Study of Anti-Inflammatory and Analgesic Activity of Scorpion Toxins DKK-SP1/2 from Scorpion Buthus martensii Karsch (BmK).

Buthus martensii Karsch (BmK), is a kind of traditional Chinese medicine, which has been used for a long history for the treatment of many diseases, such as inflammation, pain and cancer. In this study, DKK-SP1/2/3 genes were screened and extracted from the cDNA library of BmK. The DKK-SP1/2/3 were expressed by using plasmid pSYPU-1b in E. coli BL21, and recombinant proteins were obtained by column chromatography. In the xylene-induced mouse ear swelling and carrageenan-induced rat paw swelling model, DKK-SP1 exerted a significant anti-inflammatory effect by inhibiting the expression of Nav1.8

Scorpion Peptides in Antiviral Drug Discovery: An In-depth Review.

Despite remarkable progress in the fields of vaccinology and antiviral therapy, viral infections persist as a leading cause of morbidity and mortality worldwide. Therefore, it is crucial to identify new and effective antiviral drug candidates. Scorpion venoms are viewed as a rich source of structurally diverse and biologically active peptides, which may present opportunities for diagnostic, preventive, and therapeutic applications. This review intends to provide a contemporary and detailed analysis of scorpion-derived peptides, with a particular focus on their structural diversity, cytotoxic e

Endemic scorpion species and subspecies in Morocco: A comprehensive overview.

Scorpion fauna is abundant in regions with arid and semi-arid climates, exposing these areas to a risk of envenomation, especially for child … ecies in this region. Ongoing climate change is likely to expand the population and distribution of scorpions, as well as the number of envenomations. The results underscore the importance of implementing control and prevention strategies to reduce the risks of envenomation, particularly in the context of climate change. Therefore, to develop management and …

Scorpion envenoming by Parabuthus is a public health concern in Namibia.

Scorpion stings by members of the genus Parabuthus (Buthidae) can lead to life-threatening envenoming. Namibia harbours a rich diversity of scorpions, including seventeen species of the highly venomous genus Parabuthus, although the biology and medical relevance of most species are poorly documented. Severe envenoming is caused by at least P. granulatus, P. villosus, P. kraepelini and P. schlechteri, which are considered to be of particular importance. Data on epidemiology, venom composition and variation, and clinical effects remain scarce. A lack of effective antivenom and long distances to …

Deciphering the Enigma: Lignocaine Resistance After Scorpion Envenomation.

Lignocaine, an amide-type local anesthetic (LA), is routinely used in dentistry. Resistance to lignocaine is rare, and it can occur due to various reasons, such as genetic mutations or variations in the local environment where it is injected. One reason might be scorpion envenomation. We report a case of a 39-year-old female patient with a chief complaint of a gradual reduction in mouth opening for the past five years, with a history of scorpion bite. The patient had a scorpion bite 18 years back. She started developing symptoms of oral submucous fibrosis five years ago. The patient exhibited

Scorpion-Induced Acute Coronary Syndrome: A Stinging Complication.

The Tityus trinitatis, a black scorpion species endemic to the fauna of Trinidad, has been implicated in envenomation with devastating clinical sequelae such as acute pancreatitis and major adverse cardiovascular events. We present the first in-Caribbean case of a 59-year-old Caribbean South Asian male with human immunodeficiency virus who presented with a non-ST-segment-elevation acute coronary syndrome after being stung, which was managed with comprehensive, guideline-directed medical therapy. The clinician should be cognizant of scorpion-induced acute coronary syndrome (ACS) as a potential

Cytokines and their regulators in rat lung following scorpion envenomation.

Nowadays, more than two billion inhabitants of underdeveloped tropical and subtropical countries are at risk of being stung by scorpions. Scorpion stings annually cause 2000-3000 deaths as they can lead to the respiratory and/or cardiovascular complications. Pathogenesis of lung damage under scorpion envenomation is often comprehensive. Respiratory failure can have a cardiogenic origin, associated with venom neurotoxin action. However, some venom components can stimulate pro-inflammatory signaling cascades followed by cytokines synthesis, recruit and activate immune cells, participating in the…

Optimizing Scorpion Toxin Processing through Artificial Intelligence.

Scorpion toxins are relatively short cyclic peptides (<150 amino acids) that can disrupt the opening/closing mechanisms in cell ion channels. These peptides are widely studied for several reasons including their use in drug discovery. Although improvements in RNAseq have greatly expedited the discovery of new scorpion toxins, their annotation remains challenging, mainly due to their small size. Here, we present a new pipeline to annotate toxins from scorpion transcriptomes using a neural network approach. This pipeline implements basic neural networks to sort amino acid sequences to find those

Severity of scorpion envenomation in Saudi Arabia: A systematic review.

Over the course of five years, 72,168 scorpion stings were recorded in Saudi Arabia. Though a number of articles have been published on scorpion stings in the country, no review studies have sought to determine the severity of scorpion envenomation using a standardized scale. The purpose of this systematic review is, thus, to explore the signs and symptoms of scorpion envenomation victims in Saudi Arabia and classify them using the Abroug's severity scale. This review identified 49 articles published between January 1953 and September 2024 on scorpion sting incidents in Saudi Arabia. Out of th

Revisiting the scorpion central nervous system using microCT.

The central nervous system (CNS) of Chelicerata has remained conserved since the Cambrian, yet few studies have examined its variability within chelicerate orders including Scorpiones. The scorpion CNS comprises the prosomal ganglion and opisthosomal ventral nerve cord. We visualize the scorpion CNS with microCT, explore morphological variation across taxa, compare the scorpion CNS to other arachnids, and create a terminology glossary and literature review to assist future studies. Six scorpion species were microCT scanned. Scan quality varied and most structures in the prosomal ganglion could

A Fluorescent Compound from the Exuviae of the Scorpion, Liocheles australasiae.

Most scorpions fluoresce under UV light. To date, two types of fluorescent compounds have been identified in scorpions, but it has been assumed that other unknown compounds may be responsible for the fluorescence. In this study, we isolated a fluorescent compound from the exuviae of the scorpion Liocheles australasiae identified as a macrocyclic diphthalate ester with a molecular mass of 496.2 Da. The same compound was also detected in extracts from four other scorpion species. This suggests that this compound is shared by multiple scorpion species, although its contribution to the cuticle flu…

Scorpion toxins targeting Kv1.3 channels: insights into immunosuppression.

Scorpion venoms are natural sources of molecules that have, in addition to their toxic function, potential therapeutic applications. In this source the neurotoxins can be found especially those that act on potassium channels. Potassium channels are responsible for maintaining the membrane potential in the excitable cells, especially the voltage-dependent potassium channels (Kv), including Kv1.3 channels. These channels (Kv1.3) are expressed by various types of tissues and cells, being part of several physiological processes. However, the major studies of Kv1.3 are performed on T cells due its

Application of the Scorpion Neurotoxin AaIT against Insect Pests.

Androctonus australis Hector insect toxin (AaIT), an insect-selective toxin, was identified in the venom of the scorpion Androctonus australis. The exclusive and specific target of the toxin is the voltage-gated sodium channels of the insect, resulting in fast excitatory paralysis and even death. Because of its strict toxic selectivity and high bioactivity, AaIT has been widely used in experiments exploring pest bio-control. Recombinant expression of AaIT in a baculovirus or a fungus can increase their virulence to insect pests and diseases vectors. Likewise, transgenic plants expressing AaIT
