Genetic characterization of immune adaptor molecule MyD88 in Culex pipiens complex (Diptera: Culicidae) mosquitoes from China.

Mosquitoes of the Culex (Cx.) pipiens complex are vectors of severe diseases including West Nile fever by West Nile virus, Japanese encephalitis by Japanese encephalitis virus, and Lymphatic filariasis by filarial nematode Wuchereria bancrofti. As a major portion of mosquito immune system, the Toll pathway implicates in response against infections of mosquito-borne pathogens and biocontrol agents. The genetic diversity of immune-related molecules is expected to be a feasible and effective introduction to expand our knowledge of the mosquito-microbe interplay. However, a comprehensive descripti

Characterization of the CD28 signaling pathway related genes (CD28, Grb2 and SLP-76) of golden pompano (Trachinotus ovatus).

CD28 signaling pathway plays critical roles in the T cell activation. The golden pompano (Trachinotus ovatus, Tro) is a commercially important marine fish that widely cultured in southern China. Frequently pathogenic disease outbreaks greatly hinder the industry of golden pompano. Understanding the T cell activation signaling pathway of golden pompano contributes to prevent pathogenic diseases using immunological strategies. In the present study, three molecules of CD28 signaling pathway (TroCD28, growth factor receptor-bound protein 2 (TroGrb2) and SH2 domain-containing leukocyte phosphoprote

The seven human coronavirus main proteases exhibit differential modulation of NF-κB suppression via TAB1 cleavage.

The divergence in pathogenicity among human coronaviruses (HCoVs), from causing common cold to lethal pneumonia, may indicate distinct viral immune evasion strategies. However, the molecular basis of their differential immune evasion strategies remains unclear. In this study, we find that the innate immune adapter protein TGF-β activated kinase 1 (TAB1) is a universal target of the main proteases (Mpros) of all seven HCoVs. We show that Mpros from coronaviruses that cause severe disease (SARS-CoV-2, SARS-CoV, and MERS-CoV) efficiently cleave TAB1 at two distinct sites (Q10 and Q444), disruptin

Post-infarction KLHL40-mediated regulation of cardiac sarcomeric integrity and function.

BACKGROUND: Cardiac sarcomeric remodeling after myocardial infarction (MI) plays a pivotal role in post-injury cardiac dysfunction, yet the molecular mechanisms governing this process remain incompletely understood. OBJECTIVE: We aimed to elucidate the role of Kelch-like protein 40 (KLHL40) in sarcomeric protein remodeling and calcium signaling regulation after MI. METHODS: By integrating transcriptomic and proteomic datasets with experimental validation, we characterized dynamic changes in KLHL40 expression and its effects on sarcomeric components and myocyte state following hypoxia. KLHL40

Tetrabromophthalate bis(2-ethylhexyl) ester (TBPH) exposure induces renal inflammation injury by activating MAVS-IRF3 signaling pathway in vitro and in vivo models.

Tetrabromophthalate bis(2-ethylhexyl) ester (TBPH) is a common brominated flame retardant, which exhibits environmental persistence, the potential for bioaccumulation, and concerning toxicological effects. But until now, there is limited data on TBPH-induced nephrotoxicity. Therefore, this study assessed the impact of TBPH exposure on renal cells and tissues through in vitro and in vivo models. A series of biochemical experiments, including Western-blot, indirect immunofluorescence, ELISA, SiRNA, H&E and immunohistochemistry were carried out. In in vitro models, TBPH exposure dose-dependently

Aggregatin is a mitochondrial regulator of MAVS activation to drive innate immunity.

Mitochondrial antiviral-signaling protein (MAVS) is a key adapter protein required for inducing type I interferons (IFN-Is) and other antiviral effector molecules. The formation of MAVS aggregates on mitochondria is essential for its activation; however, the regulatory mitochondrial factor that mediates the aggregation process is unknown. Our recent work has identified the protein Aggregatin as a critical seeding factor for β-amyloid peptide aggregation. Here we show that Aggregatin serves as a cross-seed for MAVS aggregates on mitochondria to orchestrate innate immune signaling. Aggregatin is

Novel Aptamers Targeting Sclerostin Loop3 Improve Skeletal and Muscle Properties Without Adverse Cardiovascular Effects in Orchiectomized Mice.

BACKGROUND: The Wnt/β-catenin pathway and its bone-specific inhibitor, sclerostin, play important roles in skeletal development and homeostasis. The humanized sclerostin antibody, romosozumab, can significantly increase bone mineral density (BMD) of patients with osteoporosis, but it may also increase cardiovascular adverse events, particularly in male patients. We try to investigate the effects of novel aptamers targeting the sclerostin loop3 on the skeleton and muscle of orchiectomized (ORX) mice. METHODS: After 12 weeks of ORX surgery, mice were randomly assigned to receive treatment with

WDR77 inhibits prion-like aggregation of MAVS to limit antiviral innate immune response.

RIG-I-MAVS signaling pathway plays a crucial role in defending against pathogen infection and maintaining immune balance. Upon detecting viral RNA, RIG-I triggers the formation of prion-like aggregates of the adaptor protein MAVS, which then activates the innate antiviral immune response. However, the mechanisms that regulate the aggregation of MAVS are not yet fully understood. Here, we identified WDR77 as a MAVS-associated protein, which negatively regulates MAVS aggregation. WDR77 binds to MAVS proline-rich region through its WD2-WD3-WD4 domain and inhibits the formation of prion-like filam

FYN-TRAF3IP2 induces NF-κB signaling-driven peripheral T cell lymphoma.

Angioimmunoblastic T cell lymphoma (AITL) and peripheral T cell lymphoma not-otherwise-specified (PTCL, NOS) have poor prognosis and lack driver actionable targets for directed therapies in most cases. Here we identify FYN-TRAF3IP2 as a recurrent oncogenic gene fusion in AITL and PTCL, NOS tumors. Mechanistically, we show that FYN-TRAF3IP2 leads to aberrant NF-κB signaling downstream of T cell receptor activation. Consistent with a driver oncogenic role, FYN-TRAF3IP2 expression in hematopoietic progenitors induces NF-κB-driven T cell transformation in mice and cooperates with loss of the Tet2

The role of BAG3 in dilated cardiomyopathy and its association with Charcot-Marie-Tooth disease type 2.

Bcl2-associated athanogene 3 (BAG3) is a multifunctional cochaperone responsible for protein quality control within cells. BAG3 interacts with chaperones HSPB8 and Hsp70 to transport misfolded proteins to the Microtubule Organizing Center (MTOC) and degrade them in autophagosomes in a process known as Chaperone Assisted Selective Autophagy (CASA). Mutations in the second conserved IPV motif of BAG3 are known to cause Dilated Cardiomyopathy (DCM) by inhibiting adequate removal of non-native proteins. The proline 209 to leucine (P209L) BAG3 mutant in particular causes the aggregation of BAG3 and

A novel homozygous truncating variant in PPFIBP1 further delineates PPFIBP1-associated neurodevelopmental disorder.

Neurodevelopmental disorders (NDDs) are classified as a group of disorders affecting function and development of the brain and having wide clinical variability. Herein, we describe two affected individuals segregating a recessive NDD. The affected individuals exhibited phenotypes such as global developmental delay (GDD), intellectual disability (ID), microcephaly and speech delay. Whole-exome sequencing (WES) followed by bidirectional Sanger sequencing techniques identified a homozygous nonsense variant (c.466C > T; p.Gln156*) in the PPFIBP1 gene (NM_003622.4) that segregated with the disease

Adenoviral protein E4orf4 interacts with the polarity protein Par3 to induce nuclear rupture and tumor cell death.

The tumor cell-selective killing activity of the adenovirus type 2 early region 4 ORF4 (E4orf4) protein is poorly defined at the molecular level. Here, we show that the tumoricidal effect of E4orf4 is typified by changes in nuclear dynamics that depend on its interaction with the polarity protein Par3 and actomyosin contractility. Mechanistically, E4orf4 induced a high incidence of nuclear bleb formation and repetitive nuclear ruptures, which promoted nuclear efflux of E4orf4 and loss of nuclear integrity. This process was regulated by nucleocytoskeletal connections, Par3 clustering proximal t

Reciprocal Negative Regulation Between Lmx1a and Lmo4 Is Required for Inner Ear Formation.

LIM-domain containing transcription factors (LIM-TFs) are conserved factors important for embryogenesis. The specificity of these factors in transcriptional regulation is conferred by the complexes that they form with other proteins such as LIM-domain-binding (Ldb) proteins and LIM-domain only (LMO) proteins. Unlike LIM-TFs, these proteins do not bind DNA directly. LMO proteins are negative regulators of LIM-TFs and function by competing with LIM-TFs for binding to Ldb's. Although the LIM-TF Lmx1a is expressed in the developing mouse hindbrain, which provides many of the extrinsic signals for

Innate immune signaling relays environmental cues to mitophagy for metabolic adaptation to obesity.

The mechanisms by which pancreatic β cells sense and communicate environmental cues to mitochondria to maintain insulin secretion remain poorly understood. We identified that β cells transduce metabolic stress, such as during obesity or glucolipotoxicity, through the innate immune signaling adaptor TRAF6 to regulate mitochondrial quality control. TRAF6 was dispensable for β-cell function at baseline yet was essential for glucose-stimulated insulin secretion in mouse or human β cells following nutrient excess. TRAF6 was required for the recruitment of components of both ubiquitin- and receptor-

NEDD4 promotes reactive astrogliosis by enhancing K63-linked ubiquitination and inhibiting chaperone-mediated autophagy degradation of YAP1.

Following spinal cord injury (SCI), the transcriptional regulator yes-associated protein 1 (YAP1) is upregulated and accumulates in the nuclei of astrocytes, where it promotes reactive astrogliosis-a process that critically influences wound healing and neurological function recovery. However, the mechanisms regulating YAP1 in reactive astrocytes after SCI remain largely unclear. This study, we identify the E3 ubiquitin ligase NEDD4 as a critical regulator of astrocyte reactive proliferation. NEDD4 enhances astrogliosis by suppressing YAP1 degradation. Conditional deletion of Nedd4 in astrocyte

CDH-3/cadherin, YAP-1/YAP, and EGL-44/TEAD promote SYX-2/syntaxin and EFF-1 fusogen-mediated phagosome closure.

Physical interactions between cells can profoundly impact cell fate. A vital cell fate for normal development and homeostasis is programmed cell death. Cells fated to die must be efficiently cleared via phagocytosis, with defects associated with a variety of diseases. How cell-cell physical associations affect programmed cell elimination is not fully understood. Here we describe, in vivo, a cell-cell adhesion-driven signaling pathway that ensures compartment-specific cell clearance. We previously described the specialized cell death program "Compartmentalized Cell Elimination" (CCE) in the Cae

Sclerostin deficient mice rapidly heal bone defects by activating β-catenin and increasing intramembranous ossification.

We investigated the influence of the osteocyte protein, sclerostin, on fracture healing by examining the dynamics and mechanisms of repair of single-cortex, stabilized femoral defects in sclerostin knockout (Sost(-/-); KO) and sclerostin wild-type (Sost(+/+); WT) mice. Fourteen days following generation of bone defects, Sost KO mice had significantly more bone in the healing defect than WT mice. The increase in regenerating bone was due to an increase in the thickness of trabecularized spicules, osteoblast numbers and surfaces within the defect. Enhanced healing of bone defects in Sost KO mice
