Comparative reproducibility of dermal microvascular blood flow changes in response to acetylcholine iontophoresis, hyperthermia and reactive hyperaemia.

Laser Doppler fluxmetry (LDF) can non-invasively measure skin microvascular changes in response to acetylcholine (ACh), local heating of the skin and reactive hyperaemia following arterial occlusion. Various studies have used microvascular changes in response to these stimuli, especially ACh iontophoresis and local heating, as a surrogate marker of endothelial function. There are few data in the literature regarding the comparative reproducibility of microvascular perfusion changes induced by the three stimuli. The aim of this study was to systematically assess and compare the reproducibility

The eye in cerebral malaria: what can it teach us?

The pathophysiology of coma in cerebral malaria (CM) is not well understood. Obstruction of microcirculatory flow is thought to play a central role, but other hypotheses include roles for parasite- and host-derived factors such as immune mediators, and for increased blood-brain barrier permeability leading to raised intracranial pressure. The retinal vasculature is a direct extension of the cerebral vasculature. It is the only vascular bed easily accessible for visualisation and provides a unique opportunity to observe vascular pathology and its effect on neurological tissue. A specific retino

PET-based assessment of coronary microvascular dysfunction and its relation to plaque burden and vascular remodeling: integration with microvascular resistance reserve.

BACKGROUND: Coronary microvascular dysfunction (CMD) contributes to myocardial ischemia in patients with angina and non-obstructive coronary artery disease. Quantitative positron emission tomography (PET) allows noninvasive assessment of myocardial blood flow and identification of CMD. OBJECTIVES: To investigate the association between PET-assessed coronary microvascular dysfunction (CMD), coronary plaque burden, and vascular remodeling, integrating microvascular resistance reserve (MRR). METHODS: This post hoc analysis of the PACIFIC-1 trial included coronary arteries from symptomatic patie

Endothelial IGF-1/IGF-1R signaling supports young-blood-induced restoration of blood-brain barrier integrity and microvascular architecture in aged mice: evidence from the heterochronic parabiosis model.

Aging is associated with blood-brain barrier (BBB) breakdown and microvascular rarefaction, key contributors to cerebral neuroinflammation, hypoperfusion, and vascular cognitive impairment and dementia (VCID). Exposure to a young systemic milieu through heterochronic parabiosis has been shown to restore BBB integrity and enhance cerebrovascular function in aged mice, suggesting that circulating factors can rejuvenate the aging brain. However, the molecular mediators responsible for these effects remain poorly defined. Circulating insulin-like growth factor-1 (IGF-1) declines markedly with age

Exercise-induced redox modulation of red blood cell function in health and disease: Mechanistic implications for hemoglobin cycling and microvascular regulation.

Exercise generates transient systemic redox and hemodynamic signals that influence oxygen delivery, vascular tone, and metabolic adaptation in both health and disease. Red blood cells (RBCs) are increasingly recognized as regulated redox-responsive cells rather than passive oxygen carriers. This narrative review integrates mechanistic and translational evidence linking RBC deformability, microvascular transit, and perfusion matching to exercise-induced reactive oxygen and nitrogen species (RONS), hemodynamic shear, hemoglobin redox cycling, antioxidant buffering, and nitric oxide (NO)-related

Super-Resolution Contrast-Enhanced Ultrasound Detects Renal Microvascular Impairment in Early Chronic Kidney Disease with Preserved Estimated Glomerular Filtration Rate: A Comparative Study with Conventional Contrast-enhanced Ultrasound and Serum Biomarkers.

OBJECTIVE: Renal microvascular rarefaction is an early pathological event in chronic kidney disease (CKD) that may precede a measurable decline in renal function. However, currently available non-invasive tools remain limited in detecting these early structural alterations. This study aimed to evaluate the value of super-resolution contrast-enhanced ultrasound (SR-CEUS) for assessing renal cortical microvascular impairment in early CKD with preserved estimated glomerular filtration rate (eGFR), and to compare its performance with conventional contrast-enhanced ultrasound (CEUS) and serum bioma

Hemodynamic and microvascular abnormalities in P450 oxidoreductase deficiency: evidence for COX-dependent dysfunction.

OBJECTIVE: Cytochrome P450 oxidoreductase deficiency (PORD) is a rare form of congenital adrenal hyperplasia caused by impaired electron transfer to multiple microsomal enzymes. Hypertension in PORD has been attributed to mineralocorticoid excess; however, experimental evidence suggests that POR may also regulate endothelial vasodilatory pathways. The vascular effects of PORD in humans remain poorly defined. We evaluated blood pressure regulation, microvascular structure, and serum vasoactive mediators in patients with PORD. DESIGN: Observational, cross-sectional study. METHODS: Ten patients

Dl-3-n-Butylphthalide Protects Human Brain Microvascular Endothelial Cells Against Ischemic Injury Through Dual Modulation of HIF1α/VEGF-Mediated Angiogenesis and COX2-Mediated Inflammation.

BACKGROUND: Stroke remains a leading cause of death and disability worldwide. Endothelial dysfunction plays a central role in both acute ischemic injury and subsequent recovery. In this study, we investigated the protective effects and underlying mechanisms of Dl-3-n-butylphthalide (NBP) on human brain microvascular endothelial cells (HBMECs) subjected to oxygen-glucose deprivation (OGD), an in vitro model of ischemic stroke. METHODS: HBMECs were divided into three groups: control (normoxia), OGD (10 h hypoxia, followed by reoxygenation for different durations (0-24 h), and then a subsequent

Panax notoginseng Saponins Attenuate Cerebral Ischemia-Reperfusion Injury by Suppressing Neutrophil Extracellular Traps and Ferroptosis in Brain Microvascular Endothelial Cells.

Panax notoginseng saponins (PNS), the chief saponin components of the traditional Chinese medicine Panax notoginseng, are known to ameliorate the outcomes of ischemic stroke (IS), primarily by mitigating inflammatory responses, enhancing microcirculation, and modulating oxidative stress. Damage to brain microvascular endothelial cells (BMECs) and BMECs' ferroptosis are critical steps in cerebral ischemia-reperfusion injury (CIRI), a process in which neutrophil extracellular traps (NETs) have been shown to induce BMECs injury, thereby aggravating CIRI. We aimed to investigate the regulatory eff

ALKBH5 deficiency attenuates oxygen-glucose deprivation-induced injury in mouse brain microvascular endothelial cells in an m6A dependent manner.

Structural and functional alterations in brain microvascular endothelial cells (BMECs) caused by oxygen-glucose deprivation (OGD) are involved in the pathogenesis of various brain disorders. AlkB homolog 5 (ALKBH5) is a primary m6A demethylase that regulates various cell processes, but its distinct roles in BMEC function remain to be clarified. In the present study, in mouse middle cerebral artery occlusion (MCAO) model, knockout of ALKBH5 reduced neurological deficits, infarct volumes and tissue apoptosis caused by ischemia/reperfusion injury. Evans blue leakage and decreased expression of th

Noninvasive assessment of human microvascular function in health and disease using incident dark-field microscopy.

Reduced peripheral microvascular reactivity is associated with an increased risk for major adverse cardiac events (MACEs). Tools for noninvasive assessment of peripheral microvascular function are limited, and existing technology is poorly validated in both healthy populations and patients with cardiovascular disease (CVD). Here, we used a handheld incident dark-field imaging tool (CytoCam) to test the hypothesis that, compared with healthy individuals (no risk factors for CVD), subjects formally diagnosed with coronary artery disease (CAD) or those with ≥2 risk factors for CAD (at risk) would
