In vitro inhibitory effects on free radicals, pigmentation, and skin cancer cell proliferation from Dendrobium hybrid extract: A new plant source of active compounds.

Orchidaceae are diverse plants whose bioactive compounds have various biological activities. New hybrids of Dendrobium have been generated to gain characteristics shared with their ancestors. Dendrobium Pearl Vera (designated as DH) is derived from parents used for dermatological treatments and cosmetics. However, the phytoconstituents and biological properties of DH have not been reported. The current study investigated extracts from DH plants using four solvents (water, methanol, ethanol, or 2-propanol). The propanolic extract (DH-P) contained the highest phenolic and flavonoid contents, alo

Plant Hsp90 is a novel adjuvant that elicits a strong humoral and cellular immune response against B- and T-cell epitopes of a Toxoplasma gondii SAG1 peptide.

BACKGROUND: The 90-kDa heat-shock protein (Hsp90) from Nicotiana benthamiana (NbHsp90.3) is a promising adjuvant, especially for those vaccines that require a T cell-mediated immune response. Toxoplasma gondii SAG1 is considered one of the most important antigens for the development of effective subunit vaccines. Some epitopes located in the SAG1 C-terminus region have showed a strong humoral and cellular immune response. In the present study, we aimed to assess the efficacy of NbHsp90.3 as carrier/adjuvant of SAG1-derived peptide (SAG1HC) in a T. gondii infection murine model. METHODS: In th

In vivo anti-arthritic effect and repeated dose toxicity of standardized methanolic extracts of Buddleja cordata Kunth (Scrophulariaceae) wild plant leaves and cell culture.

ETHNOPHARMACOLOGICAL RELEVANCE: Buddleja cordata Humb. Bonpl. & Kunth, known by the population as Tepozán blanco, is a shrub plant used in traditional herbal medicine in Mexico for the treatment of tumors, cancer, sores, skin burns, rheumatic pains and diseases related to inflammatory processes such as arthritis; authors adjudicate this etno-medicinal effect to the presence of secondary metabolites in the plant such as verbascoside, however due to its low concentration in recent years biotechnological tools are applied as cell culture to biosynthesize these pharmacological active metabolites i

Polysaccharides isolated from plant cell cultures of Echinacea purpurea enhance the resistance of immunosuppressed mice against systemic infections with Candida albicans and Listeria monocytogenes.

Polysaccharides (EP) isolated from large scale plant cell cultures of Echinacea purpurea, have been shown to activate human and murine phagocytes. In this study we investigated the influence of EP on the nonspecific immunity in immunodeficient mice. EP was effective in activating peritoneal macrophages isolated from animals after administration of cyclophosphamide (CP) or cyclosporin A (CsA). EP-treated macrophages exhibited increased production of tumor necrosis factor-alpha (TNF) and enhanced cytotoxicity against tumor target WEHI 164 as well as against the intracellular parasite Leishmania

Navigating Guaiacyl and Syringyl Lignin Structural Complexity in Intact Plant Cell Walls Through 13C Chemical Shift Mapping by Two-Dimensional Solid-State NMR.

Lignin is a structurally complex and heterogeneous biopolymer whose in situ architecture remains difficult to resolve because of extensive spectral overlap and structural perturbations introduced during isolation and dissolution. These challenges have limited direct interrogation of native lignin within intact plant cell walls and underscore the need for analytical approaches capable of resolving lignin structure in its native context. Here, we present a framework that assists systematic 13C solid-state NMR identification of guaiacyl- and syringyl-based lignin motifs, including their associate

A fraction of diet-derived microRNAs present in plant extracellular vesicles remain detectable after gastrointestinal digestion and shows epithelial uptake in a CaCo-2 cell model.

Diet constitutes a source of exogenous miRNAs that are detectable in humans. Source, stability to digestion, and cellular uptake of diet-derived miRNAs are debated topics. Here, we studied the stability of cauliflower miRNAs to cooking (100 °C, 20 min) and gastrointestinal digestion (static INFOGEST 2.0), their possible cellular uptake/secretion by CaCo-2 cells, and the role of plant extracellular vesicles (PEVs). miRNAs were assessed by qPCR. The role of PEVs was evaluated via dynamic light scattering, electron microscopy, and immunoblotting. Most RNA was degraded after digestion (99.1%). How

Cellular and molecular effects of Baccharis dracunculifolia D.C. and Plectranthus barbatus Andrews medicinal plant extracts on retinoid metabolism in the human hepatic stellate cell LX-2.

BACKGROUND: Chronic hepatic diseases are serious problems worldwide, which may lead to the development of fibrosis and eventually cirrhosis. Despite the significant number of people affected by hepatic fibrosis, no effective treatment is available. In the liver, hepatic stellate cells are the major fibrogenic cell type that play a relevant function in chronic liver diseases. Thus, the characterization of components that control the fibrogenesis in the hepatic stellate cells is relevant in supporting the development of innovative therapies to treat and/or control liver fibrosis. The present stu

Screening of promising chemotherapeutic candidates from plants against human adult T-cell leukemia/lymphoma (IV): phenanthroindolizidine alkaloids from Tylophora tanakae leaves.

Adult T-cell leukemia/lymphoma (ATL) is a malignancy of mature peripheral T lymphocytes caused by human T-cell lymphotropic virus type 1 (HTLV-1). There are an estimated 5 million to 20 million HTLV-1-infected individuals worldwide; their lifetime risk of developing ATL is 3-5 %, and high HTLV-1 proviral loads have been shown to be an independent risk factor. Although conventional chemotherapeutic regimens used against other malignant lymphomas have been administered to ATL patients, the prognosis is often poor. In previous studies, we screened 459 extracts from 344 plants to isolate component

Unprecedentedly tough, strong, water-resistant, and multifunctional lignin-based resin adhesives inspired by loose connective tissue, plant cell walls, and mussel foot proteins.

Utilizing lignin as a complete substitute for petroleum-derived phenol to synthesize non-toxic lignin-based resin adhesives with bio-based aldehydes holds substantial significance for advancing sustainable development in the wood-based panel industry. Nevertheless, the poor bonding strength, toughness, and water resistance of phenol-free lignin-based resins hinder their practical application. Herein, inspired by the hyperbranched structures of loose connective tissue, borate chemistry of plant cell walls, and Fe3+-phenol coordination bonds of mussel foot proteins, a tough, strong, water-resist

Screening of Promising Chemotherapeutic Candidates from Plants against Human Adult T-Cell Leukemia/Lymphoma (VII): Active Principles from Thuja occidentalis L.

During the screening of novel chemotherapeutic candidates from plants against adult T-cell leukemia/lymphoma, we identified that the extracts of Thuja occidentalis (Cupressaceae) showed potent anti-proliferative activity in MT-1 and MT-2 cells. Therefore, we attempted to isolate the active components from this plant. We isolated and identified 32 compounds (1-32; eight lignans, 18 terpenoids, and six flavonoids) from the extracts of the leaves and cones. Their structures were determined by spectroscopic analysis. Several of the isolated compounds inhibited the growth of both cell lines. Lignan

Plasmodium falciparum raf kinase inhibitor is a lipid binding protein that interacts with and regulates the activity of PfCDPK1, an essential plant-like kinase required for red blood cell invasion.

Raf Kinase Inhibitor Protein (RKIP) is an important regulator of the MAPK signaling pathway in multicellular eukaryotes. Plasmodium falciparum RKIP (PfRKIP) is a putative phosphatidylethanolamine binding protein (PEBP) that shares limited similarity with Homo sapiens RKIP (HsRKIP). Interestingly, critical components of the MAPK pathway are not expressed in malaria parasites and the physiological function of PfRKIP remains unknown. PfRKIP is expressed throughout the asexual schizogony with maximum expression in late schizonts. Interestingly, PfRKIP and HsRKIP show pH-dependent differential inte

Simulated acid rain (SAR) induced alterations in plant morpho-physiology, leaf ultrastructure, cell viability, and yield - A case study on Mentha arvensis L. (Wild mint).

Acid rain (AR), a secondary pollutant, a product of sulfur and nitrogen oxides fusion with the atmosphere's water vapor, comes on earth in the form of rain, poses a significant environmental threat, and affects soil characteristics and plant morphology, ultrastructure, physiology, and yield. However, its effects on aromatic and medicinal plants remain insufficiently studied. Due to their low side effects and high efficacy, the use of herbal plants for disease remedy preparation is found globally and their consumption increases year after year. Mentha arvensis is such an aromatic crop, having g

Plant-based whole-food diets are feasible during autologous stem cell transplantation and are associated with dose-dependent microbiome modulation: Results from a pilot clinical trial.

Plant-based dietary strategies may offer a tractable approach to mitigating microbiome disruption and improving outcomes in patients undergoing autologous hematopoietic cell transplantation (auto-HCT) for multiple myeloma, a population in whom intestinal dysbiosis has been linked to infectious complications and inferior survival. We conducted a single-arm study to test the feasibility and biological activity of a high-fiber, plant-based, whole-food meal delivery intervention during the peri-transplant period. Adults with multiple myeloma (n = 22) received fully prepared, plant-based meals for

PaDef (Persea americana var. drymifolia), a Plant Antimicrobial Peptide, Triggers Apoptosis, and Induces Global Epigenetic Modifications on Histone 3 in an Acute Lymphoid Leukemia Cell Line.

In recent years, it has been recognized that epigenetic alterations play an important role in the development and maintenance of cancer, including leukemias. Furthermore, it is known that these alterations are involved in the emergence of resistance to conventional chemotherapeutics. Consequently, molecules with an anticancer activity whose activity is ruled by epigenetic modifications are attractive to search for new therapies against cancer. The plant antimicrobial peptides have been widely evaluated as molecules with anticancer activity; however, the analysis of the epigenetic regulation in

Enhancement of glutathione and g-glutamylcysteine synthetase, the rate limiting enzyme of glutathione synthesis, by chemoprotective plant-derived food and beverage components in the human hepatoma cell line HepG2.

Glutathione (GSH) is an important antioxidant and cofactor of detoxifying metabolism. Therefore, elevation of GSH as achieved by inducing g-glutamylcysteine synthetase (GCS), the limiting enzyme of GSH synthesis, may contribute to chemoprevention against cancer. In previous animal studies, increases in GCS were mainly found in liver and other organs that are not easily accessible in humans. Thus, employment and evaluation of alternative systems such as human-derived cell lines are encouraged. In the present experiment, we used the hepatoma cell line HepG2 to investigate the response of GCS and

Enterolobium contortisiliquum trypsin inhibitor (EcTI), a plant proteinase inhibitor, decreases in vitro cell adhesion and invasion by inhibition of Src protein-focal adhesion kinase (FAK) signaling pathways.

Tumor cell invasion is vital for cancer progression and metastasis. Adhesion, migration, and degradation of the extracellular matrix are important events involved in the establishment of cancer cells at a new site, and therefore molecular targets are sought to inhibit such processes. The effect of a plant proteinase inhibitor, Enterolobium contortisiliquum trypsin inhibitor (EcTI), on the adhesion, migration, and invasion of gastric cancer cells was the focus of this study. EcTI showed no effect on the proliferation of gastric cancer cells or fibroblasts but inhibited the adhesion, migration,
