Since the discovery of x-rays, many investigations of radiation effects on humans have taken place. From the discovery of severe x-ray dermatitis near the end of 1896, the multiple radiation burns endured by Thomas Edison's assistant, Clarence Dally, to the establishment of early radiation regulation in 1929, radiation exposure has been a continuing concern. Studies from the 1940s to the 1960s looked at effects such as leukemia and other cancers that appeared to be caused by exposure to radiation; outcomes of these studies resulted in increased regulation, establishment of dose tolerances and

Childhood radiation exposure is a known thyroid cancer risk factor. This study evaluated the effects of age on radiation-induced thyroid carcinogenesis in rats irradiated with 8 Gy X-rays. We analyzed cell proliferation, cell death, DNA damage response, and autophagy-related markers in 4-week-old (4W) and 7-month-old (7M) rats and the incidence of thyroid tumors in 4W, 4-month-old (4M), and 7M rats 18 months after irradiation. Cell death and DNA damage response were increased in 4W rats compared to those in controls at 1 month post-irradiation. More Ki-67-positive cells were observed in 4W rat

BACKGROUND: Malignant pleural mesothelioma is a rare disease with poor prognosis. Surgery is used as a radical treatment modality but the risk of local relapse is very high. Therefore, radiation therapy is used in postoperative setting to improve local control. However, owing to elliptical shape, treating the entire pleura requires a large radiation field which increases toxicity. Precision radiation is mandatory to optimally irradiate the tumor area while sparing critical neighboring normal organs.

Ultraviolet-B radiation has a dual effect on plants, acting both as a developmental cue and a source of genetic stress. Plants must balance growth, DNA repair, cell division, and damage mitigation. Oxidation Resistance (OXR) proteins are evolutionarily conserved, TLDc-domain-containing mitochondrial proteins that link redox homeostasis to nuclear stress responses, which makes them strong candidates to coordinate the plant response to UV-B. This study highlights that the Arabidopsis protein AtOXR2 and its sunflower orthologue, HaOXR2, are critical for enhancing plant tolerance to UV-B radiation

Chenopodium quinoa Willd. is a betalainic crop with remarkable tolerance to extreme environmental conditions. Despite numerous varieties grow at high altitudes, where UV-B radiation is intense, research on the effects of UV-B stress on this and related species is very scarce. In the present work we aimed to determine whether UV-B radiation induces the production of betalains, and evaluated the role of these pigments and other key antioxidants in preventing oxidative damage, in seedlings of C. quinoa (ecotypes CICA and Villarrica) and its close relative C. berlandieri Moq., grown in darkness an

The lung cancer incidence and mortality rates have risen steadily each year. Survival rates during the same period have improved marginally even by the best treatment approaches. A small proportion of patients with limited disease can be offered surgical resection as the primary curative treatment. In a significant number of cases, complete resection of cancer is not possible either because the patient could not tolerate radical surgical approach or because the tumor is attached to important structures in the mediastinum precluding any surgical intervention. Intraoperative brachytherapy by var

Tolerance to major histocompatibility antigens as well as the ability to mount a cytotoxic response to hapten-modified cells of bone marrow donor and host origin was studied in allogeneic radiation chimeras. Lethally irradiated (C57BL/6 X DBA/2)F1 hosts reconstituted with anti-Thy 1.2 + C-treated bone marrow from (C57BL/6 X CBA)F1 mice showed tolerance to the MHC antigens of the three parental strains as measured by MLC and CML assay. The chimeras responded normally to unrelated allogeneic cells. Chimeric animals generated a cytotoxic response to hapten-modified cells of both donor (CBA) and h

INTRODUCTION: Chronic radiation proctitis is a common chronic complication of malignant pelvic diseases after pelvic radiation therapy. Although, the incidence has decreased after advent of intensity-modulated radiotherapy due to better control of radiation dose to rectum. In the era of conventional two-field radiotherapy to pelvis, this was a common complication usually presenting after 1-2 years of treatment completion. Rectal bleeding caused by radiation proctitis is difficult to manage. Argon plasma coagulation (APC) is an electrocoagulation technique that appears to be an effective and lo

Acquired radioresistance remains a major clinical obstacle in triple-negative breast cancer (TNBC) radiotherapy. Here, we first report the combined use of an ultrasmall copper-doped hafnium oxide nanoradiosensitizer (US-HfO2:Cu, ∼10 nm) and 192Ir brachytherapy (BT) to address this issue. BT triggers radiation-responsive Cu+ release from US-HfO2:Cu, inducing intracellular copper overload and cuproptosis, while the high-Z HfO2 core enhances radiation energy deposition to aggravate DNA damage. This synergistic biophysical strategy impairs mitochondrial function and activates the cGAS-STING pathwa

Radiotherapy is used to treat over 50 % of cancer patients. It is often used in combination with surgery, chemotherapy, and immunotherapy, for cancers of the breast, lung, oesophagus, and rectum. Ionising radiation predominantly exerts its anti-cancer effect through both direct DNA damage and indirectly via water radiolysis and the production of reactive oxygen species. This DNA damage is made permanent in the presence of molecular oxygen; however, it is reversible under hypoxia. Therefore, hypoxia confers significant radiotherapy resistance and given that it is a common feature of most solid

Reproductive cell death (RCD) occurs after one or more cell divisions resulting from an insult such as radiation exposure or other treatments with carcinogens or mutagens. The radioadaptive response for RCD is usually investigated by in vitro or in vivo clonogenic assay. To date, this has not been demonstrated in the vulval tissue in Caenorhabditis elegans ( C. elegans ), which is a well established in vivo model for radiation-induced RCD. In this study to determine whether radioadaptive response occurs in the vulval tissue model of C. elegans , early larval worms were gamma irradiated with lo

BACKGROUND: A promising approach to kill antibiotic-resistant bacteria uses light in combination with a photosensitizer to induce a phototoxic reaction. A major concern with the use of any non-antibiotic antimicrobial treatment is that exposure of bacteria to sub-lethal concentrations will lead to the development of resistance to antibiotics. This study aimed to determine the effect of sub-lethal photodynamic inactivation (PDI) on the antibiotic susceptibility and biofilm formation of clinical Staphylococcus aureus isolates.
METHODS: Forty clinical S. aureus isolates were exposed to PDI with

PURPOSE: Despite its prominent contribution to cancer cure and palliation, around 1% to 5% of cancer patients suffer serious side effects from radiotherapy. A cardinal goal in the fields of radiobiology and oncology is to predict normal tissue radiosensitivity of a cancer patient before radiotherapy. Higher tumor control rates are likely if radiotherapy individualization could be achieved by applying predictive approaches.
EXPERIMENTAL DESIGN: Here, we make use of the cytokinesis block micronucleus assay to assess radiosensitivity in cell lines derived from two different cell lineages obtaine

Cellular stress responses are upregulated following exposure to radiation and other DNA-damaging agents. Therefore radiation response can be dose dependent so that small acute exposures (and possibly exposures at very low dose rates?) are more lethal per unit dose than larger exposures above a threshold (typically 10-40 cGy) where induced radioprotection is triggered. We have termed these interlinked phenomena low-dose hypersensitivity (HRS) and induced radioresistance (IRR) as the dose increases. HRS/IRR has been recorded in cell-survival studies with yeast, bacteria, protozoa, algae, higher

PURPOSE: The purpose of this study was to analyse donor and time dependent variations in the frequencies of radiation-induced aberrations in chromosomes 1 and 2.
MATERIALS AND METHODS: Human lymphocytes from two donors were irradiated with 1 and 2 Gy of X-rays. Chromosomal aberrations were scored in chromosomes 1 and 2 painted with different fluorochromes and in Giemsa stained cells. Two time displaced experiments were performed with lymphocytes of each donor.
RESULTS: In cells of both donors chromosome 1 was generally more frequently involved in translocations than chromosome 2. This result

The thyroid is the purest endocrine gland in the body and is likely to produce clinically significant abnormalities after external radiotherapy. Functional clinical modifications after direct irradiation exceeding 30 Gy are essentially related to hypothyroidism which may be clinically overt or subclinical with normal serum free thyroxine levels and high thyrotropin concentrations; the risk of hyperthyroidism, silent thyroiditis and Hashimoto's disease is also increased. Secondary hypothyroidism related to irradiation of the hypothalamus and the pituitary gland may arise with doses over 40-50 G

Hyperthermia combined with 60Co gamma irradiation was studied using V79 hamster cells cultured in vitro. Modest hyperthermia (41 degrees C for 6 hrs.) enhanced the cell killing produced by acute exposure to radiation. The same treatment enhanced the effect of low dose-rate irradiation (200 rads/hr.) even more. The sequence in which modest hyperthermia was combined with low dose-rate irradiation was important. Maximal enhancement was observed when hyperthermia was followed by irradiations. The probable explanation is that, by damaging the repair system, prior heat renders the cells unable to re

BACKGROUND: When arsenic trioxide (ATO) was combined with radiation for treatment of transplanted murine gliomas in the brain, tumor response improved with disrupted tumor blood flow and survival was significantly prolonged.
METHODS: Total of 31 patients with newly diagnosed glioblastoma were accrued to a multi-institutional, NCI-funded, phase I study to determine the maximum tolerated dose (MTD) of ATO administered with radiation. Secondary objectives were survival and pharmacodynamic changes in perfusion on magnetic resonance imaging (MRI). Patients (unknown MGMT and IDH status) received AT

Liver damage upon exposure to ionizing radiation, whether accidental or because of therapy can contribute to liver dysfunction. Currently, radiation therapy is used for various cancers including hepatocellular carcinoma; however, the treatment dose is limited by poor liver tolerance to radiation. Furthermore, reliable biomarkers to predict liver damage and associated side-effects are unavailable. Here, we investigated fibrinogen-like 1 (FGL1)-expression in the liver and plasma after radiation exposure. We found that 30 Gy of liver irradiation (IR) induced cell death including apoptosis, necros

Radioligand therapies have opened new treatment avenues for cancer patients. They offer precise tumor targeting with a favorable efficacy-to-toxicity profile. Specifically, the kidneys, once regarded as the critical organ for radiation toxicity, also show excellent tolerance to radiation doses as high as 50-60 Gy in selected cases. However, the number of nephrons that form the structural and functional units of the kidney is determined before birth and is fixed. Thus, loss of nephrons secondary to any injury may lead to an irreversible decline in renal function over time. Our primary understan
