[Studies on the regulatory mechanism of Staphylococcus aureus virulence].

Staphylococcus aureus causes various diseases against humans, including skin infection, pneumonia, food poisoning, and meningitis. Methicillin-resistant S. aureus (MRSA) is resistant to a broad range of antibiotics, causing serious clinical problems. In this review, I summarize our studies to evaluate S. aureus virulence and identify novel virulence regulators. First, we utilized silkworms as an infection model of S. aureus and identified novel virulence factors of S. aureus. Some of the virulence factors interact with RNA in bacterial cells and regulate the expression of virulence factors. Se…

The exopolysaccharide matrix: a virulence determinant of cariogenic biofilm.

Many infectious diseases in humans are caused or exacerbated by biofilms. Dental caries is a prime example of a biofilm-dependent disease, r … microenvironments/niches. Further understanding on how the matrix modulates microbial activity and virulence expression could lead to new approaches to control cariogenic biofilms.

The virulence of Streptococcus mutans and the ability to form biofilms.

In some diseases, a very important role is played by the ability of bacteria to form multi-dimensional complex structure known as biofilm. The most common disease of the oral cavity, known as dental caries, is a top leader. Streptococcus mutans, one of the many etiological factors of dental caries, is a microorganism which is able to acquire new properties allowing for the expression of pathogenicity determinants determining its virulence in specific environmental conditions. Through the mechanism of adhesion to a solid surface, S. mutans is capable of colonizing the oral cavity and also of fo…

Cytoadherence and virulence - the case of Plasmodium knowlesi malaria.

BACKGROUND: Cytoadherence of infected red blood cells to brain endothelium is causally implicated in malarial coma, one of the severe manifestations of falciparum malaria. Cytoadherence is mediated by specific binding of variant parasite antigens, expressed on the surface of infected erythrocytes, to endothelial receptors including, ICAM-1, VCAM and CD36. In fatal cases of severe falciparum malaria with coma, blood vessels in the brain are characteristically congested with infected erythrocytes. Brain sections from a fatal case of knowlesi malaria, but without coma, were similarly congested wi

Hookworm virulence factors: making the most of the host.

Hookworm disease from Necator americanus and Ancylostoma duodenale affects approximately 700 million people, with N. americanus being the predominant species. Unlike other pathogens (e.g., bacterial infections), where "virulence" is described in regards to acute pathogenesis and case-fatality, hookworms are well-evolved, multicellular parasites that establish long-term infections in their human hosts with a subtle and chronic, but insidious, pathogenesis, usually in the form of iron deficiency anemia from parasite blood feeding that, over time, has devastating effects on the human host especia…

Brucella adaptation and survival at the crossroad of metabolism and virulence.

"In vivo" bacterial nutrition, i.e. the nature of the metabolic network and substrate(s) used by bacteria within their host, is a fundamental aspect of pathogenic or symbiotic lifestyles. A typical example are the Brucella spp., facultative intracellular pathogens responsible for chronic infections of animals and humans. Their virulence relies on their ability to modulate immune response and the physiology of host cells, but the fine-tuning of their metabolism in the host during infection appears increasingly crucial. Here we review new insights on the links between Brucella virulence and meta…

Molecular analysis of virulent determinants of enterovirus 71.

Enterovirus 71 (EV71) is the most important causative agent of hand, foot and mouth disease (HFMD) in children. In most cases, it is a self-limiting illness. However some EV71 infectious cases can develop severe clinical outcomes, such as encephalitis, meningitis, poliomyelitis like paralysis, and even death. To identify the determinants of virulence, the deduced amino acid sequence of polyprotein and nucleotide sequence of 5'-NTR and 3'-NTR in 25 SC-EV71 strains (strains from severe cases) and 31 MC-EV71 strains (strains from mild cases) were analyzed. Results showed four amino acids on two p…

[Melanin and its role on the virulence of Cryptococcus neoformans].

Melanins can be produced by some species of pathogenic bacteria, helminths and fungi. The production of melanin appears to contribute to microbial pathogenesis owing to its potential for protection against host defence systems. Melanin synthesis in Cryptococcus neoformans has been associated with virulence by the ability to protect against phagocytosis, oxidative injury and phagocytic killing. Moreover, it was shown that, melanization of C.neoformans has involved in protection against some antifungal compounds. The detection of the association between melanin and microbial virulence might prov…

Characterization of virulence of Leptospira isolates in a hamster model.

Effort has been made to identify protective antigens in order to develop a recombinant vaccine against leptospirosis. Several attempts failed to conclusively demonstrate efficacy of vaccine candidates due to the lack of an appropriate model of lethal leptospirosis. The purposes of our study were: (i) to test the virulence of leptospiral isolates from Brazil, which are representative of important serogroups that cause disease in humans and animals; and (ii) to standardize the lethal dose 50% (LD(50)) for each of the virulent strains using a hamster (Mesocricetus auratus) model. Five of seven Br…

A role of Burkholderia pseudomallei flagella as a virulent factor.

Burkholderia pseudomallei is an agent of melioidosis and is closely related to avirulent B. thailandensis. Burkholderia thailandensis has a 15-bp deletion within the variable region of the flagellin gene fliC compared with B. pseudomallei. The difference in the fliC gene might be related to virulence. In the present study, the invasion, internalization and intracellular replication of both phagocytic (mouse macrophage cell line RAW264.7) and non-phagocytic cells (human lung epithelial cell line A549) of B. pseudomallei fliC knockout mutant (MM35) complemented with its own fliC (Cp) or with B. …

Comparison of Listeria monocytogenes virulence in a mouse model.

Listeriosis results from exposure to the foodborne pathogen Listeria monocytogenes. Although many different strains of L. monocytogenes are isolated from food, no definitive tests currently predict which isolates are most virulent. The objectives of this study were to address two major data gaps for risk assessors, variability among L. monocytogenes strains in pathogenicity and virulence. Strains used in our monkey clinical trial or additional food isolates were evaluated for their virulence and infectivity in mice. All strains were equally pathogenic to immunocompromised mice, causing deaths …

The alpha-toxin of Clostridium septicum is essential for virulence.

Clostridium septicum is the causative agent of spontaneous gas gangrene or atraumatic myonecrosis, a sudden and frequently fatal infection that is increasingly associated with malignancy of the colon. Little is known about the disease process although the focus of virulence studies has been the alpha-toxin, a pore-forming cytolysin that is encoded by the csa gene and secreted as an inactive protoxin. Until now a lack of techniques for the genetic manipulation of C. septicum has hindered the use of molecular approaches to understand pathogenesis. By introducing plasmids by conjugation from Esch…

Role of HtrA in the virulence and competence of Streptococcus pneumoniae.

HtrA is a major virulence factor of Streptococcus pneumoniae (the pneumococcus). Deletion of the gene for HtrA from strain D39 of the pneumococcus completely abolished its virulence in mouse models of pneumonia and bacteremia, while the virulence of a second strain (TIGR4) was dramatically reduced. HtrA-negative mutants induced much less inflammation in the lungs during pneumonia than the wild type. HtrA is involved in the ability of the pneumococcus to grow at high temperatures, to resist oxidative stress, and to undergo genetic transformation. The expression and cellular location of several …

Pathogen survival in the external environment and the evolution of virulence.

Recent studies have provided evolutionary explanations for much of the variation in mortality among human infectious diseases. One gap in this knowledge concerns respiratory tract pathogens transmitted from person to person by direct contact or through environmental contamination. The sit-and-wait hypothesis predicts that virulence should be positively correlated with durability in the external environment because high durability reduces the dependence of transmission on host mobility. Reviewing the epidemiological and medical literature, we confirm this prediction for respiratory tract pathog…

Variants of Sporothrix schenckii with attenuated virulence for mice.

Strains of Sporothrix schenckii preserved under mineral oil were examined for virulence in BALB/c mice. The mice were inoculated with S. schenckii conidia and development of cutaneous lesions, signs of inactivity, weight loss, survival rates, number of viable yeast cells in lung and spleen, splenomegaly and organ lesions were evaluated. After intravenous injection of 7.5 x 10(6) conidia, two of five S. schenckii strains were unable to induce systemic disease and to kill the mice, only producing cord-like lesions on the tail that regressed with mouse maturation. Very small numbers of viable cel…

Is streptolysin S of group A streptococci a virulence factor?

The possible role played by streptolysin S (SLS) of group A streptococci in the pathophysiology of streptococcal infections and in post-streptococcal sequelae is discussed. The following properties of SLS justify its definition as a distinct virulence factor: 1) its presence on the streptococcus surface in a cell-bound form, 2) its continuous and prolonged synthesis by resting streptococci, 3) its non-immunogenicity, 4) its extractability by serum proteins (albumin, alpha lipoprotein), 5) its ability to become transferred directly to target cells while being protected from inhibitory agents in…

A porcine model for the evaluation of virulence of Bordetella bronchiseptica.

Studies of virulence factors of Bordetella bronchiseptica require a suitable system. Such a system was devised in colostrum-deprived, caesarean-derived pigs, aged 7 d. In two different experiments, pigs (n = 11) were inoculated intranasally with 10(6) colony-forming units of the virulent strain 4609. In the same way, further pigs (n = 11) were inoculated with a strain (B133) of unknown virulence. No significant differences between 4609 and B133 colonization were seen. However, colonization of the turbinates was significantly higher than that of the trachea, lung and tonsil, and a significantly…

Iron and virulence of Haemophilus ducreyi in a primate model.

BACKGROUND AND OBJECTIVES: The pathogenesis of chancroid is currently unclear. There are discrepancies between the virulence of Haemophilus ducreyi in the human and animal experiments. Iron availability influences the virulence of many pathogens. The objective of this study was to investigate the role of iron in virulence of H. ducreyi. STUDY DESIGN: Intradermal inoculation of the primate Macaca mulatta was studied as an animal model for pathogenesis of chancroid. This model then was used to study the influence of iron availability on virulence of H. ducreyi. Eleven strains of H. ducreyi with…

Short-sighted evolution and the virulence of pathogenic microorganisms.

For some microorganisms, virulence may be an inadvertent consequence of mutation and selection in the parasite population, occurring within a host during the course of an infection. This type of virulence is short-sighted, in that it engenders no advantage to the pathogen beyond the afflicted host. Bacterial meningitis, poliomyelitis and AIDS are three candidates for this model of the evolution of virulence.
