark departure from the classical Hofmeister series, Ca2+ initiates an anomalous salting-in effect that contrasts with the Na+-induced salting-out. DFT/MD calculation

Effect of freeze-thaw cycle induced oligomers on alpha-synuclein liquid-liquid phase separation and aggregation.

Protein aggregation in the form of amyloid fibrils is linked with the onset and progression of several widespread and incurable disorders, including neurodegenerative Alzheimer's and Parkinson's diseases. Despite decades of research and countless attempts to discover potential aggregation-altering treatment modalities, there are still no effective cures for the majority of amyloid-related disorders. This is, in part, due to our incomplete understanding of the underlying factors that influence the phase separation and aggregation of these various disease-related proteins. Over the last few year

Gene Expression Patterns in Liquid-Liquid Phase Separation: A Prognostic and Predictive Tool for Colorectal Cancer.

To illuminate the role of liquid-liquid phase separation (LLPS) in colorectal cancer (CRC) and its potential as a therapeutic target, we conducted an in-depth analysis focusing on LLPS-related genes (LLPSRGs). Our approach involved a predictive model with LLPSRGs levels a s basis, with intent to prognosticate patient survival and assist in immunotherapy decision-making. Employing univariate Cox regression equipped with LASSO for initial screening, we identified LLPSRGs linked to CRC survival. A subsequent multivariate Cox regression analysis led to the establishment of a prognostic model that

Sulfobutylether-(hydroxypropyl-β-cyclodextrin) bonded silica as stationary phase for enhancing chiral separation in liquid chromatography.

The present study addresses the limited recognition modes and poor universality of conventional cyclodextrin-based chiral stationary phases (CSPs) by proposing a rational design strategy based on "functional group synergy" concept. Following this strategy, sulfobutylether-(hydroxypropyl-β-cyclodextrin), hereafter referred to as SBE-(HP-β-CD), was successfully synthesized and, for the first time, chemically bonded to silica matrix to fabricate a SBE-(HP-β-CD) CSP. Comprehensive evaluation demonstrated that this CSP exhibits superior chiral selectivity, higher column efficiency, and shorter rete

Hydrophobic cellulose scaffolds for oil-water separation via high internal phase emulsion templating and surface modification.

The increasing frequency of oil contamination in aquatic environments necessitates the development of efficient, sustainable, and reusable sorbent materials for oil-water separation. This study reports the fabrication and evaluation of a hydrophobically modified cellulose-based interpenetrating polymer network (IPN) scaffold, synthesized via high internal phase emulsion (HIPE) templating and chemically modified using trimethoxy(methyl)silane (MTMS) through chemical vapour deposition (CVD). The resulting scaffold exhibits a highly porous structure (porosity ≈ 96.5%) and hydrophobic characterist

Blocking the Phase Separation of hnRNP L Attenuates Pulmonary Fibrogenesis by Inhibiting Fibroblast-to-myofibroblast Differentiation.

INTRODUCTION: Pulmonary fibrosis is an interstitial lung disease characterized by diverse etiologies and unknown mechanisms. Phase separation has been implicated in the development of numerous diseases, but its role in the pathogenesis of pulmonary fibrogenesis remains unclear. In this study, we explored whether heterogeneous nuclear ribonucleoprotein L (hnRNP L) can form phase-separation condensates that contribute to the progression of pulmonary fibrosis. METHODS: Pulmonary fibrosis was modeled in vitro using TGF-β1-treated fibroblasts and in vivo by intratracheal bleomycin administration i

Physical Simulation of Phase Separation at the Slag-Metal Interface During Pellet Melting: A Phenomenological Study.

Metallized pellets are spherical iron-bearing burden materials obtained by treating iron ore pellets through processes such as direct reduction. They contain a certain proportion of metallic iron and mainly consist of metallic iron, incompletely reduced oxides, gangue, and other nonmetallic components. They are one of the commonly used iron-bearing materials in electric smelting furnaces. The melting process of metallized pellets not only affects the melting efficiency of the charge but is also accompanied by slag-metal separation and gangue separation, which is directly related to mass transf

Liquid-liquid phase separation enables chromatography-free purification and high-performance spidroin-amyloid hybrid silk fibers.

Large-scale production of artificial spider silk fibers requires heterologous expression of spider silk proteins (spidroins), yet current methods remain limited by low yields and costly purification processes. To overcome these challenges, we engineered mini-spidroins in which the poly-alanine motifs of the repetitive region were replaced with the non-natural amyloidogenic β16 peptide, significantly enhancing expression yields and solubility. Furthermore, we developed a simple, chromatography-free purification method for these constructs based on NaCl-induced liquid-liquid phase separation (LL

Thermally-induced phase separation of alkali lignin into mesoscale pigments for rapid mordant-free cotton dyeing.

Industrial lignin is a promising sustainable textile colorant, but its weak adhesion to fibers limits practical use. Here we report a metal-free dyeing strategy based on mesoscale lignin pigments (MLPs) formed through thermally induced phase separation. Alkali lignin treated in 50% (v/v) ethanol-water at 200 °C underwent partial depolymerization and, upon cooling, reorganized into irregular ~4 μm flocs driven by hydrophobic interactions and π-π stacking. The resulting metastable suspension mobilized ~52% of the initial lignin and remained dyeing-active for ~24 h, with coarser flocs settling ou

Phosphate anions-induced liquid-liquid phase separation in silk fibroin systems to produce biologically stable patches.

Silk fibroin (SF), the primary protein component of Bombyx mori silkworm cocoons, undergoes liquid-liquid phase separation (LLPS), followed by coacervation into fibers, in the silkworm glands. The molecular mechanisms underlying LLPS remain to be revealed. Here, we show that phosphate buffer (PB), a less commonly utilized, but more biomimetic route towards triggering SF assembly, induces LLPS of water-soluble SF by increasing hydrophobic interactions between SF chains. We demonstrate the ability of phosphate anions to promote self-assembly of silk fibroin through LLPS, resulting in protein-ric

The ITGB2-COPS3-SOX2 Axis and SOX2 Liquid-Liquid Phase Separation: Dual Mechanisms Governing Osteosarcoma Stemness.

Osteosarcoma is a highly malignant bone tumor prevalent in adolescents, whose therapeutic challenges stem largely from cancer stem cell (CSC)-mediated chemoresistance and pulmonary metastasis. High-frequency amplification of the 17p11.2 chromosomal region in osteosarcoma represents a key molecular event driving malignant progression. This study focuses on the key gene COPS3 in this region, delving into its molecular mechanisms regulating cancer stemness. The findings show that elevated COPS3 expression strongly correlates with stemness features. Mechanistically, COPS3 binds directly to transcr

FUS controls muscle differentiation through phase separation-mediated recruitment of the transcription factors MEF2 and ETV5.

FUS is an RNA-binding protein mutated in amyotrophic lateral sclerosis (ALS), a neurodegenerative disease characterized by progressive muscle weakness. We show in this work that a heterozygous knock-in mutation in the mouse Fus gene leads to cell-autonomous ultrastructural defects in skeletal muscle, with disruption of sarcomeres and mitochondria. Studies in mouse and Drosophila models demonstrate an evolutionarily conserved cell-autonomous function of FUS in muscle development. Mechanistically, FUS is required for the transcription of MEF2 target genes, binds to the promoter of genes bound by

Distribution and Relative Size of Protein Binding Domains Cooperatively Influence Phase Separation of Protein-RNA Mixtures.

Multivalent protein-protein interactions play essential roles in mediating liquid-liquid phase separation (LLPS) that drives biomolecular condensate formation. Here, we systematically investigate how the spatial distribution and relative size of protein binding domains (PBDs) would influence LLPS in a mixture of spherical proteins and RNA single strands by using a patchy-particle polymer model, wherein each protein contains a fixed number of PBDs on the surface distributed closely or sparsely. Intriguingly, we find that LLPS behavior exhibits a nontrivial dependence on the cooperative interpla
