In a comparative histopathologic autopsy study, 100 patients with diabetes mellitus were compared to 100 without this condition with respect to the presence of chronic pancreatopathy. No qualitative differences in specific alterations could be identified. In diabetic patients, however, intralobular and interacinar fibrosis, lipomatosis and atrophy of the parenchyma occurred more frequently and tended to be more severe. We adapted a previously described procedure to quantitate collagen in corneal tissue sections prepared from paraffin-embedded samples. The method entailed staining the deparaffinized tissue sections with Sirius red, eluting the bound dye with NaOH-methanol, and estimating the color in a spectrophotometer as an indication of the collagen content. Thirteen normal subjects and thirteen subjects with bona fide keratoconjunctivitis sicca were examined employing wide-field colour specular microscopy of the corneal epithelium. The prevalence of small, medium and large cells is different in these two groups with a shift to small cells in patients with keratoconjunctivitis. Increased rose bengal dye uptake filaments, coarse mucus plaques and pre-exfoliative sheets are also seen in keratoconjunctivitis sicca. These findings suggest an accelerated epithelial cell loss from the surface of the eye in keratoconjunctivitis sicca.

A macroscopic staining method was used to assess the incidence and extent of squamous metaplasia of the subglottic portion of the larynx. A total of 226 postmortem specimens were studied, 105 from nonsmokers and 121 from smokers. More than half of the specimens stained showed some metaplasia, but the incidence and extent of metaplasia was greater in smokers than in nonsmokers, and women were more affected than men. The incidence of metaplasia was greater in pipe and cigar smokers than in nonsmokers, but previous smokers did not differ from nonsmokers.

The cytologic findings in two histologically confirmed cases of granuloma inguinale are presented. The Papanicolaou staining method used in routine cytology screening proved to be suitable for the detection of the characteristic Donovan bodies in smears of the vulvar lesions.

An ovarian tumour of mixed thecomatous and lipid cell structure is described and the clinical and pathological features of this and eleven previously reported cases are reviewed. Functionally the tumours resemble lipid cell tumours in being mainly androgenic and are similar in size range and age incidence to the 'adrenal' type. Three tumours however contained hilus (Leydig) cells with Reinke crystalloids. The tumours furnish additional evidence of the stromal origin of lipid cell tumours and the term 'lipid cell thecoma' is proposed for them as indicating their main morphological affinities. I

A large firm 12.5 cm mass at the tail of pancreas was surgically resected in a 73-year-old white man. The tumor was composed of solid sheets of cells with abundant finely granular eosinophilic cytoplasm. Electron microscopic study revealed tumor cells which were packed with mitochondria, a feature characteristic of the oncocyte. Perineural invasion and direct extension into the parenchyma of the spleen was present, as well as metastasis to a lymph node. There appears not to be a report of a similar case.

Sets of three Isolator blood culture tubes were seeded with low numbers of 96 strains of 26 bacterial species (fresh and stock clinical isolates). One tube was processed immediately, and the other two were held at 22 and 34 degrees C for 15 h before processing. Organism recovery was 99, 99, and 98%, respectively. Diagnosis of Campylobacter enteritis by direct smear examination of stool specimens, using 1% aqueous basic fuchsin, was compared with a conventional cultural method (Campy-BAP). After examination of 485 stool specimens the direct smear method produced a sensitivity and specificity of 94% and 99.5%, respectively.

In 87 cases of lobular carcinoma in situ, 64 contained mucous globules in more than 25% of the cells. Eighteen, however, had no or only a few mucous globules. In cancerization of lobules by duct carcinoma and in benign hyperplastic lesions, only a few cells contained mucous globules. The presence of many of these globules may be helpful in the diagnosis of lobular carcinoma in situ. On the other hand, the quantitation of mucous globules in lobular carcinoma in situ is not of prognostic significance.

On the basis of histochemical characteristics it was possible to demostrate a sexual dimorphism of the tree shrew sublingual gland. Although numerous staining methods for the demonstration of mucosubstances were used in this study, only methods for the demonstration of sulfated glycoproteins (sulfomucins) were effective in demonstrating the sexual dimorphism. Two methods, the carbol fuchsin with acetic acid differentiation and the periodic acid-carbol pararosaniline, were used for demonstrating leprosy bacilli in skin smears. Bacillary smears from 200 long-treated patients with tuberculoid, borderline and lepromatous leprosy were stained with periodic acid-carbol pararosaniline. Electron microscopy processing and staining of nuclei were used to localize reaction products of acridine orange staining in actinic keratosis of human skin. Electron-dense granules about 10-100 nm in diameter were seen exclusively in the euchromatin portion of the nucleus. Almost all tumor cells had granules (mean = 65; SD = 26). These granules were also occasionally observed in the dermal connective tissue cells in the lesion.
