The Borrelia burgdorferi sensu lato (s.l.) group comprises genetically related spirochetes, mostly associated with tick species belonging to the Ixodes ricinus complex in the Northern Hemisphere. The present study evaluated borrelial infection in the tick Ixodes pararicinus, which is the only representative species of the I. ricinus complex in Uruguay. A total of 137 I. pararicinus ticks were collected from deer, cattle, or vegetation in 2 Uruguayan Departments. A part of these ticks was tested directly by PCR targeting the borrelial gene flagellin (fla), whereas another part of the ticks was …

The ever-increasing number of Lyme borreliosis patients led us to consider more effective procedures for disease prevention. The aim of our study was to monitor the annual activity and infectivity of Ixodes ricinus ticks in the Pisárky region, City of Brno, CR, and to test the responses of the locally-captured ticks to selected repellents. The result of regular one-hour-per-week monitoring in 2011 was the collection of ticks that directly reflected the highest number of Lyme disease patients (4,835) detected throughout the period of recording in the Czech Republic. The ticks were examined for …

Lyme borreliosis (LB) is the most common arthropod-borne disease of humans in the Northern hemisphere. In Europe, the causative agent, Borrelia burgdorferi sensu lato complex, is principally vectored by Ixodes ricinus ticks. The aim of this study was to identify environmental factors influencing questing I. ricinus nymph abundance and B. burgdorferi s.l. infection in questing nymphs using a large-scale survey across Scotland. Ticks, host dung and vegetation were surveyed at 25 woodland sites, and climatic variables from a Geographical Information System (GIS) were extracted for each site. A to…

In Sweden, the geographical distribution of Lyme borreliosis corresponds to that of its vector Ixodes ricinus. Both tick activity and the length of the vegetation period are determined by daily mean temperatures ≥5°C. We analysed the correspondence between the distribution of I. ricinus in Sweden, the start date, end date, and length of the vegetation period, and the distributions of tick habitat-associated plant species. The geographical distribution of I. ricinus in Sweden corresponds to a vegetation period averaging approximately 170 days, an early start (before May 1st) of spring, and to t…

The risk to humans of contracting tick-borne zoonotic diseases depends on the risk of a bite from an infected tick, which can be broken down into its component parts as the number of host-seeking ticks in the environment, in particular nymphs, and the prevalence of tick-borne pathogens they are carrying. In turn, the prevalence of tick-borne pathogens is dependent upon tick biting intensity on hosts that support transmission between ticks; namely rodents. These ticks once fed moult into the next life stage and search for the next blood meal, thus posing a zoonotic risk. Here, we analyse tick b

Saliva-activated transmission (SAT) of Borrelia burgdorferi sensu stricto was demonstrated using real-time PCR and salivary gland extract (SGE) from partially fed Ixodes ricinus ticks. C3H/HeN mice were injected intradermally with 1.5 x 10(3) spirochetes mixed with 40 microg of SGE per mouse. The control group was inoculated with the same dose of spirochetes without SGE. The accelerating effect of SGE on spirochete proliferation was demonstrated on day 1 post infection, when a 4.2-fold increase in spirochetes was found in the skin and a 10-fold increase in the blood, compared with control mice…

Borrelia burgdorferi sensu lato (sl), the agent of Lyme disease, is transmitted to the host during the blood meal of Ixodes ticks. In most unfed ticks, spirochetes are present in the midgut and migrate during blood feeding to the salivary glands, from which they are transmitted to the host via saliva. In the present study, the efficiency of Ixodes ricinus ticks to transmit B. afzelii and B. burgdorferi sensu stricto (ss) and their infectivity for mice were examined in relation to the duration of the blood meal. In addition, we investigated whether these two Borrelia species can penetrate intac…

In a controlled study, we analyzed the efficiency of cleansing out the colon and the best contrast medium of two different regimens (total n = 237) for preparing the colon for double-contrast examination. The recommendations regarding diet and liquid intake, contrast medium and examination technique were identical in both groups. The combination laxative Prepacol without cleansing enema resulted in a more thoroughly cleaned colon that ricinol with cleansing enema (very good/good cleanliness: Prepacol, 92.4%; ricinol, 83.2%, p less than 0.05). However, the quality of contrast medium (good: Prep

INTRODUCTION: Borrelia burgdorferi sensu lato, the causative agents of Lyme borreliosis, are transmitted by Ixodes ticks. Tick saliva proteins are instrumental for survival of both the vector and spirochete and have been investigated as targets for vaccine targeting the vector. In Europe, the main vector for Lyme borreliosis is Ixodes ricinus, which predominantly transmits Borrelia afzelii. We here investigated the differential production of I. ricinus tick saliva proteins in response to feeding and B. afzelii infection.

Tick saliva is a rich source of pharmacologically and immunologically active molecules. These salivary components are indispensable for successful blood feeding on vertebrate hosts and are believed to facilitate the transmission of tick-borne pathogens. Here we present the functional and structural characterization of Iripin-3, a protein expressed in the salivary glands of the tick Ixodes ricinus, a European vector of tick-borne encephalitis and Lyme disease. Belonging to the serpin superfamily of protease inhibitors, Iripin-3 strongly inhibited the proteolytic activity of serine proteases kal…

This study aimed to reveal whether green lizards (Lacerta viridis), common hosts of tick larvae and nymphs, might be involved in the transmission cycle of Borrelia burgdorferi sensu lato in the Czech Republic. Green lizards were sampled in two areas at the Tiché Údolí Nature Reserve (site A: 50.1482 N, 14.3669E; site B: 50.1476 N, 14.3745 E), Central Bohemian Region, Czech Republic. The skin biopsy specimens and attached ticks (if any) were collected from 52 captured lizards. Also, questing ticks from both areas were collected by flagging. The touchdown polymerase chain reaction and gel electr

This study addresses the modifications that future climate conditions could impose on the transmission cycles of Borrelia burgdorferi s.l. by the tick Ixodes ricinus in Europe. Tracking the distribution of foci of a zoonotic agent transmitted by vectors as climate change shapes its spatial niche is necessary to issue self-protection measures for the human population. We modeled the current distribution of the tick and its predicted contact rates with 18 species of vertebrates known to act as reservoirs of the pathogen. We approached an innovative way for estimating the possibility of permanent…

For the first time in the world literature, the presented study describes oral-anal contact between two sympatric Ixodes ricinus and Dermacentor reticulatus ticks, which are the vectors of many pathogens in Europe. Among the specimens collected in nature, an I. ricinus male was found with the hypostome and chelicerae inserted in the anal aperture of a D. reticulatus female. Given the biological and physiological traits of both species and microorganisms colonising these arthropods, it seems that the interspecific oral-anal contacts between ticks may be an unknown alternative route of transmiss…

BACKGROUND: Tick-borne encephalitis is the most common tick-borne viral infection in Europe with 3,000 human cases reported each year. In Western Europe, the castor bean tick, Ixodes ricinus, is the principal vector of the tick-borne encephalitis virus (TBEV). TBEV appears to be spreading geographically and was recently detected for the first time in Canton Valais in the southern part of Switzerland. The purpose of the present study was to survey the I. ricinus tick populations of Canton Valais for TBEV.
METHODS: We collected a total of 19,331 I. ricinus ticks at 45 different sites in Canton …

Ticks use their energy reserves to maintain their water balance, search for hosts and transmit tick-borne pathogens. However, the influence of tick-borne pathogens on the energy reserves of the tick vector has not been well studied. The relationship between Borrelia burgdorferi sensu lato (s.l.) infection status and fat content in questing Ixodes ricinus nymphs was examined. Nymphs were sampled from the field. Their body mass and fat content were measured, and their Borrelia genospecies infection status (using reverse line blot analysis), and spirochete load (using quantitative PCR) were analy…
