Effect of feeding on the tissue free amino acid concentrations in rainbow trout (Oncorhynchus mykiss Walbaum).

This study investigates whether tissue free amino acid (FAA) pools in rainbow trout,Oncorhynchus mykiss (Walbaum), are altered following feeding and the relationships between the amount of food consumed and the FAA pool size. Trout were starved for 7 days to provide baseline data and then refed on day 8. Individual food intake was measured by radiography and the consumption of amino acids (AA) calculated from dietary protein consumption. Total FAA concentrations in the stomach, liver and white muscle were little changed at various times after the meal and this pattern was repeated for the majo…

Gonadotropin releasing hormone (GnRH) neurons project to growth hormone and somatolactin cells in the steelhead trout.

Analysis of gene expression using gonadotropin-releasing hormone (GnRH) antisense oligonucleotide confirmed by immunocytochemical localization the occurrence of GnRH neurons along the nervus terminalis in the steelhead trout (Oncorhynchus mykiss). Double-label immunocytochemistry revealed the distribution of mammalian (m), salmon (s) and chicken II (cII)-type GnRHs and various pituitary hormones. Both sGnRH and mGnRH appeared to be colocalized in the same cells of the nervus terminalis. Chicken GnRH II-immunoreactivity was found only in fibers and terminals. In the younger fish [73 and 186 day…

Cholinergic control of stanniocalcin release in the rainbow trout, Oncorhynchus mykiss, and the American eel, Anguilla rostrata.

An in vivo whole animal 45Ca influx bioassay was used to study the cholinergic control of the release of stanniocalcin (STC) in the rainbow trout (Oncorhynchus mykiss) and the American eel (Anguilla rostrata). In both species calcium influx (JinCa2+) was lowered in response to hypercalcemia induced by intravascular (trout) or intraperitoneal (eel) injections of CaCl2. In trout, this response was blocked by the cholinergic antagonist atropine (0.25 mumol kg-1) and mimicked by the cholinoceptor agonist carbachol (0.25 mumol kg-1). These observations are consistent with a cholinergic stimulation …

Regulation of melatonin production by pineal photoreceptor cells: role of cyclic nucleotides in the trout (Oncorhynchus mykiss).

The light/dark cycle influences the rhythmic production of melatonin by the trout pineal organ through a modulation of the serotonin N-acetyltransferase (NAT) activity. In static organ culture, cyclic AMP (cAMP) levels (in darkness) and NAT activity (in darkness or light) were stimulated in the presence of forskolin, isobutylmethylxanthine, or theophylline. Analogues of cAMP, but not of cyclic GMP, induced an increase in NAT activity. Light, applied after dark adaptation, inhibited NAT activity. This inhibitory effect was partially prevented in the presence of drugs stimulating cAMP accumulati…

Distribution of vasoactive intestinal polypeptide-like immunoreactivity in the olfactory bulb of the rainbow trout (Salmo gairdneri).

The distribution of vasoactive intestinal polypeptide-like structures in the olfactory bulb of the rainbow trout was studied using an indirect-immunoperoxidase technique. Olfactory fibres were very strongly labelled, whereas the fibres or cell bodies in the remaining strata of the olfactory bulb showed no immunoreactivity. In addition, the olfactory nerve fibres were not immunoreactive for methionine- and leucine-enkephalins, motilin, neuropeptide Y, substance P, cholecystokinin-8 and tyrosine-hydroxylase.

Gonadotropin hormone (GtH) receptors in the ovary of the brown trout Salmo trutta L. in vitro studies.

A method has been developed suitable for the study of the binding of salmon (Oncorhynchus tsawytscha) gonadotropic hormone (GtH) to brown trout (Salmo trutta) ovarian particulate fractions. Several proteolytic enzyme inhibitors were tested to preserve the binding capacity of particulate fractions (1000, 20,000, and 110,000g) from yolky oocytes. The best preservation was obtained with trypsin inhibitor. The effects on binding of the pH, time, and temperature of the incubation were investigated. Binding increased linearly with amount of particulate fraction, and is saturable. In both the 20,000 …

In vitro effects of acephate on carbonic anhydrase activity in the blood and gills of rainbow trout, Salmo gairdneri.

Acephate, a water-soluble organophosphate pesticide used to control terrestrial insect pests, may enter aquatic ecosystems in the course of its use and adversely affect fish populations. The in vitro effects of this insecticide on gill and red blood cell (RBC) carbonic anyhdrase (CA) activity in rainbow trout were investigated over a range of 100 mg/1 (0.55 mM) to 50,000 mg/l (273 mM) to assess the manner in which acephate might affect respiratory capacity in exposed fish. Concentrations required to produce 50% inhibition of CA activity in the gill and RBC preparations were 38,000 mg/l (207 mM…

Gross and histological signs of dietary deficiencies of biotin and pantothenic acid in lake trout, Salvelinus namaycush.

We describe the pathologic changes of single or simultaneous dietary deprivations of biotin (B) and pantothenic acid (PA) in lake trout, Salvelinus namaycush. A deficiency of PA produced gross signs of anorexia, inanition, emaciation, gill abnormalities and high mortality. In B-deficient fish, growth retardation reached statistically significant levels (P less than 0.05) after week 10, but gill and liver lesions were observed earlier. Anorexia and reduced weight gain were observed earlier in fish deprived of both nutrients than in those deficient in B alone. All B-deficient trout fed PA surviv…

Field potentials in the olfactory bulb of the rainbow trout (Salmo gairdneri): evidence for a dendrodendritic inhibitory pathway.

Field potentials have been recorded from the olfactory bulb of the teleost fish, Salmo gairdneri. Stimulation of the olfactory nerve generates a complex wave pattern, which when recorded near the surface in the centre of the bulb consists of one positive and three negative components, P, N1, N2 and N3. The N1 has been attributed to depolarization of mitral cell dendrites, N2 to depolarization of the superficial dendrites of granules cells, and P to subsequent hyperpolarization of mitral cells. Possible origins of the N3 wave are discussed. Spikes from single units recorded in the mitral cell l

Bridging a barrier: the role of three-spined stickleback in the trophic transmission of Eubothrium crassum to brown trout.

Helminth parasites often have complex life cycles that include trophic transmission and multiple hosts. Comprehensive knowledge of parasite life cycles and host identity is crucial for understanding their interactions and the ecological role of parasites, however detailed information is available only from a small fraction of species despite a lengthy history of parasitological studies. This study addresses the cestode Eubothrium crassum, a trophically transmitted parasite commonly found in lake-dwelling brown trout (Salmo trutta). The life cycle is considered to involve one intermediate host,…

Bacillus subtilis spores influence the capacity of rainbow trout leukocytes to respond to a posterior viral encounter.

Although the positive effects of probiotics on the general immune status of aquacultured fish has been widely demonstrated using different probiotic strains and fish species, whether probiotics specifically stimulate antiviral responses has only been scarcely addressed. To provide more insights into this matter, we have investigated the capacity of Bacillus subtilis to modulate the transcription of a range on genes related to antiviral responses in rainbow trout (Oncorhynchus mykiss) through two independent experiments. In the first one, fish were fed a B. subtilis-supplemented diet or a contr…

In vitro effects of 11-deoxycorticosterone on hepatocytes and gill epithelial cells of rainbow trout (Oncorhynchus mykiss).

The 11-deoxycorticosterone (DOC) corticosteroid has been recently described as having a potential role in fish, complementing cortisol action through distinct physiological effects. Although systemic effects of 11-deoxycorticosterone (DOC) have been reported, the specific mechanisms mediated through glucocorticoid (GR) and mineralocorticoid (MR) receptors remain poorly understood. Therefore, we evaluated the DOC effects through both receptors in rainbow trout hepatoma-derived (RTH-149) and gill epithelial (RTgill-W1) cell lines. Cultures were pretreated with GR (Mifepristone) or MR (Eplerenone…

Trout Kidney Extracellular Matrix as a Noncytotoxic Scaffold for Promoting Mesenchymal Stem Cell Growth and Specialization.

PURPOSE: The mesonephric kidney of fish retains its capacity for nephrogenesis and regeneration throughout the organism's lifetime. It is hypothesized that the extracellular matrix (ECM) of trout plays a pivotal role in supporting the proliferation and differentiation of resident cells. The ECM is a complex three-dimensional network of macromolecules secreted by cells during early development and is essential for regulating key cellular functions, including adhesion, migration, proliferation, differentiation, and survival. This study aimed to evaluate the effects of decellularized trout kidney…

Detection and Localization of IL-8 and CXCR1 in Rainbow Trout Larvae in Response to Pseudomonas aeruginosa Lipopolysaccharide.

The salmonid industry faces challenges due to the susceptibility of fish to opportunistic pathogens, particularly in early developmental stages. Understanding the immunological capacity during these stages is crucial for developing effective disease control strategies. IL-8R, a member of the G-protein-coupled receptor family, acts as a receptor for Interleukin 8 (IL-8). The binding of IL-8 to IL-8R plays a major role in the pathophysiology of a wide spectrum of inflammatory conditions. This study focused on the immune response capacity of rainbow trout (Oncorhynchus mykiss) larvae by analyzing…

Evidence for infection influencing survival of the freshwater copepod Salmincola californiensis, a parasite of Pacific salmon and trout.

OBJECTIVE: We explore apparent infection of Salmincola californiensis arising during investigations involving this lernaeopodid copepod parasitic on Pacific salmon and trout Oncorhynchus spp. METHODS: We noted occasional unusual coloration of adult female copepods collected from the wild. These females were bright blue and pink in contrast to the cream white coloration characteristic of the copepod. We also observed that similar color patterns developed under laboratory settings when copepod eggs were held for hatching. In paired egg cases, we found consistent hatching failure of blue and pin…

Involvement of miR-495 in the skin pigmentation of rainbow trout (Oncorhynchus mykiss) through the regulation of mc1r.

MicroRNAs (miRNAs) play crucial roles in skin pigmentation in animals. Rainbow trout (Oncorhynchus mykiss) is a key economic fish species worldwide, and skin color directly affects its economic value. However, the functions of miRNAs in rainbow trout skin pigmentation remain largely unknown. Herein, we overexpressed and silenced miR-495 in vitro and in vivo to investigate its functions. The analysis of spatial and temporal expression patterns suggested that miR-495 is a potential regulator during the process of skin pigmentation. In vitro, mc1r was validated as a direct target for miR-495 by d…

Telomerase Activity in Somatic Tissues and Ovaries of Diploid and Triploid Rainbow Trout (Oncorhynchus mykiss) Females.

Telomerase activity has been found in the somatic tissues of rainbow trout. The enzyme is essential for maintaining telomere length but also assures homeostasis of the fish organs, playing an important role during tissue regeneration. The unique morphological and physiological characteristics of triploid rainbow trout, when compared to diploid specimens, make them a promising model for studies concerning telomerase activity. Thus, in this study, we examined the expression of the Tert gene in various organs of subadult and adult diploid and triploid rainbow trout females. Upregulated Tert mRNA …
