The effect of butenolide on behavioral and morphological changes in two marine fouling species, the barnacle Balanus amphitrite and the bryozoan Bugula neritina.

Butenolide [5-octylfuran-2(5H)-one] is a very promising antifouling compound. Here, the effects of butenolide on larval behavior and histology are compared in two major fouling organisms, viz. cypris larvae of Balanus amphitrite and swimming larvae of Bugula neritina. Butenolide diminished the positive phototactic behavior of B. amphitrite (EC50=0.82 μg ml(-1)) and B. neritina (EC50=3 μg ml(-1)). Its effect on the attachment of cyprids of B. amphitrite was influenced by temperature, and butenolide increased attachment of larvae of B. neritina to the bottom of the experimental wells. At concent

The hepatoprotective activity of kinsenoside from Anoectochilus formosanus.

Carbon tetrachloride (CCl(4)) causes chronic hepatitis, featuring an increase in hepatic hydroxyproline, spleen weight and serum GPT levels and a decrease in plasma albumin levels. Crude extracts of fresh whole plants of Anoectochilus formosanus showed inhibition of chronic hepatitis induced by CCl(4) in mice. Bioactivity-guided fractionation and spectroscopic analysis revealed that kinsenoside was the most active compound. In an in vitro study, the LD(50) values for H(2)O(2)-induced cytotoxicity in BALB/c normal liver cells were significantly higher after kinsenoside pretreatment than after v

A thirteen week dietary toxicity study with 7-hydroxymatairesinol potassium acetate (HMRlignan) in rats.

Plant lignan 7-hydroxymatairesinol (7-HMR) is a novel precursor of the mammalian lignan enterolactone. A 13 week toxicity study at dietary levels of 0, 0.25, 1, and 4% (w/w) of potassium acetate complex of 7-HMR (HMRlignan) was conducted in the Wistar rat. These dietary levels resulted in an average daily intake of 160, 640, and 2600 mg HMRlignan/kg body weight/day, respectively. A considerable systemic exposure of HMRlignan was verified by dose-related increases in plasma total (conjugated and unconjugated) concentration of 7-HMR and metabolites enterolactone, 7-hydroxyenterolactone, and mata

Natural Product 4-Acetylantroquinonol B Enhances NK Cell-Mediated Immunity Against Glioblastoma.

Glioblastoma (GBM) recurs rapidly despite surgery and radiochemotherapy and is associated with profound natural killer (NK) cell dysfunction. Adoptive NK cell transfer has emerged as a plausible immunotherapy for GBM and is being tested clinically; however, its efficacy is limited by the suppressive tumor microenvironment, low NK infiltration, and metabolic exhaustion. 4-Acetylantroquinonol B (4-AAQB), a ubiquinone derivative purified from Antrodia cinnamomea, has demonstrated antitumor and anti-inflammatory properties. These properties suggest that 4-AAQB may rescue or enhance NK cell activit

Cassane butenolides from the fruits of Caesalpinia pulcherrima.

Seven undescribed cassane diterpenes, caeslpulrins A-G (1, 3, 4, and 6-9), the C-12 ethoxy derivatives of caeslpulrins A and C (2 and 5), along with five artificial or known cassane diterpenes (10-14) and eight known sesquiterpenoids (15-22) were isolated from the fruits of Caesalpinia pulcherrima. Their structures were elucidated by extensive spectroscopic analyses, including HR-ESI-MS, 1D and 2D NMR, and electronic circular dichroism (ECD) calculations. Caeslpulrins A-F were identified as a group of rare cassane butenolides possessing a C-3 benzoyloxy unit. This is the first report of a seri

Effects of phenylbutazone, firocoxib, and dipyrone on the diuretic response to furosemide in horses.

BACKGROUND: Treatment with phenylbutazone (nonselective COX inhibitor) decreases the diuretic and natriuretic effects of furosemide by nearly 30% but the effects of COX-2 specific inhibitors (firocoxib) and atypical NSAIDs (dipyrone) are unknown. HYPOTHESIS: Furosemide-induced diuresis after pretreatment with firocoxib or dipyrone is diminished to a lesser extent than after pretreatment with phenylbutazone. ANIMALS: Eight healthy mares. METHODS: Each mare received 4 treatments in a prospective experimental crossover study using a replicated 4 × 4 Latin Square design: furosemide alone (FU),

Comparing and Contrasting the Multiple Roles of Butenolide Plant Growth Regulators: Strigolactones and Karrikins in Plant Development and Adaptation to Abiotic Stresses.

Strigolactones (SLs) and karrikins (KARs) are both butenolide molecules that play essential roles in plant growth and development. SLs are phytohormones, with SLs having known functions within the plant they are produced in, while KARs are found in smoke emitted from burning plant matter and affect seeds and seedlings in areas of wildfire. It has been suggested that SL and KAR signaling may share similar mechanisms. The α/β hydrolases DWARF14 (D14) and KARRIKIN INSENSITIVE 2 (KAI2), which act as receptors of SL and KAR, respectively, both interact with the F-box protein MORE AXILLARY GROWTH 2

N-(3-oxododecanoyl)-l-homoserine-lactone, a quorum sensing molecule, affects cellular content of nonprotein thiol content in rat lymphocytes: Its relation with intracellular Zn2.

Cellular actions of N-(3-oxododecanoyl)-l-homoserine-lactone (ODHL), a quorum sensing molecule of bacteria, were studied on rat thymocytes using a flow cytometer with appropriate fluorescent dyes to elucidate the effects of ODHL on host cells. A bell-shaped concentration-response relation was observed in the ODHL-induced changes in cellular glutathione content ([GSH]i). ODHL concentration-dependently increased intracellular Zn2+ levels ([Zn2+]i) and cellular O2- content ([O2-]i). The bell-shaped relation induced by ODHL can be explained as follows: a low concentration of ODHL is expected to in

Unusual presentation of a patient with GBL withdrawal: a case report.

GBL (gamma-butyro-lactone) is converted to Gamma hydroxyl butyrate (GHB) in the body. GBL and GHB are available in liquid form and powder form. Once categorised under "legal highs", these two are not associated with any dependence or withdrawal in animal studies. But there are case reports indicating their high dependence potential in humans. We here present a case of a 29 year old who came to the attention of psychiatric services with very bizarre presentation and needed a host of investigations and expert views from various medical disciplines. He was treated mainly symptomatically followed

Antiproliferative compounds of Artabotrys madagascariensis from the Madagascar rainforest.

Bioassay-guided fractionation of an ethanol extract of Artabotrys madagascariensis led to the isolation of the new compound artabotrene (1), two butenolides (2 and 3), and the tetracyclic triterpene polycarpol (4). Structure elucidation was determined on the basis of one and two-dimensional NMR, and absolute configuration of compounds 2-4 was verified by analysis of CD and optical rotation spectra. Two of the isolates, melodorinol (2) and acetylmelodorinol (3), were found to display antiproliferative activity against five different tumour cell lines with IC50 values ranging from 2.4 to 12 micr

The global posttranscriptional regulator RsmA modulates production of virulence determinants and N-acylhomoserine lactones in Pseudomonas aeruginosa.

Posttranscriptional control is known to contribute to the regulation of secondary metabolism and virulence determinants in certain gram-negative bacteria. Here we report the isolation of a Pseudomonas aeruginosa gene which encodes a global translational regulatory protein, RsmA (regulator of secondary metabolites). Overexpression of rsmA resulted in a substantial reduction in the levels of extracellular products, including protease, elastase, and staphylolytic (LasA protease) activity as well as the PA-IL lectin, hydrogen cyanide (HCN), and the phenazine pigment pyocyanin. While inactivation o

Endogenous gamma-hydroxybutyric acid levels in postmortem blood.

The goal of this study was to determine how the postmortem interval and duration of storage of blood at 4 degrees C affect endogenous gamma-hydroxybutyric acid (GHB) levels in blood. Forty-three autopsy cases of non-users of GHB were involved. The postmortem interval ranged from 8 to 132 h. Blood samples were collected and stored without any preservatives at 4 degrees C for 1 day up to 15 months until analysis. In some cases, samples were also stored at -20 degrees C for 10 days to 7 months to determine GHB levels at autopsy. Blood GHB concentrations were measured by headspace gas chromatograp

Design of Novel Furan β-Butenolide Compounds with Low Bee Toxicity Based on the Binding Pharmacophore Model to AChBP.

Developing novel-structured nAChR modulators is urgent due to the high bee toxicity of current neonicotinoids. In this work, a pharmacophore model of an nAChR modulator was generated, and a series of novel furan β-butenolide compounds were designed and synthesized. The bioassay study demonstrated that the insecticidal activity of compound 8c was the best (LC50 = 54.68 μg/mL) against Myzus persicae, possibly due to its binding mode being more similar to that of flupyradifurone, as well as its interactions with specific key residues (ARG55, TRP143, and TYR89) within Ls-AChBP. Notably, compound 8

Ligustilide Ameliorates Traumatic Brain Injury in Aged Mice by Attenuating Microglia-Mediated Neuroinflammation.

Traumatic brain injury (TBI) is common in the aged population and rapidly triggers a pro-inflammatory response in microglia, leading to severe secondary damage. Ligustilide (LIG), a natural compound with excellent blood-brain barrier (BBB) penetration, shows great potential in neuroprotection, primarily due to its anti-inflammatory, antioxidant, and pro-autophagic properties. However, the role of LIG in elderly TBI remains unclear. This study aims to investigate the effects of LIG on elderly TBI and explore its mechanisms of action. In vivo, we assessed the impact of LIG on behavioral outcomes

Ligustilide improves functional constipation by non-covalently activating TRPA1 in colon tissue.

ETHNOPHARMACOLOGICAL RELEVANCE: Angelica sinensis (Oliv.) Diels (AS), a medicinal plant renowned for its constipation-relieving properties, lacks comprehensive studies on its active pharmaceutical ingredients (APIs) and underlying mechanisms. In the gastrointestinal tract, TRP channels enhance colonic mucus secretion, expedite intestinal motility, and regulate gastrointestinal hormones; however, few reports have systematically established the relationship between TRPs and ligustilide (Lig), a key API of AS. AIM OF THE STUDY: This study aimed to explore the pharmacodynamic properties of AS in

Flexible 2,4-diaminopyrimidine bearing a butyrolactone as Plasmodium falciparum dihydrofolate reductase inhibitors.

Recently, P218, a new flexible antifolate targeting Plasmodium falciparum dihydrofolate reductase (PfDHFR), has entered its clinical trial with good safety profile and effective Pf infection prevention. However, it carries a free carboxyl terminal, which is hydrophilic and prone to metabolic glucuronidation. Here, a new series of P218 analogues carrying butyrolactone has been synthesized with the purpose of enhancing lipophilicity and minimizing metabolic instability. The inhibition constants against the mutant PfDHFR enzymes are in sub-nanomolar level and the antimalarial activity against ant
