The ubiquitin-proteasome system (UPS) is a major proteolytic system that regulates the degradation of intracellular proteins in the heart. The UPS regulates the turnover of misfolded and damaged proteins, in addition to numerous cellular processes, by affecting the stability of short-lived proteins such as transcription factors and cell signaling pathways. The UPS is tightly regulated by the specificity of ubiquitin ligases that recognize specific substrates and direct the addition of ubiquitin, targeting the substrates for degradation by the 26S proteasome. An increasing number of cardiac ubi

Huntington disease and its related autosomal-dominant polyglutamine (pQ) neurodegenerative diseases are characterized by intraneuronal accumulation of protein aggregates. Studies on protein aggregates have revealed the importance of the ubiquitin-proteasome system as the front line of protein quality control (PQC) machinery against aberrant proteins. Recently, we have shown that the autophagy-lysosomal system is also involved in cytoplasmic aggregate degradation, but the nucleus lacked this activity. Consequently, the nucleus relies entirely on the ubiquitin-proteasome system for PQC. Accordin

Huntington's disease and several of the spinocerebellar ataxias are caused by the abnormal expansion of a CAG repeat within the coding region of the disease gene. This results in the production of a mutant protein with an abnormally expanded polyglutamine tract. Although these disorders have a clear monogenic cause, each polyglutamine expansion mutation is likely to cause the dysfunction of many pathways and processes within the cell. It has been proposed that the ubiquitin proteasome system is impaired in polyglutamine expansion disorders and that this contributes to pathology. However, this

Accurate semen analysis is an important issue in the swine industry. We evaluated two candidate fertility marker proteins associated with sperm cytoplasmic droplet (CD), including 15-lipoxygenase (15-LOX) and ubiquitin (UBI) in a controlled single-sire artificial insemination (AI) trial. Ejaculates (n=116) were collected from 18 fertile Large White boars monthly for 8 mo, and analyzed by semi-quantitative, densitometry-based Western blotting and flow cytometry with antibodies against 15-LOX and UBI. Data were correlated with farrowing rates (FR) and total numbers of piglets born (TNB) from 175

Dementia with ubiquitinated neuronal inclusions has been described only with frontotemporal dementia (FTD). The authors report a patient with progressive FTD accompanied by prominent impairments in visuospatial cognitive functions. Pathology was characterized by ubiquitin-positive intranuclear and cytoplasmic neuronal inclusions. Cortical pathology was widespread and posteriorly accentuated but spared the hippocampal dentate gyrus.

Intraneuronal inclusions containing ubiquitylated filamentous protein aggregates are a common feature of many of the major human neurodegenerative disorders, including Alzheimer's and Parkinson's disease. Loss of function mutations in enzymes of the ubiquitin conjugation/deconjugation pathway are sufficient to cause familial forms of neurodegenerative diseases, suggesting that failure of ubiquitin-mediated proteolysis could also be central to inclusion formation in the more common sporadic cases. Examination of ubiquitin-positive inclusions at the protein level provides evidence of attempted p

alpha-Synuclein is one of the major components of intracellular fibrillary aggregates in the brains of a subset of neurodegenerative disorde … itin. These results strongly suggest that phosphorylated alpha-synuclein is targeted to mono- and diubiquitination in synucleinopathy brains, which may have implications for mechanisms of these diseases.

An 8 kDa ubiquitin-like peptide (ULP) was isolated by high performance liquid chromatography from the rabbit vitreous humor, and the N-terminal amino acid sequence of this peptide showed complete homology with ubiquitin. Western blot revealed the presence of free ULP in both the iris-ciliary (IC) complex and the aqueous humor extracts. In the IC complex, fluorescence and immunoelectron microscopy detected high concentrations of ULP in the posterior epithelial cells, suggesting this tissue as a possible source of ULP in the ocular fluids. Significantly, this is the first time that the presence

Eosinophilic hyaline inclusions were consistently seen in the perinuclear cytoplasm of suprabasal keratinocytes in lichen amyloidosus. The inclusions, negative with amyloid staining, were immunoreactive for ubiquitin and cytokeratin, and ultrastructurally showed aggregations of fine filaments of two sizes (central thin and peripheral thick). The thin filaments were the main component in the upper epidermal layer. Four monoclonal antibodies (AE1, AE3, KL1 and CAM5.2) and one antiserum (WSS) were used for characterizing cytokeratin expression. The AE1 antibody normally stained the basal cells, b

Colorectal cancer (CRC) progression is profoundly influenced by the tumor immune microenvironment (TME), which orchestrates the interplay be … mponents of the immune regulatory network in CRC, modulating their stability and activity through deubiquitination. This interaction ultimately influences the balance between pro-tumor and anti-tumor immune responses within the TME. Our findings provide novel insights into the complex interplay between USP11 and the TME in CRC, suggesting that USP11 functions as …

BACKGROUND: Although both RNF214 and CAV1 are implicated in tumor progression and signaling regulation, the role of the RNF214-CAV1 axis in prostate cancer (PCa) remains unclear. This study aims to explore how RNF214 modulates CAV1 stability via ubiquitination and its influence on PCa progression and mTOR signaling.
METHODS: The expression patterns and functional impact of RNF214 were assessed using clinical PCa tissues, cultured cell lines, and mouse xenograft models. Gene silencing experiments were conducted to evaluate effects on cell proliferation, migration, and signaling activity. Prote…

Amyloid precursor protein (APP), a type I transmembrane protein, is closely related to the pathogenesis of Alzheimer's disease (AD). Amyloid … (VHL) protein, a subunit of an E3 ligase, recognizes the cytoplasmic domain of APP and mediates its ubiquitination. VHL-mediated ubiquitination facilitates the sorting of membrane APP into intraluminal vesicles of multivesicular bodies (MVBs) and subsequent degradation in lysosomes. Therefore, the loss of VHL accelerates Aβ plaque deposition and memory deficits …

Temozolomide (TMZ) resistance in glioblastoma (GBM) is often linked to high MGMT protein levels, but MGMT-deficient tumors can still acquire resistance, implicating MGMT-independent repair pathways. Here, we identify an oxidative stress-responsive LMO7-ALKBH2 ubiquitination mechanism that regulates ALKBH2 stability and modulates alkylation damage tolerance in GBM. The E3 ligase LMO7 directly binds ALKBH2 and promotes K48-linked polyubiquitination and proteasomal degradation of ALKBH2, with Lys7 serving as a major ubiquitin acceptor site. Oxidative stress enhances LMO7-dependent ALKBH2 ubiquiti…

UNLABELLED: Methylenetetrahydrofolate dehydrogenase 2 (MTHFD2), a pivotal enzyme in one-carbon metabolism, is significantly overexpressed in tongue squamous cell carcinoma (TSCC) and correlates with poor patient prognosis. Functional studies demonstrate that MTHFD2 drives TSCC proliferation and migration in vitro and in vivo, whereas its inhibition suppresses tumor progression. Mechanistically, MTHFD2 orchestrates a dual posttranslational modification cascade: Through its enzymatic activity, it simultaneously induces forkhead box O1 (FOXO1) hypermethylation and ubiquitination, ultimately trigg…

Small-molecule inhibitors represent a novel potential therapeutic for tumors. Our previous work identified SIC-19, a small molecule compound that induces degradation of salt-inducible kinase 2 (SIK2), leading to apoptosis and reduced proliferation in tumor, expecting to be a promising clinical treatment for ovarian cancer. While we initially observed that SIC-19 triggers SIK2 ubiquitination, the underlying mechanism was unknown. Here we elucidate this process by demonstrating that SIC-19 promotes the interaction between SIK2 and the CUL4B-DDB1 E3 ubiquitin ligase. Mass spectrometry discovered …

Alcoholic liver fibrosis (ALF) is a chronic liver disease caused by long-term excessive alcohol consumption. Increasing evidence indicates that ferroptosis is a key contributor to the development and progression of liver fibrosis; however, its precise molecular mechanisms in ALF remain unclear. In this study, we investigated the role of TRIM21 in regulating the ubiquitination of IDO1 to modulate ferroptosis and thereby alleviate ALF. An ALF mouse model was established using the Lieber-DeCarli ethanol diet combined with ethanol gavage for 8 weeks, and liver fibrosis was evaluated via histologic…
