CLAC: a novel Alzheimer amyloid plaque component derived from a transmembrane precursor, CLAC-P/collagen type XXV.

We raised monoclonal antibodies against senile plaque (SP) amyloid and obtained a clone 9D2, which labeled amyloid fibrils in SPs and reacted with approximately 50/100 kDa polypeptides in Alzheimer's disease (AD) brains. We purified the 9D2 antigens and cloned a cDNA encoding its precursor, which was a novel type II transmembrane protein specifically expressed in neurons. This precursor harbored three collagen-like Gly-X-Y repeat motifs and was partially homologous to collagen type XIII. Thus, we named the 9D2 antigen as CLAC (collagen-like Alzheimer amyloid plaque component), and its precurso

Biological activity of soluble CD100. I. The extracellular region of CD100 is released from the surface of T lymphocytes by regulated proteolysis.

CD100 is the first semaphorin described in lymphoid tissues, where it has been shown to be associated with a serine kinase activity. Semaphorins are molecules involved in axon pathfinding during nerve development and act as repellent guidance cues. In the nervous system semaphorins exist as either membrane-bound or secreted forms. We report here a spontaneous processing of membrane CD100, suggesting that it is also produced as a diffusable semaphorin from lymphoid cells. Monomeric and homodimeric forms of CD100 are expressed by T lymphocytes and CD100-transfected fibroblasts. We demonstrate th

Augmentation of L-type calcium current by hypoxia in rabbit carotid body glomus cells: evidence for a PKC-sensitive pathway.

Previous studies have suggested that voltage-gated Ca(2+) influx in glomus cells plays a critical role in sensory transduction at the carotid body chemoreceptors. The purpose of the present study was to determine the effects of hypoxia on the Ca(2+) current in glomus cells and to elucidate the underlying mechanism(s). Experiments were performed on freshly dissociated glomus cells from rabbit carotid bodies. Ca(2+) current was monitored using the whole cell configuration of the patch-clamp technique, with Ba(2+) as the charge carrier. Hypoxia (pO(2) = 40 mmHg) augmented the Ca(2+) current by 24

Manometric changes during retrograde biliary infusion in mice.

The manometric, ultrastructural, radiographic, and physiological consequences of retrograde biliary infusion were determined in normostatic and cholestatic mice. Intraluminal biliary pressure changed as a function of infusion volume, rate, and viscosity. Higher rates of constant infusion resulted in higher peak intraluminal biliary pressures. The pattern of pressure changes observed was consistent with biliary ductular and/or canalicular filling followed by leakage at a threshold pressure. Retrograde infusion with significant elevations in pressure led to paracellular leakage of lanthanum chlo

Interstitial carbonic anhydrase (CA) activity in brain is attributable to membrane-bound CA type IV.

We tested the hypothesis that extracellular membrane-bound carbonic anhydrase (CA) type IV is responsible for the regulation of interstitial pH (pH(o)) transients in brain. Rat hippocampal slices were incubated in phosphatidylinositol-specific phospholipase C (PI-PLC), which cleaves the link of CA IV to the external face of plasma membranes. Then evoked alkaline pH(o) shifts were studied in a recording chamber, using pH microelectrodes. Incubation fluid was saved for later analysis. The ability to buffer a rapid alkaline load was reduced markedly in PI-PLC-treated tissue as compared with adjac

Activity and calcium-dependent mechanisms maintain reliable interneuron synaptic transmission in a rhythmic neural network.

Inputs from glutamatergic excitatory interneurons (EIN) to motor neurons in the lamprey spinal cord locomotor network exhibit activity-dependent depression during spike trains. The mechanism underlying this depression has been examined here, and its relevance to transmitter release during rhythmic activity has been investigated. The depression of EIN inputs was greater after larger initial EPSPs and reduced in low-calcium Ringer's solution, effects that are consistent with depression caused by depletion of releasable transmitter stores. However, the depression was greater at lower stimulation

Riboflavin uptake by the human-derived liver cells Hep G2: mechanism and regulation.

The water-soluble vitamin riboflavin (RF) plays a critical role in many metabolic reactions, and thus, is essential for normal cellular functions and growth. The liver plays a central role in normal RF metabolism and is the site of maximal utilization of the vitamin. The mechanism of liver uptake of RF has been studied in animals, but no information is available describing the mechanism of the vitamin uptake in the human situation and its cellular regulation. In this study, we used the human-derived liver cells Hep G2 as an in vitro model system to address these issues. Uptake of RF by Hep G2

Umbilical cord blood gas analysis at delivery.

BACKGROUND: Umbilical cord blood gas values are better indicators of perinatal asphyxia than Apgar scores. However, the reported normal range of umbilical blood gas values vary greatly in the literature. The aim of this prospective study was to establish the normal range of umbilical cord blood gas values in our labour ward. METHODS AND RESULTS: Umbilical cord blood gas from 153 vaginal deliveries and 52 Caesarean sections for indication other than fetal distress were evaluated. In our labour ward, the mean and standard deviation of umbilical artery pH were 7.21 and 0.08 for vaginal deliverie

Presynaptic inhibition of calcium-dependent and -independent release elicited with ionomycin, gadolinium, and alpha-latrotoxin in the hippocampus.

1. Presynaptic inhibition of synaptic transmission in the hippocampus was investigated by comparing the effects of several agonists on miniature excitatory and inhibitory postsynaptic currents (mEPSCs and mIPSCs). 2. The Ca2+ ionophore ionomycin increased the frequency of mEPSCs and mIPSCs but did not affect their amplitude. Ionomycin-induced release required extracellular Ca2+ and was prevented by pretreatment with botulinum neurotoxin serotype F, like evoked synaptic transmission. Unlike evoked transmission, however, this increase did not involve activation of voltage-dependent Ca2+ channels

Age-related changes in the NMDA receptor/nitric oxide/cGMP pathway in the hippocampus and cerebellum of freely moving rats subjected to transcerebral microdialysis.

The N-methyl-D-aspartate (NMDA) receptor/nitric oxide synthase/guanylate cyclase pathway was studied during aging by monitoring extracellular cGMP in the rat hippocampus and cerebellum during in vivo microdialysis. In the hippocampus the basal cGMP efflux decreased by 50% from 3 to 12 months of age, whereas it remained constant with age in the cerebellum. Locally perfused NMDA (1 mM) evoked remarkable cGMP responses in 3-month-old rats; in the hippocampus the cGMP production was already dramatically reduced at 12 months, whereas in the cerebellum a similar impairment occurred much later (24 mo

Microstructure imaging of prostate cancer by diffusion MRI.

Prostate cancer remains one of the most prevalent malignancies in men worldwide, and early detection, accurate characterization, and staging are critical for optimal patient management. Diffusion magnetic resonance imaging (dMRI) has emerged as a powerful technique for non-invasive in vivo imaging of tissue microstructure, promising to enable unprecedented insights into the pathophysiology of prostate cancer. In this review, we examine the current state and future directions of dMRI techniques for microstructure MRI in prostate cancer applications. We describe the technical challenges of prost

Association of glymphatic dysfunction, free water, white matter integrity and long-term memory performance in aging autistic adults.

BACKGROUND: Autistic adults are at elevated risk of accelerated cognitive aging and Alzheimer’s disease and related dementias (ADRD), yet the underlying neurobiological mechanisms remain poorly understood. Dysfunction in the glymphatic system—a brain-wide network responsible for clearing waste via interstitial fluid flow—may contribute to this vulnerability by promoting extracellular free water (FW) accumulation and white matter (WM) degeneration. METHODS: A total of 113 autistic and 90 age- and sex-matched neurotypical (NT) adults (aged 18–71 years) underwent multimodal MRI scanning and episo

GLT-1 and alcohol.

Glutamate transporter 1 (GLT-1) is responsible for the uptake of glutamate from the extracellular space, predominantly into astrocytes that insulate glutamatergic synapses. This transport is the primary method by which glutamate is removed from the synapse, as, unlike several other neurotransmitters, there is very little reuptake of glutamate by presynaptic neurons following release. As such, GLT-1 is crucial for homeostatic functioning of glutamatergic synapses, with disruptions such as excessive downregulation contributing to excitotoxicity. Alcohol is one example of an exogenous agent that …

A Methodological Approach to Relate Pentapeptide Sequence, Atomistic Self-Assembly, and Gelation Behavior.

The development of biomaterials that mimic the extracellular matrix of the native tissue represents an exciting frontier for tissue engineering and regenerative medicine. Injectable hydrogels made of short, self-assembling peptides offer a promising platform for the delivery and directed differentiation of therapeutic stem cells. However, the rational design of peptide hydrogels remains a significant challenge in tissue engineering due to our lack of understanding of the molecular mechanisms that underlie self-assembly. Although these materials hold great promise, most computational design eff

Neutrophil extracellular trap-related pathology in sporadic brain arteriovenous malformations after radiosurgery: An exploratory histopathological study.

OBJECTIVES: Brain arteriovenous malformations (bAVMs) shunt arterial blood directly into veins, bypassing capillaries and predisposing young patients to hemorrhagic stroke. Stereotactic radiosurgery (SRS) can result in curative bAVM obliteration through delayed thrombosis, yet hemorrhage risk persists during the latency period. The mechanisms underlying this delayed response remain unclear. Neutrophil extracellular traps (NETs), web-like structures of chromatin DNA and granule proteins, have been identified in bAVMs. We investigated whether citrullinated histone H3 (CitH3)-positive cells, used
