Molecular Identification of Cytochrome b Mutations in Buparvaquone-Unresponsive Theileria annulata Isolates from Khorasan-Razavi, Iran.

INTRODUCTION: The failure of conventional therapies for bovine theileriosis in Khorasan-Razavi province and other regions of Iran has led to significant economic losses and raised concerns about rural livestock sustainability and national food security. This study identified specific cytochrome b mutations in Theileria annulata isolates collected from cattle showing clinical unresponsiveness to buparvaquone treatment in the Khorasan-Razavi province of Iran. MATERIALS AND METHODS: Blood samples were collected, and parasite DNA was extracted and amplified via PCR using specific primers. RESULT

Microscopy-PCR discordance and shifting epidemiology in South India: advancing diagnostic precision for control of morphologically similar bovine hemopathogens (Babesia, Theileria, Anaplasma) from a hospital-based study.

Advancing epidemiological insights is instrumental in optimizing region-specific hemopathogen control. A retrospective study in Chennai, South India screened 2,434 cattle over one year, revealing a 13.93% hospital-based prevalence of morphologically similar hemopathogen (Babesia, Theileria, Anaplasma) diseases (HPDs) by light microscopy (LM). Anaplasmosis predominated (8.63%), increasing during monsoon; theileriosis rose to 4.44%, babesiosis declined to 0.86%, with highest prevalence in multiparous Jersey crossbreds. Relative sensitivity, specificity, PPV and NPV of LM were prospectively estim

Theileria luwenshuni and Novel Babesia spp. Infections in Humans, Yunnan Province, China.

Piroplasmid parasites such as Theileria luwenshuni protozoa pose a global threat to both animal and human health. However, human theileriosis remains underexplored compared to infections caused by Plasmodium and Babesia species parasites. We investigated potential hemoparasite infections among 1,721 persons with fever, anemia, or both in Yunnan Province, China. Molecular detection identified 13 cases positive for T. luwenshuni protozoa, of which 5 patients were further confirmed by Western blot antibody analysis. We also identified 6 babesiosis cases, 3 infections with B. microti and 3 with no

Sequence-based analysis of Theileria annulata reveals high Tams1 gene diversity and conservation of 18 S rRNA in Türkiye.

BACKGROUND: Theileria annulata, a tick-borne apicomplexan parasite, causes tropical theileriosis in cattle, leading to significant economic losses in endemic regions. In our previous study, T. annulata was detected in various bovine blood samples via sequencing, despite negative results from conventional species-specific PCR assays, suggesting potential sequence variation in diagnostic target regions. Building upon this observation, the present study aimed to investigate the genetic diversity and population structure of T. annulata isolates from Türkiye by analyzing two molecular markers with

Molecular detection of Theileria cervi in equids from México.

Equine piroplasmosis is a parasitic illness caused by various protozoa of the Babesia and Theileria genera, which parasitize within red blood cells. The transmission of these pathogens occurs through certain genus of ticks, including Amblyomma, Haemaphysalis, Hyalomma, and Rhipicephalus. In recent times, an increase in the identification of new Theileria species and genotypes has been observed. This is further complicated by the presence of mixed Theileria infections in both mammals and tick vectors, particularly in regions where wildlife and livestock share habitats and vectors. Therefore, th

18S rRNA Gene-Based Piroplasmid PCR: An Assay for Rapid and Precise Molecular Screening of Theileria and Babesia Species in Animals.

PURPOSE: The parasites of genera such as Babesia and Theileria are called piroplasmids due to the pear-shaped morphology of the multiplying parasite stages in the blood of the vertebrate host. Because of the enormous number of parasite species and the challenges of multiplex PCR, initial screening of samples using piroplasmid-specific PCR may be a more cost-effective and efficient technique to identify parasite species, especially during epidemiological studies. Accordingly, 18S rRNA PCR was standardized and optimized on common piroplasmids of different animals like cattle, buffaloes, sheep, g

Molecular detection of Theileria species and Babesia caballi from horses in Nigeria.

Equine piroplasmosis (EP) is an infectious, tick-borne disease caused by the hemoprotozoan parasites, Theileria equi, Babesia caballi, and a recently reported new species, T. haneyi. Infections by these apicomplexan parasites limit performance and cause economic losses for the horse industry. Equine piroplasmosis is widespread in the northern regions of Nigeria, where an increasing portion of the animal population is composed of horses. This disease has remained epidemiologically challenging, especially as the movement of horses increases across Nigeria. In this study, blood samples from 300 h

Evaluation of the in vitro and in vivo inhibitory effect of thymoquinone on piroplasm parasites.

BACKGROUND: Developing new antibabesial drugs with a low toxic effect to the animal and with no resistance from Babesia parasites is in urgent demand. In this concern, the antimalarial, anticancer and antioxidant effect of thymoquinone (TQ), a phytochemical compound found in the plant Nigella sativa, has been reported. Therefore, in the present study, the antibabesial effect of this compound was evaluated on the growth of piroplasm parasites. RESULTS: Significant inhibition (P < 0.05) of the in vitro growth of piroplasm parasites were observed after treatment by TQ with IC50 values of 35.41 ±

History and critical review of Theileria parva (Boleni), the vaccine stock against Zimbabwean cattle theileriosis.

The paper reviews the infectivity, cross-immunization experiments, and cattle vaccination of Zimbabwean cattle-derived Theileria parva (Boleni) sporozoite stabilates produced at the Central Veterinary Laboratory (CVL) in Harare between 1980 and 2003. The Boleni stock was first isolated in July 1978 during a theileriosis outbreak and was shown to be virulent in susceptible cattle. Thereafter, the reactions observed in susceptible cattle produced by different tick stabilates derived from the original have been mostly severe (76%) or moderate (24%). The parasite concentrations in the Boleni vacci

Seroprevalence of Babesia caballi and Theileria equi in the Swiss horse population.

In Switzerland, the prevalence and incidence of equine piroplasma parasite (EPP) infections are unknown. In order to obtain a first insight into the prevalence, a representative sample of 689 sera of horses from Switzerland was serologically tested for the presence of antibodies directed against T. equi and B. caballi using the Indirect Fluorescence Antibody Test (IFAT). A total of 50 (7.3%) horses were seropositive for EPP: overall, the seroprevalence of T. equi was significantly higher than that of B. caballi (p=0.002). The seropositivities in indigenous horses (animals bred and raised in Sw

A PCR study of piroplasms in 166 dogs and 111 horses in France (March 2006 to March 2008).

Positive PCR results for piroplasms were obtained on EDTA blood samples from 166 dogs and 111 horses between March 2006 and March 2008. The organisms were initially identified using common primers, followed by restriction enzyme profiles to determine the species and types of Babesia/Theileria. In 27 dogs and eight horses with positive results, the size of the specimen was insufficient to separate species/types. We identified Babesia canis canis in 105 of the 166 dogs (63%) and Theileria equi in 89 of the 111 horses (80%). Babesia caballi was also present, but rare, in only two Babesia/Theileri

Occurrence of Theileria parva and other haemoprotozoa in cattle at the edge of Hluhluwe-iMfolozi Park, KwaZulu-Natal, South Africa.

Theileria parva, the most important bovine theilerial species in sub-Saharan Africa, causes widespread mortality and morbidity in endemic areas. A survey was conducted using buffy-coat specimens from 60 apparently healthy adult communally herded Nguni-type cattle at the northeastern edge of the Hluhluwe-iMfolozi Park to determine, by means of PCR and Reverse Line Blot (RLB) hybridisation, the occurrence of Theileria and Babesia species. The presence of Trypanosoma species was determined using PCR-RFLP. Results showed that 6.7 % of the specimens were positive for Theileria parva. This significa

Allograft responses can interfere with the development of immunity against Theileria annulata following vaccination with parasite infected cell lines.

Theileria annulata macroschizont-infected cell lines are successfully used as vaccines in several countries. The inoculated animals produce a strong allogeneic response against the MHC antigens of the immunizing cell line followed by an anti-parasite response. Immunity against the parasite wanes in the absence of challenge and re-immunization is sometimes recommended. However, it is not known if allogeneic responses generated by the first immunization with a T. annulata infected cell line will interfere with the boosting of immunity against the parasite at the time of re-immunization with the

Genetic diversity and phylogeography of Hyalomma scupense, the main vector of tropical theileriosis: Global analysis with novel data from Tunisia.

Hyalomma scupense, the main vector of tropical theileriosis, presents taxonomic challenges that impede vector surveillance. This study combines targeted sampling of 20 morphologically-confirmed specimens from El Hessiene, Tunisia (36°50'N 10°12'E) with global GenBank analysis of mitochondrial markers 16S rRNA, 12S rRNA, and COI to assess genetic diversity, phylogeographic structure, and regional gene flow. All Tunisian sequences showed 99.88-100% identity to H. scupense references. Global analysis revealed moderate genetic diversity: 8 genotypes for 16S rRNA (Hd=0.551, π=0.0036), 13 for 12S rR
