Identification of sulfenylation patterns in trophozoite stage Plasmodium falciparum using a non-dimedone based probe.

Plasmodium falciparum causes the deadliest form of malaria. Adequate redox control is crucial for this protozoan parasite to overcome oxidative and nitrosative challenges, thus enabling its survival. Sulfenylation is an oxidative post-translational modification, which acts as a molecular on/off switch, regulating protein activity. To obtain a better understanding of which proteins are redox regulated in malaria parasites, we established an optimized affinity capture protocol coupled with mass spectrometry analysis for identification of in vivo sulfenylated proteins. The non-dimedone based prob

Palmitoylated Proteins in Plasmodium falciparum-Infected Erythrocytes: Investigation with Click Chemistry and Metabolic Labeling.

The examination of the complex cell biology of the human malaria parasite Plasmodium falciparum usually relies on the time-consuming generation of transgenic parasites. Here, metabolic labeling and click chemistry are employed as a fast transfection-independent method for the microscopic examination of protein S-palmitoylation, an important post-translational modification during the asexual intraerythrocytic replication of P. falciparum. Applying various microscopy approaches such as confocal, single-molecule switching, and electron microscopy, differences in the extent of labeling within the

The enzymatic mechanisms of lactylation and its role in gynecological diseases: a comprehensive review.

Lysine lactylation (Kla) is a novel form of post-translational modification. It utilizes lactate as its core substrate. Through an enzymatic regulatory network, it mediates modifications of both histones and non-histone proteins. This modification is thought to regulate gene transcription, reprogramming cellular metabolism, and remodeling the immune microenvironment. Studies on lactylation in gynecological diseases have progressively advanced. Its dysregulation appears to be associated with the pathogenesis of multiple conditions, including gynecological cancers, endometriosis, premature ovari

Base-modified factor VIII mRNA delivery with galactosylated lipid nanoparticles as a protein replacement therapy for haemophilia A.

The bleeding disorder hemophilia A (HemA) requires systemic functional factor VIII protein infusions on prophylactic schedules. Recently, chemically modified mRNAs have emerged as promising protein replacement therapies to reduce repeated infusions and improve safety profiles. However, the influence of base modifications on mRNA translation kinetics to specific cell types remains unexplored. In this study, towards developing mRNA therapeutics for haemophilia A, we synthesized chemically modified mRNAs with commercially available base modifications of adenine, guanine, uridine, and cytidine, an

Clinical Significance of Mucin Signatures in Inflammatory Bowel Diseases: A Systematic Review of Their Expression Patterns, Polymorphisms, and Post-translational Modifications.

BACKGROUND: The intestinal mucosal barrier plays an important role in the pathophysiology of inflammatory bowel disease (IBD), with mucins being key components of this barrier. Classified as either transmembrane or secreted, these highly glycosylated proteins protect the mucosal barrier while also influencing barrier integrity. Given their indispensable role in maintaining the intestinal mucosal barrier, mucins are compelling candidates for the evaluation of barrier dysfunction. Numerous studies have investigated mucins in the context of IBD, but a clear consensus regarding their expression, p

β-Hydroxybutyrate suppresses M1 macrophage polarization through β-hydroxybutyrylation of the STAT1 protein.

β-Hydroxybutyrate (β-OHB), the primary ketone body, is a bioactive metabolite that acts as both an energy substrate and a signaling molecule. Recent studies found that β-OHB inhibits the production of pro-inflammatory cytokines in macrophages, but its underlying molecular mechanisms have not yet been fully elucidated. Lysine β-hydroxybutyrylation (Kbhb), a post-translational modification mediated by β-OHB, plays a key role in regulating the expression and activity of modified proteins. However, whether macrophages undergo protein Kbhb and whether Kbhb modification regulates macrophage polariza

SUMOylation and anticancer drug resistance.

Therapeutic resistance is a major obstacle in cancer treatment, often leading to recurrence and death. SUMOylation is a dynamic and reversible process of post-translational modification through a strict enzyme cascade that regulates the modification of target proteins by SUMO1-5 and SENP family proteins, which are involved in regulating protein stability, function, and localization to influence biological pathways, including cancers. SUMOylation contributes significantly to cancer drug resistance, undermining the efficacy of clinical treatment. Therefore, studying the role of SUMOylation in th

[Ubiquitination modification of NLRP3 inflammasome and its role in inflammatory bowel disease].

Inflammatory bowel disease (IBD) belongs to a group of idiopathic intestinal inflammatory disorders that affect the ileum, rectum, and colon. There is still no effective treatment methods, and with the increasing number of patients, IBD has brought a heavy burden to the medical system. Chronic inflammation mediated by abnormal activation of the intestinal mucosal immune system plays an important role in the occurrence and development of IBD. As a common intracellular sensor that activates the innate immune system, inflammasomes have a signaling cascade regulated by ubiquitination/deubiquitinat

Functional S-persulfidation exploration reveals a novel regulatory mechanism of HNF1α.

Protein S-persulfidation, a post-translational modification mediated by hydrogen sulfide (H2S), plays an essential role in regulating protein function. However, current profiling methods, mainly based on liquid chromatography-tandem mass spectrometry (LC-MS/MS), tend to detect abundant proteins and often miss low-abundance but important regulators like transcription factors (TFs). To better understand the S-persulfidation landscape, we integrated datasets from three species-Homo sapiens, Mus musculus, and Arabidopsis thaliana-and found that TFs are strikingly underrepresented. To address this

SUMOylation modulates mitochondrial dynamics in an in vitro rotenone model of Parkinson's disease.

SUMOylation is a post-translational modification essential for various biological processes. SUMO proteins bind to target substrates in a three-step enzymatic pathway, which is rapidly reversible by the action of specific proteases, known as SENPs. Studies have shown that SUMOylation is dysregulated in several human disorders, including neurodegenerative diseases that are characterized by the progressive loss of neurons, mitochondrial dysfunction, deficits in autophagy, and oxidative stress. Considering the potential neuroprotective roles of SUMOylation, the aim of this study was to investigat

Protein Persulfidation in Plants: Function and Mechanism.

As an endogenous gaseous transmitter, the function of hydrogen sulfide (H2S) has been extensively studied in plants. Once synthesized, H2S may be involved in almost all life processes of plants. Among them, a key route for H2S bioactivity occurs via protein persulfidation, in which process oxidizes cysteine thiol (R-SH) groups into persulfide (R-SSH) groups. This process is thought to underpin a myriad of cellular processes in plants linked to growth, development, stress responses, and phytohormone signaling. Multiple lines of emerging evidence suggest that this redox-based reversible post-tra

TRPC5 channel instability induced by depalmitoylation protects striatal neurons against oxidative stress in Huntington's disease.

Protein S-palmitoylation, the covalent lipid modification of the side chain of Cys residues with the 16‑carbon fatty acid palmitate, is the most common acylation, and it enhances the membrane stability of ion channels. This post-translational modification (PTM) determines a functional mechanism of ion channel life cycle from maturation and membrane trafficking to localization. Especially, neurodevelopment is regulated by balancing the level of synaptic protein palmitoylation/depalmitoylation. Recently, we revealed the pathological role of the transient receptor potential canonical type 5 (TRPC

RNA modifications and cancer.

RNA plays essential roles in not only translating nucleic acids into proteins, but also in gene regulation, environmental interactions and many human diseases. Nature uses over 150 chemical modifications to decorate RNA and diversify its functions. With the fast-growing RNA research in the burgeoning field of 'epitranscriptome', a term describes post-transcriptional RNA modifications that can dynamically change the transcriptome, it becomes clear that these modifications participate in modulating gene expression and controlling the cell fate, thereby igniting the new interests in RNA-based dru

The Expanding Landscape of the Thiol Redox Proteome.

Cysteine occupies a unique place in protein chemistry. The nucleophilic thiol group allows cysteine to undergo a broad range of redox modifications beyond classical thiol-disulfide redox equilibria, including S-sulfenylation (-SOH), S-sulfinylation (-SO(2)H), S-sulfonylation (-SO(3)H), S-nitrosylation (-SNO), S-sulfhydration (-SSH), S-glutathionylation (-SSG), and others. Emerging evidence suggests that these post-translational modifications (PTM) are important in cellular redox regulation and protection against oxidative damage. Identification of protein targets of thiol redox modifications i

Kinases Associated with Herpes Zoster Virus Infection Unveiled by Phosphoromics Profiling.

INTRODUCTION: Reactivation of varicella-zoster virus (VZV) causes herpes zoster (HZ) in humans and elicits a VZV-specific immune response. However, the effect of VZV on host protein post-translational modifications (PTMs) remains largely unknown. OBJECTIVE: In this study, we investigated global changes in phosphorylation levels in HZ patients with postherpetic neuralgia (PHN) compared to healthy controls. METHODS: Using publicly available datasets, we found that the serine/threonine protein kinase and phosphatase pathways are significantly regulated by VZV infection, suggesting that VZV infe

Dynamic regulation of RNA m6A methylation in the ribonucleoprotein complexes in apicomplexan and kinetoplastid parasites.

m6A, or N6-methyladenosine, is the most abundant modification of mRNA transcripts. These modifications are known to influence mRNA transcript stability, transcription, translation, alternative splicing and decay, with undoubtedly more functions to be discovered. In this review, we explored the roles of m6A modifications in two groups of protozoan parasites: the apicomplexans, which include Plasmodium and Toxoplasma species, and kinetoplastids, which include Trypanosoma and Leishmania species. We also compared the key players of m6A epitranscriptomic machinery in creating, interpreting, and rem

Modulation of Rab GTPase function by a protein phosphocholine transferase.

The intracellular pathogen Legionella pneumophila modulates the activity of host GTPases to direct the transport and assembly of the membrane-bound compartment in which it resides. In vitro studies have indicated that the Legionella protein DrrA post-translationally modifies the GTPase Rab1 by a process called AMPylation. Here we used mass spectrometry to investigate post-translational modifications to Rab1 that occur during infection of host cells by Legionella. Consistent with in vitro studies, DrrA-mediated AMPylation of a conserved tyrosine residue in the switch II region of Rab1 was detec

Deep coverage and quantification of the bone proteome provides enhanced opportunities for new discoveries in skeletal biology and disease.

Dysregulation of cell signaling in chondrocytes and in bone cells, such as osteocytes, osteoblasts, osteoclasts, and an elevated burden of senescent cells in cartilage and bone, are implicated in osteoarthritis (OA). Mass spectrometric analyses provides a crucial molecular tool-kit to understand complex signaling relationships in age-related diseases, such as OA. Here we introduce a novel mass spectrometric workflow to promote proteomic studies of bone. This workflow uses highly specialized steps, including extensive overnight demineralization, pulverization, and incubation for 72 h in 6 M gua

Glycosylation in malaria parasites: what do we know?

In malaria parasites, although post-translational modification of proteins with N-. O-, and C-glycosidic bond-linked glycans is limited, it is confined to relatively fewer proteins in which the glycans are present at significant levels and may have important functions. Furthermore, several proteins are modified with glycosylphosphatidylinositols (GPIs) which represent the predominant glycan synthesized by parasites. Modification of proteins with GPIs is obligatory for parasite survival as GPI-anchored proteins (GPI-APs) play essential roles in all life cycle stages of the parasites, including

It takes two to tango: the intersection of deubiquitinases and m6A modification in cancer.

The dynamic and reversible N6-methyladenosine (m6A) RNA modification has emerged as a pivotal layer of epitranscriptomic regulation in cancer, governing RNA metabolism and function. Concurrently, deubiquitylating enzymes (DUBs), key components of the post-translational modifications (PTMs) system, orchestrate protein stability and signaling pathways crucial for tumorigenesis. Recent evidence reveals extensive and intricate crosstalk between these two regulatory systems, forming a complex network that can influence cancer progression. This review systematically explores the bidirectional interp
