Effect of a very low negative dietary cation-anion difference (DCAD) diet on plasma and urine metabolomics of prepartum Holstein cows.

The objectives of this cross-sectional, nonintervention, observational study were to compare urine and blood parameters between cows consuming a positive dietary cation-anion difference (DCAD) diet [early dry cows, DCAD + 250 mEq/kg of dry matter (DM), n = 15] with the same cows consuming a negative DCAD diet (-220 mEq/kg of DM) 10 d after moving them from the early dry to the prepartum group. The most remarkable finding was that cows consuming the anionic diet had very low urine pH and very low base excess in blood, suggestive of uncompensated metabolic acidosis. Importantly, the metabolomics

Pathway analysis using (13) C-glycerol and other carbon tracers reveals a bipartite metabolism of Legionella pneumophila.

Amino acids represent the prime carbon and energy source for Legionella pneumophila, a facultative intracellular pathogen, which can cause a life-threatening pneumonia termed Legionnaires' disease. Genome, transcriptome and proteome studies indicate that L. pneumophila also utilizes carbon substrates other than amino acids. We show here that glycerol promotes intracellular replication of L. pneumophila in amoeba or macrophages (but not extracellular growth) dependent on glycerol-3-phosphate dehydrogenase, GlpD. An L. pneumophila mutant strain lacking glpD was outcompeted by wild-type bacteria

Comparative Analysis of Meat Quality and Flavor-Forming Volatile Compounds in Longissimus Dorsi from Different Beef Breeds.

This study aimed to evaluate breed-specific differences in meat quality, nutritional compo sition, and volatile flavor compounds of beef, and to discuss their implications for animal breeding and health. Longissimus dorsi muscles were collected from four cattle breeds-Dabieshan cattle (DBS), local water buffalo (LWB), Simmental (SM), and Angus (AG)-with six animals per breed, all reared under identical feeding conditions. Meat quality parameters (color, cooking loss, shear force, and water-holding capacity), proximate composition, fatty acid and amino acid profiles, and volatile compounds were

Choroid plexus LAT2 and SNAT3 as partners in CSF amino acid homeostasis maintenance.

BACKGROUND: Cerebrospinal fluid (CSF) is mainly produced by the choroid plexus (CP) located in brain ventricles. Although derived from blood plasma, it is nearly protein-free (~ 250-fold less) and contains about 2-20-fold less free amino acids, with the exception of glutamine (Gln) which is nearly equal. The aim of this study was to determine which amino acid transporters are expressed in mouse CP epithelium in order to gain understanding about how this barrier maintains the observed amino acid concentration gradient. METHODS: Expression of amino acid transporters was assessed in isolated cho

Evolution of viscera and muscle fractional protein synthesis rate in lean meat selected hybrids and castrated Duroc pigs fed under moderate crude protein restriction.

Differences in producing performance and organoleptic meat characteristics among pig genotypes and/or producing types are widely known. These parameters are also subjected to the animal's development, feeding and management. Detailed knowledge of the effects of production phase (PP), pig producing type (PT), dietary protein availability and their interactions on nutrient digestibility, nitrogen balance and protein metabolism is essential information to improve precision feeding techniques. The experiment was a 2 (PP) × 2 (PT) × 2 (diet) factorial design conducted with 32 male pigs, 16 entire F

Human monoclonal autoantibody fragments from combinatorial antibody libraries directed to the U1snRNP associated U1C protein; epitope mapping, immunolocalization and V-gene usage.

To study the localization and function of the U1snRNP associated U1C protein, so far only human sera from systemic lupus erythematosus (SLE) overlap syndrome patients have been used. Here we report for the first time the isolation of human monoclonal anti-UIC autoantibody fragments from IgG derived combinatorial and semi-synthetic human antibody libraries. Two classes of human monoclonal anti-UIC (auto)antibodies were found: specific anti-U1C autoantibodies, recognizing U1C only, and cross-reactive antibodies which also react with U1A and Sm-B/B'proteins. The heavy chains (V(H)genes) of all fi

Listeria monocytogenes folate metabolism is required to generate N-formylmethionine during infection.

Folic acid and its derivatives are required for the synthesis of purines, pyrimidines, and some amino acids. Antifolate antibiotics that target the folic acid metabolism pathway are commonly used for the treatment of listeriosis caused by the intracellular pathogen Listeria monocytogenes (Lm). In recent work in mBio, Feng et al. sought to understand the role of folic acid metabolism in Lm virulence (Y. Feng, S. Chang, D. A. Portnoy, 2023, mBio https://doi.org/10.1128/mbio.01074-23). The authors discovered that N-formylmethionine, an amino acid utilized by bacteria to initiate protein synthesis

The rubella virus nonstructural protease recognizes itself via an internal sequence present upstream of the cleavage site for trans-activity.

The substrate requirement for rubella virus protease trans-activity is unknown. Here, we analyzed the cleavability of RV P200-derived substrates varying in their N-terminal lengths (72-475 amino acids) from the cleavage site by the RV protease trans-activity. Only substrates with at least 309 amino acid residues N-terminal to the cleavage site were able to undergo cleavage. Further, rubella sequence was found to be necessary in the N-terminal region of the substrate, whereas a heterologous sequence C-terminal to the cleavage site was tolerated. These results demonstrated a requirement for resi

Diet-dependent chemical fingerprinting in Tenebrio molitor larvae reared on tuna by-products.

Diet composition significantly influences the chemical quality of edible insect ingredients. This study compared Tenebrio molitor larvae reared on wheat bran (WB) and tuna by-products (TB) using proximate analysis and metabolomics. On a dry matter basis, crude protein was similar at about 45 g per 100 g, whereas TB showed higher lipid and lower carbohydrate contents. Fatty acid methyl ester analysis revealed higher oleic and linoleic acids and a shift toward C18 unsaturated fatty acids in TB. Total hydrolyzable amino acids and essential amino acid proportion were conserved, although proline wa

Alanine decreases the protein requirements of infants with inborn errors of amino acid metabolism.

Supplementation with alanine was found to increase growth in weight and nitrogen balance in 5 infants with a variety of inborn errors of amino acid metabolism receiving diets restricted in protein. The addition of alanine to the regimen led to a mean increase in weight of 15 g/day. This and the increased nitrogen balance of 15 mg/kg/day were highly significant statistically. In addition a dose-response effect of alanine was observed. The effect of alanine was compared with that of a supplemental mixture of essential and non-essential amino acids, lacking only those considered to be toxic in th

Nutrition and infectious diseases in developing countries and problems of acquired immunodeficiency syndrome.

Infectious diseases are the major causes of death and morbidity in underdeveloped countries, particularly in children. Increasing evidence suggests that malnutrition-both Protein-Energy type Malnutrition (PEM) and essential micronutrient (vitamins, trace minerals, essential amino acids, polyunsaturated fatty acids) type-is the underlying reason for increased susceptibility to infections. On the other hand, certain infectious diseases also cause malnutrition, which results in a vicious cycle. Before its viral origin was known, acquired immunodeficiency syndrome (AIDS) had been termed the thin d

Hartnup disorder: unraveling the mystery.

Hartnup disorder is an autosomal recessive disease that can be associated with neurological, psychiatric and dermatological abnormalities or be asymptomatic. Excessive intestinal and urinary loss of neutral amino acids is an essential feature of this disorder, which had been presumed to be due to hereditary abnormalities in an apical membrane-situated amino acid transporter. As anticipated, recently, mutations in the cytoplasmic and transmembrane domains of SLC6A19, the recently cloned neutral amino acid transporter, were detected in members of families with Hartnup disorder. Presumably, defic

Similar Muscle Recovery but Greater Soreness in Active Vegans on Low-Protein Diets Compared to High-Protein Omnivores Following Exercise-Induced Damage.

To assess whether a low-protein vegan diet is associated with differences in recovery from exercise-induced muscle damage (EIMD) in physically active individuals. Physically active vegans (n = 7) and omnivores (n = 7) underwent a lower-body muscle damage protocol consisting of five sets of repetitions to failure using the Smith machine squat and leg press at 85% of one-repetition maximum (1RM). Muscle damage and recovery were assessed at baseline, immediately post-exercise, and at 24, 48, and 72 h post-EIMD. Outcomes included serum concentrations of creatine kinase (CK) and lactate dehydrogena

HLA-EMMA: A user-friendly tool to analyse HLA class I and class II compatibility on the amino acid level.

In renal transplantation, polymorphic amino acids on mismatched donor HLA molecules can lead to the induction of de novo donor-specific antibodies (DSA), which are associated with inferior graft survival. To ultimately prevent de novo DSA formation without unnecessarily precluding transplants it is essential to define which polymorphic amino acid mismatches can actually induce an antibody response. To facilitate this, we developed a user-friendly software program that establishes HLA class I and class II compatibility between donor and recipient on the amino acid level. HLA epitope mismatch al

Effect of parenteral glutamine supplementation on plasma amino acid concentrations in extremely low-birth-weight infants.

BACKGROUND: Glutamine is one of the most abundant amino acids in both plasma and human milk and may be conditionally essential in premature infants. However, glutamine is not provided by standard intravenous amino acid solutions. OBJECTIVE: We assessed the effect of parenteral glutamine supplementation on plasma amino acid concentrations in extremely low-birth-weight infants receiving parenteral nutrition (PN). DESIGN: A total of 141 infants with birth weights of 401-1000 g were randomly assigned to receive a standard intravenous amino acid solution that did not contain glutamine or an isoni

Impacts of maternal dietary protein intake on fetal survival, growth, and development.

Maternal nutrition during gestation, especially dietary protein intake, is a key determinant in embryonic survival, growth, and development. Low maternal dietary protein intake can cause embryonic losses, intra-uterine growth restriction, and reduced postnatal growth due to a deficiency in specific amino acids that are important for cell metabolism and function. Of note, high maternal dietary protein intake can also result in intra-uterine growth restriction and embryonic death, due to amino acid excesses, as well as the toxicity of ammonia, homocysteine, and H2S that are generated from amino

Nutritional potential of invasive Rugulopteryx okamurae, of Gelidium corneum industrial by-product, and of five native Atlantic macroalgae as feedstocks for alternative protein production.

Macroalgae are increasingly considered sustainable resources for protein-oriented food and bioprocessing applications due to their rapid growth, wide availability, low freshwater and fertilizer requirements, and potential as natural sources of proteins and bioactive compounds. However, integrated assessments comparing native, invasive, and industrial side-stream macroalgal biomasses under the same nutritional and safety framework remain scarce, limiting the identification of suitable candidates for protein-oriented valorization. In this context, this study assessed seven Atlantic macroalgal bi
