Pineal involvement in the alimentary behavior and taste preferences in the rat.

The purpose of this experiment was to explore the effects of pinealectomy or sham pinealectomy on circadian rhythms of taste preferences, food and fluid intake and body weight gain in rats. We compared the body weight gain, the amounts of food eaten and that of deionized water, total fluid, salt, sour, sweet and bitter near-threshold solutions consumed by rats before all surgical manipulations and after pinealectomy or sham pinealectomy. The results showed that the pineal gland does not exert a major influence on circadian organization of taste preferences, drinking, feeding and body weight mo…

Corticofugal influence on taste responses in the parabrachial pons of the rat.

Although the anatomy of centrifugal input to gustatory neural structures has been described, little is known of the physiological mechanisms that convey this influence or of their functional significance. As a first step in the investigation of these issues, the effect of a reversible lesion in the gustatory neocortex (GN) on the neural code for taste in the parabrachial nucleus of the pons (PbN) was studied in rats. Electrophysiological responses to taste stimuli bathed over the tongue were recorded from single units in the PbN before, after and following recovery from an infusion of procaine…

Taste and smell problems: validation of questions for the clinical history.

Complaints of taste and smell dysfunction unaccompanied by symptoms of neurological or nasal problems are not uncommon. However, "I can't taste" is not necessarily an accurate symptom description. Complaints tend to reflect the common confusion between taste sensations (that is, salt, sour, sweet, bitter) and flavor sensations (including taste, smell, temperature, and texture). A number of questions have been identified that help classify symptoms according to the type of dysfunction (taste, smell, or both): whether the problem is quantitative (reduced or absent sensation) or qualitative (dist…

Impairment of taste and olfaction in patients with cirrhosis: the role of vitamin A.

Taste and olfaction sensory modalities and the response to vitamin A replenishment were studied in 37 vitamin A deficient patients (determined by low serum levels and abnormal dark adaptation) with stable alcoholic cirrhosis. Among this group of patients, 11 also had low serum zinc levels which ranged from 45 to 63 micrograms/dl. Initial taste and olfaction mean detection thresholds for all test substances were significantly higher (less sensitive) in all patients as compared to 21 sex-matched healthy subjects of the same age range (P less than 0.05). Twenty-seven patients including the 11 pat…

Taste responses to neohesperidin dihydrochalcone in rats and baboon monkeys.

Preference-aversion behavior to solutions containing neohesperidin dihydrochalcone (NHDHC) was studied rats and baboon monkeys. Electrophysiological responses evoked by application of NHDHC solutions to taste receptors innervated by the chorda tympani and the glossopharyngeal nerves were also measured. As a group, rats were indifferent to solutions containing up to 1.2 x 10(-3) M NHDHC in short and long-term preference tests. A solution containing the very high concentration of 8.2 x 10(-3) M NHDHC was consumed less than water by all rats. The aversive behavior of rats to the 8.2 x 10(-3) M NH…

Mechanistic insights into taste modulation of Xanthoceras sorbifolium leaf tea fermented by Aspergillus cristatus and Aspergillus neoniger: an integrated component-taste relationship analysis.

Xanthoceras sorbifolium leaf tea has notable pharmacological potential, but its taste limits broader acceptance. In this study, the tea was fermented with Aspergillus cristatus (AC) or Aspergillus neoniger (AN), and taste modulation was investigated by electronic tongue analysis and untargeted metabolomics. AC fermentation increased bitterness and sweetness but reduced umami and saltiness, whereas AN increased sourness and reduced saltiness and bitterness. Integrated metabolite-taste analysis identified 34 and 48 key differential metabolites in AC- and AN-fermented teas, respectively. Redundan…

Normal tissue complication probability model predicting taste impairment in head and neck cancer patients: Evaluating the taste bud bearing tongue mucosa as a predictor.

BACKGROUND/PURPOSE: Taste impairment is a common yet complex toxicity of head and neck cancer (HNC) radiotherapy treatment that may affect quality of life of survivors. This study aimed to predict acute and late taste impairment using taste bud bearing tongue mucosa as a new taste-specific organ-at-risk compared to full oral cavity as identified in previous studies. MATERIALS/METHODS: Included HNC patients were treated with curative radiotherapy between 2007 and 2022. The endpoint was patient-rated moderate-to-severe taste loss scored with the EORTC QLQ-H&N35. The new tongue mucosa structure …

Covid-19 affects taste independent of taste-smell confusions: results from a combined chemosensory home test and online survey from a large global cohort.

People often confuse smell loss with taste loss, so it is unclear how much gustatory function is reduced in patients self-reporting taste loss. Our pre-registered cross-sectional study design included an online survey in 12 languages with instructions for self-administering chemosensory tests with 10 household items. Between June 2020 and March 2021, 10,953 individuals participated. Of these, 5,225 self-reported a respiratory illness and were grouped based on their reported COVID test results: COVID-positive (COVID+, N = 3,356), COVID-negative (COVID-, N = 602), and COVID unknown for those wai…

Identification and structural characterization of stereochemical promiscuity in a taste receptor.

Taste receptor type 1 (TAS1R), which consists of sweet and umami receptors in humans, senses nutrients (such as sugars and amino acids) with substrate specificity that is often broad and varies among animals and subtypes. However, the structural basis for achieving diverse specificities remains largely elusive. Here, we present the crystal structure of the ligand-binding domain (LBD) of Tas1r1/Tas1r3 heterodimer from pufferfish, an ortholog of the human umami taste receptor. The overall structure of Tas1r1/Tas1r3LBD resembles previously reported TAS1R structures, indicating a conserved core ar…

The taste for the beautiful: influence of diet on the evolution of plumage in tanagers.

The diversity of animal shapes and colors raises questions about the mechanisms by which species acquire complex phenotypes considering that they evolve under both natural and sexual selection. Tanagers employ a diversity of signals in social interactions which, coupled with diverse diets that include fruits and flowers, make them an excellent group for studying how diet influences the evolution of communication signals. Using Bayesian linear mixed models, plumage reflectance data for 291 tanager species (∼77% of species), and appropriate techniques for compositional diet data, we show that pe

Intracellular and dual-site inhibition of a bitter taste GPCR.

Bitter taste receptors (TAS2Rs) are G protein-coupled receptors expressed in both gustatory and extraoral tissues and activated by a broad range of compounds. TAS2R14 is among the most promiscuous members of this family, responding to many structurally diverse ligands. Cryo-electron microscopy structures of TAS2R14 have revealed agonists binding in an intracellular pocket, raising the question of the main sites of interaction for known TAS2R14 antagonists. To address this, we examined the effects of mutations at residues located in the extracellular and intracellular regions on receptor inhibi…

Association of COVID-19 Vaccination With Changes in Smell and Taste.

Since the introduction of vaccines for severe acute respiratory syndrome coronavirus 2 in the United States, there has been significant vaccine hesitancy, in part due to fear of adverse effects. We sought to investigate the rates of smell and taste changes after COVID-19 vaccination compared to other common vaccines. Our study cohort included individuals identified by Current Procedural Terminology code in the TriNetX database receiving the COVID-19 first series, COVID-19 booster, influenza, tetanus, diphtheria, pertussis (TDAP), or pneumococcal vaccines between December 15, 2020, and August 1…

Research and development of taste sensors as a novel analytical tool.

Gustatory and olfactory receptors receive multiple chemical substances of different types simultaneously, but they can barely discriminate one chemical species from others. In this article, we describe a device used to measure taste, i.e., taste sensors. Toko and colleagues developed a taste sensor equipped with multiarray electrodes using a lipid/polymer membrane as the transducer in 1989. This sensor has a concept of global selectivity to decompose the characteristics of a chemical substance into taste qualities and to quantify them. The use of taste sensors has spread around the world. More…

Effect of salivary gland removal on taste preference in mice.

To evaluate the effect of decreased salivary secretion on taste preference, we investigated taste preference for five basic tastes by a 48 h two-bottle preference test using a mouse model (desalivated mice) that underwent surgical removal of three major salivary glands: the parotid, submandibular, and sublingual glands. In the desalivated mice, the avoidance behaviors for bitter and salty tastes and the attractive behaviors for sweet and umami tastes were significantly decreased. We confirmed that saliva is necessary to maintain normal taste preference. To estimate the cause of the preference …

The Effect of Genetic Taste Status on Swallowing: A Literature Review.

PURPOSE: Swallowing and taste share innervation pathways and are crucial to nutritive intake. Individuals vary in their perception of taste due to factors such as genetics; however, it is unclear to what extent genetic taste status influences swallowing physiology and function. The purpose of this review article is to provide background on genetic taste status, review the evidence on the association between genetic taste status and swallowing, and discuss research and clinical implications. METHOD: A comprehensive literature review was conducted using search terms related to swallowing and ge…

The druggability of bitter taste receptors for the treatment of neurodegenerative disorders.

The delivery of therapeutic drugs to the brain remains a major pharmacology challenge. A complex system of chemical surveillance to protect the brain from endogenous and exogenous toxicants at brain barriers hinders the uptake of many compounds with significant in vitro and ex vivo therapeutic properties. Despite the advances in the field in recent years, the components of this system are not completely understood. Recently, a large group of chemo-sensing receptors, have been identified in the blood-cerebrospinal fluid barrier. Among these chemo-sensing receptors, bitter taste receptors (TAS2R…
