Age-Related Inflammation and Oxidative Stress in the Cochlea Are Exacerbated by Long-Term, Short-Duration Noise Stimulation.

We have previously reported that young adult rats exposed to daily, short-duration noise for extended time periods, develop accelerated presbycusis starting at 6 months of age. Auditory aging is associated with progressive hearing loss, cell deterioration, dysregulation of the antioxidant defense system, and chronic inflammation, among others. To further characterize cellular and molecular mechanisms at the crossroads between noise and age-related hearing loss (ARHL), 3-month-old rats were exposed to a noise-accelerated presbycusis (NAP) protocol and tested at 6 and 16 months of age, using aud…

Serum homocysteine and folate concentrations are associated with prevalent age-related hearing loss.

Elevated total serum homocysteine (tHcy) concentrations associated with vitamin B-12 or folate deficiencies may adversely affect blood flow to the cochlea, leading to age-related hearing loss (presbycusis). However, only 2 small cross-sectional studies have assessed the link between folate, vitamin B-12, or tHcy and presbycusis. We aimed to determine both the cross-sectional and longitudinal association between serum concentrations of folate, vitamin B-12, or tHcy and risk of age-related hearing loss. The Blue Mountains Hearing Study is a population-based survey of age-related hearing loss (19…

Anti-epileptic drugs delay age-related loss of spiral ganglion neurons via T-type calcium channel.

Loss of spiral ganglion neurons is a major cause of age-related hearing loss (presbycusis). Despite being the third most prevalent condition afflicting elderly persons, there are no known medications to prevent presbycusis. Because calcium signaling has long been implicated in age-related neuronal death, we investigated T-type calcium channels. This family is comprised of three members (Ca(v)3.1, Ca(v)3.2, and Ca(v)3.3), based on their respective main pore-forming alpha subunits: α1G, α1H, and α1I. In the present study, we report a significant delay of age-related loss of cochlear function and…

Sound lateralization and speech discrimination in patients with sensorineural hearing loss.

Sound lateralization and speech discrimination abilities are both closely related to central auditory system function. In this study, we investigated the relationship between sound lateralization and speech discrimination in patients with sensorineural hearing loss (SNHL) of unknown aetiology or presbycusis. Interaural intensity difference (IID) and interaural time difference (ITD) discriminations were measured using a self-recording apparatus for dichotic sound presentation, and the means of bilateral maximum scores in speech discrimination tests were calculated. Subjects with normal sound la…

The influence of aging on auditory brainstem response and electrocochleography in the elderly.

Auditory brainstem responses and simultaneous transtympanic electrocochleographies were investigated in 92 subjects aged between 50 and 89 years with normal hearing or mild presbycusis. Subjects were classified into four age-related groups: 16 subjects in the sixth decade, 28 in the seventh decade, 32 in the eighth decade and 16 in the ninth decade. Thirty young subjects with normal hearing aged 20-29 years were chosen as controls. The result showed progressive delay of wave I (or action potential: AP), wave III and wave V and lengthening of the interpeak intervals (IPIs) of waves III-V and I-…

The cognitive cost of age-related hearing loss.

BACKGROUND: Age-related hearing loss (ARHL) or presbycusis is a bilateral sensorineural hearing impairment associated with aging of the structures of the inner ear and auditory pathways and is the most common cause of acquired hearing loss in older adults. Presbycusis is a disabling condition that affects communication and quality of life, and has been associated with poorer cognitive performance. This study aimed to examine the association between ARHL, cognitive screening performance, and hearing-related quality of life in older adults. METHODS: This single-center prospective observational …

Reduced expression of Connexin26 and its DNA promoter hypermethylation in the inner ear of mimetic aging rats induced by d-galactose.

Connexin26 (Cx26), one of the major protein subunits forming gap junctions (GJs), is important in maintaining homeostasis in the inner ear and normal hearing. Cx26 mutation is one of the most common causes for inherited nonsyndromic deafness, but the relationship between Cx26 and presbycusis is unknown. Our study aimed at exploring the expression and the aberrant methylation of the promoter region of Cx26 gene in the cochlea of inner ear mimetic aging rats. We applied a mimetic aging of inner ear rat model with mtDNA common deletion by d-gal injection for 8weeks. Real-time RT-PCR and Western b…

[Difficulties understanding compressed speech in elderly listeners with mild-to-moderate hearing loss].

OBJECTIVE: In this work we evaluated the difficulties in understanding rapid speech (normal, fast and very fast rates) in elderly listeners with and without hearing loss (presbycusis and moderate hearing loss). Rapid speech is common in daily communication, yet few studies have been conducted to assess this problem in Spanish-speaking listeners, as has been done for English speakers. MATERIAL AND METHOD: We compared the recognition of sentences presented at normal, fast, and very fast speech rates in 3 groups of listeners. Recognition scores were correlated to their audiological measures and …

Estrogen and the inner ear: megalin knockout mice suffer progressive hearing loss.

Megalin, the largest member of the low-density lipoprotein receptor-related protein family, functions as an endocytic receptor for a variety of essential lipophilic metabolites, including the steroid hormone estrogen. In the cochlea, megalin is strongly expressed within the marginal cells of the stria vascularis, and previous studies demonstrated that beta-estrogen receptors are also expressed in megalin-expressing marginal cells. In the present study, we demonstrate that homozygous megalin mutant mice exhibit profound hearing loss at 3 months of age associated with features of presbycusis, en…

How do aging and age-related hearing loss affect the ability to communicate effectively in challenging communicative conditions?

This study investigated the relation between the intelligibility of conversational and clear speech produced by older and younger adults and (a) the acoustic profile of their speech (b) communication effectiveness. Speech samples from 30 talkers from the elderLUCID corpus were used: 10 young adults (YA), 10 older adults with normal hearing (OANH) and 10 older adults with presbycusis (OAHL). Samples were extracted from recordings made while participants completed a problem-solving cooperative task (diapix) with a conversational partner who could either hear them easily (NORM) or via a simulated…

Age-related hearing loss: GABA, nicotinic acetylcholine and NMDA receptor expression changes in spiral ganglion neurons of the mouse.

Age-related hearing loss - presbycusis - is the number one communication disorder and most prevalent neurodegenerative condition of our aged population. Although speech understanding in background noise is quite difficult for those with presbycusis, there are currently no biomedical treatments to prevent, delay or reverse this condition. A better understanding of the cochlear mechanisms underlying presbycusis will help lead to future treatments. Objectives of the present study were to investigate GABAA receptor subunit α1, nicotinic acetylcholine (nACh) receptor subunit β2, and N-methyl-d-aspa…

Changes of ribbon synapses number of cochlear hair cells in C57BL/6J mice with age(Δ).

To investigate the changes of ribbon synapses (RS) number in cochlear hair cells in C57BL/6J mice with age. Basilar membranes within the coc … l turn increased slightly. Reduction of RS probably takes place in the early stage of C57BL/6J mice presbycusis. Early prevention of presbycusis can be achieved through measures to mitigate the reduction of RS.

Case history: asking the right questions.

The importance of asking the right question is highlighted in this case study. The patient was a 76-yr-old man with hearing loss that initially appeared to be a classic case of presbycusis. Without asking the right question this patient might have been managed in a manner inappropriate to his true hearing sensitivity.

Autophagy regulates the degeneration of the auditory cortex through the AMPK-mTOR-ULK1 signaling pathway.

Presbycusis is the most common sensory impairment associated with aging; however, the underlying molecular mechanism remains unclear. Autophagy has been demonstrated to serve a key role in diverse diseases; however, no studies have examined its function in central presbycusis. The aim of the present study was to investigate the changes of autophagy in the physiological processes of the auditory cortex and its role in the degeneration of the auditory cortex, as well as the related mechanisms using naturally aging rats and a D‑galactose (D‑gal)‑induced mimetic rat model of aging. The present stu…

Identification of Hub Genes and Molecular Pathways in Age-Related Hearing Loss: An Integrated Bioinformatics and Machine Learning Approach.

BACKGROUND: Age-related hearing loss (ARHL), or presbycusis, is a prevalent sensory impairment in the elderly, driven by multifactorial degenerative changes in cochlear structures such as the inner and outer hair cells, stria vascularis, and auditory nerve. Despite its significant impact on quality of life, including associations with social isolation, depression, and cognitive decline, the molecular mechanisms underlying ARHL remain incompletely understood. This study leverages bioinformatics and machine learning to identify hub genes and pathways from gene expression data, aiming to uncover …

Linking anatomical and physiological markers of auditory system degeneration with behavioral hearing assessments in a mouse (Mus musculus) model of age-related hearing loss.

Age-related hearing loss is a very common sensory disability, affecting one in three older adults. Establishing a link between anatomical, physiological, and behavioral markers of presbycusis in a mouse model can improve the understanding of this disorder in humans. We measured age-related hearing loss for a variety of acoustic signals in quiet and noisy environments using an operant conditioning procedure and investigated the status of peripheral structures in CBA/CaJ mice. Mice showed the greatest degree of hearing loss in the last third of their lifespan, with higher thresholds in noisy tha…
