Effects of furosemide, exercise, and atropine on tracheal mucus transport rate in horses.

Effects of furosemide, exercise, and atropine on tracheal mucus transport rate (TMTR) in horses were investigated. Atropine (0.02 mg/kg of body weight) administered IV or by aerosolization significantly (P < 0.05) decreased TMTR at 60, but not at 30 minutes after its administration in standing horses. Furosemide (1.10 mg/kg, IV) did not have any significant effect on TMTR when measured at 2 or 4 hours after its administration in standing horses. Exercise alone or furosemide (1.10 mg/kg, IV) administration followed 4 hours later by exercise did not alter TMTR, compared with values for standing …

Intravesical atropine and suppression of detrusor hypercontractility in the neuropathic bladder. A preliminary study.

Twelve patients with detrusor hypercontractility secondary to spinal cord lesions were treated with intravesical atropine sulphate. Five patients expelled the solution almost immediately. In the seven patients who retained the solution the cystometric capacity increased by a mean of 301 ml (P < 0.01). The volume at which an unstable contraction occurred increased by a mean of 190 ml (P < 0.05) and the volume at which a urinary leak occurred increased by a mean of 297 ml (P < 0.05). The maximum hyperreflexic detrusor contraction pressure was reduced by a mean of 24 cm water (P < 0.05). No signi…

Toxicokinetics of soman stereoisomers after subcutaneous administration to atropinized guinea pigs.

The toxicokinetics of the four stereoisomers of the nerve agent C(+/-)P(+/-)-soman were investigated after subcutaneous administration of a 6 LD50 dose (148 micrograms/kg) to anaesthetized, atropinized, and artificially ventilated guinea pigs. Whereas the relatively nontoxic C(+/-)P(+)-isomers were not detected in blood, the highly toxic C(+/-)P(-)-isomers appeared within 1 min in the general circulation and reached maximum levels of 10-15 ng/ml blood within a period of ca. 7 min. In this absorption phase the blood levels of the C(+)P(-)-isomer lag clearly behind those of the C(-)P(-)-isomer.

Atropine-resistant submandibular responses to stimulation of the parasympathetic innervation in the anaesthetized ferret.

1. Submandibular salivary and vascular responses to stimulation of the peripheral end of the chorda-lingual nerve at 20 Hz continuously for 60 min were investigated in anaesthetized ferrets, in which the sympathetic innervation to the gland was cut, in the presence and absence of atropine (2.0 mg kg-1). 2. Both the increase in submandibular salivary flow and protein output, which occurred in response to nerve stimulation, were substantially reduced following the administration of atropine, the latency was greatly increased thereby, and both responses were more transient but neither was abolish…

Evaluation of acepromazine/meperidine/atropine premedication followed by thiopental anesthesia in the cat.

Cardiopulmonary function was assessed in healthy cats premedicated with intramuscular acepromazine, meperidine, atropine combination (premix), followed by induction and maintenance with intravenous thiopental for 30 min. Cardiac output by thermodilution, heart rate, blood pressure and blood gas analysis were evaluated over 120 min. A minor degree of respiratory depression was noted in the cats. Cardiac index (cardiac output/kg) was significantly depressed following thiopental induction and for the entire 120 min studied. Stroke volume was significantly reduced after premix administration and f…

The effects of a single intramuscular injection of atropine sulphate on visual performance in man.

The effects of a single intramuscular injection of 2 mg atropine sulphate on visual function were studied in volunteer subjects. The well-known effects of increased heart rate, dryness of the mouth, increased pupil diameter and reduced accommodative range were confirmed. Visual acuity, stereoacuity, red-green colour balance and reaction time to a visual stimulus were unaffected by atropine, while extraocular muscle balance (horizontal heterophoria and cyclophoria) underwent a transient change. There was no significant change in contrast sensitivity measurements to stationary sinusoidal grating…

Comparative effects of atropine and oxotremorine on neuromuscular transmission at the rat hemidiaphragm preparation.

The effects of atropine (0.001-10 microM) and oxotremorine (0.51-51.3 microM) on neuromuscular transmission and muscle contraction were studied by analysing their effects on the twitch, tetanic and post-tetanic twitch responses and on the phenomenon of post-tetanic twitch potentiation (PTP) in a rat phrenic nerve-diaphragm preparation. In addition, the effects of atropine (0.1 microM) and oxotremorine (0.6 microM) were studied on the contractions produced in rat ileum by superfusate from repetitively stimulated phrenic nerve diaphragm preparation. The results showed that atropine had a dual ac…

An atropinized heat-stressed rat model: dose response effects and pharmacokinetics.

Atropine and other anticholinergic drugs are widely used in common medications and in the treatment of organophosphate poisoning. Man dissipates heat by the evaporation of sweat. Analogously, rats spread saliva over their bodies for evaporative cooling. Atropine inhibits both sweating and salivation. Therefore, we sought to quantitate the effects of atropine in our rat heatstroke model. While heat-stressing adult male rats of 500 or 250 g at 41.5 degrees C, we measured the effects of i.v. atropine (10-4000 micrograms X kg-1) on the following: heating rate (HR), % wt loss (saliva production), a…

Effects of atropine, acepromazine, meperidine, and xylazine on gastroesophageal sphincter pressure in the dog.

Gastroesophageal sphincter (GES) pressure was 47.9 +/- 1.2 mm of Hg in nontreated dogs. Treatment with atropine, acepromazine, and xylazine reduced GES pressure to 13.2 +/- 2.03, 18.6 +/- 2.14, and 11.7 +/- 1.19 mm of Hg, respectively. Treatment with meperidine resulted in phasic contractions with minimum and maximum pressures of 27.9 +/- 4.55 and 98.9 +/- 9.16 mm of Hg, respectively. Drugs used in anesthetic procedures can reduce GES pressure in dogs.

Effect of topically applied atropine, methysergide and chlorpheniramine on nasal challenge with serotonin.

Serotonin challenge in the nose produces sneezing and rhinorrhoea. To study the mode of action of serotonin in the nose, 14 normal persons, in a double-blind, randomized study, were provoked with serotonin in three concentrations. Fifteen minutes before challenge, they were pretreated in the nose with either a cholinoceptor antagonist (atropine), a serotonin antagonist (methysergide), an H1-histamine antagonist (chlorpheniramine) or isotonic saline. The number of sneezes was noted, and the amount of secretions measured. Serotonin induced dose-related increase in sneezing and hypersecretion (P …

Influence of metiamide and atropine on acid and pepsin secretion in the anaesthetized Heidenhain cat.

The inhibition of acid and pepsin secretion by metiamide and atropine was studied in the anaesthetized Heidenhain-pouch cat. Metiamide strongly inhibited acid secretion induced by histamine, pentagastrin or carbachol. Metiamide had not effect on pepsin secretion stimulated by pentagastrin and carbachol but was effective on histamine-stimulated pepsin secretion. Atropine strongly inhibited pepsin secretion but was not as effective as metiamide in the inhibition of acid secretion. Reperfusion of the pouch with acid + salicylic acid also increased pepsin secretion. This increased pepsin output wa…

Comparison of glucagon, atropine, and placebo as premedication for endoscopy of the upper gastrointestinal tract.

The effects of 0.5 mg glucagon and 0.5 mg atropine given intravenously as premedication in upper gastrointestinal endoscopy have been examined and compared to those of placebo (0.9% NaCl) in a double-blind study involving 36 patients and 1 endoscopist. The results showed no difference between atropine, glucagon, and placebo with regard to vomiting, opening of the pylorus, feeling of discomfort, or the success of the examination. Glucagon significantly reduced peristalsis compared with both atropine (p less than 0.01) and placebo (p less than 0.01). The reflux was also significantly reduced by …

The effects of acetylcholine and atropine on the 22Na+ permeability of intestinal smooth muscle vesicles.

A technique is described which has enabled us to measure change in 22Na+ efflux from smooth muscle plasma membrane vesicles. The resting 22N … d by measurement of the half-life of 22Na+ efflux (t1/2). The acetylcholine effect was inhibited by atropine, but unlike the situation in the whole muscle, this inhibition was noncompetitive. Tubocuraine (a nicotinic antagonist) had no effect on this acetyl-choline response. Atropine has no effect by itself on the resting 22Na+ efflux, neither did tetrodot …

Effect of atropine and oxotremorine on the release of acetylcholine from the frog spinal cord.

The in vitro frog spinal cord has been used to study acetylcholine (ACh) release and spinal root potentials. The preparation bathed in an eserine-containing medium spontaneously released ACh into the bathing fluid. This release was enhanced by atropine in a dose-related and long-lasting manner and transiently by oxotremorine. The release rate of ACh was increased by low frequency ventral root stimulation; this increase was found to be proportionally the same after application of atropine. Oxotremorine did not modify the elctrically-evoked ACh release but blocked or reduced the effect of atropi…
