ZBTB16 promotes the transcription of glycolytic enzyme PFKFB2 to participate in fibrogenesis in septic lung injury.

BACKGROUND: Sepsis is a severe systemic inflammatory response syndrome that leads to acute lung injury (ALI), which subsequently progresses to lung fibrosis. This study aims to investigate the role of zinc finger and BTB domain containing 16 (ZBTB16), a transcription factor, in septic lung injury. METHODS: The expression of Zbtb16 in the injured lung was evaluated using single-cell sequencing. The signaling pathways that can be regulated by Zbtb16 were predicted using gene set enrichment analysis (GSEA). Then, the functions of ZBTB16 and its potential molecular mechanism in fibrogenesis were

Pp6-Pfkfb1 axis modulates intracellular bacterial proliferation by orchestrating host-pathogen metabolic crosstalk.

Intracellular bacterial pathogens exhibit heterogeneous replication rates within host macrophages, but the mechanisms by which they manipulate host factors for survival remain incompletely understood. Using a fluorescence-dilution reporter system in Salmonella Typhimurium (Salmonella)-infected macrophages, we found that Protein Phosphatase 6 (Pp6) was downregulated in macrophages harboring growing bacteria. Conditional knockout of Pp6 elevated host susceptibility to Salmonella-mediated lethality due to compromised antimicrobial defenses. MicroRNA-31 (miR-31) was identified as a negative regula

Inhibition of 6-phosphofructo-2-kinase activity by mercaptopurines.

The activity of 6-phosphofructo-2-kinase (PFK-2), the enzyme that catalyses the synthesis of fructose 2,6-bisphosphate (Fru-2,6-P2), was inhibited by mercaptopurines in vitro. Inhibition was observed with the purified enzyme from rat liver and bovine heart, and in extracts from rat lymphocytes and hepatoma cells, chick embryo fibroblasts, and human HeLa and lymphoblastoid cells. Half-maximal effect was obtained with 0.1-0.2 mM mercaptopurine and maximal inhibition ranged between 50 and 90% depending on the enzyme preparation. The inhibition resulted from a decrease in Vmax with no change in Km

Single cell multi-omics guided hydrogel enables closed-loop therapy for renal osteodystrophy.

Fracture repair in renal osteodystrophy remains challenging because impaired bone quality compromises fixation stability, while current systemic therapies regulating bone metabolism do not sufficiently address the disease-specific mechanisms underlying defective repair. An integrated strategy combining early fracture stabilization with mechanism-guided local treatment is therefore needed. Here, integrated single-cell RNA sequencing and multi-omics analyses identify inflammatory macrophages as a key pathogenic population, characterized by glycolytic reprogramming driven by hypoxia-inducible fac

Orientin alleviates severe inflammation via regulating macrophage glycolysis and immune function in sepsis.

Sepsis is a heterogeneous syndrome triggered by a dysregulated host response to infection, with glycolysis playing a vital role in maintaining macrophage immune function, which is critical for immune homeostasis and host survival during severe sepsis. Targeting glycolytic enzymes may offer effective strategies to mitigate macrophage-mediated inflammatory responses during sepsis. This study investigated the anti-inflammatory and metabolic modulatory effects of Orientin (Ori) in murine models of endotoxemia and sepsis, with a particular focus on its interaction with the glycolytic enzyme phospho

Adaptive Evolution of the PFK Gene Family in Chinese Longsnout Catfish, Leiocassis longirostris.

The Chinese longsnout catfish is a typical carnivorous fish with a relatively weak ability to utilize glucose. However, the genomic basis for its glucose metabolic adaptation remains unclear. In this study, we used comparative genomics methods to systematically analyze the evolutionary characteristics of glucose metabolism-related genes in the Chinese longsnout catfish, focusing on gene family evolution, patterns of expansion and contraction, and selective pressures. The results indicate that glucose metabolism-related genes have undergone significant reshaping during evolution. Genes involved

Exploring the effects of lifelong aerobic exercise on early skeletal muscle remodeling between 8 and 14 months in the rat.

The insidious onset and progression of sarcopenia make it vital to understand the early skeletal muscle changes and explore therapies to slow its progression. This study explored the gastrocnemius remodeling at an early stage of aging (14 months of age) and the effects of lifelong aerobic exercise. For that, 2-month-old male Wistar rats underwent a 12-month treadmill exercise program. Sedentary age-matched, young sedentary, and young exercised for 6 months rats were considered. The results highlighted an age-related decrease in the relative gastrocnemius muscle mass, suggestive of loss or atro

Glycolysis-dependent sulfur metabolism orchestrates morphological plasticity and virulence in fungi.

Fungi exhibit remarkable morphological plasticity, which allows them to undergo reversible transitions between distinct cellular states in response to changes in their environment. This phenomenon, termed fungal morphogenesis, is critical for fungi to survive and colonize diverse ecological niches and establish infections in a variety of hosts. Despite significant advancements in the field with respect to understanding the gene regulatory networks that control these transitions, the metabolic determinants of fungal morphogenesis remain poorly characterized. In this study, we uncover a previous

LPS and β-Glucan Induce Differential Memory-like Inflammatory Responses with Enhanced Reactivity in Neonatal Neutrophils Compared to Adults.

Neutrophils are key effectors of innate immunity. Although traditionally regarded as short-lived effector cells, recent evidence suggests they can undergo functional reprogramming after microbial stimulation, leading to trained immunity. We investigated whether lipopolysaccharide (LPS) or Candida albicans β-glucan induces memory-like inflammatory responses in adult peripheral and neonatal cord blood neutrophils. We used complementary molecular and functional approaches to characterize neutrophil responses following in vitro priming with LPS or β-glucan and subsequent LPS restimulation. Cytokin

P4HA2 promotes the progression of thyroid cancer by regulating PFKP-mediated glycolysis.

Emerging evidence suggests that prolyl-4-hydroxylase α subunit 2 (P4HA2) plays critical roles in cancer progression through multiple mechanisms. Notably, P4HA2 has been implicated in modulating glycolytic pathways in malignancies. Phosphofructokinase (PFKP), a key glycolytic enzyme, exhibits significant overexpression in thyroid cancer. This study investigates the functional of P4HA2 in thyroid cancer and elucidates the P4HA2/PFKP axis in regulating cancer cell glycolysis. Bioinformatics analysis using GEPIA website revealed P4HA2 expression patterns in thyroid cancer samples. P4HA2 protein le

Mindful Eating: A Deep Insight Into Fructose Metabolism and Its Effects on Appetite Regulation and Brain Function.

Fructose, a common sweetener in modern diets, has profound effects on both metabolism and brain function, primarily due to its distinct metabolic pathways. Unlike glucose, fructose bypasses critical regulatory steps in metabolism, particularly the phosphofructokinase-1 (PFK-1) feedback inhibition, leading to uncontrolled metabolism and increased fat storage. This review delves into the metabolic consequences of fructose consumption, including its limited role in directly stimulating insulin secretion, which affects satiety signaling and contributes to increased food intake. The small intestine

Transcriptomic and in vitro analysis of red pitahaya betacyanin fraction as a natural anti-biofilm agent against oral Streptococcus mutans.

Dental plaque biofilms, primarily formed by Streptococcus mutans, contribute significantly to oral diseases, necessitating effective anti-biofilm strategies. This study investigates the anti-biofilm potential of red pitahaya betacyanin fraction (BF) using transcriptomic analysis and an in vitro denture plaque model. Stringent differential gene expression analysis (p < 0.01, log2 fold change > ± 2) revealed that BF significantly downregulates genes involved in sucrose metabolism, energy production, and cell wall biosynthesis, impairing bacterial adhesion and biofilm integrity. Notably, the supp

Research on the mechanism of resistance exercise in promoting glucose metabolic shift to regulate muscle satellite cell proliferation in type 2 diabetic rats.

Skeletal muscle atrophy is a common complication in patients with type 2 diabetes (T2D) and is associated with dysfunction of muscle satellite cells. The activation and proliferation of muscle satellite cells involve a switch in glucose metabolism, which is regulated by driving the acetylation of histones to control the expression of related genes. Studies have confirmed that resistance exercise can improve insulin resistance and activate muscle satellite cells, but the specific molecular mechanisms are not yet clear. This study aims to investigate whether resistance exercise can promote the p

PFKP inhibition protects against pathological cardiac hypertrophy by regulating protein synthesis.

Metabolic reprogramming precedes most alterations during pathological cardiac hypertrophy and heart failure (HF). Recent studies have revealed that Phosphofructokinase, platelet (PFKP) has a wealth of metabolic and non-metabolic functions. In this study, we explored the role of PFKP in cardiac hypertrophic growth and HF. The expression level of PFKP was elevated both in pathological cardiac remodeling mouse model challenged by transverse aortic constriction (TAC) surgery and in the neonatal rat cardiomyocytes (NRCMs) stimulated by phenylephrine (PE). In global PFKP knockout (PFKP-KO) mice, car
