Pheromone receptor of the globally invasive quarantine pest of the palm tree, the red palm weevil (Rhynchophorus ferrugineus).

Palm trees are of immense economic, sociocultural, touristic, and patrimonial significance all over the world, and date palm-related knowledge, traditions, and practices are now included in UNESCOs list of the Intangible Cultural Heritage of Humanity. Of all the pests that infest these trees, the red palm weevil (RPW), Rhynchophorus ferrugineus (Olivier), is its primary enemy. The RPW is a category-1 quarantine insect pest that causes enormous economic losses in palm tree cultivation worldwide. The RPW synchronizes mass gathering on the palm tree for feeding and mating, regulated by a male-pro

Unveiling sugarcane defense response to Mythimna separata herbivory by a combination of transcriptome and metabolic analyses.

BACKGROUND: Sugarcane is the most important sugar crop in the world. Like other crops, sugarcane suffers from herbivorous insect attack. The oriental armyworm Mythimna separata is a devastating pest of various crops in northeast Asia and an outbreak of this pest can result in substantial yield loss for sugarcane. However, the plant defense response situation is widely acquisition in model crops, but there is little information about how sugarcane plants defend themselves against this herbivore at the molecular and biochemical levels. RESULTS: We combined transcriptome and metabolomic analysis

Characterization of HIV-1 virus-like particles and determination of Gag stoichiometry for different production platforms.

The importance of developing new vaccine technologies towards versatile platforms that can cope with global virus outbreaks has been evidenced with the most recent severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic. Virus-like particles (VLPs) are a highly immunogenic, safe, and robust approach that can be used to base several vaccine candidates on. Particularly, HIV-1 Gag VLPs is a flexible system comprising a Gag core surrounded by a lipid bilayer that can be modified to present diverse types of membrane proteins or antigens against several diseases, like influenza, dengue

Biochemical response of Sonneratia alba Sm. branches infested by a wood boring moth (Gazi Bay, Kenya).

Infestation by a moth woodborer species is causing mortality of Sonneratia alba Sm. mangrove by tunneling through the inner bark, cambium and conductive tissue. Infestation leads to death of some infested branches, whereas in other cases infested branches have been observed to recover from infestation. We have used Fourier transform infrared spectroscopy (FTIR) to investigate the differences in macromolecule (polysaccharide and lignin) content present in branches that died (D) of the infestation, those that recovered (R) from the infestation and control branches (C) that were not subject to an

Evaluating Flight Performance of Mass-Reared and Irradiated Navel Orangeworm (Lepidoptera: Pyralidae) for Sterile Insect Technique.

Navel orangeworm (Pyralidae: Amyelois transitella) is a key pest of almonds and pistachios in California. Moths directly infest nuts which leads to reduced crop yield and quality, and infestation can predispose nuts to fungal pathogens that produce aflatoxins. While several integrated pest management strategies have been developed for A. transitella, studies have recently been initiated to explore the use of sterile insect technique (SIT) as an additional control tool. Mass-rearing, sterilization, and transportation methods originally developed for Pectinophora gossypiella (Lepidoptera: Gelech

An ovary-specific mucin is associated with choriogenesis mediated by prostaglandin signaling in Spodoptera exigua.

Polytrophic ovarioles of Spodoptera exigua, a lepidopteran insect, begins with the development of oocytes and differentiation of nurse cells followed by vitellogenesis and choriogenesis. Compared with previtellogenic and vitellogenic developments, choriogenesis has not been clearly understood yet in endocrine control. This study investigated the expression and function of a mucin-like structural protein of S. exigua called Se-Mucin1 in choriogenesis. It was highly expressed in ovarioles containing chorionated oocytes. The expression level of Se-Mucin1 was increased during adult stage as early

Prevalence of skin infections, infestations, and papular urticaria among adolescents in secondary schools in Calabar, Nigeria.

BACKGROUND: Acne is an inflammatory disorder of the pilosebaceous gland, and the most common dermatosis in adolescents globally. Infectious dermatoses are common in the tropics, but due to the paucity of epidemiologic surveys, not much is known about the prevalence and common types found in different sub-populations including adolescents. It is however presumed that the prevalence will be high and the pattern diverse. We therefore conducted a school-based survey to ascertain the prevalence and pattern of infectious dermatoses, infestations, and papular urticaria (insect bite reactions) in teen

The evolutionary origins of pesticide resistance.

Durable crop protection is an essential component of current and future food security. However, the effectiveness of pesticides is threatened by the evolution of resistant pathogens, weeds and insect pests. Pesticides are mostly novel synthetic compounds, and yet target species are often able to evolve resistance soon after a new compound is introduced. Therefore, pesticide resistance provides an interesting case of rapid evolution under strong selective pressures, which can be used to address fundamental questions concerning the evolutionary origins of adaptations to novel conditions. We ask:

The Polymorphic Variant P24T of UDP-Glucuronosyltransferase 1A4 and Its Unusual Consequences.

The P24T polymorphic variant of the human UDP-glucuronosyltransferase 1A4 (UGT1A4*2, 70C>A) occurs within the signal peptide, five amino acids upstream of the cleavage site and the start of the mature protein. Bioinformatic analysis of the variant suggested that the signal peptide of part of the translated protein is cleaved two residues upstream of the regular site, whereas the rest is cleaved as usual. To test this, recombinant UGT1A4-P24T, with a C-terminal His-tag, was expressed in sf9 insect cells and affinity-purified for N-terminal protein sequencing. The results were in agreement with

Fenitrothion, an organophosphorous insecticide, impairs locomotory function and alters body temperatures in Sminthopsis macroura (Gould 1845) without reducing metabolic rates during running endurance and thermogenic performance tests.

Endemic Australian mammal species are exposed to pesticides used for locust control as they occupy the same habitat as the target insect. The authors examined the impact of an ultra-low volume formulation of the organophosphorous insecticide fenitrothion (O,O-dimethyl-O-[3-methyl-4-nitrophenol]-phosphorothioate) on a suite of physiological measures that affect the ability of animals to survive in free-living conditions: locomotory and thermogenic functions, metabolic performance, body mass, and hematocrit and hemoglobin levels. Plasma and brain cholinesterase activity in relation to time since

[Epidemiology of imported malaria and entomological study of breeding sites of potential risk areas in the province of Khemisset (Morocco)].

No case of autochthonous malaria has been detected in Morocco since 2004. This achievement is due to a national strategy to combat the disease by appropriate and well-organized disease detection and treatment, as well as control of the mosquito vector of the disease, the female Anopheles mosquito. Nonetheless, imported malaria cases have been increasing (75 in 2007), due to the rise in international travel and migration from countries where the disease is endemic. This work is divided into two parts: the first part is a retrospective study of the cases of imported malaria identified by optical

The Cry4B toxin of Bacillus thuringiensis subsp. israelensis kills Permethrin-resistant Anopheles gambiae, the principal vector of malaria.

Resurgence of malaria has been attributed, in part, to the development of resistance by Anopheles gambiae, a principal vector of the disease, to various insecticidal compounds such as Permethrin. Permethrin, a neurotoxicant, is widely used to impregnate mosquito nets. An alternative strategy to control mosquitoes is the use of Bacillus thuringiensis subsp. israelensis (Bti) because there is no observable resistance in the field to the bacterium. Bti kills mosquitoes by targeting cadherin molecules residing in the midgut epithelium of larvae of the insect. Cry proteins (Cry4A, Cry4B, Cry10A and

Naturally occurring culturable aerobic gut flora of adult Phlebotomus papatasi, vector of Leishmania major in the Old World.

BACKGROUND: Cutaneous leishmaniasis is a neglected, vector-borne parasitic disease and is responsible for persistent, often disfiguring lesions and other associated complications. Leishmania, causing zoonotic cutaneous leishmaniasis (ZCL) in the Old World are mainly transmitted by the predominant sand fly vector, Phlebotomus papatasi. To date, there is no efficient control measure or vaccine available for this widespread insect-borne infectious disease. METHODOLOGY/PRINCIPAL FINDINGS: A survey was carried out to study the abundance of different natural gut flora in P. papatasi, with the long-

Development of CE methods to analyze potential components of the angiogenic glycoprotein vascular endothelial growth factor 165.

The vascular endothelial growth factor 165 (VEGF(165)) is the predominant form of the complex VEGF family. This glycoprotein has, among others, an angiogenic effect in many physiological and pathological events. For this reason, its roles as a biomarker and as a therapeutic drug have been considered. However, very little is known about the existence of different forms of VEGF(165) arising from glycosylation and other potential PTMs. This aspect is crucial because it is known that for other glycoproteins the ratio between these isoforms actually acts as a biomarker for certain diseases and othe

Phylogenetic analysis of New World screwworm fly, Cochliomyia hominivorax, suggests genetic isolation of some Caribbean island populations following colonization from South America.

Larval infestations of the New World screwworm (NWS) fly, Cochliomyia hominivorax, cause considerable economic losses through the direct mortality and reduced production of livestock. Since the 1950s, NWS populations in North and Central America have been the target of virtually continuous eradication attempts by sterile insect technique (SIT). Nevertheless, in some areas, such as Jamaica, SIT-based control programmes have failed. Reasons for the failure of SIT-based programmes in some locations are unknown, but it is hypothesized that failure may be related to the mating incompatibility betwe

Multinational, freshwater biomonitoring programs in the developing world: lessons learned from African and Southeast Asian river surveys.

Biomonitoring programs are widely used in developed countries. They also offer many advantages in assessing ecological consequences of perturbations in developing countries, including reducing the equipment-operation, maintenance, and training costs associated with physicochemical monitoring. Three case histories of river biomonitoring using freshwater organisms (fish, benthic macroinvertebrates, diatoms, zooplankton) are described that involve (1) documentation of environmental effects from long-term, large-scale applications of insecticides to control insect-vectors of river blindness (oncho

Cold shock and regulation of surface protein trafficking convey sensitization to inducers of stage differentiation in Trypanosoma brucei.

Transmission of a protozoan parasite from a vertebrate to invertebrate host is accompanied by cellular differentiation. The signals from the environment that trigger the process are poorly understood. The model parasite Trypanosoma brucei proliferates in the mammalian bloodstream and in the tsetse fly. On ingestion by the tsetse, the trypanosome undergoes a rapid differentiation that is marked by replacement of the variant surface glycoprotein (VSG) coat with GPI-anchored EP and GPEET procyclins. Here we show that a cold shock of DeltaT > 15 degrees C is sufficient to reversibly induce high-le

Electrophysiological analysis of the neurotoxic action of a funnel-web spider toxin, delta-atracotoxin-HV1a, on insect voltage-gated Na+ channels.

The effects of delta-ACTX-Hv1a, purified from the venom of the funnel-web spider Hadronyche versuta, were studied on the isolated giant axon and dorsal unpaired median (DUM) neurones of the cockroach Periplaneta americana under current- and voltage-clamp conditions using the double oil-gap technique for single axons and the patch-clamp technique for neurones. In parallel, the effects of the toxin were investigated on the excitability of rat dorsal root ganglion (DRG) neurones. In both DRG and DUM neurones, delta-ACTX-Hv1a induced spontaneous repetitive firing accompanied by plateau potentials.

Diastereomer-selective developmental toxicity of cis- and trans-permethrin in zebrafish (Danio rerio) embryos: AI-assisted analysis of seizure-like behaviors.

Permethrin (PM) has been widely used to control insect pests, including mosquitoes, flies, scabies, and mites. Its use for agricultural purposes is strongly restricted because of its high toxicity to aquatic organisms. Nevertheless, PM continues to be detected in aquatic environments, and formulations containing different diastereomeric ratios of cis-PM and trans-PM (40:60 or 25:75) are still used in the field. In this study, the developmental toxicities associated with the diastereomeric properties of PM were evaluated using zebrafish (Danio rerio) embryos. Although neither PM diastereomer si

Sustainable and affordable pest control: In silico study and toxicological effects of black soldier fly, Hermetia illucens (Diptera: Stratiomyidae), oil, and its nanoemulsion against Agrotis ipsilon (Lepidoptera: Noctuidae) larvae.

Black soldier fly, Hermetia illucens (Linnaeus, 1758) (Diptera: Stratiomyidae) larvae decompose several organic wastes into valuable biomass (oil, protein, chitin, and frass) for promising applications. Black Soldier Fly Oil (BSFO) is extracted using a mechanical pressing method, an affordable technique that yields about 24.56% oil. Gas chromatography-mass spectrometry analysis of oil showed a high content of fatty acids, with 3 predominant fatty acids, oleic acid (40.14%), palmitic acid (24.53%), and lauric acid (10.00%). Black soldier fly oil nanoemulsion (BSFO-NE) exhibited a droplet size o
