Application of Fmoc-SPPS, Thiol-Maleimide Conjugation, and Copper(I)-Catalyzed Alkyne-Azide Cycloaddition "Click" Reaction in the Synthesis of a Complex Peptide-Based Vaccine Candidate Against Group A Streptococcus.

Fmoc solid-phase peptide synthesis (SPPS) is the most common approach used to synthesize natural and unnatural peptides. However, the synthesis of sequences longer than 30-60 amino acids is associated with a drastic reduction in peptide quality. Thus, large and complex peptides are normally synthesized as fragments, which are then conjugated together. Here, we describe the synthesis of a large, branched peptide, a multi-epitope vaccine candidate against Group A Streptococcus, with the help of microwave-assisted Fmoc-SPPS, thiol-maleimide conjugation, and copper(I)-catalyzed alkyne-azide cycloa

Characterization of folding-sensitive nanobodies as tools to study the expression and quality of protein particle immunogens.

Vaccination is as one of the most beneficial biopharmaceutical interventions against pathogens due to its ability to induce adaptive immunity through targeted activation of the immune system. Each vaccine needs a tailor-made set of tests in order to monitor its quality throughout the development and manufacturing. The analysis of the conformational state of protein nanoparticles is one of the key steps in vaccine quality control. The enzyme lumazine synthase from Brucella spp. (BLS) acts as a potent oral and systemic immunogen. BLS has been used as a carrier of foreign peptides, protein domain

Design and Pharmacological Characterization of a Novel Antithrombotic P2Y1 Receptor-Based Vaccine.

Platelet activation processes begin when injury to blood vessels exposes the subendothelial matrix, leading platelets to attach to it, where they become activated and exert their hemostatic function. Excessive platelet aggregation is associated with thrombotic disorders such as arterial thrombosis. To manage such diseases, medications that inhibit thrombosis are continuously sought, despite potential drawbacks that include hemorrhage. This study described the development of a novel peptide-based vaccine that targets the purinergic ADP P2Y1 receptor (abbreviated EL2Vac) and its pharmacological

Immunoinformatic Approach for Rational Identification of Immunogenic Peptides Against Host Entry and/or Exit Mpox Proteins and Potential Multiepitope Vaccine Construction.

COVID-19 has intensified humanity's concern about the emergence of new pandemics. Since 2018, epidemic outbreaks of the mpox virus have become worrisome. In June 2022, the World Health Organization declared the disease a global health emergency, with 14 500 cases reported by the Centers for Disease Control and Prevention in 60 countries. Therefore, the development of a vaccine based on the current virus genome is paramount in combating new cases. In view of this, we hypothesized the obtainment of rational immunogenic peptides predicted from proteins responsible for entry of the mpox virus into

IgG subclass distribution of anti-hCG and anti-diphtheria toxoid antibodies in women immunized with a beta-hCG based immunocontraceptive vaccine.

IgG subclass distribution of antibodies against human chorionic gonadotropin (hCG) and diphtheria toxoid (DT) was determined in the sera of women recruited into Phase II clinical trials of a beta-hCG based immunocontraceptive vaccine. Serum samples from 30 different individuals were analyzed. Anti-hCG antibody response was predominantly of IgG1 subclass, with a mean titer of 537.94 +/- 560 antibody units (AU). The other 3 subclasses showed considerably lower mean levels (IgG2, 16.46 +/- 8.33; IgG3, 3.22 +/- 8.48; IgG4, 56.65 +/- 82.60 AU). A good correlation was observed between the anti-hCG I

Designing a multi-epitope vaccine candidate to combat MERS-CoV by employing an immunoinformatics approach.

Currently, no approved vaccine is available against the Middle East respiratory syndrome coronavirus (MERS-CoV), which causes severe respiratory disease. The spike glycoprotein is typically considered a suitable target for MERS-CoV vaccine candidates. A computational strategy can be used to design an antigenic vaccine against a pathogen. Therefore, we used immunoinformatics and computational approaches to design a multi-epitope vaccine that targets the spike glycoprotein of MERS-CoV. After using numerous immunoinformatics tools and applying several immune filters, a poly-epitope vaccine was co

The Mycoplasma hyopneumoniae protein Mhp274 elicits mucosal and systemic immune responses in mice.

BACKGROUND: Mycoplasma hyopneumoniae is the etiological agent of mycoplasmal pneumonia of swine (MPS). Commercial vaccines provide partial protection and do not prevent the colonization and transmission of M. hyopneumoniae. The bottleneck in the development of more effective vaccines for MPS is the stimulation of effective immune responses in the host. The purpose of the present study was to evaluate the immune responses of immunodominant proteins Mhp170, Mhp274 and Mhp336 in BALB/c mice. METHODS: The recombinant Mhp170 (rMhp170), Mhp274 (rMhp274), and Mhp336 (rMhp336) proteins were purified

Reduction in Herpes Zoster Antiviral Use Since the Introduction of the Live-Attenuated Zoster Vaccine on Australia's National Immunisation Program: A Population-Based Study from 1994 to 2019.

INTRODUCTION: Zostavax, the live-attenuated vaccine used to prevent herpes zoster (HZ), has been available to individuals aged 70 and 71-79 years (phased catch-up) via Australia's National Immunisation Program (NIP) since 2016. There are limited data characterising the incidence of HZ at the level of the Australian population. National prescription data for antivirals used to treat HZ may be used as a proxy for HZ incidence. We aimed to examine trends in antiviral prescriptions supplied for the treatment of HZ in Australia pre- and post-2016, and to assess whether Zostavax's inclusion on the N

Unveiling promising immunogenic targets in Coxiella burnetii through in silico analysis: paving the way for novel vaccine strategies.

BACKGROUND: Coxiella burnetii, an intracellular pathogen, serves as the causative agent of zoonotic Q fever. This pathogen presents a significant threat due to its potential for airborne transmission, environmental persistence, and pathogenicity. The current whole-cell vaccine (WCV) utilized in Australia to combat Q fever exhibits notable limitations, including severe adverse reactions and limited regulatory approval for human use. This research employed the reverse vaccinology (RV) approach to uncover antigenic proteins and epitopes of C. burnetii, facilitating the development of more potent

BACKGROUND: Acquired immune deficiency syndrome (HIV/AIDS) has been a major global health concern for over 38 years. No safe and effective preventive or therapeutic vaccine has been developed although many products have been investigated. Computational methods have facilitated vaccine developments in recent decades. Among HIV-1 proteins, p24 and Nef are two suitable targets to provoke the cellular immune response. However, the fusion form of these two proteins has not been analyzed in silico yet. OBJECTIVE: This study aimed at the evaluation of possible fusion forms of p24 and Nef in order to

Real-World Effectiveness of Recombinant Zoster Vaccine in Allogeneic Hematopoietic Cell Transplant Recipients.

Herpes zoster (HZ) is a frequent and clinically significant complication following allogeneic hematopoietic cell transplantation (allo-HCT), arising from delayed immune reconstitution and impaired varicella-zoster virus-specific immunity. Although acyclovir prophylaxis reduces early risk, prolonged use is often limited by toxicity and poor adherence. The recombinant zoster vaccine (RZV), a non-live adjuvanted subunit vaccine, has demonstrated high efficacy in immunocompetent adults and in several immunocompromised populations; however, real-world data in allo-HCT recipients remain scarce. Immunologic assessment of the impact of SARS-CoV-2 vaccine booster doses on humoral immunity: a cross-sectional study in morocco.

To assess the impact of the SARS-CoV-2 booster dose on the immune response against COVID-19, we conducted a cross-sectional study in the Casablanca-Settat region of Morocco. The study included 2,802 participants from 16 provinces, all of whom had received three doses of a SARS-CoV-2 vaccine. IgG antibodies targeting the S1 RBD subunit of the SARS-CoV-2 spike protein were quantified using the SARS-CoV-2 IgG II Quant assay and measured on the Abbott Architect i2000SR instrument. Adjusted seroprevalence of anti-RBD antibodies showed that participants who received two doses of ChAdOx1-S followed b

Antigenic analysis for vaccines and diagnostics.

Coxiella burnetii infection is frequently unrecognized or misdiagnosed, as symptoms generally mimic an influenza-like illness. However, the disease (Q fever) may result in chronic infection, usually manifesting as potentially fatal endocarditis. The development of a chronic fatigue-like sequela may also occur. Infected ruminants are the major reservoir for infection in humans, primarily through exposure to birth products or aerosols that transmit the bacterium over wide regions. A vaccine against C. burnetii infection has been in use in Australia for abattoir and agricultural workers for many

Engineering Komagataella phaffii for secreted and surface-displayed production of SARS-CoV-2 receptor-binding domain and its immunogenicity evaluation.

BACKGROUND: The receptor-binding domain (RBD) of the SARS-CoV-2 spike protein represents a key antigen for vaccine development due to its critical role in the ACE2 receptor recognition. Yeast-based expression systems, particularly Komagataella phaffii, offer scalable and cost-effective platforms for recombinant protein production. In addition to secretion, cell surface display provides an alternative strategy enabling direct antigen delivery, including applications via mucosal routes. RESULTS: In this study, we engineered K. phaffii strains for the production of SARS-CoV-2 RBD in two formats:

Design of a Multi-Epitope Vaccine using β-barrel Outer Membrane Proteins Identified in Chlamydia trachomatis.

Chlamydia trachomatis is an obligate intracellular Gram-negative pathogen that causes sexually transmitted infections (STIs) and trachoma. Current interventions are limited due to the widespread nature of asymptomatic infections, and the absence of a licensed vaccine exacerbates the challenge. In this study, we predicted outer membrane β-barrel (OMBB) proteins and designed a multi-epitope vaccine (MEV) construct using identified proteins. We employed a consensus-based computational framework on the C. trachomatis D/UW-3/CX proteome and identified 17 OMBB proteins, including well-known Pmp fami

An insight into differential protein abundance throughout Leishmania donovani promastigote growth and differentiation.

Leishmania donovani causes anthroponotic visceral leishmaniasis, responsible for about 50,000 annual deaths worldwide. Current therapies have considerable side effects. Drug resistance has been reported and no vaccine is available nowadays. The development of undifferentiated promastigotes in the sand fly vector's gut leads to the promastigote form that is highly infective to the mammalian host. Fully differentiated promastigotes play a crucial role in the initial stages of mammalian host infection before internalization in the host phagocytic cell. Therefore, the study of protein levels in th
