Peptidomic Characterization and Amino Acid Availability after Intake of Casein vs. a Casein Hydrolysate in a Pig Model.

It is known that casein hydrolysis accelerates gastrointestinal transit in comparison to intact casein, although the effect of the protein hydrolysis on the composition of the digests is not fully understood. The aim of this work is to characterize, at the peptidome level, duodenal digests from pigs, as a model of human digestion, fed with micellar casein and a previously described casein hydrolysate. In addition, in parallel experiments, plasma amino acid levels were quantified. A slower transit of nitrogen to the duodenum was found when the animals received micellar casein. Duodenal digests

Decode the specific flavor and spectral fingerprint of the proteolytic products of milk-derived defatted high protein.

Proteolysis is crucial for generating functional ingredients that enhance milk quality, yet the associated odor and taste profiles of the proteolytic hydrolysates remain poorly characterized. This study utilized a multidimensional approach to analyze the volatile compounds, the peptidome, and spectroscopic properties of defatted high-protein enzymatic hydrolysates under varying conditions. Sensory evaluation integrated with facial expression was applied to comprehensively assess flavor profiles across different enzymatic hydrolysis treatments. Results showed significant differences between enz

Continuous production of velvet bean-based bioactive peptides in membrane reactor with dual enzyme system.

One of the main challenges hindering the commercialization of bioactive peptides is the lack of scalable and consistent production methods. To overcome this obstacle, an automated enzyme membrane reactor was used to continuously produce bioactive peptides from velvet bean (Mucuna pruriens). The optimum operating conditions were [E]/[S] = 5%, pH = 7.5, and τ = 12 h. The long-term continuous operation of the EMR system demonstrated its ability to maintain steady-state conditions. To minimize membrane fouling, an industrially viable strategy was employed, which combines operation at threshold flu

Amino acid requirements in the early post-implantation rat conceptus.

Amino acid analysis has been performed on hydrolysates of embryos/fetuses, visceral yolk sacs and ectoplacental cones/placentae from early post-implantation rat conceptuses. The increments in each amino acid between 10.5 and 11.5 days, between 11.5 and 12.5 days, and between 12.5 and 13.5 days, are expressed as percentages of the total amino acid increment. These three profiles are very similar to each other and also strongly resemble the amino acid composition of hydrolysates of rat serum. The results are discussed in the context of the hypothesis that a transudate of plasma is responsible fo

[Do breast-feeding and "diet" milks have any preventive or curative effect in the management of atopic dermatitis in children?].

A) The preventive interest of infants' food in the onset of atopic dermatitis. Measures of prevention of atopic dermatitis concern predisposed children. Most studies agree on the protective effect of breast feeding for at least 3 to 4 months, compared with industrial milk products, on the onset of atopic dermatitis. Partial breast feeding is not protective. There is no preventive effect of breast feeding on the onset of atopic dermatitis in the absence of a family history of atopy. However, a few studies have raised the question of the aggravation of eczema during prolonged breast feeding. The

Identification of Angiotensin I-Converting Enzyme-Inhibitory and Anticoagulant Peptides from Enzymatic Hydrolysates of Chicken Combs and Wattles.

Peptides from protein hydrolysate of a mixture of chicken combs and wattles (CCWs) were obtained through enzymatic hydrolysis, and their anticoagulant and inhibitory effects on angiotensin I-converting enzyme (ACE) were investigated. The protein hydrolysate exhibited anticoagulant capacity by the intrinsic pathway (activated partial thromboplastin time) and potent ACE-inhibitory activity. The peptides were sequenced by LC-MS to identify those with higher inhibitory potential. From the pool of sequenced peptides, the following three peptides were selected and synthesized based on their low mole

Effects of bromelain-induced protein hydrolysis of mealworm (Tenebrio molitor larva) flour on protein characteristics, proximate composition, and glycemic response of crackers.

Diabetes management requires effective dietary strategies. Mealworm (Tenebrio molitor larva) is a sustainable protein source, but its digestibility is limited by chitin and complex structures. Bromelain enzymatic hydrolysis can improve its digestibility for functional food applications like crackers. This study aimed to develop functional crackers substituted with mealworm protein hydrolysate and evaluate their protein characteristics, proximate composition, and glycemic response. To achieve this, mealworm flour was enzymatically treated with bromelain, and the optimal hydrolysate was used to

The development of a novel ferric phytate compound for iron fortification of bouillons (part I).

In a series of two studies, we report the development (this study) and evaluation (part II) of a novel ferric phytate compound designed as a condiment iron fortificant. Condiments are used as iron fortification vehicles to reduce the prevalence  of iron deficiency. The challenge for iron fortificants in e.g. a bouillon matrix is to avoid undesired sensory effects and to ensure a reasonable cost. We added phytic acid to chelate iron, and hydrolysed protein to counteract the inhibiting effect of phytic acid on iron bioaccessibility. We characterised four novel ferric phytate compounds, destabili

Exploration of Potentially Bioactive Peptides Generated from the Enzymatic Hydrolysis of Hempseed Proteins.

The seed of industrial hemp is an underexploited protein source. In view of a possible use in functional foods, a hempseed protein concentrate was hydrolyzed with pepsin, trypsin, pancreatin, or a mixture of these enzymes. A detailed peptidomic analysis using data-dependent acquisition showed that the numbers of peptides identified ranged from 90 belonging to 33 parent proteins in the peptic hydrolysate to 9 belonging to 6 proteins in the pancreatin digest. The peptic and tryptic hydrolysates resulted to be the most efficient inhibitors of 3-hydroxymethyl-coenzyme A reductase activity when tes

A metabolomics approach to commercial protease enzyme hydrolysis of gourami meat.

The enzymatic hydrolysis of fish proteins has gained increasing attention to enhance nutritional quality and generate bioactive peptides with functional properties. In this study, we investigated the metabolomic consequences of gourami (Osphronemus goramy) meat hydrolysis using two commercial proteases, papain and bromelain. Hydrolysis was conducted over 0-240 min, and the resulting metabolites were profiled using ultra-performance liquid chromatography coupled with high-resolution mass spectrometry (UPLC-HRMS). Multivariate statistical analyses revealed a clear separation of hydrolysates by e

Utilizing brewer's spent grain as a sustainable antioxidant peptides source to enhance yeast tolerance under multiple stresses.

This study explored utilizing nitrogen sources derived from brewer's spent grain (BSG) to enhance brewer's yeast tolerance, with the aim of reducing carbon emissions in breweries and promoting BSG's value-added applications. Results demonstrated that preculture with BSG protein hydrolysates (BSGPH) and purified antioxidant peptides improved yeast tolerance to ethanol, oxidative and osmotic stresses. Specifically, both the ethanol precipitated hydrolysates (BH-95S) and its chromatographically separated G15-B fraction, exhibited superior biological activity in improving yeast physiological prope

Milk Protein-Derived Antioxidant Tetrapeptides as Potential Hypopigmenting Agents.

Excessive accumulation of melanin can cause skin pigmentation disorders, which may be accompanied by significant psychological stress. Although many natural and synthetic products have been developed for the regulation of melanogenesis biochemistry, the management of unwanted skin pigmentation remains challenging. Herein, we investigated the potential hypopigmenting properties of peptide sequences that originated from milk proteins such as ĸ-casein and β-lactoglobulin. These proteins are known to inhibit melanogenesis and their hydrolysates are reported as antioxidant peptides. We synthesize t

Electro-activation of sweet defatted whey: Impact on the induced Maillard reaction products and bioactive peptides.

Electro-activation was used to add value to sweet defatted whey. This study aimed to investigate and to characterize the bioactive compounds formed under different electro-activation conditions by molecular and proteomic approaches. The effects of electric current intensity (400, 500 or 600mA) and whey concentration (7, 14 or 21% (w/v)) as a function of the electro-activation time (0, 15, 30 or 45min) were evaluated. The targeted dependent variables were the formation of Maillard reaction products (MRPs), protein hydrolysates and glycated compounds. It was shown that the MRPs derived from elec

Generation of a mutagenized organophosphorus hydrolase for the biodegradation of the organophosphate pesticides malathion and demeton-S.

AIMS: The bacterial organophosphorus hydrolase (OPH) enzyme hydrolyses and detoxifies a broad range of toxic organophosphate pesticides and warfare nerve agents by cleaving the various phosphorus-ester bonds (P-O, P-F, P-CN, P-S); however, OPH hydrolyses these bonds with varying efficiencies. The aim of this study was to generate a variant OPH enzyme with improved hydrolytic efficiency against the poorly hydrolysed P-S class of organophosphates. METHODS AND RESULTS: The gene encoding OPH was sequentially mutated at specific codons by saturation mutagenesis and screened for improved activity a

Genetically Engineering Escherichia coli to Produce Xylitol from Corncob Hydrolysate without Lime Detoxification.

Before fermentation with hemicellulosic hydrolysate as a substrate, it is generally necessary to detoxify the toxic substances that are harmful to microorganism growth. Cyclic AMP receptor protein (CRP) is a global regulator, and mutation of its key sites may have an important impact on E. coli virulence tolerance. Using corncob hydrolysate without ion-exchange or lime detoxification as the substrate, shake flask fermentation experiments showed that CRP mutant IS5-dG (I112L, T127G, A144T) produced 18.4 g/L of xylitol within 34 h, and the OD600 was 9.7 at 24 h; these values were 41.5% and 21.3%

Tracking Gluten throughout Brewing Using N-Terminal Labeling Mass Spectrometry.

Gluten-containing grains cause adverse health effects in individuals with celiac disease. Fermentation of these grains results in gluten-derived polypeptides with largely uncharacterized sizes and sequences, which may still trigger an immune response. This research used N-terminal labeling mass spectrometry to characterize protein hydrolysates during each stage of bench-scale brewing, including malting, mashing, boiling, fermentation, and aging. Gluten hydrolysates from each brewing step were tracked, and the immunotoxic potential was evaluated by sequence comparison with peptides known to sti

Preparation, characterization and in vitro stability of iron-chelating peptides from mung beans.

Mung bean protein was enzymatically hydrolyzed with either alcalase, neutral protease, or papain. The mung bean protein hydrolysates (MPH) showed good ability to chelate ferrous ions, and the chelates had high stability in vitro. The hydrolysates prepared by alcalase showed the highest degree of hydrolysis and the highest ferrous chelating rate. Single factor tests showed that the pH and the material ratio had significant effects on ferrous chelating rates. The optimal MPH to FeCl2·4H2O material ratio was 8:1 (w/w) and the optimal pH of the reaction was 7.0, which yielded a chelating rate of 9
