BACKGROUND: This study aimed to explore the relationship between self-reported sleep disturbances (e.g., insomnia, REM sleep behavior disorder [RBD]) and cortical thickness, brainstem volume, amyloid accumulation, and inflammatory markers in Parkinson's disease (PD) patients.
METHODS: We conducted a cross-sectional study comparing 100 PD patients (observation group) with 100 age-matched controls. Sleep quality was assessed using the Pittsburgh Sleep Quality Index (PSQI), and serum levels of amyloid-beta (Aβ1-42), α-synuclein, and inflammatory markers (CRP, TNF-α, IL-1β) were quantified.

Incense burning is a widespread indoor combustion practice, yet its neurotoxic potential and impact on α-synuclein (α-Syn) proteostasis remain poorly defined. Using SH-SY5Y cells overexpressing α-Syn as a cellular Parkinson's disease model, we exposed cells to size-fractionated incense aerosol extracts (IAE) prepared as organic-phase (OP) or water-soluble phase (WP). α-Syn overexpression augmented vulnerability to IAE, producing greater losses in viability and pronounced increases in intracellular hydrogen peroxide (H2O2), mitochondrial membrane potential depolarization, and engagement of prog

Different morphological and pathological features of abnormal α-synuclein in synucleinopathies-multiple system atrophy (MSA) and Lewy body diseases, including Parkinson's disease (PD) and dementia with Lewy bodies (DLB)-may underlie disease-specific synaptic dysfunction. To compare synaptic profiles associated with abnormal α-synuclein, we examined temporal lobe tissues from patients with MSA (N = 4), DLB (N = 5), and controls (N = 9). We further analysed cerebrospinal fluid (CSF) samples from patients with early-stage MSA-parkinsonism (MSA-P: N = 8) and PD (N = 8) to determine whether synapti

There are no approved treatments that slow Parkinson's disease (PD) progression and therefore it is important to identify novel pathogenic mechanisms that can be targeted. Loss of the epigenetic marker, Tet2 appears to have some beneficial effects in PD models, but the underlying mechanism of action is not well understood. We performed an unbiased transcriptomic analysis of cortical neurons isolated from patients with PD to identify dysregulated pathways and determine their potential contributions to the disease process. We discovered that genes associated with primary cilia, non-synaptic sens

Parkinson's disease (PD) is a widespread degenerative disorder of the central nervous system. The gradual degeneration of dopaminergic neurons in the substantia nigra region is one of the primary pathological features of PD. Glial cells in SN are also linked to the pathological PD alterations. To discern the role of neurons and glial cells as well as their corresponding genetic modifications in PD, we utilized diverse bioinformatics techniques and performed biological experiments on cell and animal models. Several transcriptome datasets of the substantia nigra region were collected from the Ge

Autophagic decline accompanies age and causes a deterioration in proteostasis, rendering neuronal demise. Rab27 functions as a vesicle regulator for macroautophagic/autophagic degradation and exocytosis. Loss of Drosophila Rab27 in αβp brain neurons enhances longevity, underscoring its neuronal role and systemic effect. To understand the underlying mechanisms, we characterized the cell autonomous and non-autonomous functions of Rab27. Rab27 expression increased in midlife, providing a temporal manipulation window. Depleting Rab27 at that timepoint activated autophagy and sustained neuronal mai

Recent epidemiologic studies have shown that patients with age-related macular degeneration (AMD) have a considerably higher risk of developing Parkinson disease (PD) later in life, suggesting a possible link between these diseases. However, the common mechanisms between these two diseases remain obscure, although the pathophysiology of each has been well investigated. This study was designed to explore the shared pathologic features of AMD and PD by focusing on leucine-rich repeat kinase 2 (LRRK2) and α-synuclein, both of which play crucial roles in PD pathogenesis. Sex differences in dementia with Lewy bodies (DLB) remain understudied despite growing evidence in neurodegenerative diseases. In this retrospective multicentric study of 310 patients with probable DLB, we assessed whether sex-related clinical differences could be explained by Alzheimer's disease (AD) cerebrospinal fluid biomarkers. While, in univariate analysis, females had lower Mini-Mental State Examination (MMSE) scores and more frequent AD profiles, males presented more REM sleep disorders. Adjusting for AD biomarkers attenuated some differences (REM sleep disorder, MMSE), but not all lik

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by the loss of dopaminergic neurons, aggregation of α-synuclein (α-Syn), lysosomal dysfunction, and mitochondrial impairment. Curcumin has demonstrated neuroprotective effects against PD pathology; however, its poor bioavailability, rapid systemic clearance, and limited blood-brain barrier permeability remain significant challenges to be overcome. BACKGROUND: Multiple system atrophy (MSA) is a neurodegenerative disease pathologically characterized by the presence of glial cytoplasmic inclusions (GCI) composed of α-synuclein aggregates. In Parkinson's disease, increases in monounsaturated fatty acids (MUFA) in phospholipid membranes promote α-synuclein binding, aggregation, and toxicity, and the inhibition of stearoyl-CoA desaturase (SCD), the enzyme responsible for synthesizing MUFA, alleviates α-synuclein toxicity. However, little is known about phospholipid MUFA or SCD in the context of MSA pathology.
OBJECTIVES: To determine whether

BACKGROUND: Autophagy dysfunction in glial cells is implicated in the pathogenesis of Parkinson's disease (PD). The previous study reported that α-synuclein (α-Syn) disrupted autophagy in cultured microglia. However, the mechanism of microglial autophagy dysregulation is poorly understood.
METHODS: Two α-Syn-based PD models were generated via AAV-mediated α-Syn delivery into the mouse substantia nigra and striatal α-Syn preformed fibril (PFF) injection. The levels of microglial UNC-51-like kinase 1 (Ulk1) and other autophagy-related genes in vitro and in PD mice, as well as in the peripheral

The dynamic equilibrium between acetylation and deacetylation is vital for cellular homeostasis. Parkinson's disease (PD), a neurodegenerative disorder marked by α-synuclein (α-syn) accumulation and dopaminergic neuron loss in the substantia nigra, is associated with a disruption of this balance. Therefore, correcting this imbalance with histone deacetylase (HDAC) inhibitors represents a promising treatment strategy for PD. CAY10603 (CAY) is a potent and selective HDAC6 inhibitor. However, because of its poor water solubility and short biological half-life, it faces clinical limitations. Herei

BACKGROUND: Parkinson's disease (PD) is a progressive neurodegenerative disorder thought to be caused by accumulation of α-synuclein (α-syn) within the brain, autonomic nerves, and the enteric nervous system (ENS). Involvement of the ENS in PD often precedes the onset of the classic motor signs of PD by many years at a time when severe constipation represents a major morbidity. Studies conducted in vitro and in vivo, have shown that squalamine, a zwitterionic amphipathic aminosterol, originally isolated from the liver of the dogfish shark, effectively displaces membrane-bound α-syn.

Protein aggregation into amyloid fibrils is a key feature of a multitude of neurodegenerative diseases such as Alzheimer's, Parkinson's, and Prion disease. To detect amyloid fibrils, fluorophores with high sensitivity and better efficiency coupled with the low toxicity are in high demand even to date. In this pursuit, we have unveiled two benzimidazole-based fluorescence sensors ([C15 H15 N3 ] (C1) and [C16 H16 N3 O2 ] (C2), which possess exceptional affinity toward different amyloid fibrils in its submicromolar concentration (8 × 10-9 M), whereas under a similar concentration, the gold stand

Gaucher disease (GD) patients and carriers of GD mutations have a higher propensity to develop Parkinson's disease (PD) in comparison to the non-GD population. This implies that mutant GBA1 allele is a predisposing factor for the development of PD. One of the major characteristics of PD is the presence of oligomeric α-synuclein-positive inclusions known as Lewy bodies in the dopaminergic neurons localized to the substantia nigra pars compacta. In the present study we tested whether presence of human mutant GCase leads to accumulation and aggregation of α-synuclein in two models: in SHSY5Y neur

The self-assembly of proteins into structured fibrillar aggregates is associated with a range of neurodegenerative diseases, including Alzheimer's and Parkinson's diseases, in which an important cytotoxic role is thought to be played by small soluble oligomers accumulating during the aggregation process or released by mature fibrils. As the structural characteristics of such species and their links with toxicity are still not fully defined, we have compared six examples of preformed misfolded protein oligomers with different β-sheet content, as determined using Fourier transform infrared spect

Oxidative stress and neuroinflammation play key roles in the initiation and progression of Parkinson's disease (PD), a neurodegenerative disorder, associated with the loss of nigrostriatal dopaminergic pathway. Thus, compounds that can mitigate oxidative stress and neuroinflammation are being investigated as promising agents for the treatment of PD. This study was designed to evaluate the effects of methyl jasmonate (MJ), a potent antioxidant and anti-inflammatory compound on parkinsonian-like symptoms and the underlying biochemical changes induced by rotenone (Rot) in mice. To this end, the e

Alzheimer's dementia is significantly more common in women than in men. However, few pathological studies have addressed sex difference in Alzheimer's disease (AD) and other brain pathologies. We leveraged postmortem data from 1453 persons who participated in one of two longitudinal community-based studies of older adults, the Religious Orders Study and the Rush Memory and Aging Project. Postmortem examination identified AD pathologies, neocortical Lewy bodies, DNA-binding protein 43 (TDP-43), hippocampal sclerosis, gross and micro infarcts, atherosclerosis, arteriolosclerosis, and cerebral am

Parkinson's disease (PD), the most common neurodegenerative movement disorder, is characterized by the progressive loss of nigral dopamine neurons. The deposition of fibrillary aggregated α-synuclein in Lewy bodies (LB), that is considered to play a causative role in the disease, constitutes another key neuropathological hallmark of PD. We have recently described that synapsin III (Syn III), a synaptic phosphoprotein that regulates dopamine release in cooperation with α-synuclein, is present in the α-synuclein insoluble fibrils composing the LB of patients affected by PD. Moreover, we observed
