Genomic comparison and phenotypic profiling of small colony variants of Burkholderia pseudomallei.

Burkholderia pseudomallei (B. pseudomallei) is an intracellular pathogen that causes melioidosis, a life-threatening infection in humans. The bacterium is able to form small colony variants (SCVs) as part of the adaptive features in response to environmental stress. In this study, we characterize the genomic characteristics, antimicrobial resistance (AMR), and metabolic phenotypes of B. pseudomallei SCV and wild type (WT) strains. Whole-genome sequence analysis was performed to characterize the genomic features of two SCVs (CS and OS) and their respective parental WT strains (CB and OB). Phylo

Patterns of Sphaeridiotrema pseudoglobulus infection in sympatric and allopatric hosts (Bithynia tentaculata) originating from widely separated sites across the USA.

In circumstances where populations of invasive species occur across variable landscapes, interactions among invaders, their parasites, and the surrounding environment may establish local coevolutionary trajectories for the participants. This can generate variable infection patterns when parasites interact with sympatric versus allopatric hosts. Identifying the potential for such patterns within an invasive-species framework is important for better predicting local infection outcomes and their subsequent impacts on the surrounding native community. To begin addressing this question, we exposed

Evidence for divergent patterns of local selection driving venom variation in Mojave Rattlesnakes (Crotalus scutulatus).

Snake venoms represent an enriched system for investigating the evolutionary processes that lead to complex and dynamic trophic adaptations. It has long been hypothesized that natural selection may drive geographic variation in venom composition, yet previous studies have lacked the population genetic context to examine these patterns. We leverage range-wide sampling of Mojave Rattlesnakes (Crotalus scutulatus) and use a combination of venom, morphological, phylogenetic, population genetic, and environmental data to characterize the striking dichotomy of neurotoxic (Type A) and hemorrhagic (Ty

Differential miRNA expression in the three-spined stickleback, response to environmental changes.

miRNAs play important role in the various physiological and evolutionary processes, however, there is no data allowing comparison of evolutionary differences between various ecotypes adapted to different environmental conditions and specimen demonstrating immediate physiological response to the environmental changes. We compared miRNA expression profiles between marine and freshwater stickleback populations of the three-spined stickleback to identify the evolutionary differences. To study the immediate physiological response to foreign environment, we explored the changes induced by transfer o

The king cobra genome reveals dynamic gene evolution and adaptation in the snake venom system.

Snakes are limbless predators, and many species use venom to help overpower relatively large, agile prey. Snake venoms are complex protein mixtures encoded by several multilocus gene families that function synergistically to cause incapacitation. To examine venom evolution, we sequenced and interrogated the genome of a venomous snake, the king cobra (Ophiophagus hannah), and compared it, together with our unique transcriptome, microRNA, and proteome datasets from this species, with data from other vertebrates. In contrast to the platypus, the only other venomous vertebrate with a sequenced gen

Upregulation of the mitochondrial Lon Protease allows adaptation to acute oxidative stress but dysregulation is associated with chronic stress, disease, and aging.

The elimination of oxidatively modified proteins is a crucial process in maintaining cellular homeostasis, especially during stress. Mitochondria are protein-dense, high traffic compartments, whose polypeptides are constantly exposed to superoxide, hydrogen peroxide, and other reactive species, generated by 'electron leakage' from the respiratory chain. The level of oxidative stress to mitochondrial proteins is not constant, but instead varies greatly with numerous metabolic and environmental factors. Oxidized mitochondrial proteins must be removed rapidly (by proteolytic degradation) or they

Production and partial characterization of bioemulsifier from a chromium-resistant actinobacteria.

Surface-active compounds such as synthetic emulsifiers have been used for several decades, both for the degradation of hydrocarbons and increasing desorption of soil-bound metals. However, due to their high toxicity, low degradability, and production costs unaffordable for use in larger ecosystems, synthetic emulsifiers have been gradually replaced by those derived from natural sources such as plants or microbes. In previous studies, the bacterium Streptomyces sp. MC1 has shown the ability to reduce and/or accumulate Cr(VI), a highly promising advance in the development of methods for environm

The role of the striatum in adaptation learning: a computational model.

To investigate the functional role of the striatum in visuo-motor adaptation, we extend the DIRECT-model for visuo-motor reaching movements formulated by Bullock et al.(J Cogn Neurosci 5:408-435,1993) through two parallel loops, each modeling a distinct contribution of the cortico-cerebellar-thalamo-cortical and the cortico-striato-thalamo-cortical networks to visuo-motor adaptation. Based on evidence of Robertson and Miall(Neuroreport 10(5): 1029-1034, 1999), we implement the function of the cortico-cerebellar-thalamo-cortical loop as a module that gradually adapts to small changes in sensori

Living in a changing environment: insights into host adaptation in Neisseria meningitidis from comparative genomics.

Neisseria meningitidis (the meningococcus) colonizes the human nasopharynx of about 10% of the human population. However, for reasons that are still mostly unknown meningococci occasionally enter the cerebrospinal fluid leading to often fatal bacterial meningitis especially in children and young adults. The genetic basis for the observed differences in the pathogenic potential of different strains has only partially been unravelled so far. With the advent of whole genome sequencing technologies, complete genome sequences from three pathogenic meningococcal strains have become available and all

Idiopathic osteoporosis: an evolutionary dys-adaptation?

Osteoporosis is characterised by simultaneous net bone growth and net resorption on different surfaces, suggesting that systemic factors are not the sole explanation for the findings. The main clinical consequence is fracturing in the largely trabecular bones of the spine, hip, and radius, and the key problem in these areas is finding an explanation for the preferential loss of transverse trabeculae. In normal bone, local maintenance depends on a negative feedback response to intermittent compression strain, and it is concluded, from biomechanical analysis of the response required to achieve n

The security circuit: a proposed construct for the central nervous system.

The theory of the Security Circuit offers a new conceptual framework for the numerous brain functions pertaining to survival. Many such activities are related to the coordination of the conventional physiological systems in the maintenance of homeostasis and the defense of the internal environment. This work, however, focuses on that aspect of brain function that makes possible man's interaction with external environment, thereby facilitating individual and group adaptation and survival. In our schema we greatly enlarge the conceptualization of the autonomic nervous system. We postulate the ex

Mechanical optimization of bone.

In bending, the mechanical strength of tubular bone can be estimated by the area moment of inertia (I) = 1/4 x pi x (R4 - r4) (R: external radius, and r: internal radius). Mechanical strength of bone is dissociated from bone density, since radiological density is different from inertia. When the cross-sectional area is constant (C = R2 - r2) in this equation, inertia can be expressed by the equation of (I) = 1/4 x pi x (2Cr2 + C2). Inertia increases with increases of the external and internal radii. According to the above equation, increase of inertia depending on the expansion of radii is ine

The Myelin Sheath as a Multilamellar Electromechanical System: Bilayer Stack Analogs, Energy Buffering, and Biological Memory.

The myelin sheath has traditionally been viewed as a passive electrical insulator that accelerates nerve impulse propagation. Recent experimental studies, however, indicate that myelin is a dynamic biological material whose structure and hydration state adapt to neuronal activity, metabolic conditions, and environmental perturbations. Building on these observations, we propose a conceptual framework that treats myelin as an adaptive electromechanical multilamellar interface, in which coupled lipid bilayers, hydration layers, and interlayer interactions influence energy dissipation, structural

Emerging roles of vascular brassinosteroid receptors of the BRI1-like family.

Brassinosteroids (BRs) are essential hormones for plant growth and development that are perceived at the plasma membrane by a group of Leucine-Rich Repeat Receptor-Like Kinases (LRR-RLKs) of the BRASSINOSTEROID INSENSITIVE 1 (BRI1) family. The BRI1 receptor was first discovered by genetic screenings based on the dwarfism of BR-deficient plants. There are three BRI1 homologs, named BRI1-like 1, 2 and 3 (BRLs), yet only BRL1 and BRL3 behave as functional BR receptors. Whereas the BRI1 pathway operates in the majority of cells to promote growth, BRL receptor signaling operates under specific spat

Invariant template matching in systems with spatiotemporal coding: A matter of instability.

We consider the design principles of algorithms that match templates to images subject to spatiotemporal encoding. Both templates and images are encoded as temporal sequences of samplings from spatial patterns. Matching is required to be tolerant to various combinations of image perturbations. These include ones that can be modeled as parameterized uncertainties such as image blur, luminance, and, as special cases, invariant transformation groups such as translation and rotations, as well as unmodeled uncertainties (noise). For a system to deal with such perturbations in an efficient way, they

[Sensitivity and adaptation of various Brucella species to acid shock].

The sensitivity to acid shock (pH 2.5, 2.8, 3.0, and 3.3) and the capacity to adaptive acid tolerance response (ATR) is studied in the reference Brucella strains differing by the origin, biological, and virulent properties. The ability of Brucella to survive under conditions of acid pH depended on the medium composition, growth phase, and preliminary adaptation at pH 5.6. B. suis biovar 1 and B. melitensis biovar 3 were more resistant to low pH than other Brucella species. Adaptation by overnight growth at pH 5.6 (stationary phase) induced ATR to the acid shock (pH 3.3-3.0) in all Brucella spe

Cancer cell plasticity and therapeutic resistance: mechanisms, crosstalk, and translational perspectives.

Resistance to targeted cancer therapies is a significant barrier to favorable treatment outcomes. Malignant cells can tolerate and resist drug treatments due to their biological flexibility. Specifically, slow-cycling drug-resistant cells may achieve permanent resistance to the treatment or restore sensitivity upon cessation of therapy. Enhancing cancer treatment methodologies necessitates a deeper understanding of the adaptability of tumor cells. Drug resistance and cellular heterogeneity are closely associated with cancer cell adaptability. Alterations in cellular signaling, interactions wit

Dosimetric performance of cone beam CT-guided adaptive carbon-ion radiotherapy with daily replanning for pancreatic cancer.

PURPOSE: We investigated whether an ultra-hypofractionated carbon-ion radiotherapy (CIRT) protocol for pancreatic cancer (PC) could produce satisfactory dosimetric results with or without cone-beam CT-guided adaptive replanning and explored the potential dosimetric advantages of the adapted protocol. METHODS: Eleven PC patients who underwent CBCT-guided online adaptive photon radiotherapy were selected. Data were imported into a CIRT treatment planning software to develop new plans for an ultra-hypofractionated CIRT protocol. Prescriptions and constraints were recalculated for a five-fraction
