Subretinal neovascularization from the retina in radiation retinopathy.

In a 66-year-old woman with radiation retinopathy, subretinal neovascularization was present, originating from telangiectatic retinal vessels in the macular area. The patient showed no clinical or histologic evidence of age-related macular degeneration or other conditions that may have contributed to the subretinal neovascularization.

Spontaneous reattachment of the retina despite proliferative vitreoretinopathy.

In six cases of spontaneous retinal reattachment despite severe proliferative vitreoretinopathy, the retina was initially attached after a buckling procedure or vitreous surgery for proliferative vitreoretinopathy. However, each retina suddenly detached several weeks later. Surprisingly, the retinas settled slowly during the following weeks without further treatment. One case was marked by repeated episodes of detachment and reattachment. In none of the cases could an open retinal break be detected. In certain cases of proliferative vitreoretinopathy in which no open break is apparent and the …

Endothelial protein C receptor-mediated endogenous and therapeutic transport of activated protein C across the blood-retina barrier protects the retina.

BACKGROUND: Activated protein C (APC) is a serine protease known for its anticoagulant, anti-inflammatory, and cytoprotective properties. Although clinical evidence links protein C (PC) deficiency to various retinal pathologies, the physiological role of APC in the retina remains unexplored. OBJECTIVES: To determine whether PC and APC are synthesized locally in the healthy retina or are transported systemically across the blood-retina barrier (BRB) and to evaluate the contribution of the endothelial protein C receptor (EPCR) to APC transport and the therapeutic potential of APC in the retina …

[Remodeling of the internal retina-Implications for targeted optogenetics].

BACKGROUND: For the design of optogenetic treatment approaches for degenerative retinal disorders two factors are of key relevance: firstly, the treatment targets a diseased retina where morphology and function may already be severely altered. These alterations are referred to as remodeling. The second factor is the complex signal processing in the retina, which may be altered by this remodeling or short-circuited by optogenetic therapy. RESEARCH QUESTION: This article presents the current state of knowledge on disease-related morphological and functional remodeling processes in the retina an…

Primary Cilia of Astrocytes in the Retina in Development and Disease.

PURPOSE: Defects in primary cilia and ciliary signaling are associated with an array of neurodegenerative diseases. Primary cilia are sensory organelles that transmit and regulate various cellular communication pathways, including the significant Sonic hedgehog and Wnt signal pathways. Extensive studies have shown that neuronal cilia in the central nervous system contribute to early patterning, neuronal maturation, and survival. The behavior of cilia on astrocytes in glaucoma and aging remains poorly understood. This study used rodent and human retinal astrocytes to examine primary cilia and t…

A standardized nomenclature for the rods and cones of the vertebrate retina.

Vertebrate photoreceptors have been studied for well over a century, but a fixed nomenclature for referring to orthologous cell types across diverse species has been lacking. Instead, photoreceptors have been variably-and often confusingly-named according to morphology, presence/absence of 'rhodopsin', spectral sensitivity, chromophore usage, and/or the gene family of the opsin(s) they express. Here, we propose a unified nomenclature for vertebrate rods and cones that aligns with the naming systems of other retinal cell classes and that is based on the photoreceptor type's putative evolutionar…

Laser-induced histopathological changes to the retina: A review.

Although the indications for retinal laser have diminished over the past 2 decades due to the rise of intravitreal injection therapy, retinal laser remains a useful clinical tool, with novel laser technologies continuing to emerge. Continuous wave photocoagulation remains the most well-characterized laser in terms of its histopathological effects. Information about the histological effects of newer retinal laser modalities is relatively scarce. Available evidence supports the notion that short pulse applications result in less collateral damage to the neurosensory retina, but that at sufficien…

Prototype Hardware for Closed-loop Stimulation of the Retina.

Since the first human trial demonstrated the potential of retinal prostheses in 1956, research on retinal stimulation for vision restoration has been intensive. Compared to the advancements in stimulating electrodes and electronics, relatively less progress has been made in stimulation strategies. Current devices largely rely on manually tuned stimulation generated solely from camera inputs. As electrode pitch continues shrinking to improve visual acuity, manually tuning stimulation parameters becomes increasingly impractical. Moreover, the manual approach fails to account for time-dependent p…

Light as a Mediator of Acute and Chronic Retina Degeneration.

In age-related macula dystrophy (AMD) and some forms of inherited retinal dystrophies (IRDs), blindness is caused by the loss of photoreceptors and retinal pigment epithelium (RPE) cells. This process can be exacerbated by genetic and environmental risk factors, including exposure of the retina to bright light. Several light damage models have been developed and have proved to be powerful tools to study retinal degeneration. These models have enabled the investigation of common mechanisms of cell death and inflammation, as well as the identification of therapeutic targets and the assessment of…

Effects of a Hyperbaric Normoxic Environment on the Retina and Choroid.

PURPOSE: Divers and individuals working in submarines and other underwater environments are exposed to normoxic hyperbaric conditions frequently. The aim of this study was to evaluate the effects of hyperbaric normoxia on the retina and choroid. METHODS: Healthy participants with no prior diving experience were exposed to 2.4 atmospheres absolute pressure in a normoxic hyperbaric chamber (HC). Optical coherence tomography was used to measure retinal thickness, the peripapillary retinal nerve fiber layer, and choroidal thickness (CT) before and within 30 minutes after HC exposure. RESULTS: Th…

The Surprising Nonlinear Effects of S100A9 Proteins in the Retina.

Age-related macular degeneration (AMD) is a complex disease in which inflammation is implicated as a key factor but the precise molecular mechanisms are poorly understood. AMD lesions contain an excess of the pro-inflammatory S100A9 protein, but its retinal significance was yet unexplored. S100A9 was shown to be intrinsically amyloidogenic in vitro and in vivo. Here, we hypothesized that the retinal effects of S100A9 are related to its supramolecular conformation. ARPE-19 cultures were treated with native dimeric and fibrillar S100A9 preparations, and cell viability was determined. Wild-type r…

C9ORF72 Deficiency Results in Neurodegeneration in the Zebrafish Retina.

Hexanucleotide repeat expansions within the gene C9ORF72 are the most common cause of the neurodegenerative diseases amyotrophic lateral scl … or increased levels of neuroinflammation. However, detailed examination of adult female c9orf72-/- retinas showed prominent neurodegenerative features, including a decrease in retinal thickness, gliosis, and an overall reduction in neurons of all subtypes. Analysis of rod and cone cells within the photoreceptor layer showed a disturbance in their outer …

A circuit motif for color in the human foveal retina.

The neural pathways that start human color vision begin in the complex synaptic network of the foveal retina where signals originating in long (L), middle (M), and short (S) wavelength-sensitive cone photoreceptor types are compared through antagonistic interactions, referred to as opponency. In nonhuman primates, two cone opponent pathways are well established: an L vs. M cone circuit linked to the midget ganglion cell type, often called the red-green pathway, and an S vs. L + M cone circuit linked to the small bistratified ganglion cell type, often called the blue-yellow pathway. These pathw…
