Prenatal alcohol exposure blunts interleukin-1-induced ACTH and beta-endorphin secretion by immature rats.

The functional relationship between the immune and the hypothalamic-pituitary-adrenal (HPA) axis [and in particular the release of pro-opiomelanocortin (POMC)-related peptides and corticosteroids induced by interleukins (ILs)] is essential for coordinating the appropriate immune responses to pathogens. Exposure of pregnant mammalian females to alcohol results in abnormal immune functions in the offspring, as well as in altered HPA axis activity. We therefore tested the hypothesis that prenatal alcohol exposure might modify the stimulatory action of ILs on the HPA axis of the pups, thus providi

Visual expectation and dimensions of infant information processing.

Visual expectation was assessed in 103 black 6.5-month-olds using Haith, Hazan, and Goodman's paradigm and related to performance on standard developmental assessments and tests of information processing skill. As expected, percent anticipations was higher and RT lower than in 3.0-month-olds previously tested. Split-half and left-right correlations for the RT measures were moderate and similar to those previously reported, as was split-half reliability for percent anticipations. The 2 RT measures were related to fixation duration on both visual recognition memory (VRM) and cross-modal transfer

Permanent neuronal deficits in rats exposed to alcohol during the brain growth spurt.

The purpose of this study was to determine whether developmental alcohol exposure could induce permanent neuronal deficits, whether the peak blood alcohol concentration (BAC) influences the severity of the effects, and whether the effects are gender related. Rat pups were reared artificially over postnatal days (PD) 4 through 11 (a period of rapid brain growth, comparable to part of the human third trimester). Alcohol treatments were administered on PD 4 through 9. Patterns of alcohol exposure that produce different peak BACs have been shown to affect differentially the amount of brain weight

Effects of prenatal ethanol and long-chain n-3 fatty acid supplementation on development in mice. 2. Fatty acid composition of brain membrane phospholipids.

Pregnant mice were fed equivalent daily amounts of a liquid diet containing 25% (kcal) ethanol, or with maltose dextrin substituted isocalorically for ethanol. The diet also contained 20% oil; this was either of two mixtures, one comprised of predominantly n-6 (18:2n-6) fatty acids, and the other containing an equivalent amount of n-6, but supplemented with a source of long chain n-3 (20:5n-3, 22:6n-3) fatty acids. An additional control group was fed lab chow ad libitum. The treatment was implemented from day 7 to 17 of gestation, whereafter all groups were fed lab chow. Birth occurred on day

[Pharmacological correction of experimental alcoholic embryopathy].

In the experiments on the progeny of ethanol-exposed rats it was shown that consumption of 15% alcohol instead of water during pregnancy resulted in the worsening of shuttlebox avoidance learning and decrease in succinate dehydrogenase activity in the visual and sensorimotor cortex. These data are indicative of cerebral hypoxia during embryogenesis. The injection of synthetic opioid peptide dalargin during critical periods of development (at the end of embryogenesis and early ontogeny) prevented partially the disturbances of higher nervous activity and tissue breathing which were induced by al

Neuropathological effects of alcohol on the developing nervous system.

The formation of functional neuronal networks in the developing nervous system is dependent on three mechanisms which have been shown to be susceptible to disturbance by alcohol exposure. These are cell acquisition, cell migration and cellular maturation. Cell acquisition can be reduced by either impaired proliferation or increased cell deletion. Effects of alcohol on cell proliferation, both early in development and in the postnatal cerebellum, are overshadowed by cell loss, which in the cerebellum may affect both small and large neurones. Disturbed cell migration in the developing nervous sy

Prevention possibility for brain dysfunction in rat with the fetal alcohol syndrome--low-zinc-status and hypoglycemia.

As a treatable cause of CNS dysfunctions in the fetal alcohol syndrome (FAS), low-zinc-status in addition to hypoglycemia has been investigated in experimental rat models. During the premating period female rats of an ethanol group and a control group received 30% ethanol (E) and water (W), respectively. During pregnancy, some of both groups received zinc or nicotinamide-adenine dinucleotide (NAD) or nicotinamide throughout pregnancy and glucose for gestational day (gd) 15 to 19 with E or W. Independent of maternal blood glucose levels, maternal insulin levels were lower on gd 15 and 18 in the

Long-term protective effect of PACAP in a fetal alcohol syndrome (FAS) model.

Prenatal ethanol exposure provokes teratogenic effects, due to oxidative stress and massive neuronal apoptosis in the developing brain that result in lifelong behavioral abnormalities. PACAP exerts anti-oxidative and neuroprotective activities on neuronal cells, and prevents ethanol neurotoxicity. The present study focused on the ability of PACAP to protect the brain of 30-day-old mice (P30) from prenatal alcohol exposure induced oxidative damage and toxicity. Pregnant mice were divided randomly into 4 groups, i.e. control group, ethanol group (1.5 g/kg ip daily injection), PACAP group (5 μg i

Clinical profile and prevalence of fetal alcohol syndrome in an isolated community in British Columbia.

The authors were invited by the band council to carry out a study to determine the prevalence of alcohol embryopathy among children in a native Indian community in British Columbia. The mothers of the 123 children aged 18 years or less who lived in the community were interviewed. In addition, educational screening was carried out for children in grades 1 through 12, and 116 of the children underwent medical examination. A diagnosis of fetal alcohol syndrome or fetal alcohol effects (FAS/FAE) was made in 22 children aged 3 to 18 years. Each of these children was matched for age and sex with an

Magnetic resonance microscopy defines ethanol-induced brain abnormalities in prenatal mice: effects of acute insult on gestational day 8.

BACKGROUND: Magnetic resonance microscopy (MRM), magnetic resonance imaging (MRI) at microscopic levels, provides unprecedented opportunities to aid in defining the full spectrum of ethanol's insult to the developing brain. This is the first in a series of reports that, collectively, will provide an MRM-based atlas of developmental stage-dependent structural brain abnormalities in a Fetal Alcohol Spectrum Disorders (FASD) mouse model. The ethanol exposure time and developmental stage examined for this report is gestational day (GD) 8 in mice, when the embryos are at early neurulation stages; s…

Distinct neurobehavioral dysfunction based on the timing of developmental binge-like alcohol exposure.

Gestational exposure to alcohol can result in long-lasting behavioral deficiencies generally described as fetal alcohol spectrum disorder (FASD). FASD-modeled rodent studies of acute ethanol exposure typically select one developmental window to simulate a specific context equivalent of human embryogenesis, and study consequences of ethanol exposure within that particular developmental epoch. Exposure timing is likely a large determinant in the neurobehavioral consequence of early ethanol exposure, as each brain region is variably susceptible to ethanol cytotoxicity and has unique sensitive per

Ethanol increases fetal human neurosphere size and alters adhesion molecule gene expression.

BACKGROUND: Ethanol (ETOH) consumption by pregnant women can result in Fetal Alcohol Spectrum Disorder (FASD). To date, the cellular targets and mechanisms responsible for FASD are not fully characterized. Our aim was to determine if ETOH can affect fetal human brain-derived neural progenitor cells (NPC). METHODS: Neural progenitor cells were isolated by positive selection from normal second trimester fetal human brains (n = 4) and cultured, for up to 72 hours, in mitogenic media containing 0, 1, 10, or 100 mM ETOH. From 48 to 72 hours in culture, neurospheres generated in these conditions we

Fetal alcohol syndrome and population level health care usage in British Columbia, Canada.

BACKGROUND: The literature indicates that youth with Fetal Alcohol Syndrome (FAS) may experience high rates of both physical and mental health issues compared to youth without FAS. However, there is little population level health data available for youth with FAS, particularly for youth transitioning from pediatric to adult healthcare services. OBJECTIVE: The objective of this study was to compare health care usage of youth with Fetal Alcohol Syndrome to youth without any intellectual/developmental disabilities (IDD). METHODS: This study used a retrospective cohort design and population-leve

Erythropoietin improves spatial and nonspatial memory defects by suppressing oxidative damage, inflammation and apoptosis against ethanol neurotoxicity in the developing male rat hippocampus.

Excessive prenatal exposure to ethanol leads to a condition called fetal alcohol spectrum disorder (FASD). The neurotoxicity of alcohol causes changes in the hippocampus of animals during this time, resulting in impaired hippocampus-related functions, including memory/learning and cognition. The liver and kidneys produce erythropoietin (EPO). The synthesis of EPO by immature neurons also plays a decisive role in the embryonic stage. Also, exogenous EPO exerts its neurocognitive effects in the developing brain under pathophysiological conditions. The aim of this study was to investigate the prot

CB1R-Mediated Activation of Caspase-3 Causes Epigenetic and Neurobehavioral Abnormalities in Postnatal Ethanol-Exposed Mice.

Alcohol exposure can affect brain development, leading to long-lasting behavioral problems, including cognitive impairment, which together is defined as fetal alcohol spectrum disorder (FASD). However, the fundamental mechanisms through which this occurs are largely unknown. In this study, we report that the exposure of postnatal day 7 (P7) mice to ethanol activates caspase-3 via cannabinoid receptor type-1 (CB1R) in neonatal mice and causes a reduction in methylated DNA binding protein (MeCP2) levels. The developmental expression of MeCP2 in mice is closely correlated with synaptogenesis and

Neurobiology and neurodevelopmental impact of childhood traumatic stress and prenatal alcohol exposure.

PURPOSE: Research reveals that prenatal alcohol exposure and child trauma (i.e., abuse, neglect, sexual abuse) can have deleterious effects on child development across multiple domains. This study analyzed the impact on childhood neurodevelopment of prenatal alcohol exposure and postnatal traumatic experience compared to postnatal traumatic experience alone. Although the harmful effects of both have been well documented individually, there is no research documenting the concurrent effects of prenatal alcohol exposure and postnatal trauma on a child's developmental process. METHOD: Transdiscip
