Binding of 1-(4-chloro-benzenesulfonyl)-4-hydroxy-pyrrolidine-2-carboxylic acid to human carbonic anhydrase II affects its structure and function: a multi-spectroscopic and molecular docking study.

The interaction between human carbonic anhydrase II (hCA II) and 1-(4-chloro-benzenesulfonyl)-4-hydroxy-pyrrolidine-2-carboxylic acid (CBHPCA) was inspected by applying different spectroscopic methods and molecular docking, as well. Kinetic studies revealed that CBHPCA inhibits hCA II esterase activity through a linear competitive mode. Fluorescence analyses showed that CBHPCA quenches the enzyme's intrinsic fluorescence through a static quenching mechanism. Thermodynamic analyses indicated that hydrogen bonding and van der Waals interactions stabilize the CBHPCA-enzyme complex. Protein surfac

Cerebral blood flow autoregulation during prehospital post-resuscitation care.

BACKGROUND: Successfully resuscitated out-of-hospital cardiac arrest (OHCA) patients often experience cerebral malperfusion, possibly due to impaired autoregulation. Understanding the link between arterial pressure and cerebral blood flow (CBF) is essential for optimising cerebral oxygenation. This study examined early post-resuscitation CBF autoregulation and assessed the feasibility of identifying patient-specific optimal mean arterial pressure (MAPopt) from data obtained during prehospital care. METHODS: We performed a post hoc analysis of 12 OHCA patients treated by a physician-staffed he

Structural, proteomic, and metabolomic insights into the antibacterial efficacy of nalidixic acid against Corynebacterium pseudotuberculosis.

Corynebacterium pseudotuberculosis is a facultative intracellular pathogen that causes caseous lymphadenitis, a disease responsible for substantial economic losses in the livestock industry. Although nalidixic acid (NA) has been shown to inhibit C. pseudotuberculosis biofilm formation, the molecular mechanisms underlying its efficacy remain poorly characterized. This study aimed to characterize the structural, proteomic, and metabolomic alterations induced by NA in C. pseudotuberculosis at effective concentrations. The minimum inhibitory concentration (MIC) and minimum bactericidal concentrati

Slow running benefits: Boosts in mood and facilitation of prefrontal cortex function even at very light intensity.

Although running upright has been reported to have positive effects on both physical and mental health, the minimum running intensity/speed that would benefit mood and prefrontal cognition is not yet clear. Here, we tested the hypothesis that a brief bout of very slow running, which is classified as a very light intensity exercise, would enhance positive mood and executive function and increase activation in executive-function-related prefrontal subregions. Twenty-four healthy participants completed a 10-minute very slow running session on a treadmill at 35% V̇o2peak and a resting control sess

Acute Effects of Different Inspiratory Muscle Training Intensities on Intercostal Muscle Oxygenation in Healthy Adults: A Randomized Crossover Study.

BACKGROUND: Increased inspiratory loading increases oxygen consumption and metabolic demand in the respiratory muscles, affecting muscle oxygenation. However, evidence regarding different inspiratory muscle training (IMT) intensities on intercostal muscle oxygenation remains limited. This study compared acute effects of different IMT intensities on intercostal muscle oxygenation and cardiorespiratory responses. METHODS: This randomized, single-blind, crossover study included 30 healthy individuals. Participants performed IMT at 0% (sham), 30% maximal inspiratory pressure (endurance-based) (MI

Heterogeneous Fenton reactions: oxidation of adsorbing and non-adsorbing probe molecules via H2O2-promoted reduction of iron oxides.

Hydroxyl radicals (HO•), produced through reactions between H2O2 and iron oxides, drive biogeochemical transformations, mediate organism toxicity, and facilitate advanced oxidation processes. The effectiveness of these processes depends on the spatial proximity between HO• generation and target substrates. Consequently, the oxidation mechanisms should be governed by interfacial interactions among H2O2, substrates, and iron oxide surfaces. Substrate oxidation by iron oxide/H2O2 systems were studied using two probes simultaneously: terephthalate (TPA), forming outer-sphere surface complexes, and

Rapid spatiotemporal monitoring of beach microplastics in the Galápagos Islands.

Understanding the environmental fate and behaviour of microplastics across space and time is challenging, especially in remote locations such as the Galápagos Islands. Citizen science can support plastic monitoring by extending geographic and temporal coverage, though concerns persist around data validation. This study evaluated a simple citizen science methodology, aided by a near-infrared portable spectrometer (PlasTell), for the rapid monitoring of beach microplastics (>1 mm) to investigate spatiotemporal accumulation patterns on Tortuga Bay, Santa Cruz Island, Galápagos. A total of 3377 su

Microcirculation and Cerebrovascular Autoregulation in Patients With Mechanical Circulatory Support Devices.

Acute brain injury (ABI) affects up to one-third of patients using mechanical circulatory support (MCS). In venoarterial extracorporeal membrane oxygenation (VA ECMO), ABI incidence (11%-40%) has not improved in two decades. Conversely, improvements in left ventricular assist devices (LVADs) have reduced the incidence of stroke, although it remains a major complication (10%-30%). The failure of MCS to ensure adequate cerebral protection may impair cerebrovascular autoregulation (CVAR) and disrupt microcirculatory function affected by reduced pulsatility, endothelial injury, acute perturbations

A chemical bonding adhesive for PEEK based on aromatic ring interactions.

OBJECTIVES: Reliable bonding to polyetheretherketone (PEEK) remains challenging because of its semicrystalline, chemical-resistant nature. In this study, a two-part adhesive was developed for grit-blasted and otherwise not aggressively treated PEEK that targets the phenylene backbone units of PEEK through noncovalent π-π interactions and chain-level compatibility. METHODS: Miscibility of candidate solvents and monomers with PEEK powder was measured by differential scanning calorimetry (DSC) to identify chain-level effects. A formulation based on 1-vinylimidazole and glycidyl methacrylate, wit

Physiology, monitoring, and optimisation of perioperative tissue oxygenation: a narrative review.

Maintenance of tissue oxygenation in patients having surgery is important as tissue hypoxia is a major determinant of organ failure. Tissue oxygenation follows a stepwise physiological pathway involving the macrocirculation, the microcirculation, and the cellular oxygen metabolism. This narrative review endorsed by the Network for the Advancement of Patient Blood Management, Haemostasis and Thrombosis outlines the physiology of tissue oxygenation, evaluates methods for intraoperative tissue oxygenation monitoring, and summarises therapeutic strategies to ensure adequate tissue oxygenation. Cardiopulmonary and skeletal muscle strategies underlying exhaustive exercise in adults with glycogen storage disease type III.

People with glycogen storage disease type III (GSDIII-p) have a remarkably reduced exercise tolerance. Aim of this study was to analyze the oxygen transport-utilization chain strategies adopted by GSDIII-p during exercise. Nine GSDIII-p (39.4 ± 10.0 year, 33% female) and 11 healthy controls (CTRL), age and gender matched, underwent an incremental cardiopulmonary exhaustion test (CPET) to assess peak heart rate (HR), blood lactate [La]p and vastus lateralis O2 fractional extraction (ΔHHb/isch) using near-infrared spectroscopy. Patterns of breathing (PBr) were assessed accordingly by analyzing p
