Brown crusts on infant owing to contact dermatitis associated with ethanol disinfection.

A 2-year-old girl developed dermatitis with atypical brown crusts after ethanol disinfection. Since many Oriental people have genetically reduced acetaldehyde dehydrogenase type 2 (ALDH2) activity, ethanol disinfection causes acetaldehyde to accumulate in the skin, resulting in dermatitis.

Protein adduct species in muscle and liver of rats following acute ethanol administration.

AIMS: Previous immunohistochemical studies have shown that the post-translational formation of aldehyde-protein adducts may be an important process in the aetiology of alcohol-induced muscle disease. However, other studies have shown that in a variety of tissues, alcohol induces the formation of various other adduct species, including hybrid acetaldehyde-malondialdehyde-protein adducts and adducts with free radicals themselves, e.g. hydroxyethyl radical (HER)-protein adducts. Furthermore, acetaldehyde-protein adducts may be formed in reducing or non-reducing environments resulting in distinct …

Atmospheric ethanol in London and the potential impacts of future fuel formulations.

There is growing global consumption of non-fossil fuels such as ethanol made from renewable biomass. Previous studies have shown that one of the main air quality disadvantages of using ethanol blended fuels is a significant increase in the production of acetaldehyde, an unregulated and toxic pollutant. Most studies on the impacts of ethanol blended gasoline have been carried out in the US and Brazil, with much less focus on the UK and Europe. We report time resolved measurements of ethanol in London during the winter and summer of 2012. In both seasons the mean mixing ratio of ethanol was arou…

ALDH2*2 but not ADH1B*2 is a causative variant gene allele for Asian alcohol flushing after a low-dose challenge: correlation of the pharmacokinetic and pharmacodynamic findings.

OBJECTIVE: It has been well documented that variant alleles of both ADH1B*2 of alcohol dehydrogenase (ADH) and ALDH2*2 of aldehyde dehydrogenase (ALDH) protect against the development of alcoholism in East Asians. However, it remains unclear whether ADH1B*2 contributes significantly toward the accumulation of systemic blood acetaldehyde and whether it plays a critical role in the alcohol flushing reaction. PARTICIPANTS AND METHODS: Sixty-one adult Han Chinese men were recruited and divided into six combinatorial genotypic groups: ALDH2*1/*1-ADH1B*1/*1 (12), ALDH2*1/*1-ADH1B*1/*2 (11), ALDH2*1…

Ni- and Zn-Doping Effects on Cu/SiO2 Catalysts in Nonoxidative Ethanol Dehydrogenation.

Nonoxidative ethanol dehydrogenation opens a pathway for the sustainable production of acetaldehyde and butadiene. One crucial aspect of producing butadiene by the Lebedev process is the high-temperature stability of ethanol to acetaldehyde conversion. However, copper-based catalysts, despite exhibiting high activity and selectivity, suffer from sintering and coking and need to be improved for successful industrial applications. Herein, we show Cu-based (∼2.5 wt %) catalysts doped with Ni and Zn (0.028-0.36 wt %) to improve the catalytic performance of nanoparticles. The catalysts were prepare…

Suppression of ethanol intake following administration of dopamine-beta-hydroxylase inhibitors in rats.

A comparison was made between the efficacy of action of dopamine-beta-hydroxylase and/or aldehyde dehydrogenase inhibition in suppressing voluntary ethanol consumption in rats. Calcium carbimide, which caused the largest increase in blood acetaldehyde levels following ethanol injections, was the least effective in reducing ethanol consumption, whereas FLA-63, which had a smaller effect on blood acetaldehyde levels, was the most potent suppressor of ethanol ingestion. Disulfiram, which inhibits both aldehyde dehydrogenase and dopamine-beta-hydroxylase had an intermediate effect in terms of rais…

Effects of GSH on Alcohol Metabolism and Hangover Improvement in Humans: A Randomized Double-Blind Placebo-Controlled Crossover Clinical Trial.

BACKGROUND: The definition of alcohol hangovers refers to a combination of mental and physical side effects that occur after drinking. One of the ways that hangovers can be ameliorated is by promoting the rapid and effective elimination of acetaldehyde to alleviate the discomfort it causes. This study aimed to investigate the effects of GSH (yeast extract containing 50 mg of glutathione) on the hangover-relieving effect. METHODS: A randomized double-blind placebo-controlled crossover clinical trial was conducted with 40 participants who reported experiencing hangover symptoms. Participants co…

Acute and chronic effects of ethanol and its metabolites on vascular production of prostacyclin in rats.

The cutaneous vasodilation produced by ethanol is exaggerated when acetaldehyde levels are increased after aldehyde dehydrogenase inhibition, producing a flushing reaction, the mechanism of which is unknown. The authors investigated whether ethanol and its metabolites affect the vascular release of prostacyclin, a potent vasodilator, and whether such an effect might be modified by chronic alcohol consumption. Aortic rings from rats fed Chow ad libitum or pair-fed liquid diets containing either ethanol (36% of energy) or isocaloric carbohydrate for 4 to 5 weeks were incubated in Krebs-Ringer bi…

Role of intestinal bacterial overgrowth in ethanol production and metabolism in rats.

To investigate the role of intestinal bacterial overgrowth on the production and metabolism of ethanol, rats with a jejunal self-filling diverticulum (blind-loop) were compared to controls with a self-emptying diverticulum. Both endogenous ethanol and acetaldehyde were found in the blind-loop contents. Intragastric administration of sucrose produced a marked increase in acetaldehyde and acetate in the portal venous blood, with only a modest elevation of ethanol. Blind-loop contents readily oxidized ethanol to acetaldehyde in a concentration-dependent manner and more actively under aerobic than…

Studies on carbon tetrachloride-ethanol interactions in mice.

Male C57BL/6J (C57) and DBA/2J (DBA) inbred mice were dosed with either corn oil or carbon tetrachloride (CCl4) in corn oil 24 h before a 3.25 g/kg i.p. dose of ethanol. The CCl4 doses were increased in approximate half-log intervals (5, 15, 50, 150 or 500 microliter/kg, intragastrically (i.g.)). Blood acetaldehyde concentrations were significantly increased 1, 2, 3 and 4 h after ethanol administration at CCl4 doses of 15 microliter/kg and higher, with DBA and C57 mice exhibiting similar dose-response effects up to the 150 microliter/kg dose. A CCL4 dose of 500 microliter/kg produced differenc…

Electrophysiological effects of monoamine-derived aldehydes on single neurons in neocortex and cerebellum in rats.

The electrophysiological effects of aldehydes derived from several monoamines were studied on single neurons in the cerebellum and neocortex of rats. The aldehydes derived from dopamine (3,4-dihydroxyphenylacetaldehyde) and serotonin (5-hydroxy-3-acetaldehyde) were prepared as stable disulfite complexes, from which free aldehydes were extracted. Serotonin and 5-hydroxy-3-acetaldehyde caused pronounced depression of firing rates both of cerebellar Purkinje neurons and neurons in prefrontal cortex. When locally applied from multibarrel micropipettes by pressure ejection, 5-hydroxy-3-acetaldehyde…

Lipid peroxidation in alcoholic myopathy and cardiomyopathy.

The hypothesis is presented that lipid peroxidation is responsible for the damage in skeletal and cardiac muscle of chronic alcoholic subjects. The enhanced lipid peroxidation is caused by the accumulation of oxygen radicals. Both excessive production and decreased disposal of oxygen radicals can arise from the acetaldehyde formed in the oxidation of ethanol. Although acetaldehyde from hepatic sources may contribute, muscle itself can generate significant amounts of acetaldehyde through the action of muscle catalase. The effects of alcohol on other tissues, and its known long-term effects on m…

A study of the calcium carbimide-ethanol interaction in man.

In six male alcoholic volunteers, oral administration of calcium carbimide (0.7 mg/kg) before ingestion of ethanol (0.5 g/kg) produced an interaction consisting of increased blood acetaldehyde level, tachycardia and increased pulse pressure, which was due mainly to decreased diastolic blood pressure. For these experimental conditions, calcium carbimide had a duration of action of at least 24 h to produce an interaction with ethanol. The order of intensity of the interaction with regard to the calcium carbimide pretreatment time interval was 4 greater than 8 approximately 12 greater than 24 h. …

Effects of ethanol and ethanol metabolites on intrinsic function of mesenteric resistance arteries.

Evidence suggests that ethanol-induced hypertension is associated with increased cardiovascular responsiveness to vasopressors in vivo and enhanced reactivity of isolated arteries to vasopressors ex vivo. The underlying mechanisms are not well understood and the contribution of ethanol metabolites to vascular effects induced by ethanol consumption are unclear. Mesenteric resistance arteries were harvested from Sprague-Dawley rats. Pressure myography was utilized to test effects of ethanol, acetaldehyde and phosphatidylethanol on myogenic tone and on vasoconstriction induced by phenylephrine, a…

Pathogenesis of alcoholic liver disease: role of oxidative metabolism.

Alcohol consumption is a predominant etiological factor in the pathogenesis of chronic liver diseases, resulting in fatty liver, alcoholic hepatitis, fibrosis/cirrhosis, and hepatocellular carcinoma (HCC). Although the pathogenesis of alcoholic liver disease (ALD) involves complex and still unclear biological processes, the oxidative metabolites of ethanol such as acetaldehyde and reactive oxygen species (ROS) play a preeminent role in the clinical and pathological spectrum of ALD. Ethanol oxidative metabolism influences intracellular signaling pathways and deranges the transcriptional control…

Ciliastatic components in the gas phase of cigarette smoke.

The gas phase of cigarette smoke, separated into its components by gas chromatography, was passed across a ciliated specimen. The acetaldehyde, acrolein, and hydrogen cyanide produced strong inhibition of the ciliary beat. A filter which removed most of the acetaldehyde and acrolein from smoke did not reduce the inhibitory effect of the gas phase of that smoke, whereas a filter that removed most of the hydrogen cyanide did reduce inhibition.
