Intact germinal layer of liver hydatid cysts removed after administration of albendazole.

BACKGROUND: Hydatid disease is a common health problem especially in Mediterranean and sheep-farming countries, caused by infection with the metacestode stage of the tapeworm Echinococcus. The liver is the most frequent primary site of Echinococcus granulosus infection in humans. Surgery remains the main treatment modality for cystic hepatic hydatid disease, with complete resection of the germinal layer being of major importance for recurrence. Perioperative administration of albendazole has been reported to improve surgical outcome but the results are controversial. We report here our observa…

Anti-filarial activity of novel formulations of albendazole against experimental brugian filariasis.

The study was aimed at developing better orally active albendazole (ALB) formulations. Six formulations (ALB-1 to ALB-6) were prepared and tested against Brugia malayi in Mastomys coucha and jird (Meriones unguiculatus) at 200 mg/kg, orally, for 5 consecutive days. The anti-filarial efficacy was assessed against microfilariae (mf), adult worms and female reproductive potential. Three of the 6 ALB formulations showed greatly improved female worm sterilizing potential (ALB-1: 90%; ALB-3: 63%; ALB-4: 77% of untreated control) in B. malayi - M. coucha model. Sterilization efficacy of ALB-1 was als…

Albendazole therapy of hydatid disease: 2-year follow-up of 40 cases.

Forty patients with 63 Echinococcus granulosus cysts affecting different sites were treated with albendazole and have been followed up for at least 24 months from completion of therapy. Twenty-one patients (53%) with 37 cysts (59%) showed evidence of healing. The criteria and pattern of healing are outlined. The most serious complication of albendazole therapy was hepatoxic jaundice, which occurred in 5% of patients. Recurrence during the observation period was encountered in 9.5% of patients with a positive response. It is suggested that patients suffering from uncomplicated hydatid disease s…

Developmental toxicities of albendazole and its metabolites in zebrafish (Danio rerio) embryos.

Albendazole (ABZ) is a benzimidazole anthelmintic medicine widely used in livestock and, especially, aquaculture industries. Given its widespread use, ABZ commonly enters waterways, negatively impacting aquatic organisms. Classified as Pregnancy Category C because of its teratogenic properties, ABZ poses a high risk of developmental toxicity. In this study, zebrafish (Danio rerio) embryos were used to assess the developmental toxicity of ABZ and its two metabolites, ABZ-sulfoxide and ABZ-sulfone. Among these, only ABZ exhibited strong lethality, demonstrating an LC50 value of 83.3 μg/L, and in…

Therapeutic Efficacy of Albendazole in Neonatal Calves Naturally Infected with Cryptosporidium parvum.

Cryptosporidiosis is a significant zoonotic disease caused by Cryptosporidium species, primarily Cryptosporidium parvum, leading to serious economic losses in neonatal calves; the absence of an approved vaccine or fully effective treatment necessitates alternative therapeutic approaches. This study evaluated the therapeutic efficacy of albendazole (20 mg/kg, orally, 5 days) in naturally infected neonatal calves, comparing it to paromomycin (100 mg/kg, orally, 5 days) and an albendazole + paromomycin combination. Forty-eight Simmental calves (36 naturally infected, mean age of 14 days, and 12 h…

Tea Saponins: a Novel Stabilizer for Enhancing the Oral Bioavailability of Albendazole Nanocrystals.

Albendazole serves as a broad-spectrum anthelmintic medication for treating hydatid cysts and neurocysticercosis. However, its therapeutic effectiveness is limited by poor solubility. Nanocrystals offer a promising technology to address this limitation by enhancing drug solubility. The objective of this study is to evaluate an effective stabilizer for creating an albendazole nanocrystal formulation to improve oral absorption. Among different surfactants and polymers examined, tea saponins were used as the stabilizer to develop a nanosuspension with the particle size of 180 nm through a wet gri…

Silibinin enhanced the efficacy of albendazole in treating the muscular phase of experimental trichinellosis.

Trichinella spiralis is a parasitic nematode with a special life cycle. Both adults and larvae live in two different niches in the same host (intestinal and muscular). The parasite is known to manipulate the immune system of the host to be able to survive. One of the pathways the parasite modulates is the programmed death 1/ programmed death ligand 1 (PD1/PDL1), a pathway important to maintain the immune homeostasis during chronic infections and cancers. Albendazole (ABZ) shows anti-trichinellosis efficacy, especially against the intestinal phase of the infection. In an attempt to discover a d…

Synergistic in vitro effects of albendazole and minocycline on the viability of hydatid cyst protoscoleces.

Hydatid disease poses a major zoonotic threat, affecting human health and livestock. Although albendazole is the standard treatment, it may cause adverse effects at high serum levels. This study investigated the protoscolicidal efficacy of minocycline alone and in combination with albendazole, compared to albendazole monotherapy, on hydatid cyst protoscoleces from sheep liver. Protoscoleces were aseptically aspirated from the sheep livers containing hydatid cysts. A total of 30,000 protoscoleces were suspended in 300 µl of normal saline and exposed to various concentrations of minocycline (10,…

Assessment of Oral Albendazole and Fumagillin in the Treatment of Pseudoloma neurophilia in Adult Zebrafish.

Pseudoloma neurophilia ( Pn ), the causative agent of the most commonly reported disease of zebrafish, is a microsporidian parasite that confounds research by inducing behavioral and physiologic changes in zebrafish. Although a treatment for P. neurophilia has not been documented in zebrafish, albendazole (ALB) and fumagillin (FUM) have been used to treat microsporidian infections of other fish species. To investigate the efficacy of oral ALB and FUM in the treatment of Pn, we performed a pilot study that demonstrated the safety and palatability of novel gel-based diets containing FUM or ALB i…

Synthesis, characterization and antiparasitic activity of organometallic derivatives of the anthelmintic drug albendazole.

Helminthiases, a group of neglected tropical diseases, affect more than one billion people mainly in tropical and subtropical regions. Moreover, major intestinal protozoa have a significant impact on global public health. Albendazole (ABZ) is a broad-spectrum anthelmintic recommended by the World Health Organisation (WHO). However, drug resistance is emerging due to its widespread use. In order to tackle this problem, taking into account the spectacular results obtained with ferroquine, an organometallic derivatization of the antimalarial drug chloroquine, we have prepared, in this study, a se…

Efficacy of High-Dose Albendazole with Ivermectin for Treating Imported Loiasis, Italy.

We describe the outcomes of 16 cases of imported loiasis in Italy. Patients had microfilaremia <20,000/mL and were treated with high-dose albendazole for 28 days and a single dose of ivermectin. This combination might be an effective treatment option in nonendemic areas, when diethylcarbamazine, the drug of choice, is not available.

In vitro evaluation of ivermectin, moxidectin, albendazole and pyrantel against cyathostomins of horses.

Cyathostomins are the most prevalent nematodes of horses, and multidrug resistance has been reported worldwide. There is a need to implement alternative drug monitoring analytical tests. The objective of this study was to determine the consistency (5 repetitions) of the larval migration on agar test (LMAT) using ivermectin, moxidectin, pyrantel or albendazole against cyathostomin infective-stage larvae in eight different concentrations. LMAT showed a strong coefficient of determination (R2 > 0.91), between the test repetitions (n=5). The average 50% effective concentration (EC50) for ivermecti…

[MORPHOLOGICAL CHANGES IN THE STRIATED MUSCLES AFTER ALBENDAZOLE TREATMENT FOR EXPERIMENTAL TRICHINOSIS].

Trichinosis was experimentally induced in rats. The morphology of encystation of Trichinella larvae was studied in the striated muscles. The efficacy of albendazole was evaluated in different intensity of Trichinella larvae infestation in the striated muscles of the laboratory animals. Encystation was shown to cease at 21 days. Albendazole showed high activity against intestinal and encapsulated larvae of Trichinella, leading to 100% death of T. spiralis. Capsules and Trichinella larvae were described to be destructed after treatment with albendazole 15 mg/kg twice daily for 10 days.

A review of pharmacokinetic drug-drug interactions with the anthelmintic medications albendazole and mebendazole.

Medications indicated for helminthes and other parasitic infections are frequently being used in mass populations in endemic areas. Currently, there is a lack of guidance for clinicians on how to appropriately manage drug interactions when faced with patients requiring short-term anthelmintic therapy with albendazole or mebendazole while concurrently taking other agents. The objective of this review was to systematically summarize and evaluate published literature on the pharmacokinetics of albendazole or mebendazole when taken with other interacting medications. A search of MEDLINE (1946 to O…
