Sequence-encoded tubular architectures in disordered spider silk proteins revealed by multiscale simulations and NMR.

Spider silk proteins (spidroins) are large, block-copolymer-like proteins that must remain soluble at high concentration in the spinning dope while being primed for rapid fiber formation. Understanding how these intrinsically disordered proteins organize in solution is key to explaining the transformation from soluble dope to solid fibers with exceptional strength and toughness. Here, we show that major ampullate (Ma) spidroins from the black widow spider form dynamic ensembles that include metastable tubular substructures. Multiscale molecular dynamics (MD) simulations reveal compact, anisotr…

A spider silk-inspired, transparent, anti-freezing ionic conductive hydrogel as a flexible sensor device.

As soft material ionic conductors, ionically conductive hydrogels are of great significance for the development of flexible electronics. However, it is still a great challenge to effectively design functional hydrogel structures to address various practical application scenarios (such as low temperature environments) and expand their application range (such as transparent display devices). In this paper, an anti-bacterial and ionically conductive TEMPO-oxidized cellulose nanofiber/polyvinyl alcohol/quaternary ammonium chitosan/Al3+ (CPQA-EH) hydrogel (conductivity of 7.50 ms cm-1) with high tr

Smart Silk-Based In Situ Sol-Gel Modulates Rectal Microenvironment for Effective Ulcerative Colitis Alleviation.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease, with untreated cases often progressing to colorectal cancer. Current treatments aim to induce inflammatory remission but often neglect the surrounding microenvironment, which significantly impairs mucosal healing and contributes to treatment failures. This study presents a novel silk fibroin-based fucoidan (SFU) in situ rectal gel, with sol-gel transition confirmed through rheological analysis under physiological pH and temperature conditions. The SFU gel exhibits strong antioxidant activity, achieving a DPPH radical scavenging r…

Adipose derived mesenchymal stem cell-seeded regenerated silk fibroin scaffolds reverse liver fibrosis in mice.

Liver fibrosis (LF) is an important process in the progression of chronic liver disease to cirrhosis. We have previously demonstrated that a regenerated silk fibroin scaffold loaded with adipose-derived stem cells (RSF + ADSCs) can repair acute liver injury. In this study, we established a chronic LF animal model using carbon tetrachloride (CCl4) and a high-fat diet. We then investigated the liver repair capacity after transplanting RSF + ADSC scaffolds and RSF scaffolds onto the liver surface of mice. Compared with the control group, the concentrations of ALT and AST in the serum were signifi…

Targeting local glucocorticoid metabolism with a silk fibroin microneedle for more effective hypertrophic scar repair.

Hypertrophic scars (HS) are pathological cutaneous scars characterized by excessive collagen deposition and fibrosis. Intralesional glucocorticoid injections remain the standard treatment for HS, but it is often associated with side effects. In this study, we developed a novel silk fibroin microneedle (SF-MN) loaded with prednisone for localized delivery and glucocorticoid metabolism. The SF-MNs were fabricated using 10 % (w/v) silk fibroin. Prednisone-loaded SF-MNs were characterized by SEM and evaluated for biocompatibility, mechanical strength, and preclinical efficacy. Overall, the results…

Silk fibroin and hydroxypropyl cellulose composite injectable hydrogel-containing extracellular vesicles for myocardial infarction repair.

Extracellular vesicles (EVs) have been recognized as one of the promising specific drugs for myocardial infarction (MI) prognosis. Nevertheless, low intramyocardial retention of EVs remains a major impediment to their clinical application. In this study, we developed a silk fibroin/hydroxypropyl cellulose (SF/HPC) composite hydrogel combined with AC16 cell-derived EVs targeted modification by folic acid for the treatment of acute myocardial infarction repair. EVs were functionalized by distearoylphosphatidyl ethanolamine-polyethylene glycol (DSPE-PEG-FA) via noncovalent interaction for targeti…

Silk Fibroin-Based Hydrogel Microneedles Deliver α-MSH to Promote Melanosome Delivery for Vitiligo Treatment.

Microneedles have shown great advantages in subcutaneous drug delivery and skin disease treatment. Vitiligo is a difficult-to-cure skin disease characterized by the depigmentation of the epidermis. Melanosomes produced in melanocytes are transported through dendrites to adjacent keratinocytes, where they accumulate, resulting in skin pigmentation. However, melanocytes in vitiligo patients are functionally disrupted. Silk fibroin (SF) methacrylate hydrogel microneedle can deliver α-MSH to the epidermis directly, where α-MSH helps the protection of melanocytes, extension of melanocytic dendrites…

Multi-scale, multi-level anisotropic silk fibroin/metformin scaffolds for repair of peripheral nerve injury.

Silk fibroin (SF) as a natural polymer has a long history of application in various regenerative medicine fields, but there are still many shortcomings in silk fibroin for using as nerve scaffolds, which limit its clinical application in peripheral nerve regeneration (PNR). In this work, a multi-scale and multi-level metformin (MF)-loaded silk fibroin scaffold with anisotropic micro-nano composite topology was prepared by micromolding electrospinning for accelerating PNR. The scaffolds were characterized for morphology, wettability, mechanical properties, degradability, and drug release, and S…

Evaluation of an antibacterial peptide-loaded amniotic membrane/silk fibroin electrospun nanofiber in wound healing.

Antimicrobial peptides (AMPs) are among the compounds that have significant potential to deal with infectious skin wounds. Using wound dressings or skin scaffolds containing AMPs can be an effective way to overcome infections caused by antibiotic-resistant strains. In this study, we developed an amniotic membrane-based skin scaffold using silk fibroin to improve mechanical properties and CM11 peptide as an antimicrobial peptide. The peptide was coated on the scaffold using the soaking method. The fabricated scaffold was characterised by SEM and FTIR, and their mechanical strength, biodegradati…

Prospective >12 Months Outcomes After Vocal Fold Injection Medialization With Silk Microparticle-Hyaluronic Acid Material.

OBJECTIVE: Vocal fold injection medialization (VFIM) is widely used as an initial treatment for unilateral vocal fold paralysis (UVFP). Current practices employ materials that share the limitation of temporary clinical effect from variable resorption rates. A novel silk protein microparticle-hyaluronic acid-based material (silk-HA) has demonstrated cellular infiltration and tissue deposition that may portend a durable medialization effect. We report on ≥12 months outcomes after VFIM with silk-HA. METHODS: Prospective open-label study of patients with UVFP that elected treatment with VFIM with…

Silk-inspired fiber implant with multi-cues enhanced bionic microenvironment for promoting peripheral nerve repair.

Successful repair and desirable functional recovery of large-gap nerve injuries using artificial nerve implants remains a significant clinical challenge. The beneficial bionic microenvironment within scaffolds can significantly promote the outgrowth of newborn nerve tissues after implantation. Herein, we developed an aligned silk-inspired fiber scaffold (RGD@ASFFs) with a synergistic effect of an extracellular matrix mimicking physical cues and RGD (Arg-Gly-Asp) signals to provide an enhanced cell-friendly microenvironment for repairing large-gap peripheral nerve injuries. The topographic alig…

Marginal sealing around integral bilayer scaffolds for repairing osteochondral defects based on photocurable silk hydrogels.

Osteochondral repair remains a major challenge in current clinical practice despite significant advances in tissue engineering. In particular, the lateral integration of neocartilage into surrounding native cartilage is a difficult and inadequately addressed problem that determines the success of tissue repair. Here, a novel design of an integral bilayer scaffold combined with a photocurable silk sealant for osteochondral repair is reported. First, we fabricated a bilayer silk scaffold with a cartilage layer resembling native cartilage in surface morphology and mechanical strength and a BMP-2-…

Enhanced dual network hydrogels consisting of thiolated chitosan and silk fibroin for cartilage tissue engineering.

Thiolated chitosan (CS-NAC) was synthesized and the selected CS-NAC was used together with silk fibroin (SF) to produce dual network CS-NAC/SF hydrogels. The CS-NAC/SF solutions with formulated compositions were able to form hydrogels at physiological temperature and pH. Rheological measurements showed that elastic modulus of some CS-NAC/SF gels could reach around 3 kPa or higher and was much higher than their respective viscous modulus, indicating that they behaved like strong gels. Deformation measurements verified that CS-NAC/SF gels had well-defined elasticity. The optimized CS-NAC/SF gels…

Bilayer silk fibroin grafts support functional oesophageal repair in a rodent model of caustic injury.

Surgical repair of caustic oesophageal injuries with autologous gastrointestinal segments is often associated with dysmotility, dysphagia and donor site morbidity, and therefore alternative graft options are needed. Bilayer silk fibroin (BLSF) scaffolds were assessed for their ability to support functional restoration of damaged oesophageal tissues in a rat model of onlay oesophagoplasty. Transient exposure of isolated oesophageal segments with 40% NaOH led to corrosive oesophagitis and a 91% reduction in the luminal cross-sectional area of damaged sites. Oesophageal repair with BLSF matrices …

Identification of Wet-Spinning and Post-Spin Stretching Methods Amenable to Recombinant Spider Aciniform Silk.

Spider silks are outstanding biomaterials with mechanical properties that outperform synthetic materials. Of the six fibrillar spider silks, aciniform (or wrapping) silk is the toughest through a unique combination of strength and extensibility. In this study, a wet-spinning method for recombinant Argiope trifasciata aciniform spidroin (AcSp1) is introduced. Recombinant AcSp1 comprising three 200 amino acid repeat units was solubilized in a 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP)/water mixture, forming a viscous α-helix-enriched spinning dope, and wet-spun into an ethanol/water coagulation ba…
