Development of amorphous solid dispersion of delamanid, an extremely water-insoluble drug, by hot melt extrusion applying acid-base supersolubilization (ABS) principle.

Delamanid, an antituberculosis drug used for multidrug-resistant tuberculosis, has extremely poor water solubility and a high melting point of ∼195 °C. The current commercial product, Deltyba®, shows incomplete dissolution and food-dependent bioavailability. This study is the first to report the preparation of amorphous solid dispersions (ASD) of delamanid by hot-melt extrusion (HME), made possible by applying the acid-base supersolubilization (ABS) principle using malic acid. Malic acid increased the aqueous solubility of delamanid to 46 mg/mL at pH 1.8, approximately 660 times higher than it

Establishment of a novel asymmetric force degenerative lumbar scoliosis aging bipedal rat model.

The present study developed a long-term asymmetric force (AF)-induced degenerative lumbar scoliosis (DLS) model using bipedal upright rats and aimed to elucidate the role of AF in DLS pathogenesis. Sprague-Dawley rats underwent humeral head resection and tail amputation, with elevated food placement to encourage bipedal standing and feeding. The present training protocol continued for 16 weeks to establish the bipedal rat model. Subsequently, an AF-induced DLS rat model was established by implanting a nickel-titanium spring with anchor screws for 24, 36 and 48 weeks, respectively. X-ray imagin

The Metabolic Revolution in Dermatology: Obesity, Insulin Resistance, and Ultra-Processed Diets as Drivers of Skin Inflammation.

Inflammatory dermatoses are increasingly linked to systemic metabolic factors. Obesity and insulin resistance create a pro-inflammatory milieu that affects the skin. Adipose tissue functions as an endocrine organ secreting adipokines and cytokines that drive chronic inflammation with notable skewing toward T-helper 1 (Th1)/Th17 signaling. Hyperinsulinemia, elevated insulin-like growth factor-1 promote keratinocyte proliferation, sebum production, and autoinflammation, contributing to a multitude of skin diseases including hidradenitis suppurativa, acne, psoriasis, atopic dermatitis (AD), acant

Enhanced pancreatic toxicity induced by cadmium and a high-fat diet: attenuation by naringin through the modulation of oxidative stress, apoptosis, and inflammatory signaling.

Increasing exposure to cadmium, an environmental pollutant from industrial and agricultural activities, and changing lifestyles and a tendency to consume high-fat foods, has raised serious concerns about metabolic health. We designed this study to investigate the combined effects of cadmium and a high-fat diet on pancreatic function. We also evaluated the protective potential of naringin, a natural flavonoid with antioxidant and anti-inflammatory properties. In our 12-week experimental study, we exposed 64 female NMRI mice to a high-fat diet (58.8 % calories from fat) and different levels of c

Ultrasound-assisted ionic liquid microextraction and colorimetric analysis for trace-level gold detection in environmental and geological samples.

A novel, precise, and targeted method has been developed for isolating, preconcentrating, and analyzing gold ions in water, ore, and food samples. This approach utilizes ultrasound-assisted dispersive liquid-liquid microextraction based on ionic liquids, combined with colorimetric detection of a distinct deep red hue. The compound 2-amino-4-(m-tolylazo)pyridine-3-ol (ATAP), introduced for the first time as a gold-specific chelating agent, was employed alongside the ionic liquid 1-octyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide in an ultrasonic bath maintained at 25 ± 2 °C for extra

Effect of time-restricted feeding on cognitive dysfunction in Alzheimer's disease and the role of ApoE polymorphism: protocol for a randomised controlled trial.

INTRODUCTION: With the global ageing population accelerating, the prevalence of Alzheimer's disease (AD) continues to rise annually. However, the underlying mechanisms of AD remain unclear, and effective treatments are still lacking. Apolipoprotein E (ApoE) gene polymorphism and dietary habits are critical risk factors for AD. Time-restricted feeding (TRF), an intermittent fasting strategy that limits the daily window of food intake while maintaining nutritional balance, has garnered significant attention in recent years for its potential to improve cognitive dysfunction. This study aims to in

Paper-immobilized liquid-phase microextraction for direct paper spray mass spectrometry and immuno-detection of atropine in baby food, buckwheat cereals, and edible oils at regulatory levels.

BACKGROUND: Atropine is a strictly regulated natural toxin. Monitoring for atropine is thus important, but often expensive and time-consuming. Moreover, the range of relevant matrices, and corresponding differences in required detection limits for atropine vary. Therefore, we developed a more simplified and affordable method, combining immunodetection and mass spectrometry to detect atropine in buckwheat, canola oil, and baby cereals at regulatory levels. RESULTS: In this method, atropine is selectively enriched on paper using a dual-paper-immobilized liquid-phase microextraction (PI-LPME; en

Vitamin D improves diabetic cardiomyopathy by inhibiting pyroptosis through the NOX4/NLRP3 inflammasome pathway.

Diabetic cardiomyopathy (DCM) is a major cardiovascular complication of diabetes mellitus, for which effective treatment options remain unavailable. Recent studies have demonstrated that vitamin D exerts protective effects against DCM, but its role in alleviating cellular pyroptosis remains uncertain. The present study aimed to investigate the ameliorative effects of vitamin D on pyroptosis in DCM and to elucidate the underlying molecular mechanisms. In the in vivo experiments, male diabetic db/db mice were treated with or without 1,25(OH)2D3 by oral gavage for 17 weeks, while age-matched non-

Rapid, ultrasensitive, and specific RPA-THz system for pathogenic microorganism detection.

Pathogenic microorganisms responsible for infectious diseases pose a significant global threat to human health. Existing detection methods, such as qPCR and ELISA, fail to simultaneously meet the requirements for high sensitivity, high specificity, and rapid detection. This study presents an innovative approach for the rapid, specific, and highly sensitive detection of pathogenic microorganisms, particularly Escherichia coli O157:H7 (E. coli O157:H7) and varicella-zoster virus (VZV), by combining recombinase polymerase amplification (RPA) with terahertz time-domain spectroscopy (THz-TDS). Time-Restricted Feeding Reduces Atherosclerosis in LDLR KO Mice but Not in ApoE Knockout Mice.

BACKGROUND: Dyslipidemia increases cardiovascular disease risk, the leading cause of death worldwide. Under time-restricted feeding (TRF), wherein food intake is restricted to a consistent window of <12 hours, weight gain, glucose intolerance, inflammation, dyslipidemia, and hypercholesterolemia are all reduced in mice fed an obesogenic diet. LDLR (low-density lipoprotein receptor) mutations are a major cause of familial hypercholesterolemia and early-onset cardiovascular disease. METHODS: We subjected benchmark preclinical models, mice lacking LDLR-knockout or ApoE knockout to ad libitum fee

Manipulation of feeding patterns in high fat diet fed rats improves microbiota composition dynamics, inflammation and gut-brain signaling.

Chronic consumption of high fat (HF) diets has been shown to increase meal size and meal frequency in rodents, resulting in overeating. Reducing meal frequency and establishing periods of fasting, independently of caloric intake, may improve obesity-associated metabolic disorders. Additionally, diet-driven changes in microbiota composition have been shown to play a critical role in the development and maintenance of metabolic disorders. In this study, we used a pair-feeding paradigm to reduce meal frequency and snacking episodes while maintaining overall intake and body weight in HF fed rats.
