Synthesis of first trimester placental alkaline phosphatase in cultured human term placental cells.

Alkaline phosphatase activity in human placental cells transformed by a tsA mutant of simian virus 40 (SV40) can be greatly induced by growing these cells at 40 degrees C, the temperature at which the tsA transformants regain their nontransformed phenotype. The induction of alkaline phosphatase in these cells requires the synthesis of both RNA and protein. The induced alkaline phosphatase from a SV40 tsA30 mutant-transformed term placental cell line (TPA30-1) was purified, characterized, and compared with alkaline phosphatase from term placenta and first trimester placenta. The form of alkalin

Convergently evolved placental villi show multiscale structural adaptations to differential placental invasiveness.

Despite having a single evolutionary origin and conserved function, the mammalian placenta exhibits radical structural diversity. The evolutionary drivers and functional consequences of placental structural diversity are poorly understood. Humans and equids both display treelike placental villi, however these villi evolved independently and exhibit starkly different levels of invasiveness into maternal tissue (i.e. the number of maternal tissue layers between placental tissue and maternal blood). The villi in these species therefore serve as a compelling evolutionary case study to explore whet

Vitamin C supplementation improves placental function and alters placental gene expression in smokers.

Maternal smoking during pregnancy (MSDP), driven by nicotine crossing the placenta, causes lifelong decreases in offspring pulmonary function and vitamin C supplementation during pregnancy prevents some of those changes. We have also shown in animal models of prenatal nicotine exposure that vitamin C supplementation during pregnancy improves placental function. In this study we examined whether vitamin C supplementation mitigates the effects of MSDP on placental structure, function, and gene expression in pregnant human smokers. Doppler ultrasound was performed in a subset of 55 pregnant smoke

A longitudinal and cross-sectional study of placental circulation between normal and placental insufficiency pregnancies.

INTRODUCTION: To longitudinally and cross-sectionally study the differences in the uterine artery pulsatility index (UTPI), umbilical artery pulsatility index (UAPI) and placental vascularization indices (PVIs, derived from 3-dimensional power Doppler) between normal and placental insufficiency pregnancies throughout gestation. METHODS: UTPI, UAPI and PVI were measured 6 times at 4- to 5- week intervals from 11 to 13+6 weeks-36 weeks. Preeclampsia (PE) and fetal growth restriction (FGR) were defined as placental insufficiency. Comparisons of UTPI, UAPI and PVI between normal and insufficiency

MRI Assessed Placental Location as a Diagnostic Tool of Placental Invasiveness and Maternal Peripartum Morbidity.

Placenta accreta spectrum (PAS) disorder is one of the leading causes of peripartum maternal morbidity and mortality; its early identification during pregnancy is of utmost importance to ensure the optimal clinical outcome. The aim of the present study is to investigate the possible association of the presence and type/location of placenta previa on MRI with PAS and maternal peripartum outcome. One hundred eighty-nine pregnant women (mean age: 35 years; mean gestational age: 32 weeks) at high risk for PAS underwent a dedicated placental MRI. All women underwent a C-section within 6 weeks from

Intra-placental arterial Doppler: A marker of fetoplacental vascularity in late-onset placental disease?

INTRODUCTION: Late-gestation adverse pregnancy outcome is associated with reduced placental villous vascularity but rarely with a frankly abnormal umbilical artery Doppler waveform. The clinical utility of umbilical artery Doppler velocimetry in late gestation is limited by poor understanding of what aspect(s) of placental structure and function the impedance reflects. We hypothesized that placental arterial circulation impedance reflects placental vascularity and arterial function. MATERIAL AND METHODS: This was a secondary analysis of data from the FEMINA2 study, a study of pregnancy outcom

The association between first trimester placental biomarkers and placental lesions of maternal vascular malperfusion.

INTRODUCTION: Abnormal levels of first trimester placental biomarkers are associated with the development of placental syndrome (PS). However, prediction performance is moderate, possibly explained by the clinical heterogeneity of PS. Aim of this study is to investigate the association between first trimester biomarkers and the presence of maternal vascular malperfusion (MVM), as a marker for placental insufficiency. METHODS: This retrospective study included 195 women with available first trimester blood sample and placenta histological sections for examination at the Maastricht University M

Extensive shift in placental transcriptome profile in preeclampsia and placental origin of adverse pregnancy outcomes.

One in five pregnant women suffer from gestational complications, prevalently driven by placental malfunction. Using RNASeq, we analyzed differential placental gene expression in cases of normal gestation, late-onset preeclampsia (LO-PE), gestational diabetes (GD) and pregnancies ending with the birth of small-for-gestational-age (SGA) or large-for-gestational-age (LGA) newborns (n = 8/group). In all groups, the highest expression was detected for small noncoding RNAs and genes specifically implicated in placental function and hormonal regulation. The transcriptome of LO-PE placentas was clear

Placental endoglin levels in diamniotic-monochorionic twin gestations: correlation with clinical and placental characteristics.

OBJECTIVE: While endoglin has been implicated in the pathogenesis of various complications in singleton pregnancies, its potential contribution to complications of monochorionic twinning remains largely undetermined. The aim of this study was to determine the correlation between relevant clinical and pathological variables and placental endoglin levels in diamniotic-monochorionic twin pregnancies. METHODS: Endoglin expression was studied by immunohistochemistry and Western blot in a prospective cohort of 68 non-TTTS and 7 TTTS monochorionic twin placentas. Placental endoglin levels were corre

Placental membrane and placental disc microscopic chorionic cysts share similar clinicopathologic associations.

Placental membrane microscopic chorionic cysts (MCC) are associated with clinical conditions and placental features of in utero hypoxia. Those occurring in the cell islands, chorionic plate, placental septa, and maternal floor of the chorionic disc have been thought to have degenerative/liquefactive or secretory etiology and no clinical significance. To study the clinical and placental associations of MCC, 24 clinical (maternal and fetal) and 46 gross and microscopic placental features were statistically compared among 266 consecutive cases with MCC (study group, SG) and 656 consecutive placen

Human placental gonadotrophin-releasing hormone-like factors: an artefact of human placental peptidases?

Non-denatured human placental cytosol fractions displaced tracer binding in parallel with gonadotrophin-releasing hormone (GnRH) isoform and agonist peptides in GnRH-specific radioimmunoassays and radioreceptor assays. However, placental immuno- and receptor binding-GnRH-like activity was highly correlated with inactivation of GnRH tracers, suggesting that placental GnRH-like factors may be an artefact of ligand degradation during assay. The properties and inhibitor sensitivities of the major (125)I-labelled GnRH-degrading enzymes of term placental cytosol were studied using a dextran-coated c

The antenatal assessment of utero-placental and feto-placental blood flow using Doppler ultrasound.

A brief discussion of the normal anatomy and physiology relevant to the understanding of blood flow is presented. After a review of previous attempts to measure blood flow in the utero-placental and feto-placental circulation, a description is given of the methods of obtaining Doppler signals from both maternal and fetal vessels. Some of the problems are presented. Flow velocity waveform (FVW) analysis is then presented in normal and in complicated pregnancies. FVW in pregnancies complicated by proteinuric hypertension have arcuate FVWs that demonstrate increased pulsatility and decreased velo

Placental function, fetal distress, and the fetal/placental weight ratio in normal and gestotic pregnancies.

The ratio between fetal and placental weight is often thought to be a measure of the reserve capacity of the placenta. The aim of our study was to investigate the relationship between 1) endocrinologic parameters during pregnancy (serum placental lactogen - HPL, urinary estriol - E3), 2) the occurrence of fetal distress during labor, and 3) the severity of EPH gestosis, and the fetal/placental weight ratio at various gestational ages. The data from a total of 4911 consecutive pregnancies and deliveries were evaluated. Up to 37 weeks the mean fetal/placental weight ratio was significantly lower

Placental Autophagy: Causes and Consequences.

Suboptimal intake of dietary vitamin C (ascorbate) increases the risk of several chronic diseases but the exact metabolic pathways affected are still unknown. In this study, we examined the metabolic profile of mice lacking the enzyme gulonolactone oxidase (Gulo) required for the biosynthesis of ascorbate. Gulo-/- mice were supplemented with 0%, 0.01%, and 0.4% ascorbate (w/v) in drinking water and serum was collected for metabolite measurements by targeted mass spectrometry. We also quantified 42 serum cytokines and examined the levels of different stress markers in liver. The metabolic profi

The curious case of the placental genome.

The human placenta is among the few, if not the only, healthy mammalian tissues that may exploit a cancer-like somatic genome for its function. Acting simultaneously as the lungs, kidneys, liver and gastrointestinal tract, the placenta mediates nutrient and gas exchange, blocks out toxins and pathogens, removes fetal waste, and secretes hormones essential for pregnancy. Recent research has revealed that the placenta is a genomically distinctive organ with an unusually high mutational load, abundant structural variations, prevalent aneuploidy and genome amplification, as well as a tissue-sized

KRAS4B is required for placental development.

Beyond its well-established role in cancer, KRAS is also crucial for embryogenesis, as its absence leads to embryonic lethality. However, the precise mechanisms underlying the developmental functions of KRAS, as well as the respective roles of its two splicing isoforms, KRAS4A and KRAS4B, remain incompletely characterized. To address these issues, we generated Kras4A knock-out (Kras4A-/-) and Kras4B-/- mouse models using CRISPR/Cas9 technology, and compared their phenotypes to those of a Kras-/- model, in which both isoforms are simultaneously inactivated. We observed that Kras-/- and Kras4B-/

Familial risk of placental abruption.

BACKGROUND: Placental abruption (PA), the partial or complete separation of the placenta before delivery, occurs in 1-2 % of pregnancies and is associated with severe maternal and perinatal morbidity and mortality. The etiology of PA is complex, and the increased recurrence risk in subsequent pregnancies suggests a potential genetic contribution. Investigating PA in large families may reveal important insights into inherited risk and the extent to which genetic factors contribute to PA. To guide future gene-discovery studies, we investigated the familial risk of PA using data from 1.2 million
